1Effects of Seed Soaking with Paclobutrazol on Tillering and Physio-logical Characteristics of Sugarcane Seedlings显示文摘In order to study the effects of seed soaking with Paclobutrazol( PP333) on the tillering and physiological characteristics of sugarcane seedlings,sugar cane variety ROC22 was used as experimental material. The effects of seed soaking with different concentrations of PP333 were studied. The tiller number and growth of sugarcane were surveyed at the 6-leaf stage,the physiological indicators such as chlorophyll content,soluble protein content,proline content and peroxidase activity were measured at the 2-leaf,4-leaf,and 6-leaf stages. The results showed that the seed soaking with PP333 can effectively improve the tillering of sugarcane seedlings. The suitable concentration for the tillering of sugarcane and the growth and development of sugarcane was of 50 mg/L; the seed soaking with PP333 significantly increased the chlorophyll,soluble protein,proline,and peroxidase activity of leaves of sugarcane seedlings; the content of chlorophyll and soluble protein in leaves of sugarcane seedlings treated with 90 mg/L PP333 was the highest,and the proline content and peroxidase activity of sugarcane treated with 50 mg/L PP333 were the highest.Junxian LIU Song LI Fang TAN Xin LIU Yibo HE Kaichao WU Faqian XIONG Limin LIU 2017Asian Agricultural Research2017,9,1:1

3Effect of Foliar Application of Selenium Fertilizer on Yield,Selenium Content and Heavy Metal Contents of Waxy Maize显示文摘[Objectives]This study aimed to explore the effect of different concentrations of selenium fertilizer on the yield,selenium content and heavy metal contents of waxy maize,thereby providing reference for the scientific application of selenium fertilizer on waxy maize.[Methods]Different varieties of waxy maize(Guitiannuo 611,Guinuo 615 and Guiheinuo 609)were used as test materials.They were sprayed with different concentrations(0,0.5,1.0,1.5 and 2.0 g/L)of water-soluble selenium fertilizer(Xinxibao)at the big flare stage,and the effect on the yield,selenium content and heavy metal contents of the waxy maize was analyzed.[Results]Foliar application of different concentrations of selenium fertilizer showed no significant effect on the yield and yield components of waxy maize.As the concentration of selenium fertilizer increased,the grain selenium content of different varieties of waxy maize increased.Among them,the increase of Guinuo 615 was the largest,of which the selenium content reached 0.265 mg/kg,2.82 times that of the control(water).The cadmium and arsenic contents of different varieties of waxy maize decreased with the increase of the concentration of selenium fertilizer sprayed.Among the waxy maize varieties,the cadmium and arsenic contents of Guitiannuo 611 dropped the most significantly,up to 52.94%and 77.78%.In waxy maize,the correlation coefficients between selenium content and cadmium,arsenic contents were-0.5509(P<0.05)and-0.8530(P<0.01),respectively.[Conclusions]Spraying exogenous selenium fertilizer had no obvious effect on the yield of waxy maize.However,the increase of the concentration of exogenous selenium fertilizer could significantly increase the selenium content and reduce the cadmium and arsenic contents in grains of different varieties of waxy maize.Chenglin ZOU Ruining ZHAI Kaijian HUANG Hua TAN Debo ZHENG Aihua HUANG Xinxing WEI Runxiu MO Faqian XIONG Hui WEI Shanzhu WEI 2020Asian Agricultural Research2020,12,8:0
4A Non-toxic and Efficient Method for Extracting DNA and RNA from Peanut显示文摘[Objectives]To establish a non-toxic and efficient method for extracting DNA and total RNA from peanuts and laying a solid foundation for the molecular biology study of peanuts.[Methods]Based on the principle and method of purifying nucleic acids by silica gel adsorption at high salt and low pH condition,a non-toxic and efficient method to extract peanut DNA and total RNA using cetyltrimethyl ammonium bromide(CTAB)extraction solution was designed.The quality and purity of nucleic acids were detected by agarose gel electrophoresis and nucleic acids protein analyzer,respectively.The quality of DNA was further verified by enzyme digestion and PCR amplification using molecular marker techniques.The quality of total RNA was further verified by reverse transcription(RT)-PCR of actin gene and cDNA-SCoT gene differential display technique.[Results]The agarose gel electrophoresis test showed that the peanut DNA extracted by a low-toxic and effective method is free of contamination and degradation.Through the detection by the nucleic acid protein analyzer,the DNA concentration,yield,A260/A280 and A260/A230 of 5 peanut varieties were 419.6-498.2 ng/μL,20.98-24.91μg/g,1.89-1.96 and 2.03-2.28,respectively.The DNA was of high quality and can be completely digested by EcoRI restriction enzymes,and also can be used for SCoT and SRAP molecular marker technology analysis.The RNA extracted from different tissues of peanuts showed no visible DNA bands by non-denaturing agarose gel electrophoresis.The separated 28S bands were brighter than 18S.The ratio of A260/A280 and A260/A230 showed that the RNA quality was good and can be used for reverse transcription,RT-PCR of actin gene and amplification of cDNA-SCoT gene differential display technique.[Conclusions]This experiment established a low-toxic and effective method for extracting DNA and total RNA from peanuts.Compared with traditional methods,this method is more time-saving and cheaper than commercial kits.The most important point is that this method does not use toxic reagents such as phenol,chloroform and isopropanol.Thus,it is expected to be widely applied in molecular biology research.Jing LIU Congcong WANG Jing JIANG Zhuqiang HAN Xiumei TANG Lihang QIU Liangqiong HE Ruichun ZHONG Zhipeng HUANG Haining WU Ronghua TANG Junxian LIU Faqian XIONG 2021Asian Agricultural Research2021,13,1:0
5Effects of Seed Soaking with Paclobutrazol on Early Tillering and Endogenous Hormone Contents of Sugarcane Seedlings显示文摘[Objective] The paper was to explore the effect of seed soaking with paclobutrazol( PP333) on early tillering and endogenous hormone content of sugarcane seedlings.[Method] With the sugarcane variety ROC22 as the experimental material,seeds were soaked in different concentrations of PP333. The number and growth of tiller were investigated,and the contents of endogenous hormones IAA,CTK,ABA and GA3 at two-leaf,four-leaf and six-leaf stages of sugarcane seedlings were measured,to study the effects of seed soaking with PP333 on early tillering and endogenous hormone contents of sugarcane seedlings.[Result] Seed soaking with PP333 could effectively improve and advance tillering occurrence of sugarcane seedlings. The suitable PP333 concentration for sugarcane tillering occurrence,growth and development was 50 mg/L. Seed soaking with PP333 significantly increased the CTK and ABA contents in leaves of sugarcane seedlings,and decreased IAA and GA3 contents. 6Construction of EMS-Induced Peanut Mutant Libraries and Identification of Pod-Related Traits Mutant Lines显示文摘Peanut(Arachis hypogaea L.)is an oil and economic crop of vital importance,and peanut pod is the key organ influencing the yield and processing quality.Hence,the Pod-related traits(PRTs)are considered as important agronomic traits in peanut breeding.To broaden the variability of PRTs in current peanut germplasms,three elite peanut cultivars were used to construct Ethyl methane sulfonate(EMS)-induced mutant libraries in this study.The optimal EMS treatment conditions for the three peanut varieties were determined.It was found that the median lethal dose(LD50)of EMS treatment varied greatly among different genotypes.Finally,the EMS-induced peanut mutant libraries were constructed and a total of 124 mutant lines for PRTs were identified and evaluated.Furthermore,“M-8070”,one of the mutant lines for pod constriction,was re-sequenced via high-throughput sequencing technology.The genome-wide variations between“M-8070”and its wild parent“Fuhua 8”(FH 8)were detected.2994 EMS-induced single nucleotide polymorphisms(SNPs)and 1188 insertion-deletions(InDels)between“M-8070”and its wild parent were identified.The predominant SNP mutation type was C/G to T/A transitions,while the predominant InDel mutation type was“1-bp”.We analyzed the distribution of identified mutations and annotated their functions.Most of the mutations(91.68%of the SNPs and 77.69%of the InDels)were located in the intergenic region.72 SNPs were identified in the exonic region,leading to 27 synonymous,43 nonsynonymous and 2 stop-gain variation for gene structure.13 Indels were identified in the exonic region,leading to 4 frame-shift,8 non-frame-shift and 1 stop-gain variations of genes.These mutations may lead to the phenotypic variation of“M-8070”.Our study provided valuable resources for peanut improvement and functional genomic research.Hao Chen Faqian Xiong Rilong Xu Xiangyu Chen Haifeng Zhong Yumei Zhang Xinlong Lan Hong Zhang Yuhua Chen Runfang Hu Guoqiang Lin Zhaoxiu Tang 2023Phyton-International Journal of Experimental Botany2023,92,2:0
7Effect of Gibberellin,Light and Genotype on Somatic Embryogenesis and Plantlet Regeneration of Peanut显示文摘[Objective]Peanut in vitro regeneration system was optimized to provide technical support for its genetic transformation and mutagenesis.[Method]The effect of gibberellin,light and genotype on somatic embryogenesis and plantlet regeneration of peanut was studied using Guihua peanut cultivar as materials,leaflets as explants,MS+10 mg/L 2,4-D as somatic embryo induction medium,and MS+3 mg/L 6-BA+0.8 mg/L NAA+(0-15 mg/L)GA3as plantlet induction medium.[Result]The somatic embryo induction rate of five peanut varieties under light condition of light:dark=14 h:10 h was significantly higher compared with that under dark condition by 7.5-37.5 percent points.Among different peanut varieties,Guihua 26 represented the highest somatic embryo induction rate of62.8%,while Guihua 833 represented the lowest somatic embryo induction rate of 21.7%.Adding 5-15 mg/LGA3in plantlet induction medium was conducive to improving the induction rate of plantlets derived from somatic embryos.With addition of 5 mg/L GA3,Guihua 26 and Guihua 771 represented the highest plantlet induction rate of 42.8%and 35.3%,respectively;with addition of 15 mg/L GA3,Guihua 836,Guihua 1026 and Guihua 833 represented the highest plantlet induction rate of 38.7%,33.3%and 26.4%,respectively.Among different combinations of peanut variety and GA3concentration,plantlet induction rate reached the highest in the combination Guihua 26+5 mg/L GA3and reached the lowest in the combination Guihua 836+15 mg/L GA3.[Conclusion]Appropriate light conditions are conducive to peanut somatic embryogenesis in vitro.Adding GA3in plantlet induction medium is conducive to promoting plantlet regeneration.Guihua26 and Guihua 836 are the best peanut varieties among the experimental genotypes for somatic embryogenesis and plant regeneration.Jing JIANG Faqian XIONG Xiumei TANG Ruichun ZHONG Liangqiong HE Zhong LI Zhuqiang HAN Ronghua TANG 2013Agricultural Biotechnology2013,2,6:0
