| 1 | Advancement in separation materials for blood purification therapy显示文摘Blood purification refers to the extra corporeal therapies of removing potentially toxic substances, in which blood is circulated through an adsorption system loading separation materials. High-efficient inexpensive separation materials are critical to success. In this review, separation materials such as polymers and nanomaterials are summarized and compared. Combining the advantages of the adsorptive membranes and nanomaterials, organic–inorganic hybrid/blend membranes have been developed explosively. These hybrid/blend membranes have both the characteristics of high permeability, easy fabrication, good biocompatibility of adsorptive membranes, and characteristics of fast adsorption rate and high adsorption capacity of nanomaterials. The preparation and modification methodology of the separation materials is reviewed. For affinity separation materials, the relationship of ligand chemistry, ligand density and pores of the matrix is discussed. | 2 | Effects of Pseudoalteromonas sp. BC228 on Digestive Enzyme Activity and Immune Response of Juvenile Sea Cucumber(Apostichopus japonicus)显示文摘A marine bacterium, Pseudoalteromonas sp. BC228 was supplemented to feed in a feeding experiment aiming to determine its ability of enhancing the digestive enzyme activity and immune response of juvenile Apostichopus japonicus. Sea cucumber individuals were fed with the diets containing 0(control), 105, 107 and 109 CFU g-1 diet of BC228 for 45 days. Results showed that intestinal trypsin and lipase activities were significantly enhanced by 107 and 109 CFU g-1 diet of BC228 in comparison with control(P < 0.01). The phagocytic activity in the coelomocytes of sea cucumber fed the diet supplemented with 107 CFU g-1 diet of BC228 was significantly higher than that of those fed control diet(P < 0.05). In addition, 105 and 107 CFU g-1 diet of BC228 significantly enhanced lysozyme and phenoloxidase activities in the coelomic fluid of sea cucumber, respectively, in comparison with other diets(P < 0.01). Sea cucumbers, 10 each diet, were challenged with Vibrio splendidus NB13 after 45 days of feeding. It was found that the cumulative incidence and mortality of sea cucumber fed with BC228 containing diets were lower than those of animals fed control diet. | 4 | Expression of S100A10 gene and its regulation by sex hormones in mouse uterus显示文摘S100A10 belongs to the S100 calcium binding protein superfamily, and functions as one of the mediators of calcium-dependent signaling pathway. Recently, S100A10 gene was proved to be significantly up-regulated at the implantation site. In the present study, semi-quantitative reverse transcriptase-polymerase chain reaction and in situ hybridization are used to investigate the tissue-specificity of S100A10 expression and the expression pattern of S100A10 in the uteri during the estrous cycle and pregnancy. Meanwhile, the regulation of S100A10 expression by sex steroid hormones is studied in ovariectomized mice. The results show that S100A10 could be detected in various kinds of tissues, with relatively high expression in reproductive tracts including ovary, uterus, testis and epididymis.During pregnancy, the expression of S100A10 in the uteri is significantly up-regulated on the 4th day. The transcript is strongly detected in endometrial stromal cells and weakly in luminal epithelium cells at the implantation site, but almost not at the inter-implantation site.From gestational day 5 till labor, S100A10 mRNA maintains a certain level in both uteri and placentae. During the estrous cycle, expression of S100A10 is up-regulated in the uteri at proestrus and estrus. Estradiol significantly induces the expression of S100A10, while progesterone can abolish the effect of estradiol. | 7 | DDR1 promotes LoVo cell proliferation by regulating energy metabolism显示文摘Cellular energy metabolism dysregulation is associated with colorectal cancer(CRC)development and progression.Discoidin domain receptor 1a(DDR1a),one of the five DDR1 isoforms,is closely related to cell proliferation,invasion,and apoptosis in various tumors.Whether it participates in cellular metabolic reprogramming and regulates CRC initiation and progression remains unclear.In this study,we compared the expression of DDR1 in CRC tissues and adjacent tissues from 126 postoperative CRC samples.Moreover,lentivirus-mediated DDR1a overexpression and knockdown were performed in LoVo cells,and cell viability and proliferation were determined by CCK-8 and BrdU assays,respectively.Oxygen consumption rate,extracellular acidification rate,and lactate production were used to determine the effect of DDR1a on metabolic reprogramming.Clinically,CRC patients with high DDR1 expression had poor differentiation and were at an advanced TNM stage.DDR1a promoted LoVo cell proliferation,mitochondrial function,and extracellular acidification.Moreover,DDR1a knockdown inhibited intracellular lactic acid production in LoVo cells,while a pyruvate kinase inhibitor(diamide,200μM)significantly reversed this progression.Taken together,our results reveal that DDR1 plays a crucial role in maintaining intracellular environment homeostasis through metabolic reprogramming. | Bin Xiong Zehui Xie Feixue Song Huiling Chen Xiaojuan Wang Zhengxu Jin Tiyun Han Yi Li Dekui Zhang | 2022 | Acta Biochimica et Biophysica Sinica2022,54,5: | 1 |

| 19 | Efficacy of bioactive compounds of Chaihu(Radix Bupleuri Chinensis)on glaucomatous optic atrophy through interleukin-6/hypoxia inducible factor-1αsignal pathway显示文摘OBJECTIVE:To investigate the bioactive compounds of Chaihu(Radix Bupleuri Chinensis)(RB)on glaucomatous optic atrophy(GOA),and to study the pharmacological mechanism.METHODS:We collected information on the bioactive compounds of RB from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Targets related to bioactive compounds and GOA were also obtained.Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway and network analyses were performed to investigate the potential mechanism of RB against GOA.Subsequently,the main bioactive compounds of RB and targets of GOA were docked by Autodock software.Moreover,a GOA model of retinal ganglion cells(RGCs)induced by cobalt chloride was established to verify the effect of RB on GOA.RESULTS:There were 17 main bioactive compounds and 46 key targets were screened as potential players in GOA.The compound-target network mainly contained 17 compounds and 46 corresponding targets,and the key targets consisted of interleukin-6(IL-6),hypoxia inducible factor-1α(HIF1A),Caspase-3,estrogen receptor alpha(ESR1),MYC proto-oncogene(MYC),and vascular endothelial growth factor A(VEGFA).Forty-nine significantly enriched GO terms,and 134 KEGG signaling pathways were identified(P<0.05),including HIF-1,tumor necrosis factor,VEGF,prolaction,and other signaling pathways.Molecular docking results showed that the main bioactive compounds of RB exhibited the strongest binding activity with IL-6.Furthermore,experimental validation showed that the RB extract inhibited the activity and promoted apoptosis of RGCs in a dose-dependent manner.The RB extract also suppressed the expression of Bax,Caspase-3,and Caspase-9 and regulated malonaldehyde,superoxide dismutase,and glutathione peroxide by inhibiting the IL-6/HIF-1αsignaling pathway.CONCLUSIONS:The present study provided insights into the mechanism of RB on GOA.RB mainly reverses GOA by inhibiting the IL-6/HIF-1αsignaling pathway. | YANG Xirui ZHAO Hui SHAN Muhammad DONG Feixue ZHANG Dandan WANG Jixue YUAN Xingxing | 2023 | Journal of Traditional Chinese Medicine2023,43,6: | 0 |

The flow cytometry was performed to identifycirculating pDCs. The concentration of IFN-α in serum and that inthe supernatant of peripheral blood mononuclear cells (PBMCs)cultured after stimulation with CpG ODN2216 were examinedboth in CML patients and in the healthy controlsRESULTS There was significant reduction in the numberof circulating pDCs, serum concentration of IFN-α and thecapacity of IFN-α producing PBMCs in CML patients comparedwith those in healthy control individuals (P < 0.001). After theactive treatment with IFN-α and hydroxyurea, the quantity andfunction of pDCs were increased in stabilized patients, especiallythe function of pDCs in 2 patients achieving major cytogeneticresponse (MCR). The proportion and function of pDCs and theserum levels of IFN were inversely correlated with both WBC andage of the patients with CML, and positively correlated with thestate of the illness.CONCLUSION CML patients had a reduced number anddysfunction of circulating pDCs.
