5Injectable hydrogels for cartilage and bone tissue engineering显示文摘Tissue engineering has become a promising strategy for repairing damaged cartilage and bone tissue. Among the scaffolds for tissue-engineering applications, injectable hydrogels have demonstrated great potential for use as three-dimensional cell culture scaffolds in cartilage and bone tissue engineering, owing to their high water content, similarity to the natural extracellular matrix(ECM), porous framework for cell transplantation and proliferation, minimal invasive properties, and ability to match irregular defects. In this review, we describe the selection of appropriate biomaterials and fabrication methods to prepare novel injectable hydrogels for cartilage and bone tissue engineering. In addition, the biology of cartilage and the bony ECM is also summarized. 6Roux-en-Y choledochojejunostomy using novel magnetic compressive anastomats in canine model of obstructive jaundice显示文摘BACKGROUND:The traditional hand-sewn Roux-en-Y choledochojejunostomy is technically complicated,and the incidence of postoperative complications has remained high.A set of novel magnetic compressive anastomats was introduced to facilitate choledochojejunostomy and improve the prognosis of patients.METHODS:After ligating the common bile duct for 7 days,16 dogs were randomly divided into two groups (n=8 per group).Anastomats were used in the study group,and the traditional hand-sewn method was used in the control group for standard Roux-en-Y choledochojejunostomy.We compared the operation time,incidence of complications,gross appearance,and pathological disparity in stoma between the two groups in 1-month and 3-month follow-up examinations.RESULTS:The time spent on constructing the anastomosis for the study group was significantly shortened.Although no anastomotic stenosis occurred in the two groups,the narrowing rate of biliary-enteric anastomosis was much higher in the control group.There was one case of bile leakage in the control group,whereas no bile leakage occurred in the study group.A smoother surface,an improved layer apposition,and a lower local inflammatory response were identified in the anastomosis of the study group.CONCLUSION:The structures of the novel magnetic compressive anastomats are simple,and they are time-saving,safe and efficient for performing Roux-en-Y choledocho- jejunostomy procedures in a canine model of obstructive jaundice.Chao Fan,Xiao-Peng Yan,Shi-Qi Liu,Chun-Bao Wang,Jian-Hui Li,Liang Yu,Zheng Wu and Yi Lv Department of Hepatobiliary Surgery and Department of Pathology,First Affiliated Hospital,School of Medicine,Xi’an Jiaotong University,Xi’an 710061,China Department of Surgical Oncology,Third Affiliated Hospital,School of Medicine,Xi’an Jiaotong University,Xi’an 710068,China 2012Hepatobiliary & Pancreatic Diseases International2012,11,1:49
7Spatio-temporal framework of tectonic uplift stages of the Tibetan Plateau in Cenozoic显示文摘Four intensive uplift periods, i.e., 60–35, 25–17 and 12–8 Ma (but 18–13 Ma in the Himalayas of the southern Tibet), and since about 5 Ma, can be determined on the Tibetan Plateau by synthetical analysis of low-temperature thermo-chronology data, sedimentary deposit records, and structural deformation records of different areas. The strong tectonic uplift periods in different areas on the Tibetan Plateau are penecontemporaneous, except for the Himalayan area of the southern Tibet, where a rapid uplift and exhumation period, controlled by the activity of the South Tibetan Detachment System faults, occurred during 18–13 Ma. These strong uplift and exhumation periods correspond well to intensive deformation activity periods, suggesting tectonically-controlled uplift and exhumation. The deposit records, such as the distribution of coarse clastic sediments, the distribution of tectonically-controlled basins, stratigraphic discontinuousness or unconformity, and fault-controlled geomorphologic evolution, also match well with the strong uplift and exhumation periods. 9Deferoxamine promotes recovery of traumatic spinal cord injury by inhibiting ferroptosis显示文摘Ferroptosis is an iron-dependent novel cell death pathway. Deferoxamine, a ferroptosis inhibitor, has been reported to promote spinal cord injury repair. It has yet to be clarified whether ferroptosis inhibition represents the mechanism of action of Deferoxamine on spinal cord injury recovery. A rat model of Deferoxamine at thoracic 10 segment was established using a modified Allen's method. Ninety 8-week-old female Wistar rats were used. Rats in the Deferoxamine group were intraperitoneally injected with 100 mg/kg Deferoxamine 30 minutes before injury. Simultaneously, the Sham and Deferoxamine groups served as controls. Drug administration was conducted for 7 consecutive days. The results were as follows:(1) Electron microscopy revealed shrunken mitochondria in the spinal cord injury group.(2) The Basso, Beattie and Bresnahan locomotor rating score showed that recovery of the hindlimb was remarkably better in the Deferoxamine group than in the spinal cord injury group.(3) The iron concentration was lower in the Deferoxamine group than in the spinal cord injury group after injury.(4) Western blot assay revealed that, compared with the spinal cord injury group, GPX4, xCT, and glutathione expression was markedly increased in the Deferoxamine group.(5) Real-time polymerase chain reaction revealed that, compared with the Deferoxamine group, mRNA levels of ferroptosis-related genes Acyl-CoA synthetase family member 2(ACSF2) and iron-responsive element-binding protein 2(IREB2) were up-regulated in the Deferoxamine group.(6) Deferoxamine increased survival of neurons and inhibited gliosis. These findings confirm that Deferoxamine can repair spinal cord injury by inhibiting ferroptosis. 12Cenozoic sedimentary records and geochronological constraints of differential uplift of the Qinghai-Tibet Plateau显示文摘Geological mapping data (1:250000) in the Qinghai-Tibet Plateau and its adjacent regions reveal the sediment sequences, distribution and tectonic evolution of the 92 Tertiary remnant basins. Southern Tibet and the Yecheng area in Xinjiang, located at southern and northwestern margins of the Qinghai-Tibet Plateau, respectively, were parts of the Neo-Tethys remnant sea in the Paleogene. In southern Tibet, both the subabyssal and abyssal sequences occur at the Gyangze, Saga, Guoyala, and Sangmai areas. The deep-water facies successions outcrop in the west, whereas the shallow-water facies sequences in the east, indicating the east to the west retreat of the Neo-Tethys Ocean. The retreat of the Neo-Tethys Ocean in the east was contributed to the earlier tectonic uplift of the eastern Qinghai-Tibet Plateau. The uplift process of the Plateau from the Late Cretaceous to Pliocene is described as follows: During the Late Cretaceous, tectonic uplift of the Qinghai-Tibet Plateau occurred in the northeastern part and the configuration of the Qinghai-Tibet Plateau was characterized by rise in the northeast and depression in the west. In the Paleocene-Eocene interval, the Tengchong-Baingoin and Kuyake-Golmud areas experienced local tectonic uplifting, the West Kunlun uplift zone broadened easterly, the Qilian uplift zone broadened southerly, and the Songpan-Garzê uplift zone shrank easterly. The Oligocene configuration of the Qinghai-Tibet Plateau was characterized by mountain chains rising along its margins and sedimentary basins in the central part because of tectonic uplifts of the Gangdisê and the Himalaya blocks. Meanwhile, the Kunlun-Altyn-Qilian uplift zones have also broadened southerly and northerly. In contrast, the great uplift zones of the Gangdisê, the Himalaya, the Karakorum, and the Kunlun blocks characterize the paleogeographic contours of the Qinghai-Tibet Plateau during the Miocene-Pliocene. Additionally, the thermochronological data on tectonic uplift events in southern Tibet, West Kunlun Mountains, Altyn Tagh, eastern Tibet, and western Sichuan all suggest that the most intense deformation occurred at 13-8 Ma and since 5 Ma, respectively, corresponding to two great uplift periods in Neogene. As a result, turnover of paleogeographic configuration of the Qinghai-Tibet Plateau occurred during the Neogene, experiencing a change from high contours in the east in the pre-Oligocene to high contours in the west at the end-Pliocene. 13Fringe pattern analysis using deep learning显示文摘In many optical metrology techniques,fringe pattern analysis is the central algorithm for recovering the underlying phase distribution from the recorded fringe patterns.Despite extensive research efforts for decades,how to extract the desired phase information,with the highest possible accuracy,from the minimum number of fringe patterns remains one of the most challenging open problems.Inspired by recent successes of deep learning techniques for computer vision and other applications,we demonstrate for the first time,to our knowledge,that the deep neural networks can be trained to perform fringe analysis,which substantially enhances the accuracy of phase demodulation from a single fringe pattern.The effectiveness of the proposed method is experimentally verified using carrier fringe patterns under the scenario of fringe projection profilometry.Experimental results demonstrate its superior performance,in terms of high accuracy and edge-preserving,over two representative single-frame techniques:Fourier transform profilometry and windowed Fourier transform profilometry.Shijie Feng Qian Chen Guohua Gu Tianyang Tao Liang Zhang Yan Hu Wei Yin Chao Zuo 2019Advanced Photonics2019,1,2:31

15Ferroptosis,radiotherapy,and combination therapeutic strategies显示文摘Ferroptosis,an iron-dependent form of regulated cell death driven by peroxidative damages of polyunsatu-rated-fatty-acid-containing phospholipids in cellular membranes,has recently been revealed to play an important role in radiotherapy-induced cell death and tumor suppression,and to mediate the synergy between radiotherapy and immunotherapy.In this review,we summarize known as well as putative mechanisms underlying the crosstalk between radiotherapy and fer-roptosis,discuss the interactions between ferroptosis and other forms of regulated cell death induced by radiotherapy,and explore combination therapeutic strategies targeting ferroptosis in radiotherapy and immunotherapy.This review will provide important frameworks for future investigations of ferroptosis in cancer therapy.Guang Lei Chao Mao Yuelong Yan Li Zhuang Boyi Gan 2021Protein & Cell2021,12,11:29
16Geological Characteristics and Ore-forming Time of the Dexing Porphyry Copper Ore Mine in Jiangxi Province显示文摘The Dexing porphyry copper ore mine is located in the Qin-Hang metallogenic belt between the Yangtze block and the Cathaysia block. It is a giant porphyry copper mine in China, including 3 ore districts: Tongchang, Fujiawu and Zhushahong. Our analyses of Re in molybdenite indicate that the ore-forming material of the copper ore deposits in Dexing should be mainly mantle-derived. Our study fills in a gap in the study of formation time of the Dexing copper mine, and further proves that the copper ore deposits in the three ore districts should be formed simultaneously, about 170 Ma, belonging to the early Yanshan period, and that the formation time of the copper ore deposits should be consistent with the formation time of granodiorite porphyry in which the copper ore deposits are hosted. 18Construction, expression and characterization of the engineered antibody against tumor surface antigen, p185^(c-erbB-2)显示文摘The c-erbB-2 proto-oncogene encodes a 185kDa protein p185, which belongs to epidermal growth factorreceptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis forcertain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibitsproliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. Inorder to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed thesingle-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichiacoli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cellimmunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain(ECD) of p185. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion moleculewith a homodimer form and the recombinant product was expressed in mammalian cells. In a series ofsubsequent analysis this fusion protein showed identical antigen binding site and activity with the parentantibody. 19Evaluation of the 7^(th) edition of the TNM classification in patients with resected esophageal squamous cell carcinoma显示文摘AIM:To evaluate the prognostic factors and tumor stages of the 7th edition TNM classification for esophageal cancer.METHODS:In total,1033 patients with esophageal squamous cell carcinoma(ESCC)who underwent surgical resection with or without(neo)adjuvant therapy between January 2003 and June 2012 at the Thoracic Surgery DepartmentⅡof the Beijing Cancer Hospital,Beijing,China were included in this study.The following eligibility criteria were applied:(1)squamous cell carcinoma of the esophagus or gastroesophageal junction identified by histopathological examination;(2)treatment with esophagectomy plus lymphadenectomy with curative intent;and(3)complete pathologic reports and follow-up data.Patients who underwent non-curative(R1)resection and patients who died in hospital were excluded.Patients who received(neo)adjuvant therapy were also included in thisanalysis.All patients were restaged using the 7th edition of the Union for International Cancer Control and the American Joint Committee on Cancer TNM staging systems.Univariate and multivariate analyses were performed to identify the prognostic factors for survival.Survival curves were plotted using the Kaplan-Meier method,and the log-rank test was used to evaluate differences between the subgroups.RESULTS:Of the 1033 patients,273 patients received(neo)adjuvant therapy,and 760 patients were treated with surgery alone.The median follow-up time was 51.6mo(range:5-112 mo)and the overall 5-year survival rate was 36.4%.Gender,'p T'and'p N'descriptors,(neo)adjuvant therapy,and the 7th edition TNM stage grouping were independent prognostic factors in the univariate and multivariate analyses.However,neither histologic grade nor cancer location were independent prognostic factors in the univariate and multivariate analyses.The 5-year stage-based survival rates were as follows:ⅠA,84.9%;ⅠB,70.9%;ⅡA,56.2%;ⅡB,43.3%;ⅢA,37.9%;ⅢB,23.3%;ⅢC,12.9%andⅣ,3.4%.There were significant differences between each adjacent staging classification.Moreover,there were significant differences between each adjacent p N and p M subgroup.According to the p T descriptor,there were significant differences between each adjacent subgroup except between p T3 and p T4(P=0.405).However,there was no significant difference between each adjacent histologic grade subgroup and between each adjacent cancer location subgroup.CONCLUSION:The 7th edition is considered to be valid for patients with resected ESCC.However,the histologic grade and cancer location were not prognostic factors for ESCC.Jia Wang Nan Wu Qing-Feng Zheng Shi Yan Chao Lv Shao-Lei Li Yue Yang 2014World Journal of Gastroenterology2014,20,48:23
