| 1 | Blockchain: a secure, decentralized, trusted cyber infrastructure solution for future energy systems显示文摘Modern power systems are rapidly evolving into complex cyber-physical systems. The increasingly complex interaction among different energy entities calls for a secure, efficient, and robust cyber infrastructure. As an emerging distributed computing technology, Blockchain provides a secure environment to support such interactions.This paper gives a prospective on using Blockchain as a secure, distributed cyber infrastructure for the future grid.Firstly, the basic principles of Blockchain and its state-ofthe-art are introduced. Then, a Blockchain based smart grid cyber-physical infrastructure model is proposed. Afterwards, some promising application domains of Blockchain in future grids are presented. | 2 | External use of Ruyanneixiao cream efficiently blocks precancerous mammary lesions by interfering with glycolysis induced by inhibition of hypoxia inducible factor-1α, hexokinase 2, phosphofructokinase, and pyruvate kinase M2 expression显示文摘OBJECTIVE:To investigate the effect of Ruyanneixiao cream(RYNX) on the expression of hypoxia inducible factor-1α(HIF-1α), hexokinase 2(HK2),phosphofructokinase(PFK), and pyruvate kinase M2(PKM2) mRNA and protein in MCF-10 AT cells and in an animal model of precancerous mammary lesions.METHODS:Following treatment of MCF-10 AT cells with RYNX, tamoxifen(TAM) and YC-1 for 48 h,HIF-1α, HK2, PFK, PKM2 mRNA and protein expression was analyzed.Fifty female SD rats were randomly divided into control, model, TAM, and highand low-dose RYNX groups, with 10 rats in each group.A precancerous mammary lesion model was established for all groups except the control group.High-and low-dose RYNX cream containing TAM was coated on the breasts of animals in the corresponding groups.The rat mammary tissue was removed in the 10 th week and HIF-1α, HK2, PFK,PKM2 mRNA and protein was analyzed.RESULTS:In vitro analyses demonstrated that, compared with the matrix group, HIF-1α, HK2, PFK,PKM2 mRNA and protein expression was significantly decreased in the RYNX group(P < 0.05).Compared with the YC-1 + RYNX group, HK2, PFK,and PKM2 protein expression was significantly reduced in the RYNX group.HIF-1α, HK2, PFK, and PKM2 protein expression was increased significantly in the model group(P < 0.05) compared with the control group, while HIF-1α, HK2, PFK, and PKM2 mRNA and protein expression was significantly decreased in both the high-and low-dose RYNX groups(P < 0.05), with the effect being greater in the high-dose group.CONCLUSION:RYNX can block precancerous breast lesions by decreasing the expression of HK2,PFK, and PKM2 mRNA and protein via inhibition of HIF-1α mRNA and protein overexpression in a dose-dependent manner. | Li Xiaobo Ma Min Zhang Guijuan Ma Yi Liao Rui Chen Ruixue Yan Xianxin Bie Fengjie Huang Maojie Liang Shijie | 2017 | Journal of Traditional Chinese Medicine2017,37,2: | 5 |

| 5 | The gene expression level and prognosis analysis of membrane protein AP2M1 based on the protein spectrum of exosomes in hepatocellular carcinoma cells显示文摘Objective:Adopting mass spectrometry to analyze the protein spectrum of the exosomes in human hepatocellular carcinoma cells and screening the target membrane protein in order to analyze the relationship between its gene expression level and prognosis.Methods:The exosomes of hepatocellular carcinoma cells HepG2 and 7721 were isolated by ultracentrifugation,and the extracted exosomes were verified by transmission electron microscope, western blotting,and nanosight.The relative quantitative proteomics analysis was used to analyze the protein spectrum of HepG2 and 7721 cells in exosomes.The bioinformatics was used to collect the signal pathway,and the target protein AP2M1 was verified by qRTPCR.Gene Expression Profiling Interactive Analysis(GEPIA),OncoLnc,and The Cancer Genome Atlas(TCGA)database were used to analyze the expression level of AP2M1 and the survival rate of patients with liver cancer.Results:The extracellular exosomes of HepG2 and 7721 cells were isolated by ultracentrifugation.High-purity exosomes were obtained and verified by transmission electron microscopy,western blotting,and nanosight.There were 836 proteins co-expressed by the exosomes of HepG2 and 7721.By the analysis of Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway,a total of 34 pathways that were related to liver cancer were collected,and the target membrane protein AP2M1 was screened.AP2M1 highly expressed in HepG2 cells,HepG2 and Huh7 cells' exosomes were detected by qRT-PCR(P<0.05).The data of GEPIA showed that comparing to the adjacent tissues,gene AP2M1 was highly expressed in hepatocarcinoma tissues.Kaplan-Meier survival curve analysis was performed by OncoLnc.It was found that the survival rate of patients with liver cancer and high expression of AP2M1 was significantly lower than patients with liver cancer and low expression of AP2M1(P<0.01).Conclusion:The high expression of membrane protein AP2M1 in the exosomes of hepatoma cells and the high expression of its gene in liver cancer tissues predicted the low survival rate. | Lifang Liang Chunmeng Wei Yasi Li Fengjie Wan Xuejing Huang Min He | 2018 | 广西医科大学学报2018,35,11: | 1 |
| 6 | Insulin-resistance and depression cohort data mining to identify nutraceutical related DNA methylation biomarker for type 2 diabetes显示文摘Insulin-resistance(IR)is one of the most important precursors of type 2 diabetes(T2D).Recent evidence suggests an association of depression with the onset of T2D.Accumu-lating evidence shows that depression and T2D share common biological origins,and DNA methylation examination might reveal the link between lifestyle,disease risk,and potential therapeutic targets for T2D.Here we hypothesize that integrative mining of IR and depression cohort data will facilitate predictive biomarkers identification for T2D.We utilized a newly proposed method to extract gene-level information from probe level data on genome-wide DNA methylation array.We identified a set of genes associated with IR and depression in clin-ical cohorts.By overlapping the IR-related nutraceutical-gene network with depression net-works,we identified a common subnetwork centered with Vitamin D Receptor(VDR)gene.Preliminary clinical validation of gene methylation set in a small cohort of T2D patients and controls was established using the Sequenome matrix-assisted laser desorption ionization-time flight mass spectrometry.A set of sites in the promoter regions of VDR showed a signifi-cant difference between T2D patients and controls.Using a logistic regression model,the optimal prediction performance of these sites was AUC Z 0.902,and an odds ratio Z 19.76.Thus,monitoring the methylation status of specific VDR promoter region might help stratify the high-risk individuals who could potentially benefit from vitamin D dietary sup-plementation.Our results highlight the link between IR and depression,and the DNA methyl-ation analysis might facilitate the search for their shared mechanisms in the etiology of T2D. | Fengji Liang Yuan Quan Andong Wu Ying Chen Ruifeng Xu Yuexing Zhu Jianghui Xiong | 2021 | Genes & Diseases2021,8,5: | 0 |
