1Volcanism at the Permian-Triassic Boundary in South China and Its Effects on Mass Extinction显示文摘This paper discusses the clayrocks widespread at the Permian-Triassic boundary, which are mostly of volcanic origin. Volcanogenetic textures, structures and minerals such as high-temperature quartz are found in clayrocks at the Permian-Triassic boundary in many places. Thousands of microspherules have been collected from the Boundary clayrocks, many of which exhibit the typical features of the process from melting to cooling and solidification. indicating that they were formed by volcanic eruption or extraterrestrial impact. Volcanic effects on the Permian-Triassic mass extinction may be reflected in conodonts, algae and ammonoids. The Boundary clayrocks are found in many Permian-Triassic sections along the coast of Tethys. 2Advancing application of mesenchymal stem cell-based bone tissue regeneration显示文摘Reconstruction of bone defects,especially the critical-sized defects,with mechanical integrity to the skeleton is important for a patient's rehabilitation,however,it still remains challenge.Utilizing biomaterials of human origin bone tissue for therapeutic purposes has provided a facilitated approach that closely mimics the critical aspects of natural bone tissue with regard to its properties.However,not only efficacious and safe but also costeffective and convenient are important for regenerative biomaterials to achieve clinical translation and commercial success.Advances in our understanding of regenerative biomaterials and their roles in new bone formation potentially opened a new frontier in the fast-growing field of regenerative medicine.Taking inspiration from the role and multicomponent construction of native extracellular matrix(ECM)for cell accommodation,the ECM-mimicking biomaterials and the naturally decellularized ECM scaffolds were used to create new tissues for bone restoration.On the other hand,with the going deep in understanding of mesenchymal stem cells(MSCs),they have shown great promise to jumpstart and facilitate bone healing even in diseased microenvironments with pharmacology-based endogenous MSCs rescue/mobilization,systemic/local infusion of MSCs for cytotherapy,biomaterials-based approaches,cell-sheets/-aggregates technology and usage of subcellular vesicles of MSCs to achieve scaffolds-free or cell-free delivery system,all of them have been shown can improve MSCsmediated regeneration in preclinical studies and several clinical trials.Here,following an overview discussed autogenous/allogenic and ECM-based bone biomaterials for reconstructive surgery and applications of MSCsmediated bone healing and tissue engineering to further offer principles and effective strategies to optimize MSCs-based bone regeneration.Fengqing Shang Yang Yu Shiyu Liu Leiguo Ming Yongjie Zhang Zhifei Zhou Jiayu Zhao Yan Jin 2021Bioactive Materials2021,6,3:13
3Utilization of Ogura CMS germplasm with the clubroot resistance gene by fertility restoration and cytoplasm replacement in Brassica oleracea L显示文摘Clubroot disease,a major plant root disease caused by Plasmodiophora brassicae,has become one of the most destructive diseases among cultivated cruciferous vegetables.However,clubroot-resistant Brassica oleracea materials are rare.A few clubroot-resistant cabbage varieties are available on the market,but all are Ogura cytoplasmic male sterile(CMS)types.Therefore,in this study,to reutilize the clubroot-resistant Ogura CMS germplasm of cabbage,a new fertility-restored Ogura CMS material,16Q2-11,was used as a bridge to transfer the clubroot resistance(CR)gene from the Ogura CMS cytoplasm to the normal cytoplasm by a two-step method(a fertility restoration and cytoplasm replacement method).In the first cross for fertility restoration of Ogura CMS clubroot-resistant cabbage(FRCRC),16Q2-11 was used as a restorer to cross with Ogura CMS materials containing the CR gene CRb2.Eleven Rfo-positive progenies were generated,of which four contained CRb2:F8-514,F8-620,F8-732 and F8-839.After inoculation with race 4 of P.brassicae,these four CRb2-positive individuals showed resistance.Furthermore,F8-514 and F8-839 were then used as male parents in the second cross of FRCRC to cross with cabbage inbred lines,resulting in the successful introgression of the CRb2 gene into the inbred lines.All offspring produced from this step of cross,which had a normal cytoplasm,showed a high resistance to race 4 of P.brassicae and could be utilized for the breeding of clubrootresistant cabbage varieties in the future.This is the first time that the Ogura CMS restorer has been used to restore the fertility of Ogura CMS clubroot-resistant cabbages,which could improve germplasm diversity in cabbage and provide a reference method for using CMS germplasm in Brassica crops.Wenjing Ren Zhiyuan Li Fengqing Han Bin Zhang Xing Li Zhiyuan Fang Limei Yang Mu Zhuang Honghao Lv Yumei Liu Yong Wang Hailong Yu Yangyong Zhang 2020Horticulture Research2020,7,1:7
4Effect on platelet aggregation activity: extracts from 31 Traditional Chinese Medicines with the property of activating blood and resolving stasis显示文摘OBJECTIVE: To evaluate the anti-platelet aggregation effects of extracts from 31 Traditional Chinese Medicines(TCM) with the property of activating blood and resolving stasis in terms of TCM theory.METHODS: The 31 TCMs extracts were prepared using water, 90% ethanol and ethyl acetate., and the effects on anti-platelet aggregation were tested on a platelet aggregation analyzer in vitro with adenosine 5'-diphosphate, bovine thrombin and arachi-donic acid(AA) as aggregation inducers, respectively. 7Auxin response factor 6A regulates photosynthesis,sugar accumulation,and fruit development in tomato显示文摘Auxin response factors(ARFs)are involved in auxin-mediated transcriptional regulation in plants.In this study,we performed functional characterization of SlARF6A in tomato.SlARF6A is located in the nucleus and exhibits transcriptional activator activity.Overexpression of SlARF6A increased chlorophyll contents in the fruits and leaves of tomato plants,whereas downregulation of SlARF6A decreased chlorophyll contents compared with those of wild-type(WT)plants.Analysis of chloroplasts using transmission electron microscopy indicated increased sizes of chloroplasts in SlARF6A-overexpressing plants and decreased numbers of chloroplasts in SlARF6A-downregulated plants.Overexpression of SlARF6A increased the photosynthesis rate and accumulation of starch and soluble sugars,whereas knockdown of SlARF6A resulted in opposite phenotypes in tomato leaves and fruits.RNA-sequence analysis showed that regulation of SlARF6A expression altered the expression of genes involved in chlorophyll metabolism,photosynthesis and sugar metabolism.SlARF6A directly bound to the promoters of SlGLK1,CAB,and RbcS genes and positively regulated the expression of these genes.Overexpression of SlARF6A also inhibited fruit ripening and ethylene production,whereas downregulation of SlARF6A increased fruit ripening and ethylene production.SlARF6A directly bound to the SAMS1 promoter and negatively regulated SAMS1 expression.Taken together,these results expand our understanding of ARFs with regard to photosynthesis,sugar accumulation and fruit development and provide a potential target for genetic engineering to improve fruit nutrition in horticulture crops.Yujin Yuan Xin Xu Zehao Gong Yuwei Tang Mengbo Wu Fang Yan Xiaolan Zhang Qian Zhang Fengqing Yang Xiaowei Hu Qichen Yang Yingqing Luo Lihua Mei Wenfa Zhang Cai-Zhong Jiang Wangjin Lu Zhengguo Li Wei Deng 2019Horticulture Research2019,6,1:3
8Timing of the terrestrial Permian-Triassic boundary biotic crisis:Implications from U-Pb dating of authigenic zircons显示文摘The Late Permian to Early Triassic transition represents one of the most important Phanerozoic mass extinction episodes. The cause of this event is still in debate between catastrophic and gradual mechanisms. This study uses the U-Pb method on zircons from the uppermost Permian/lowermost Triassic clay deposits at Chahe (Guizhou Province, SW China) to examine time constraints for this event. The results of both this and previous studies show that the ages of Bed 68a and 68c (the upper clay bed of the terrestrial Permian-Triassic boundary (PTB)) respectively are 252.6±2.8 and 247.5±2.8 Ma. This age (within the margin of error) almost accords with the upper clay bed (Bed 28) age of Meishan and the eruption age of Tunguss Basalt, and is so far the most accurate age obtained from terrestrial PTB. The claystone of Bed 68 was formed in the earliest Triassic. The biotic crisis occurred at nearly the same time in terrestrial and marine environments during Permian-Triassic interval; however the extinction patterns and processes are different. 10Characteristics of Bacillus cereus Spore Counts in Soils w ith Underlying Gold Mineralization in Northwestern Sichuan, China: Results from a Pilot Study显示文摘A pilot survey on a microbial mineral exploration method based on so il Bacillus cereus spore counts was carried out across three different gold mi ning regions, which vary in soil type, climate condition and geological setting in northwestern Sichuan, China. B horizon soils from these sites were analyzed for B. cereus spores, Au, Ag, Cu, Pb and Zn. The results show that the numbe rs of B. cereus spores generally increased in soils overlying gold mineraliz ation. Specifically, elevated spore counts were found in samples slightly offset from the outcrops of orebodies, whereas soils directly above the outcrops of or ebodies usually contained low spore counts. 17The Songpan-Garze Massif: Its Relation to the QinlingFold Belt and Yangtze Platform and Developmental History显示文摘Whether there existed the Songpan-Garze massif is a controversial problem. This paper expounds and proves that the old basement of the massif is represented by the pre-Sinian granitic rock series. This massif and the South Qinling fold belt might both be a part of the old Yangtze platform. Rifting generated by the Caledonian orogeny in the terminal Early Palaeozoic caused the massif to be disintegrated from the northwestern part of the Yangtze platform. This disintegration, however, was not thorough, and the rift troughs were later gradually closed and filled up. The Emei taphrogeny that was initiated in the Early Permian Maokou’ an Stage involved a second disintegration of this massif from the Yangtze platform. The rift line largely goes along the Muli-Pingwu line. 18Level pinning of anti-PT-symmetric circuits for efficient wireless power transfer显示文摘Wireless power transfer(WPT) technology based on magnetic resonance(a basic physical phenomenon)can directly transfer energy from the source to the load without wires and other physical contacts, and has been successfully applied to implantable medical devices, electric vehicles, robotic arms and other fields.However, due to the frequency splitting of near-field coupling, the resonant WPT system has some unique limitations, such as poor transmission stability and low efficiency. Here, we propose anti-resonance with level pinning for high-performance WPT. By introducing the anti-resonance mode into the basic WPT platform, we uncover the competition between dissipative coupling and coherent coupling to achieve novel level pinning, and construct an effective anti-parity-time(anti-PT)-symmetric non-Hermitian system that is superior to previous PT-symmetric WPT schemes. On the one hand, the eigenvalue of the anti-PT-symmetric system at resonance frequency is always pure real in both strong and weak coupling regions, and can be used to overcome the transmission efficiency decrease caused by weak coupling, as brought about by, for example, a large size ratio of the transmitter to receiver, or a long transmission distance.On the other hand, due to the level pinning effect of the two kinds of coupling mechanisms, the working frequency of the system is guaranteed to be locked, so frequency tracking is not required when the position and size of the receiver change. Even if the system deviates from the matching condition, an efficient WPT can be realized, thereby demonstrating the robustness of the level pinning. The experimental results show that when the size ratio of the transmitter coil to the receiver coil is 4.29(which is in the weak coupling region), the transfer efficiency of the anti-PT-symmetric system is nearly 4.3(3.2) times higher than that of the PT-symmetric system when the matching conditions are satisfied(deviated). 19Investigation on the Metabolism of Salvianolic acid B, Tanshinone Ⅱ A, Ginsenoside Rg1 and Ginsenoside Rb1 from Danshen-Sanqi Herbal Pair in Zebrafish by Liquid Chromatography-Tandem Mass Spectrometry Analysis显示文摘Background:Salvianolic acid B(SAB),tanshinone IIA(TIIA),ginsenoside Rg1(GRg1)and ginsenoside Rb1(GRb1)are the main active components of Danshen-Sanqi herbal pair(DS-SQ),which is widely used in the treatment of cardiovascular diseases in Asia.The compatibility mechanisms of compound Chinese herbal preparation may be clarified to some degree by understanding the interactions of the active components based on their metabolic research.Furthermore,zebrafish is a convenient and feasible model to study the metabolism of drugs.Objective:To investigate the metabolism of SAB,TIIA,GRg1 and GRb1 in zebrafish by liquid chromatography-tandem mass spectrometry(LC-MS/MS)analysis.Methods:Zebrafish embryos after 48-hour hatching were divided into nine experimental groups.The blank group was exposed to 1 mL ultra-pure water.Eight drug-treated groups were exposed to 1 mL solution of SAB,TIIA,GRg1,GRb1,SAB and GRg1,SAB and GRb1,TIIA and GRg1,TIIA and GRb1,respectively.After homogenization,they were analyzed by LC-MS/MS.The Structures of the metabolites were determined by analyzing their corre-sponding mass spectra information and comparing the data with relevant literature.According to the change of the amount of metabolites,the metabolic effect between SAB,TIIA,GRg1 and GRb1 were investigated.Results:Eighteen metabolites and four parent drugs of SAB,TIIA,GRg1 and GRb1 in zebrafish were identified by LC-MS/MS,including methylation,degradation,reduction,dehydrogenation,hydroxylation,desugarization and glucuronidation metabolites.Furthermore,after being combined exposed of drugs,GRg1 and GRb1 could promote the metabolism of TIIA,while SAB and TIIA could promote the metabolism of GRg1 and GRb1 to different degree.Conclusion:By using LC-MS/MS analysis,zebrafish was successfully applied to study the metabolism of SAB,TIIA,GRg1 and GRb1,which is significance to explain the possible synergistic effect of components in DS-SQ through modifying the metabolism in zebrafish.This study can be helpful for the further research of DS-SQ.Shijun Yin Congpeng Zhao Guang Hu Hua Chen Fengqing Yang 2021Clinical Complementary Medicine and Pharmacology2021,1,1:0
20Map-based cloning and CRISPR/Cas9-based editing uncover BoNA1 as the causal gene for the no-anthocyanin-accumulation phenotype in curly kale (Brassica oleracea var. sabellica)显示文摘Brassica oleracea comprises several important vegetable and ornamental crops,including curly kale,ornamental kale,cabbage,broccoli,and others.The accumulation of anthocyanins,important secondary metabolites valuable to human health,in these plants varies widely and is responsible for their pink to dark purple colors.Some curly kale varieties lack anthocyanins,making these plants completely green.The genetic basis of this trait is still unknown.We crossed the curly kale inbred line BK2019(without anthocyanins)with the cabbage inbred line YL1(with anthocyanins)and the Chinese kale inbred line TO1000(with anthocyanins)to generate segregating populations.The no-anthocyanin trait was genetically controlled by a recessive gene,bona1.We generated a linkage map and mapped bona1 to a 256-kb interval on C09.We identified one candidate gene,Bo9g058630,in the target genomic region;this gene is homologous to AT5G42800,which encodes a dihydroflavonol-4-reductase-like(DFR-like)protein in Arabidopsis.In BK2019,a 1-bp insertion was observed in the second exon of Bo9g058630 and directly produced a stop codon.To verify the candidate gene function,CRISPR/Cas9 gene editing technology was applied to knock out Bo9g058630.We generated three bona1 mutants,two of which were completely green with no anthocyanins,confirming that Bo9g058630 corresponds to BoNA1.Different insertion/deletion mutations in BoNA1 exons were found in all six of the other no-anthocyanin kale varieties examined,supporting that independent disruption of BoNA1 resulted in no-anthocyanin varieties of B.oleracea.This study improves the understanding of the regulation mechanism of anthocyanin accumulation in B.oleracea subspecies.Kaiwen Yuan Xinyu Zhao Wenru Sun Limei Yang Yangyong Zhang Yong Wang Jialei Ji Fengqing Han Zhiyuan Fang Honghao Lv 2023Horticulture Research2023,10,8:0
