| 1 | Successive inverse and normal magnetocaloric effects in the Mn-vacancy compound Mn0.95Co0.75Cu0.25Ge显示文摘Ferromagnetic-structural transformation has been studied widely in MnCoGe-based materials. However, the magnetostructural transition(MST) from antiferromagnetic(AFM) orthorhombic phase to ferromagnetic(FM) hexagonal phase, which may lead to a large inverse magnetocaloric effect(MCE), has rarely been reported. Here, the introduction of Mn vacancy lowers the structural transition temperature while retains the AFM state in the orthorhombic phase, thus successfully realizing the AFM-FM MST in Mn0.95Co0.75Cu0.25Ge. Moreover, successive inverse and normal MCEs are observed around the first-order AFM-FM MST and the second-order FM-paramagnetic(PM) transition, respectively. A thermostat is proposed based on this special feature, which could release heat above the critical temperature while absorb heat below the critical temperature by simply applying the same magnetization/demagnetization cycles. | 7 | Sublytic C5b-9 triggers glomerular mesangial cell apoptosis in rat Thy-1 nephritis via Gadd45 activation mediated by Egr-1 and p300-dependent ATF3 acetylation显示文摘The apoptosis of glomerular mesangial cells(GMCs)is considered to be an important contributor to the initiation and development of rat Thy-1 nephritis(Thy-1N)and is accompanied by sublytic C5b-9 deposition.However,the mechanism by which sublytic C5b-9 triggers GMC apoptosis has not been elucidated.In this study,functional and histological examinations were performed on GMCs treated with sublytic C5b-9(in vitro)and renal tissues of Thy-1N rats(in vivo).The in vitro studies found that sublytic C5b-9 could trigger GMC apoptosis through upregulating Egr-1,ATF3,and Gadd45 expression.Egr-1-mediated post-transcriptional modulation of ATF3,Egr-1/ATF3-enhanced Gadd45 promoter activity,and p300-mediated ATF3 acetylation were all involved in GMC apoptosis.More importantly,the effective binding elements for Egr-1 and ATF3 to Gadd45β/γpromoters and the ATF3 acetylation site were identified.In vivo,silencing renal p300,Egr-1,ATF3,and Gadd45β/γsignificantly decreased GMC apoptosis,secondary GMC proliferation,and urinary protein secretion in Thy-1N rats.Together,these findings implicate that sublytic C5b-9-induced activation of Egr-1/p300–ATF3/Gadd45 axis plays a critical role in GMC apoptosis in Thy-1N rats. | Fengxia He Mengya Zhou Tianyi Yu Dan Zhao Jing Zhang Wen Qiu Yanlai Lu Yu Liu Lulu Wang Yingwei Wang | 2016 | Journal of Molecular Cell Biology2016,8,6: | 1 |

Four new sufficient and necessary conditions for global asymptotic stability and global exponential stability of the equilibrium point of GRNs are derived. Specifically, using comparing theorem and Dini derivation method, three weak sufficient conditions for global stability of GRNs with constant time delays are proposed. Finally, a general GRN model is used to illustrate the effectiveness of the proposed theoretical results. | 12 | Transformation of microstructure and phase of disodium guanosine 5′-monophosphate: Thermodynamic perspectives显示文摘Microstructure and phase transformation of disodium guanosine 5′-monophosphate(5′-GMPNa_2) are extremely important for controlling the process and understanding the mechanism of crystallization. In this work, the thermodynamic properties of polymorphous 5′-GMPNa_2 especially the solubility were studied, the solubility results show that 5′-GMPNa_2 is more soluble in ethanol–water(E–W) than in isopropanol–water(I–W). The amorphous form of 5′-GMPNa_2 is more soluble than the crystalline form at the same mole fraction and temperature. Meanwhile, the crystalline forms and morphologies of the residual solids were characterized by PXRD and SEM. The results indicate that solid forms of 5′-GMPNa_2 transformed spontaneously from amorphous to crystalline when the ethanol proportion is ≥20%. In addition, increasing the pH facilitates the dissolution of 5′-GMPNa_2 and helps to maintain the crystalline form. The associated Gibbs free energy values were calculated to verify the trend of transformation from amorphous to crystalline 5′-GMPNa_2. | 19 | PHISDetector:A Tool to Detect Diverse In Silico Phage–host Interaction Signals for Virome Studies显示文摘Phage-microbe interactions are appealing systems to study coevolution,and have also been increasingly emphasized due to their roles in human health,disease,and the development of novel therapeutics.Phage-microbe interactions leave diverse signals in bacterial and phage genomic sequences,defined as phage-host interaction signals(PHISs),which include clustered regularly interspaced short palindromic repeats(CRISPR)targeting,prophage,and protein-protein interaction signals.In the present study,we developed a novel tool phage-host interaction signal detector(PHISDetector)to predict phage-host interactions by detecting and integrating diverse in silico PHISs,and scoring the probability of phage-host interactions using machine learning models based on PHIS features.We evaluated the performance of PHISDetector on multiple benchmark datasets and application cases.When tested on a dataset of 758 annotated phage-host pairs,PHISDetector yields the prediction accuracies of 0.51 and 0.73 at the species and genus levels,respectively,outperforming other phage-host prediction tools.When applied to 125,842 metagenomic viral contigs(mVCs)derived from 3042 geographically diverse samples,a detection rate of 54.54% could be achieved.Furthermore,PHISDetector could predict infecting phages for 85.6% of 368 multidrug-resistant(MDR)bacteria and 30% of 454 human gut bacteria obtained from the National Institutes of Health(NIH)Human Microbiome Project(HMP).The PHISDetector can be run either as a web server(http://gffzz439de54f28c841f5hknxf5cfvqqqp69pq.ffgz.tsg.suse.edu.cn/PHISDetector/)for general users to study individual inputs or as a stand-alone version(http://gffzz188fe103f8f1460asknxf5cfvqqqp69pq.ffgz.tsg.suse.edu.cn/HITImmunologyLab/PHISDetector)to process massive phage contigs from virome studies. | Fengxia Zhou Rui Gan Fan Zhang Chunyan Ren Ling Yu Yu Si Zhiwei Huang | 2022 | Genomics, Proteomics & Bioinformatics2022,20,3: | 0 |

This study aims to develop a novel strategy for detecting mutations in the VGSC gene inAe. albopictus using multiplex PCR-mass spectrometry (MPCR-MS) minisequencing technology.Methods We established a new strategy for detecting mutations in the VGSC gene inAe. albopictus using MPCR-MS minisequencing technology. MPCR amplification and mass probe extension (MPE) were first used, followed by single nucleotide polymorphism (SNP) typing mass spectrometry, which allows the simultaneous detection of multiple mutation sites of the VGSC gene in 96 samples ofAe. albopictus. A total of 70 wild-collectedAe. albopictus were used to evaluate the performance of the method by comparing it with other methods.Results Three target sites (1016, 1532, 1534) in the VGSC gene can be detected simultaneously by double PCR amplification combined with matrix-assisted laser desorption ionization-time-of-flight mass spectrometry, achieving a detection limit of 20 fg/μl. We applied this method to 70 wild-collectedAe. albopictus, and the obtained genotypes were consistent with the routine sequencing results, suggesting the accuracy of our method.Conclusions MPCR-MS minisequencing technology provides a sensitive and high-throughput approach toAe. albopictus VGSC gene mutation screening. Compared with conventional sequencing, this method is economical and time-saving.
