1Estrogen and insulin synergistically promote endometrial cancer progression via crosstalk between their receptor signaling pathways显示文摘Objective: Despite evidence that estrogens and insulin are involved in the development and progression of many cancers, their synergistic role in endometrial carcinoma(EC) has not been analyzed yet.Methods: Here, we investigated how estrogens act synergistically with insulin to promote EC progression. Cell growth in vitro and in vivo, effects of estradiol and insulin on apoptosis and cell cycle distribution, and expression and activation of estrogen receptor(ER), insulin receptor(InsR), and key proteins in the PI3K and MAPK pathways were examined after combined stimulation with estradiol and insulin.Results: Compared to EC cells treated with estradiol or insulin alone, those treated with both estradiol and insulin exhibited stronger stimulation. Estradiol significantly induced phosphorylation of InsR-β and IRS-1, whereas insulin significantly induced phosphorylation of ER-α. In addition, treatment with both insulin and estradiol together significantly increased the expression and phosphorylation of Akt, MAPK, and ERK. Notably, InsR-β inhibition had a limited effect on estradiol-dependent proliferation,cell cycle, and apoptosis, whereas ER-α inhibition had a limited insulin-dependent effect, in EC cell lines. 6Sublytic C5b-9 triggers glomerular mesangial cell apoptosis in rat Thy-1 nephritis via Gadd45 activation mediated by Egr-1 and p300-dependent ATF3 acetylation显示文摘The apoptosis of glomerular mesangial cells(GMCs)is considered to be an important contributor to the initiation and development of rat Thy-1 nephritis(Thy-1N)and is accompanied by sublytic C5b-9 deposition.However,the mechanism by which sublytic C5b-9 triggers GMC apoptosis has not been elucidated.In this study,functional and histological examinations were performed on GMCs treated with sublytic C5b-9(in vitro)and renal tissues of Thy-1N rats(in vivo).The in vitro studies found that sublytic C5b-9 could trigger GMC apoptosis through upregulating Egr-1,ATF3,and Gadd45 expression.Egr-1-mediated post-transcriptional modulation of ATF3,Egr-1/ATF3-enhanced Gadd45 promoter activity,and p300-mediated ATF3 acetylation were all involved in GMC apoptosis.More importantly,the effective binding elements for Egr-1 and ATF3 to Gadd45β/γpromoters and the ATF3 acetylation site were identified.In vivo,silencing renal p300,Egr-1,ATF3,and Gadd45β/γsignificantly decreased GMC apoptosis,secondary GMC proliferation,and urinary protein secretion in Thy-1N rats.Together,these findings implicate that sublytic C5b-9-induced activation of Egr-1/p300–ATF3/Gadd45 axis plays a critical role in GMC apoptosis in Thy-1N rats.Fengxia He Mengya Zhou Tianyi Yu Dan Zhao Jing Zhang Wen Qiu Yanlai Lu Yu Liu Lulu Wang Yingwei Wang 2016Journal of Molecular Cell Biology2016,8,6:1
7Gap engineering of sandwich plasmonic gap nanostructures for boosting plasmon-enhanced electrocatalysis显示文摘Plasmonic catalysis is emerging as a dynamic field in heterogeneous catalysis and holds great promise for the efficient utilization of solar energy.Central to the development of plasmonic catalysis is the design of efficient plasmonic nanocatalysts.In this report,plasmonic gap nanostructures(PGNs)on the basis of Au@poly(o-phenylenediamine)(POPD)@Pd sandwich nanostructures are synthesized as plasmonic nanocatalysts by an in-situ reduction synthetic strategy,which allows for the precise engineering of the POPD gap size between plasmonic Au and catalytic Pd components.The introduction of conducting POPD nanogap in PGNs not only effectively enhances their light harvesting capability,but also provides an effective charge transfer channel for harnessing the photogenerated hot charge carriers.In this respect,distinct gap-dependent performances in plasmon-enhanced electrocatalysis of ethanol oxidation reactions(EOR)are demonstrated with the PGN nanocatalysts and over 2.5 folds of enhancement can be achieved.A volcano plot is derived to describe the relationship between the catalytic activities and gap size of the PGN nanocatalysts,which is well explained by the interplay of their light harvesting and charge transport capabilities.These results highlight the importance of gap engineering in PGNs for plasmonic catalysis and offer the promise of developing efficient plasmonic nanocatalysts for other heterogeneous catalytic reactions.Lu Cheng Fengxia Wu Yu Tian Xiali Lv Fenghua Li Guobao Xu Hsien-Yi Hsu Yongjun Zhang Wenxin Niu 2023Nano Research2023,16,7:1

11In vivo and in vitro development of Tibetan antelope(Pantholops hodgsonii)interspecific cloned embryos显示文摘The Tibetan antelope is endemic to the Tibetan Plateau,China,and is now considered an endangered species.As a possible rescue strategy,the development of embryos constructed by interspecies somatic cell nuclear transfer(iSCNT)was examined.Tibetan antelope fibroblast cells were transferred into enucleated bovine,ovine and caprine oocytes.These cloned embryos were then cultured in vitro or in the oviducts of intermediate animals.Less than 0.5%of the reconstructed antelope-bovine embryos cultured in vitro developed to the blastocyst stage.However,when the cloned antelope-bovine embryos were transferred to caprine oviducts,about 1.6%of the embryos developed to the blastocyst stage.In contrast,only 0.7%of the antelope-ovine embryos developed to the morula stage and none developed to blastocysts in ovine oviducts.The treatment of donor cells and bovine oocytes with trichostatin A did not improve the embryo development even when cultured in the oviducts of ovine and caprine.When the antelope-bovine embryos,constructed from oocytes treated with roscovitine or trichostatin A,were cultured in rabbit oviducts 2.3%and 14.3%developed to blastocysts,respectively.It is concluded that although some success was achieved with the protocols used,interspecies cloning of Tibetan antelope presents difficulties still to be overcome.The mechanisms resulting in the low embryo development need investigation and progress might require a deeper understanding of cellular reprogramming.Guanghua SU Lei CHENG Yu GAO Kun LIU Zhuying WEI Chunling BAI Fengxia YIN Li GAO Guangpeng LI Shorgan BOU 2014Frontiers of Agricultural Science and Engineering2014,1,1:1

13Ionic/electronic conductivity regulation of n-type polyoxadiazole lithium sulfonate conductive polymer binders for high-performance silicon microparticle anodes显示文摘Low-cost silicon microparticles(SiMP),as a substitute for nanostructured silicon,easily suffer from cracks and fractured during the electrochemical cycle.A novel n-type conductive polymer binder with excellent electronic and ionic conductivities as well as good adhesion,has been successfully designed and applied for high-performance SiMP anodes in lithium-ion batteries to address this problem.Its unique features are attributed to the stro ng electron-withdrawing oxadiazole ring structure with sulfonate polar groups.The combination of rigid and flexible components in the polymer ensures its good mechanical strength and ductility,which is beneficial to suppress the expansion and contraction of SiMP s during the charge/discharge process.By fine-tuning the monomer ratio,the conjugation and sulfonation degrees of the polymer can be precisely controlled to regulate its ionic and electronic conductivities,which has been systematically analyzed with the help of an electrochemical test method,filling in the gap on the conductivity measurement of the polymer in the doping state.The experimental results indicate that the cell with the developed n-type polymer binder and SiMP(~0.5 μm) anodes achieves much better cycling performance than traditional non-conductive binders.It has been considered that the initial capacity of the SiMP anode is controlled by the synergetic effect of ionic and electronic conductivity of the binder,and the capacity retention mainly depends on its electronic conductivity when the ionic conductivity is sufficient.It is worth noting that the fundamental research of this wo rk is also applicable to other battery systems using conductive polymers in order to achieve high energy density,broadening their practical applications.Yuanyuan Yu Huihui Gao Jiadeng Zhu Dazhe Li Fengxia Wang Chunhui Jiang Tianhaoyue Zhong Shuheng Liang Mengjin Jiang 2021Chinese Chemical Letters2021,32,1:1

16PHISDetector:A Tool to Detect Diverse In Silico Phage–host Interaction Signals for Virome Studies显示文摘Phage-microbe interactions are appealing systems to study coevolution,and have also been increasingly emphasized due to their roles in human health,disease,and the development of novel therapeutics.Phage-microbe interactions leave diverse signals in bacterial and phage genomic sequences,defined as phage-host interaction signals(PHISs),which include clustered regularly interspaced short palindromic repeats(CRISPR)targeting,prophage,and protein-protein interaction signals.In the present study,we developed a novel tool phage-host interaction signal detector(PHISDetector)to predict phage-host interactions by detecting and integrating diverse in silico PHISs,and scoring the probability of phage-host interactions using machine learning models based on PHIS features.We evaluated the performance of PHISDetector on multiple benchmark datasets and application cases.When tested on a dataset of 758 annotated phage-host pairs,PHISDetector yields the prediction accuracies of 0.51 and 0.73 at the species and genus levels,respectively,outperforming other phage-host prediction tools.When applied to 125,842 metagenomic viral contigs(mVCs)derived from 3042 geographically diverse samples,a detection rate of 54.54% could be achieved.Furthermore,PHISDetector could predict infecting phages for 85.6% of 368 multidrug-resistant(MDR)bacteria and 30% of 454 human gut bacteria obtained from the National Institutes of Health(NIH)Human Microbiome Project(HMP).The PHISDetector can be run either as a web server(http://gffzz439de54f28c841f5h90n9vbnwu5ww6o0k.ffgz.tsg.suse.edu.cn/PHISDetector/)for general users to study individual inputs or as a stand-alone version(http://gffzz188fe103f8f1460as90n9vbnwu5ww6o0k.ffgz.tsg.suse.edu.cn/HITImmunologyLab/PHISDetector)to process massive phage contigs from virome studies.Fengxia Zhou Rui Gan Fan Zhang Chunyan Ren Ling Yu Yu Si Zhiwei Huang 2022Genomics, Proteomics & Bioinformatics2022,20,3:0
17Application value of Qisexingtai hand diagnostic method in diagnosis of coronary artery disease显示文摘OBJECTIVE:To evaluate the diagnostic efficacy of Qisexingtai hand diagnostic method in the diagnosis of coronary artery disease(CAD).METHODS:This was an investigator-initiated,prospective,multi-center,cross-sectional study.All the participants from three hospitals in China had been diagnosed by both Qisexingtai hand diagnostic method and coronary angiography.We compared the two diagnostic methods to calculate the sensitivity,the specificity,the omission diagnostic rate,the mistaken diagnostic rate and accuracy in order to evaluate the diagnostic efficacy of Qisexingtai hand diagnostic method for CAD.RESULTS:A total of 326 subjects were enrolled,diagnosed by both Qisexingtai hand diagnostic method and coronary angiography.As a result,there were 166 positive cases according to Qisexingtai hand diagnostic method,and 131 positive cases according to coronary angiography.Compared with the results of coronary angiography,the sensitivity of Qisexingtai hand diagnostic method was 80.2%,the specificity was 68.7%,the omission diagnostic rate was 19.8%,the mistaken diagnostic rate was 31.3%,and the accuracy was 73.3%.Area under the receiver operating characteristic curve was estimated as 0.735 for all,and 0.718,0.735,0.783 for the three sub-centers.CONCLUSION:Qisexingtai hand diagnostic method with high accuracy and sensitivity has certain application value in the diagnosis of CAD.LIU Qian ZHAO Zhupei GUO Hua WANG Shiheng HAO Mingzhao QIN Peijie WANG Liuqing LIU Xuechun ZHANG Fengxia AN Hong LI Yu SHI Xueqin ZHU Tingyu LIU Jianfeng WANG Liying XU Tao ZHANG Limei ZOU Xu 2022Journal of Traditional Chinese Medicine2022,42,5:0

Thankfully, with 'information space' as the carrier, the 'systems science' as the guidance, and QIAN Xuesen's 'metasynthetic wisdom system' as the core, QIAN Xuesen think tank provides the decision support for China to develop an aerospace knowledge economy and to continuously enhance the 'space soft power'.
