1Ectopic expression of antifreeze protein gene from Ammopiptanthus nanus confers chilling tolerance in maize显示文摘Improved chilling tolerance is important for maize production. Previous efforts in transgenics and marker-assisted breeding have not achieved practical results. In this study, the antifreeze protein(AnAFP) from the super-xerophyte Ammopiptanthus nanus was aligned to KnS-type dehydrins.Phosphorylation in vitro and subcellular localization showed that AnAFP was phosphorylated by maize casein kinase II and translocated from nucleus to cytoplasm under chilling stress. AnAFP also increased lactate dehydrogenase activity. A parent line of commercial maize hybrids was transformed with the AnAFP gene. Based on thermal asymmetric interlaced PCR, one hemizygous and two homozygous integration sites were identified in one T_(1) line. Ectopic expression of AnAFP in transgenic lines was confirmed by quantitative real-time PCR, RNA-seq, and Western blotting. After chilling treatment, the transgenic lines showed significantly improved phenotype, higher kernel production, survival rate and biomass, and lower relative electrolyte leakage and superoxide dismutation than the untransformed line. 2Tunnel vision optimization method for VR flood scenes based on Gaussian blur显示文摘The visualization of flood disasters in virtual reality(VR)scenes is useful for the representation and sharing of disaster knowledge and can effectively improve users’cognitive efficiency in comprehending disaster information.However,the existing VR methods of visualizing flood disaster scenes have some shortcomings,such as low rendering efficiency and poor user experience.In this paper,a tunnel vision optimization method for VR flood scenes based on Gaussian blur is proposed.The key techniques are studied,such as region of interest(ROI)calculation and tunnel vision optimization considering the characteristics of the human visual system.A prototype system has been developed and used to carry out an experimental case analysis.The experimental results show that the number of triangles drawn in a flood VR scene is reduced by approximately 30%–40%using this method and that the average frame rate is stable at approximately 90 frames per second(fps),significantly improving the efficiency of scene rendering and reducing motion sickness.Lin Fu Jun Zhu Weilian Li Qing Zhu Bingli Xu Yakun Xie Yunhao Zhang Ya Hu Jingtao Lu Pei Dang Jigang You 2021International Journal of Digital Earth2021,14,7:1

4Effect of Different Thawing Methods on Post-thaw Motility of Cow Semen in Cryopreservation Straws显示文摘The study was to investigate the effects of different thawing temperatures(5,15,40,75,90 ℃) and times(1- 120 s) on properties of post-thaw cow semen by detecting frozen-thawed semen motility,acrosome integrity and tail membrane integrity,further obtaining the optimal thawing method of straw frozen semen from dairy cow. The results showed that(1) Thawing of the straw frozen semen of dairy cow at 75 ℃ for 3 s yielded the highest semen motility,followed by 40 ℃for 20 s,and the least by low temperature 5 ℃ and room temperature 15 ℃ with a semen motility of 0. 3,moreover thawing at high temperature 90 ℃ was not suitable for large scale production due to the difficult control of the temperature;(2) The acrosome intact rate and plasma membrane integrity rate of semens thawed at90 ℃ were remarkably lower than that at 40 ℃ and 75 ℃ significantly(P < 0. 05),while its semen malformation rate was significantly higher than that at 40 ℃and 75 ℃(P < 0. 05);(3) The Survival time of semens at 37 ℃ varied largely among different thawing temperature,in detail by 40 ℃ > 75 ℃ > 90 ℃. In practice,the thawing method of straw frozen semen of dairy cow should be selected according to the specific circumstance and inseminated immediately,with the recommended condition of thawing at 75 ℃ for 3 s. 6Effect of Calcium Treatment During Production on Quality of Stored Huangguan Pear Fruit显示文摘The effect of calcium treatment during production on the quality(soluble solids, fruit firmness, soluble sugar content, titratable acid content, etc.) of Huangguan pear was studied. The results showed that the calcium treatment during production improved the comprehensive quality of Huangguan pear fruit, delayed and reduced the occurrence of browning of fruit core, and enhanced its storage effect in the cold storage. 7Effect of Postharvest Heat Treatment on Pear Browning during Storage显示文摘The effect of postharvest heat treatment on browning of Huangguan pear was investigated. The results showed that heat treatment improved hardness and soluble solid content of stored pear fruit,postponed the improvement of MDA content and PPO activity in stored pear fruit and alleviated the reduction of SOD activity and the lowering of PG activity. 9Effects of Storage Temperature on the Effective Activity of Straw Frozen Semen after Thawing显示文摘The aim was to discuss the optimal storage environment and proper insemination time after thawing of 0. 25 m L straw frozen semen. Straw frozen semen was thawed at 40 ℃ for 20 s,and then stored at 0- 4 ℃,14- 16 ℃,25- 27 ℃ for 2,4,6,8 and 10 h,respectively. The sperm motility was detected. After thawing,semen was stored at 0- 4 ℃ and 14- 16 ℃ for 10 h. Their sperm motilities( 0. 434 ± 0. 016 7 and 0. 423 ± 0. 019 6) had no significant differences( P> 0. 05) with initial thawing motility( 0. 441 ± 0. 030). Sperm motility reduced as the storage time prolonged at 25- 27 ℃. Sperm motility after 6 h had significant differences with that of initial thawing motility( P < 0. 05),and sperm motilities after 8 and 10 h showed extremely significant differences( P < 0. 01).Thus,sperm motility after thawing was still very high after stored at 0- 4 ℃ and 14- 16 ℃ within 10 h,which met the requirements for insemination.
