4Combined genomic, transcriptomic, and metabolomic an alyses provide in sights into chayote (Sechium edule) evolution and fruit development显示文摘Chayote(Sechium edule)is an agricultural crop in the Cucurbitaceae family that is rich in bioactive components.To enhance genetic research on chayote,we used Nanopore third-generation sequencing combined with Hi-C data to assemble a draft chayote genome.A chromosome-level assembly anchored on 14 chromosomes(N50 contig and scaffold sizes of 8.40 and 46.56 Mb,respectively)estimated the genome size as 606.42 Mb,which is large for the Cucurbitaceae,with 65.94%(401.08 Mb)ofthe genome comprising repetitive sequences;28,237 protein-coding genes were predicted.Comparative genome analysis indicated that chayote and snake gourd diverged from sponge gourd and that a whole-genome duplication(WGD)event occurred in chayote at 25±4 Mya.Transcriptional and metabolic analysis revealed genes involved in fruit texture,pigment,fl avor,fl avonoids,antioxidants,and plant hormones during chayote fruit development.The analysis of the genome,transcriptome,and metabolome provides insights into chayote evolution and lays the groundwork for future research on fruit and tuber development and genetic improvements in chayote.Anzhen Fu Qing Wang Jianlou Mu Lili Ma Changlong Wen Xiaoyan Zhao Lipu Gao Jian Li Kai Shi Yunxiang Wang Xuechuan Zhang Xuewen Zhang Fengling Wang Donald Grierson Jinhua Zuo 2021Horticulture Research2021,8,1:6

6Traditional Chinese medicine is a useful and promising alternative strategy for treatment of Sjogren’s syndrome:A review显示文摘Primary Sjogren’s syndrome(p SS)is a chronic autoimmune disease involving exocrine glands.Current studies have found that the occurrence of the disease is closely related to genetic,environmental and neuroendocrine factors,as well as abnormal activation of T and B lymphocytes.The etiology and pathogenesis of p SS is complex,and there is a lack of specific targeted drugs.Traditional Chinese medicines(TCMs)have been comprehensively investigated for their treatment effects on p SS.Through a systematic review of the literature,we summarized the TCMs used to treat p SS,and find that there are four major ways that TCMs are used,including upregulation of aquaporin proteins,suppression of cell apoptosis,suppression of the abnormal activation of B lymphocytes and suppression of the abnormal activation of T lymphocytes(balancing T helper type[Th]1/Th 2& Th17/Treg and suppressing follicular helper T[Tfh]cells).However,there are not enough data about the active constituents,quality control,pharmacokinetics,toxicity and modern preparations of these TCMs;therefore,more investigations are needed.This paper highlights the importance of TCMs for treating p SS and provides guidance for future investigations.Shu-jun Wei Qing-man He Qing Zhang Kang-hua Fu Ruo-lan Li Wei Peng Yong-xiang Gao 2021Journal of Integrative Medicine2021,19,3:5

11Sacrificial microgel‑laden bioink‑enabled 3D bioprinting of mesoscale pore networks显示文摘Three-dimensional(3D)bioprinting is a powerful approach that enables the fabrication of 3D tissue constructs that retain complex biological functions.However,the dense hydrogel networks that form after the gelation of bioinks often restrict the migration and proliferation of encapsulated cells.Herein,a sacrificial microgel-laden bioink strategy was designed for directly bioprinting constructs with mesoscale pore networks(MPNs)for enhancing nutrient delivery and cell growth.The sacrificial microgel-laden bioink,which contains cell/gelatin methacryloyl(GelMA)mixture and gelled gelatin microgel,is first thermo-crosslinked to fabricate temporary predesigned cell-laden constructs by extrusion bioprinting onto a cold platform.Then,the construct is permanently stabilized through photo-crosslinking of GelMA.The MPNs inside the printed constructs are formed after subsequent dissolution of the gelatin microgel.These MPNs allowed for effective oxygen/nutrient diffusion,facilitating the generation of bioactive tissues.Specifically,osteoblast and human umbilical vein endothelial cells encapsulated in the bioprinted large-scale constructs(≥1 cm)with MPNs showed enhanced bioactivity during culture.The 3D bioprinting strategy based on the sacrificial microgel-laden bioink provided a facile method to facilitate formation of complex tissue constructs with MPNs and set a foundation for future optimization of MPN-based tissue constructs with applications in diverse areas of tissue engineering.Lei Shao Qing Gao Chaoqi Xie Jianzhong Fu Meixiang Xiang Zhenjie Liu Liulin Xiang Yong He 2020Bio-Design and Manufacturing2020,3,1:3
12Bioprinting of novel 3D tumor array chip for drug screening显示文摘Biomedical field has been seeking a feasible standard drug screening system consisting of 3D tumor model array for drug researching due to providing sufficient samples and simulating actual in vivo tumor growth situation,which is still a challenge to rapidly and uniformly establish though.Here,we propose a novel drug screening system,namely 3D tumor array chip with“layer cake”structure,for drug screening.Accurate gelatin methacryloyl hydrogel droplets(~0.1μL)containing tumor cells can be automatically deposited on demand with electrohydrodynamic 3D printing.Transparent conductive membrane is introduced as a chip basement for preventing charges accumulation during fabricating and convenient observing during screening.Culturing chambers formed by stainless steel and silicon interlayer is convenient to be assembled and recycled.As this chip is compatible with the existing 96-well culturing plate,the drug screening protocols could keep the same as convention.Important properties of this chip,namely printing stability,customizability,accuracy,microenvironment,tumor functionalization,are detailly examined.As a demonstration,it is applied for screening of epirubicin and paclitaxel with breast tumor cells to confirm the compatibility of the proposed screening system with the traditional screening methods.We believe this chip will potentially play a significant role in drug evaluation in the future.Mingjun Xie Qing Gao Jianzhong Fu Zichen Chen Yong He 2020Bio-Design and Manufacturing2020,3,3:3
13Low RASSF6 expression in pancreatic ductal adenocarcinoma is associated with poor survival显示文摘AIM:To analyze RASSF6 expression in pancreatic ductal adenocarcinoma(PDAC) and to determine whether RASSF6 has an independent prognostic value in PDAC.METHODS:We studied RASSF6 expression in 96 histologically confirmed PDAC samples and 20 chronic pancreatitis specimens using immunohistochemistry and real-time quantitative reverse transcription-PCR.PDAC issues were then classified as RASSF6 strongly positive,weakly positive or negative.RASSF6 mR NA and protein expression in PDAC samples with strong positive staining was further evaluated using real-time PCR and Western blot analysis.Lastly,correlations between RASSF6 staining and patients' clinicopathological variables and outcomes were assessed.RESULTS:RASSF6 was negatively expressed in 51(53.1%) PDAC samples,weakly positively expressed in 29(30.2%) and strongly positively expressed in16(16.7%),while its expression was much higher in para-tumor tissues and chronic pancreatitis tissues.Positive relationships between RASSF6 expression and T-stage(P = 0.047) and perineural invasion(P = 0.026) were observed.The median survival time of strongly and weakly positive and negative RASSF6 staining groups was 33 mo,15 mo and 11 mo,respectively.Cox multivariate analysis indicated that RASSF6 was an independent prognostic indicator of overall survival in patients with PDAC.A survival curve analysis revealed that increased RASSF6 expression was correlated with better overall survival(P = 0.009).CONCLUSION:RASSF6 expression is an independent biomarker of an unfavorable prognosis in patients with PDAC.Hui-Lin Ye Dou-Dou Li Qing Lin Yu Zhou Quan-Bo Zhou Bing Zeng Zhi-Qiang Fu Wen-Chao Gao Yi-Min Liu Rui-Wan Chen Zhi-Hua Li Ru-Fu Chen 2015World Journal of Gastroenterology2015,21,21:3
14Impact Factor:1.90 Total flavonoids from Saussurea involucrata attenuate inflammation in lipopolysaccharide-stimulated RAW264.7 macrophages via modulating p65,c-Jun,and IRF3 signaling pathways显示文摘Objective:To investigate the anti-inflammatory effects of the total flavonoids from Saussurea involucrata on lipopolysaccharides(LPS)-stimulated murine RAW264.7 macrophages and explore its underlying mechanism of action.Methods:Total flavonoids from Saussurea involucrata were extracted using chromatographic column method.Cell viability was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay.The production of nitric oxide was detected by Griess assay and the release of cytokines(IL-10 and TNF-α)and chemokines(MCP-1,MIP-1α,and CCL5/RANTES)was determined by ELISA to evaluate the anti-inflammatory activity of total flavonoids from Saussurea involucrata.Moreover,nuclear translocation of p65,c-Jun,and IRF3 was detected by immunofluorescence microscopy and Western blotting analysis was performed to determine the expression of related proteins.Results:Total flavonoids extracted from Saussurea involucrata were 751.5 mg/g and the content of rutin was 506.5 mg/g.The production of inflammatory mediators including nitric oxide,cytokines,and chemokines was effectively inhibited by total flavonoids from Saussurea involucrata.Meanwhile,total flavonoids also suppressed the nuclear translocation of p65,c-Jun,and IRF3 in LPS-stimulated RAW264.7 cells.The LPS-induced expression of iNOS and COX-2 was remarkably reduced by treatment with total flavonoids from Saussurea involucrata.Moreover,total flavonoids decreased the expression levels of p-IKKα/β,p-TBK1,p-p38,p-ERK,p-JNK,p-p65,p-c-Jun,and p-IRF3 in LPS-exposed RAW264.7 macrophages.Conclusions:Total flavonoids from Saussurea involucrata potentially inhibit the secretion of pro-inflammatory mediators,which may be related to inhibition of p65,c-Jun,and IRF3 signaling pathways in LPS-stimulated RAW264.7 cells.Li-Shan Yan Li Wang Brian Chi-Yan Cheng Yu Ding Jing Kong Qing Gao Wang Xiu-Qiong Fu Shuo-Feng Zhang Gan Luo Yi Zhang 2021Asian Pacific Journal of Tropical Biomedicine2021,11,6:3
15Transcriptomics and metabolomics analyses provide insights into postharvest ripening and senescence of tomato fruit under low temperature显示文摘Tomato is one of the most important vegetable crops in the world and is a model plant used to study the ripening of climacteric fleshy fruit.During the ripening process of tomato fruit,flavor and aroma metabolites,color,texture and plant hormones undergo significant changes.However,low temperatures delayed the ripening process of tomato fruit,inhibiting flavor compounds and ethylene production.Metabolomics and transcriptomics analyses of tomato fruit stored under low temperature(LT,5°C)and room temperature(RT,25°C)were carried out to investigate the effects of storage temperature on the physiological changes in tomato fruit after harvest.The results of transcriptomics changes revealed that the differentially expressed genes(DEGs)involved in tomato fruit ripening,including several kinds of transcription factors(TFs)(TCP,WRKY,MYB and bZIP),enzymes involved in cell wall metabolism[beta-galactosidase(β-GAL),pectinesterase(PE)and pectate lyase(PL),cellulose and cellulose synthase(CESA)],enzymes associated with fruit flavor and aroma[acetyltransferase(AT),malic enzyme(ME),lipoxygenase(LOX),aldehyde dehydrogenase(ALDH),alcohol dehydrogenase(ADH)and hexokinase(HK)],genes associated with heat stress protein 70 and genes involved in the production of plant hormones such as Ethylene responsive factor 1(ERF1),Auxin/indoleacetic acids protein(AUX/IAA),gibberellin regulated protein.Based on the above results,we constructed a regulatory network model of the effects of different temperatures during the fruit ripening process.According to the analysis of the metabolomics results,it was found that the contents of many metabolites in tomato fruit were greatly affected by storage temperature,including,organic acids(L-tartaric acid,a-hydroxyisobutyric acid and 4-acetamidobutyric acid),sugars(melezitose,beta-Dlactose,D-sedoheptulose 7-phosphate,2-deoxyribose 1-phosphate and raffinose)and phenols(coniferin,curcumin and feruloylputrescine).This study revealed the effects of storage temperature on postharvest tomato fruit and provided a basis for further understanding of the molecular biology and biochemistry of fruit ripening.Chunmei Bai Caie Wu Lili Ma Anzhen Fu Yanyan Zheng Jiawei Han Changbao Li Shuzhi Yuan Shufang Zheng Lipu Gao Xinhua Zhang Qing Wang Demei Meng Jinhua Zuo 2023Horticultural Plant Journal2023,9,1:3
16The genome and transcriptome analysis of snake gourd provide insights into its evolution and fruit development and ripening显示文摘Snake gourd(Trichosanthes anguina L.),which belongs to the Cucurbitaceae family,is a popular ornamental and food crop species with medicinal value and is grown in many parts of the world.Although progress has been made in its genetic improvement,the organization,composition,and evolution of the snake gourd genome remain largely unknown.Here,we report a high-quality genome assembly for snake gourd,comprising 202 contigs,with a total size of 919.8 Mb and an N50 size of 20.1 Mb.These findings indicate that snake gourd has one of the largest genomes of Cucurbitaceae species sequenced to date.The snake gourd genome assembly harbors 22,874 protein-coding genes and 80.0%of the genome consists of repetitive sequences.Phylogenetic analysis reveals that snake gourd is closely related to sponge gourd but diverged from their common ancestor~33–47 million years ago.The genome sequence reported here serves as a valuable resource for snake gourd genetic research and comparative genomic studies in Cucurbitaceae and other plant species.In addition,fruit transcriptome analysis reveals the candidate genes related to quality traits during snake gourd fruit development and provides a basis for future research on snake gourd fruit development and ripening at the transcript level.Lili Ma Qing Wang Jianlou Mu Anzhen Fu Changlong Wen Xiaoyan Zhao Lipu Gao Jian Li Kai Shi Yunxiang Wang Xuewen Zhang Xuechuan Zhang Zhangjun Fei Donald Grierson Jinhua Zuo 2020Horticulture Research2020,7,1:2

18Special features of the 2009 pandemic swine-origin influenza AH1N1 hemagglutinin and neuraminidase显示文摘Since the 2009 pandemic H1N1 swine-origin influenza A virus (09 S-OIV) has reminded the world about the global threat of the ever changing influenza virus,many questions regarding the detailed re-assortment of influenza viruses yet remain unanswered.Influenza A virus is the causative agent of the pandemic flu and contains 2 major antigenic glycoproteins on its surface:(i) hemagglutinin (HA);and (ii) neuraminidase (NA).The structures of the 09 S-OIV HA and NA proteins (09H1 and 09N1) have recently been resolved in our laboratory and provide some clues as to why the 09 S-OIV re-assortment virus is highly infectious with severe consequences in humans.For example,the 09H1 is highly similar to the HA of the 1918 influenza A pandemic virus in overall structure and especially in regards to its 5 defined antibody binding epitopes.For 09N1,its most distinctive feature is the lack of a 150-loop active site cavity,which was previously predicted to be present in all N1 NAs,and we hypothesize that the 150-loop may play a important role in the substrate specificity (α2,3 or α2,6 linked sialic acid receptors) and enzymatic mechanism of influenza NA.Combination of the HA and NA with special characteristics for the 09 S-OIV might contribute to its high increased transmissibility in humans.VAVRICKA Christopher John LIU Yue LI Qing SHI Yi WU Yan SUN YePing QI JianXun GAO George Fu 2011Chinese Science Bulletin2011,56,17:2
19Regulations of m^(6)A methylation on tomato fruit chilling injury显示文摘Tomato fruit are sensitive to chilling injury(CI)during cold storage.Several factors have been discovered to be involved in chilling injury of tomato fruit.Plant hormones play an important regulatory role,however,the relationship between chilling injury and N6-methyladenosine(m^(6)A)methylation of transcripts in plant hormone pathways has not been reported yet.In order to clarify the complex regulatory mechanism of m^(6)A methylation on chilling injury in tomato fruit,Nanopore direct RNA sequencing was employed.A large number of enzymes and transcription factors were found to be involved in the regulation process of fruit chilling injury,which were associated with plant hormone,such as 1-aminocyclopropane 1-carboxylate synthase(ACS),aspartate aminotransferase(AST),auxin response factor(ARF2),ethylene response factor 2(ERF2),gibberellin 20-oxidase-3(GA20ox)and jasmonic acid(JA).By conjoint analysis of the differential expression transcripts related to chilling injury andm^(6)Amethylation differential expression transcripts 41 differential expression transcripts were identified involved in chilling injury including 1-aminocyclopropane-1-carboxylate oxidase(ACO)and pectinesterase(PE)were down-regulated and heat shock cognate 70 kD protein 2(cpHSC70),HSP70-binding protein(HspBP)and salicylic acid-binding protein 2(SABP2)were up-regulated.Our results will provide a deeper understanding for chilling injury regulatory mechanism and post-harvest cold storage of tomato fruit.Chunmei Bai Minghuan Fang Baiqiang Zhai Lili Ma Anzhen Fu Lipu Gao Xiaohong Kou Demei Meng Qing Wang Shufang Zheng Jinhua Zuo 2021Horticultural Plant Journal2021,7,5:2
