1Proteomic analysis of human epithelial ovarian cancer xenografts in immunodeficient mice exposed to chronic psychological stress显示文摘This work aims at comparing alterations in the proteomes of human epithelial ovarian cancer xenografts between stressed and non-stressed immunodeficient mice as well as exploring the molecular mechanisms linking chronic psychological stress to ovarian cancer oncogenesis and progression.SK-OV-3 cells were injected subcutaneously into nude mice.The stress group was subjected to a chronic physical restraint protocol for 6 h on 35 consecutive days,while the control group was unrestrained.All mice were sacrificed on day 36 after SK-OV-3 cell injection,and tumors were excised.Tumor tissues were processed for 2D gel electrophoresis,mass spectrometry(nanoUPLC-ESI-MS/MS) and Western blotting.The expression of 20 proteins was found to be significantly altered between the stress and control groups,of which 14 were up-regulated,five were down-regulated,and one protein was found only in the stress group.All proteins were identified by UPLC-ESI-MS/MS,and Western blotting results were consistent with those of proteomic methods.The present results provide new evidence relating to the molecular mechanism underlying the relationship between psychological stress and tumor progression.GAO Jun GAO GuoLan ZHANG YanYu WANG Fen 2011Science China(Life Sciences)2011,54,2:11
2Hedgehog signaling pathway and ovarian cancer显示文摘Epithelial ovarian carcinoma (EOC) is the most common form of ovarian malignancies and the most lethal gynecologic malignancy in the United States. To date, in spite of treatment to it with the extensive surgical debulking and chemotherapy, the prognosis of EOC remains dismal. Recently, it has become increasingly clear that in many instances, the signaling and molecular players that control development are the same, and when inappropriately regulated, drive tumorigenesis and cancer development. Here, we discuss the possible involvement of Hedgehog (Hh) pathway in the cellular regulation and development of cancer in the ovaries. Using the in vitro and in vivo assays developed has facilitated the dissection of the mechanisms behind Hh-driven ovarian cancers formation and growth. Based on recent studies, we propose that the inhibition of Hh signaling may interfere with spheroid-like structures in ovarian cancers. 3Epigenetic reprogramming of embryos derived from sperm frozen at-20℃显示文摘Cryopreservation of spermatozoa is a strategy that has been used to conserve the sperm of animal species and animal strains that are valuable for biomedical research. A simple method for preserving spermatozoa after application of intracytoplasmic sperm injection (ICSI) is much needed. It has been shown previously that spermatozoa frozen at 20°C can activate oocytes and support full-term embryo development. However, epigenetic reprogramming could be affected by the environment and by the in vitro manipulation of gametes. Here, we investigated embryo epigenetic reprogramming including DNA methylation and histone modification, in embryos derived from sperm preserved at 20°C without cryoprotectants. The results showed that although both fertilization and embryo developmental competence were decreased, the dynamic epigenetic reprogramming of embryos derived from frozen sperm was similar to the reprogramming of embryos derived from fresh sperm.
