3Hedgehog signaling pathway and ovarian cancer显示文摘Epithelial ovarian carcinoma (EOC) is the most common form of ovarian malignancies and the most lethal gynecologic malignancy in the United States. To date, in spite of treatment to it with the extensive surgical debulking and chemotherapy, the prognosis of EOC remains dismal. Recently, it has become increasingly clear that in many instances, the signaling and molecular players that control development are the same, and when inappropriately regulated, drive tumorigenesis and cancer development. Here, we discuss the possible involvement of Hedgehog (Hh) pathway in the cellular regulation and development of cancer in the ovaries. Using the in vitro and in vivo assays developed has facilitated the dissection of the mechanisms behind Hh-driven ovarian cancers formation and growth. Based on recent studies, we propose that the inhibition of Hh signaling may interfere with spheroid-like structures in ovarian cancers. 4Cancer-associated adipocyte-derived G-CSF promotes breast cancer malignancy via Stat3 signaling显示文摘Adipocyte is the most predominant cell type in the tumor microenvironment of breast cancer and plays a pivotal role in cancer progression,yet the underlying mechanisms and functional mediators remain elusive.We isolated primary preadipocytes from mammary fat pads of human breast cancer patients and generated mature adipocytes and cancer-associated adipocytes(CAAs)in vitro.The CAAs exhibited significantly different gene expression profiles as assessed by transcriptome sequencing.One of the highly expressed genes in CAAs is granulocyte colony-stimulating factor(G-CSF).Treatment with recombinant human G-CSF protein or stable expression of human G-CSF in triple-negative breast cancer(TNBC)cell lines enhanced epithelial–mesenchymal transition,migration,and invasion of cancer cells,by activating Stat3.Accordantly,targeting G-CSF/Stat3 signaling with G-CSF-neutralizing antibody,a chemical inhibitor,or siRNAs for Stat3 could all abrogate CAA-or G-CSF-induced migration and invasion of breast cancer cells.The pro-invasive genes MMP2 and MMP9 were identified as target genes of G-CSF in TNBC cells.Furthermore,in human breast cancer tissues,elevated G-CSF expression in adipocytes is well correlated with activated Stat3 signal in cancer cells.Together,our results suggest a novel strategy to intervene with invasive breast cancers by targeting CAA-derived G-CSF.Li Liu Yudong Wu Cheng Zhang Chong Zhou Yining Li Yi Zeng Chunbo Zhang Rong Li Daya Luo Lieliang Wang Long Zhang Shuo Tu Huan Deng Shiwen Luo Ye-Guang Chen Xiangyang Xiong Xiaohua Yan 2020Journal of Molecular Cell Biology2020,12,9:4

9Mapping of individual sensory nerve axons from digits to spinal cord with the transparent embedding solvent system显示文摘Achieving uniform optical resolution for a large tissue sample is a major challenge for deep imaging.For conventional tissue clearing methods,loss of resolution and quality in deep regions is inevitable due to limited transparency.Here we describe the Transparent Embedding Solvent System(TESOS)method,which combines tissue clearing,transparent embedding,sectioning and block-face imaging.We used TESOS to acquire volumetric images of uniform resolution for an adult mouse whole-body sample.The TESOS method is highly versatile and can be combined with different microscopy systems to achieve uniformly high resolution.With a light sheet microscope,we imaged the whole body of an adult mouse,including skin,at a uniform 0.8×0.8×3.5μm^(3) voxel resolution within 120 h.With a confocal microscope and a 40×/1.3 numerical aperture objective,we achieved a uniform sub-micron resolution in the whole sample to reveal a complete projection of individual nerve axons within the central or peripheral nervous system.Furthermore,TESOS allowed the first mesoscale connectome mapping of individual sensory neuron axons spanning 5 cm from adult mouse digits to the spinal cord at a uniform sub-micron resolution.Yating Yi Youqi Li Shiwen Zhang Yi Men Yuhong Wang Dian Jing Jiayi Ding Qingjie Zhu Zexi Chen Xingjun Chen Jun-Liszt Li Yilong Wang Jun Wang Hanchuan Peng Li Zhang Wenjing Luo Jian Q.Feng Yongwen He Woo-Ping Ge Hu Zhao 2024Cell Research2024,34,2:0
10Aberrant expression of the extracellular matrix component Biglycan regulated by Hedgehog signalling promotes colorectal cancer cell proliferation显示文摘Hedgehog(Hh)signalling plays essential roles in regulating embryonic development and contributes to tumour initiation,growth and progression in multiple cancers.The detailed mechanism by which Hh signalling participates in tumour growth warrants thorough study,although several downstream target genes have been identified.Herein,a set of novel targets of Hh signalling was identified in multiple types of tumour cells via RNA-Seq analysis.Among these targets,the expression regulation and oncogenic function of the extracellular matrix component biglycan(BGN)were investigated.Further investigation verified that Hh signalling activates the expression of BGN via the transcription factor Gli2,which directly binds to the promoter region of BGN.Functional assays revealed that BGN facilitates tumour cell growth and proliferation in colorectal cancer(CRC)cells,and xenograft assays confirmed that BGN also promotes tumour growth in vivo.Moreover,analysis of clinical CRC samples showed that both the protein and mRNA levels of BGN are increased in CRC tissues compared to those in adjacent tissues,and higher expression of BGN is correlated with poorer prognosis of CRC patients,further confirming the function of BGN in CRC.Taken together,aberrantly activated Hh signalling increases the expression of BGN,possibly regulates the extracellular matrix,and thereby promotes tumour growth in CRC.Shaopeng Zeng Feifei Zhou Yiqing Wang Zhenyu Zhai Linlin Xu Hailong Wang Xinping Chen Shiwen Luo Minzhang Cheng 2022Acta Biochimica et Biophysica Sinica2022,54,2:0

12Designing interstitial boron-doped tunnel-type vanadium dioxide cathode for enhancing zinc ion storage capability显示文摘Chemical doping is a powerful method to intrinsically tailor the electrochemical properties of electrode materials.Here,an interstitial boron-doped tunnel-type VO_(2)(B)is constructed via a facile hydrothermal method.Various analysis techniques demonstrate that boron resides in the interstitial site of VO_(2)(B)and such interstitial doping can boost the zinc storage kinetics and structural stability of VO_(2)(B)cathode during cycling.Interestingly,we found that the boron doping level has a saturation limit peculiarity as proved by the quantitative analysis.Notably,the 2 at.%boron-doped VO_(2)(B)shows enhanced zinc ion storage performance with a high storage capacity of 281.7 mAh g^(-1) at 0.1 A g^(-1),excellent rate performance of 142.2 mAh g^(-1) at 20 A g^(-1),and long cycle stability up to 1000 cycles with the capacity retention of 133.3 mAh g^(-1) at 5 A g^(-1).Additionally,the successful preparation of the boron-doped tunneltype α-MnO_(2) further indicates that the interstitial boron doping approach is a general strategy,which supplies a new chance to design other types of functional electrode materials for multivalence batteries.Shiwen Wang Hang Zhang Kang Zhao Wenqing Liu Nairui Luo Jianan Zhao Shide Wu Junwei Ding Shaoming Fang Fangyi Cheng 2023Carbon Energy2023,5,8:0
