1The PI3K/Akt/GSK-3β/ROS/eIF2B pathway promotes breast cancer growth and metastasis via suppression of NK cell cytotoxicity and tumor cell susceptibility显示文摘Objective: To examine the effect of pSer9-GSK-3β on breast cancer and to determine whether the underlying metabolic and immunological mechanism is associated with ROS/eIF2B and natural killer(NK) cells.Methods: We employed TWS119 to inactivate GSK-3β by phosphorylating Ser9 and explored its effect on breast cancer and NK cells. The expression of GSK-3β, natural killer group 2 member D(NKG2D) ligands, eIF2B was quantified by PCR and Western blot. We measured intracellular reactive oxygen species(ROS) and mitochondrial ROS using DCFH-DA and MitoSOX^(TM) probe,respectively, and conducted quantitative analysis of cellular respiration on 4T1 cells with mitochondrial respiratory chain complex Ⅰ/Ⅲ kits.Results: Our investigation revealed that TWS119 downregulated NKG2D ligands(H60 a and Rae1), suppressed the cytotoxicity of NK cells, and promoted the migration of 4T1 murine breast cancer cells. Nevertheless, LY290042, which attenuates p-GSK-3β formation by inhibiting the PI3K/Akt pathway, reversed these effects. We also found that higher expression of p Ser9-GSK-3β induced higher levels of ROS, and observed that abnormality of mitochondrial respiratory chain complex Ⅰ/Ⅲ function induced the dysfunction of GSK-3β-induced electron transport chain, naturally disturbing the ROS level. In addition, the expression of NOX3 and NOX4 was significantly up-regulated, which affected the generation of ROS and associated with the metastasis of breast cancer. Furthermore, we found that the expression of pSer535-eIF2B promoted the expression of NKG2D ligands(Mult-1 and Rae1) following by expression of pSer9-GSK-3β and generation of ROS.Conclusions: The PI3K/Akt/GSK-3β/ROS/eIF2B pathway could regulate NK cell activity and sensitivity of tumor cells to NK cells,which resulted in breast cancer growth and lung metastasis. 2Synthesis,luminescent properties,and theoretical study of novel Sm^(3+) and Dy^(3+) complexes containing β-diketone and 1,10-phenanthroline显示文摘Two new ternary complexes of 1-(2-thienyl)-3-(p-phenylethynylphenyl)-1,3-propanedione(HTPP) and 1,10-phenanthroline(phen) with Sm3+ and Dy3+ were synthesized.The composition of the ternary complexes was characterized as Sm(TPP)3phen and Dy(TPP)3phen,respectively,by infrared(IR) spectra,chemical analysis,elemental analysis,and thermodynamic analysis.At room temperature,under UV light excitation,the Sm3+ and Dy3+ complexes exhibit characteristic emissions of the central ions.It is found that the fluorescence intensity of Sm(TPP)3phen is stronger than that of Dy(TPP)3phen.In order to explain this phenomenon,an accurate quantum chemistry calculation was carried out,and the result is in good agreement with the experiment data.LIU Xingwang GAO Saishengtai WANG Li SHEN Leijun JIANG Jiadong GAO Junfang 2011Rare Metals2011,30,1:7
3Tribological Behavior of Nano-ZrO2 Reinforced PTFE-PPS Composites显示文摘Polytetrafluoroethylene (PTFE) is a commonly used seal material for oil-free engine that is well known for its excellent tribological properties. In this work, the nano-ZrO2 particles were used as the friction modifiers to improve the friction and wear performance of PTFE-PPS composites. The friction and wear characteristics of PTFE/PPS-nano-ZrO2 composites were investigated by a block-on-ring tester under dry friction sliding condition. The worn surfaces, counterpart transfer films and wear debris were studied by scanning electron microscopy and X-ray photoelectron spectroscopy. It was found that the increase of nano- ZrO2 content could effectively reduce the coefficient of friction and enhance the anti-wear ability of PTFEPPS composites. Especially, the best tribological properties of the composites were obtained when the particle content of nano-ZrO2 was 10 vol%, the anti-wear performance of composite is 195 times better than that of the unfilled PTFE-PPS composite. 4Amyloid β-protein differentially affects NMDA receptor-and GABA_A receptor-mediated currents in rat hippocampal CA1 neurons显示文摘Although the aggregated amyloid b-protein (Ab) in senile plaques is one of the key neuropathological features of Alzheimer’s disease (AD), soluble forms of Ab also interfere with synaptic plasticity at the early stage of AD. The suppressive action of acute application of Ab on hippocampal long-term potentiation (LTP) has been reported widely, whereas the mechanism underlying the effects of Ab is still mostly unknown. The present study, using the whole-cell patch clamp technique, investigated the effects of Ab fragments (Ab25–35 and Ab31–35) on the LTP induction-related postsynaptic ligand-gated channel currents in isolated hippocampal CA1 neurons. The results showed a rapid but opposite action of both peptides on excitatory and inhibitory receptor currents. Glutamate application-induced currents weresuppressed by Ab25–35in a dose-dependent manner,andfurtherN-methyl-D-aspartate(NMDA)receptor-mediated currents were selectively inhibited. In contrast, pretreatment with Ab fragments potentiated c-aminobutyric acid (GABA)-induced whole-cell currents. As a control, Ab35–31, the reversed sequence of Ab31–35, showed no effect on the currents induced by glutamate, NMDA or GABA. 5Synthesis and luminescence properties of CdSe:Eu NPs and their surface polymerization of poly(MMA-co-MQ)显示文摘CdSe:Eu nanoparticles(NPs) were synthesized using an oil phase method and the substitution of Cd^(2+)with Eu3+ was confirmed by XRD,TEM,UV-Vis absorption and fluorescence emission analyses.The CdSe:Eu NPs are monodispersed and uniform spherical particles with a diameter of 3.2 nm,bigger than the pure CdSe NPs(2.3 nm),but with a similar cubic structure as CdSe NPs.Compared with those of pure CdSe NPs,both emission spectrum and absorption spectrum of CdSe:Eu NPs are red-shifted.The CdSe:Eu NPs are incorporated into poly(MMA-co-MQ) to afford poly(MMA-co-MQ)-CdSe:Eu NPs with the cubic structure and particle size(~3-4 nm) similar to those of CdSe:Eu NPs.The TEM imaging suggests that the CdSe:Eu NPs are uniformly dispersed in poly(MMA-co-MQ,) without any obvious aggregation.The fluorescent emission peak and absorption peak of poly(MMA-co-MQ)-CdSe:Eu NPs are between those of CdSe:Eu NPs and poly(MMA-co-MQ),possibly due to the energy transfer caused by the interactions of Cd or Eu atoms on the surfaces of CdSe:Eu NPs with the N and O atoms of poly(MMA-co-MQ).These CdSe:Eu and poly(MMA-co-MQ)-CdSe:Eu NPs with tunable photoluminescence properties can be potentially used for the fabrication of optical and optoelectronic devices.Junfang Gao Junhong Yang Shengli Yong Lijing Liu Xingwang Liu 2018Journal of Rare Earths2018,36,9:1

7The Efect of Rich Synthetic Copper-rich Solution on Antiseepage Dense Pre-hydration Geosynthetic Clay Liner显示文摘Dense pre-hydrated geosynthetic clay liners(DPH GCLs)were manufactured as innovative materials accompanied by the advantage of lower hydraulic conductivity(k).The k of DPH GCLs permeated with de-ionized water(DIW)was 9.8×10^(−12) m/s.The effect of Cu^(2+)synthetic solution on DPH GCLs was discussed.Furthermore,the effect mechanism was studied on the basis of test technologies.A significant adverse impact on hydraulic performance of DPH GCLs is found when the concentration of Cu^(2+)is greater than 1 g/L.SEM,XRD,XRF,FTIR,and XPS analyses show that the effect of Cu^(2+)on DPH GCLs includes two steps.Firstly,Cu^(2+)interacts with hydrophobic organic matter(HOM),and the adhesion of bentonite is destroyed,and some holes appear.The Cu^(2+)contacts with bentonite directly,and Cu^(2+)interacts with bentonite through ion exchange.Passivated phenomenon occurs on the surface of the bentonite,and swelling ability of bentonite is reduced,which causes permeable DPH GCLs.郭争争 GUAN Junfang 任子杰 GAO Huimin LI Peiyue 2023Journal of Wuhan University of Technology(Materials Science)2023,38,4:0
8Changes in clinical indicators among human immunodeficiency virus patients who failed in antiretroviral therapy during 2004–2016 in Yunnan, China: an observational cohort study显示文摘Background:This study aimed to investigate the changes in the clinical indicators and influencing factors of treatment duration among human immunodeficiency virus(HIV)patients in whom antiretroviral therapy(ART)was unsuccessful.Methods:In this retrospective study,a total of 9,418 HIV patients who failed in ART during 2004–2016 were included and divided into two treatment groups—Group 1(treatment time≤3 years,n1=5,218)and Group 2(treatment time>3 years,n2=4,200).Patient follow-up data,including age,cluster of differentiation 4(CD4)count,and viral load,glucose,creatinine,and triglyceride levels,were extracted from electronic health record databases.Covariance analysis for repeated measures was used to analyze the biochemical indicators,and multiple logistic regression modeling was used to compare relevant data extracted from the Group 1 and Group 2 HIV patient cohorts with different treatment time.Results:The median initial CD4 count was 175.0 cells/μl(interquartile range,77.0–282.0),while the initial CD4 counts for Group 1 were lower than those for Group 2(P<0.05).A significant interaction between group and time effects was observed(P<0.05)in total cholesterol(TC).Changes in hemoglobin level among HIV patients were also significantly associated with treatment time(P=0.001).The initial CD4 count(odds ratio[OR]=0.756),female sex(OR=0.713),Zerit(d4T)(OR=1.443),TC(OR=1.285),and aspartate aminotransferase level(OR=1.002)were significantly associated with the survival time of dead patients with HIV(P<0.05).Additionally,the initial CD4 count(OR=1.456),age(OR=1.022),time interval(OR=0.903),patient’s living status(OR=0.597),d4T(OR=2.256),and triglyceride(OR=0.930)and hemoglobin levels(OR=0.997)were significantly associated with the treatment time of HIV patients with drug withdrawal(P<0.05).Conclusion:The initial biochemical parameters can affect the survival and treatment time of HIV patients.With a comprehensive understanding of the physiological and biochemical indicators of patients,we can reduce the probability of drug withdrawal and prolong the survival time of HIV patients.Peicheng Wang Junfang Xu Bingbing Guo Jason K Wang Liangmin Gao Qianyun Wang Jun Jing Feng Cheng 2020Global Health Journal2020,4,2:0

10Antiretroviral regimen change among people infected with HIV:evidence from a crosssectional study in China显示文摘Background:The rate of death among people living with HIV/AIDS has decreased significantly as a result of treatment with highly active antiretroviral therapy(HAART).However,the issues of drug induced toxicities and complexity of current HAART regimens has remained of great concern.The aim of this study was to determine factors in uencing antiretroviral regimen changes among people living with HIV/AIDS in China.Methods:This retrospective study collected data through face-to-face interviews with people living with HIV/AIDS who were receiving HAART,and gathered relevant information from infectious disease hospitals.The following information were collected:social-demographic characteristics,antiretroviral therapies,CD4 cell counts,virus loads,reasons for changing medication and other related data.Mean and percentages were used to describe the frequency of regimen change among patients,and binary logistic regression was employed to test the factors in uencing regimen change.Results:1,123 people who had experienced regimen change were included in the analysis.On average,patients remained on HAART for 10.2 months before changing regimen,and the average CD4 cell count and viral load(VL)were 383.1 cells/ l and 28,132.4 copies/mL respectively when changing regimen.The reasons for modi cation were determined as treatment failure(52.5%),adverse reactions(32.3%),and other reasons including pregnancy(15.2%).There are significant differences in regimen change among people with different genders(P<0.001),modes of transmission(P<0.001),duration of HAART(P<0.001)and initial CD4 cell counts(P=0.0024).Males,drug users,people taking long-term medication,and those with lower initial CD4 counts when starting HAART tend to change regimen.Conclusion:Treatment failure was the main reason for the change of HAART regimen.Males,drug users,people on longterm medication and those with lower initial CD4 cell counts when starting HAART were most likely to change regimen.Junfang Xu Peicheng Wang Liangmin Gao Xinyu Peng Feng Cheng 2018Global Health Journal2018,2,3:0
11Construction of a biotin-targeting drug delivery system and its near-infrared theranostic fluorescent probe for real-time image-guided therapy of lung cancer显示文摘The therapy of non-small lung cancer(NSCLC) is limited by wide metastasis and chemotherapy resistance, herein, we present a new cancer-targeting prodrug PBG with the integration of real-time fluorescence visualization. The potent anticancer drug Gefitinib conjugates a biotin recognition ligand yielding the prodrug PBG via a GSH-activatable disulfide bond linker. Once coupling a near-infrared azo-BODIPY fluorophore into the molecular structure of PBG, we obtain its fluorescent theranostic TBG. The prodrug PBG can sustain Gefitinib release by the high level of GSH in the pathophysiological milieu. We evaluate the drug delivery of the prodrug PBG using fluorescent TBG in PC9 cancer bearing nude mice models,which indicate that TBG can be utilized to monitor the in vivo drug release process. Prodrug PBG can be targeted to accumulate in the cancer lesion with a better and efficaciously therapeutic result compared with the single Gefitinib treatment in cells and in vivo. The fluorescence images also reveal that the targeting accumulation and longitudinal retention of anticancer drug in cancer lesions will contribute to the superior therapeutic effects.
