| 1 | Process analysis of PM_(2.5) pollution events in a coastal city of China using CMAQ显示文摘US EPA's Community Multiscale Air Quality modeling system(CMAQ) with Process Analysis tool was used to simulate and quantify the contribution of individual atmospheric processes to PM_(2.5) concentration in Qingdao during three representative PM_(2.5) pollution events in the winter of 2015 and 2016. Compared with the observed surface PM_(2.5) concentrations, CMAQ could reasonably reproduce the temporal and spatial variations of PM_(2.5) during these three events. Process analysis results show that primary emissions accounted for 72.7%–93.2% of the accumulation of surface PM_(2.5) before and after the events.When the events occurred, primary emissions were still the major contributor to the increase of PM_(2.5) in Qingdao, however the contribution percentage reduced significantly,which only account for 51.4%–71.8%. Net contribution from horizontal and vertical transport to the accumulation of PM_(2.5) was also positive and its percentage increased when events occurred. Only 1.1%–4.6% of aerosol accumulation was due to PM processes and aqueous chemical processes before and after events. When the events occurred,contribution from PM processes and aqueous chemistry increased to 6.0%–11.8%. Loss of PM_(2.5) was mainly through horizontal transport, vertical transport and dry deposition, no matter during or outside the events. | 2 | NUMERICAL STUDY OF THE RELATIONSHIP BETWEEN APPARENT SLIP LENGTH AND CONTACT ANGLE BY LATTICE BOLTZMANN METHOD显示文摘The apparent slip between solid wall and liquid is studied by using the Lattice Boltzmann Method (LBM) and theShan-Chen multiphase model in this paper. With a no-slip bounce-back scheme applied to the interface, flow regimes under differentwall wettabilities are investigated. Because of the wall wettability, liquid apparent slip is observed. Slip lengths for different wallwettabilities are found to collapse nearly onto a single curve as a function of the static contact angle, and thereby a relationshipbetween apparent slip length and contact angle is suggested. | 4 | Effects of multidrug resistance, antisense RNA on the chemosensitivity of hepatocellular carcinoma cells显示文摘BACKGROUND: Multidrug resistance is a major obstacle in cancer chemotherapy. We examined whether the antisense RNA of multidrug resistance gene 1 (mdr1) could reverse multidrug resistance in the human hepatocellular carcinoma (HCC) cell line SMMC7721/ADM. METHODS: The recombinant adenoviruses pAdEasy- GFP-ASmdr1 product was produced by the adenoviral vector AdEasy system, which can express antisense RNA against the mdr1 gene. Following that, the recombinant adenovirus was transfected into the P-glycoprotein- producing multidrug resistance cell line, SMMC7721/ADM human HCC cells resistant to adriamycin (ADM) and daunorubicin (DNR). In order to investigate the reversal of multidrug resistance phenotype, we measured the expression of mdr1 mRNA by RT-PCR and the production of P-glycoprotein by flow cytometry. The sensitivities for ADM and DNR SMMC7721/ADM cells were examined by [3-(4, 5-dimethylthi-azol-2-yl)-2,5 diphenyl-terazolium bromide] (MTT) analysis. RESULTS: The low-level expression of mdr1 mRNA and P-glycoprotein production were observed in parental sensitive cells SMMC/7721 in addition to the overexpressionof mdr1 mRNA and P-glycoprotein in SMMC7721/ADM cells. The transfection of antisense-RNA into SMMC7721/ ADM cells resulted in decreases of mdr1 mRNA and P-glycoprotein, but increase of drug sensitivities. The sensitivities of transfected SMMC7721/ADM cells to ADM and DNR in IC50 reduced by 31.25% and 62.96% respectively. CONCLUSIONS: Mdr1 antisense RNA can increase the sensitivities of SMMC7721/ADM cells to anticancer drug by decreasing the expression of the mdr1 gene and inhibiting P-glycoprotein expression. | 6 | Food Frequency Questionnaire for Chinese Children Aged 12-17 Years: Validity and Reliability显示文摘Objective The primary objective of this study was to examine the validity and reliability of a semi-quantitative food frequency questionnaire(FFQ) among Chinese children aged 12-17 years. Methods A semi-quantitative 72-food item FFQ was developed for children aged 12-17 years. The reliability and validity of this FFQ were evaluated against 24-h dietary recalls(24 h DRs) to measure the consumption of foods and nutrients. We administered two FFQs and three DRs to children(N = 160) over a period of 1 month to evaluate the reliability and validity. Reliability was examined by quartile agreement and intraclass correlation coefficients(ICCs), and validity was examined by quartile agreement, Bland-Altman plots and correlation with DRs. Results For reliability, the ICCs between the two FFQs ranged from 0.21 to 0.76 for foods and nutrients, and the quartile agreement ranged from 70.0% to 95.0% in the same or adjacent quartiles. Spearman’s correlation coefficients of foods and nutrients between the second FFQ and the 24 h DRs ranged from-0.04 to 0.59. The Bland-Altman plots demonstrated good agreement across the range of intakes among nutrients. The quartile agreement ranged from 50.0% to 100.0%, with infrequent misclassification. A two-dimensional explicit dynamic finite element model with Arbitrary Lagrangian Eulerian (ALE) adaptive meshing technique has been employed to simulate asymmetrical sheet rolling, in which asymmetrical conditions are here due to different roll radii. To validate the simulation, the results of simulation and experiment are compared. Effects of asymmetry due to roll radii mismatch on the normal and shear distributions and on sheet curvature variations are discussed. An optimum roll radii ratio could be found to produce flat sheet. | 11 | Experimental analysis of decision-making for demand chasing in order behaviors显示文摘The traditional newsvendor model assumes that a decision-maker is risk-neutral. However, in actuality, a decisionmaker's order behavior is often influenced by waste-averse preference and stockout-averse preference. We extend the newsvendor model with consideration of averse preferences to investigate how the decision results of the previous period impact the order behavior of the current period, and design an inventory decision-making behavior experiment. Results from the study demonstrate that the order behavior of both a group and an individual exhibits a demand chasing phenomenon, and the former is more significant. Through the interval estimation of the decision maker's order quantity, by the maximum likelihood method we find that the stockout-averse preference has an effect on the decision-making when the prior period is insufficient, causing the current period order quantity larger than the expected profit-maximizing order quantity. In a similar way, waste-averse preference has an effect on decision-making when the prior period is surplus, resulting in the current period order quantity smaller than the expected profit-maximizing order quantity. | 13 | Clinical features and predictors of remission in children under the age of 7 years with Graves’disease显示文摘Importance:Graves’disease(GD)is rare in children under the age of 7 years.Children with this disease exhibit greater thyrotoxicity at diagnosis and require a longer course of medical therapy,compared with pubertal and postpubertal children and adults.Objective:To investigate the clinical features and identify predictors of remission in children under the age of 7 years with GD.Methods:This retrospective study included 77 children who were diagnosed with GD under the age of 7 years and were treated in the Department of Endocrinology,Beijing Children’s Hospital from 2010 to 2018.Clinical manifestations,laboratory data,and follow-up records were collected for all patients.Children who achieved remission of treatment with methimazole were compared with those who had persistent disease to identify which variables were associated with remission;multiple logistic regression and Cox regression analyses were used to evaluate interactions among predictive variables.Results:Sixty-three boys and 14 girls were included;the median age at diagnosis was 4.2 years(interquartile range:3.2-5.3 years).Forty-six(56.7%)patients had no family history of thyroid disease,17 patients had family history of thyroid disease and 14 patients with unknown family history.Of the 77 patients,18(23.4%)patients achieved remission of treatment with methimazole and 59 patients did not;moreover,51(66.2%)had Graves’ophthalmopathy.Univariate analyses revealed no significant differences between the remission group and non-remission group in terms of age at diagnosis,sex,initial goiter size,or initial thyroid hormone concentration.However,there were a trend of correlation between the initial level of thyroid peroxidase antibody(TPOAb)and remission status(univariate analysis OR 1.002,P=0.038;multivariate analysis OR 1.004,P=0.019).Similar results were observed in univariate analysis of the initial thyrotropin receptor antibody(TRAb)level,but this association was not significant in multivariate analysis.Cox regression analyses revealed that children with high TRAb level required longer duration of remission,compared with low TRAb level(OR 0.950,95%CI 0.904-0.997,P=0.037).Interpretation:Initial TRAb level was an independent predictor of remission outcome in young children under the age of 7 years with GD.Initial TRAb level may predict the likelihood of remission in patients with young-age-of-onset GD. | Yi Gu Xuejun Liang Ming Liu Di Wu Wenjing Li Bingyan Cao Yuchuan Li Chang Su Jiajia Chen Chunxiu Gong | 2020 | Pediatric Investigation2020,4,3: | 3 |
| 14 | Parametric sensitivity analysis of precipitation and temperature based on multi-uncertainty quantification methods in the Weather Research and Forecasting model显示文摘Sensitivity analysis(SA) has been widely used to screen out a small number of sensitive parameters for model outputs from all adjustable parameters in weather and climate models, helping to improve model predictions by tuning the parameters. However, most parametric SA studies have focused on a single SA method and a single model output evaluation function, which makes the screened sensitive parameters less comprehensive. In addition, qualitative SA methods are often used because simulations using complex weather and climate models are time-consuming. Unlike previous SA studies, this research has systematically evaluated the sensitivity of parameters that affect precipitation and temperature simulations in the Weather Research and Forecasting(WRF) model using both qualitative and quantitative global SA methods. In the SA studies, multiple model output evaluation functions were used to conduct various SA experiments for precipitation and temperature. The results showed that five parameters(P3, P5, P7, P10, and P16) had the greatest effect on precipitation simulation results and that two parameters(P7 and P10) had the greatest effect for temperature. Using quantitative SA, the two-way interactive effect between P7 and P10 was also found to be important, especially for precipitation. The microphysics scheme had more sensitive parameters for precipitation, and P10(the multiplier for saturated soil water content) was the most sensitive parameter for both precipitation and temperature. | 15 | Hydrological Impacts of Climate Change on Streamflow of Dongliao River Watershed in Jilin Province,China显示文摘The impacts of future climate change on streamflow of the Dongliao River Watershed located in Jilin Province,China have been evaluated quantitatively by using a general circulation model(HadCM3)coupled with the Soil and Water Assessment Tool(SWAT)hydrological model.The model was calibrated and validated against the historical monitored data from 2005 to 2009.The streamflow was estimated by downscaling HadCM3 outputs to the daily mean temperature and precipitation series,derived for three 30-year time slices,2020s,2050s and 2080s.Results suggest that daily mean temperature increases with a changing rate of 0.435℃per decade,and precipitation decreases with a changing rate of 0.761 mm per decade.Compared with other seasons,the precipitation in summer shows significant downward trend,while a significant upward trend in autumn.The annual streamflow demonstrates a general downward trend with a decreasing rate of 0.405 m3/s per decade.The streamflow shows significant downward and upward trends in summer and in autumn,respectively.The decreasing rate of streamflow in summer reaches 1.97 m 3 /s per decade,which contributes primarily to the decrease of streamflow.The results of this work would be of great benifit to the design of economic and social development planning in the study area. | ZHANG Lei LU Wenxi YANG Qingchun AN Yongkai LI Di GONG Lei | 2012 | Chinese Geographical Science2012,22,5: | 3 |

| 17 | Genome-wide analysis of differential DNA methylation in Silver-Russell syndrome显示文摘Silver-Russell Syndrome(SRS) is clinically heterogeneous disorder characterized by low birth weight, postnatal growth restriction, and variable dysmorphic features. Current evidence strongly implicates imprinted genes as an important etiology of SRS. Although almost half of the patients showed DNA hypomethylation at the H19/IGF2 imprinted domain, and approximately7%–10% of SRS patients have maternal uniparental disomy of chromosome 7(UPD(7) mat); the rest of the SRS patients shows unknown etiology. In this study, we investigate whether there are further DNA methylation defects in SRS patients. We measured DNA methylation in seven SRS patients and five controls at more than 485,000 CpG sites using DNA methylation microarrays. We analyzed methylation changes genome-wide and identified the differentially methylated regions(DMRs) using bisulfite sequencing and digital PCR. Our analysis identifies epimutations at the previously characterized domains of H19/IGF2,providing proof of principle that our methodology can detect the changes in DNA methylation at imprinted loci. In addition,our results showed a novel SRS associated imprinted gene OSBPL5 located on chromosome 11p14 with the probe cg25963939,which is hypomethylated in 4/7 patients(P=0.023, β=.0.243). We also report DMRs in other genes including TGFβ3, HSF1,GAP43, NOTCH4 and MYH14. These DMRs were found to be associated with SRS using GO pathway analysis. In this study,we identified the probe cg25963939, located at the 5′UTR of imprinted gene OSBPL5, as a novel DMR that is associated with SRS. We previously showed that a Nav1.7-targeting monoclonal antibody(SVmab) reduces Na+ currents and pain and itch responses in mice. Here, we investigated whether recombinant SVmab(rSVmab) binds to and blocks Nav1.7 similar to SVmab. ELISA tests revealed that SVmab was capable of binding to Nav1.7-expressing HEK293 cells,mouse DRG neurons, human nerve tissue, and the voltagesensor domain Ⅱ of Nav1.7. In contrast, rSVmab showed no or weak binding to Nav1.7 in these tests. Patch-clamp recordings showed that SVmab, but not rSVmab, markedly inhibited Na+ currents in Nav1.7-expressing HEK293 cells. Notably, electrical field stimulation increased the blocking activity of SVmab and rSVmab in Nav1.7-expressing HEK293 cells. SVmab was more effective than rSVmab in inhibiting paclitaxel-induced mechanical allodynia. SVmab also bound to human DRG neurons and inhibited their Na^+ currents.
