| 1 | Circular RNA circStag1 promotes bone regeneration by interacting with HuR显示文摘Postmenopausal osteoporosis is a common bone metabolic disorder characterized by deterioration of the bone microarchitecture,leading to an increased risk of fractures.Recently,circular RNAs(circ RNAs)have been demonstrated to play pivotal roles in regulating bone metabolism.However,the underlying functions of circ RNAs in bone metabolism in postmenopausal osteoporosis remain obscure.Here,we report that circ Stag1 is a critical osteoporosis-related circ RNA that shows significantly downregulated expression in osteoporotic bone marrow mesenchymal stem cells(BMSCs)and clinical bone tissue samples from patients with osteoporosis.Overexpression of circ Stag1 significantly promoted the osteogenic capability of BMSCs.Mechanistically,we found that circ Stag1 interacts with human antigen R(Hu R),an RNA-binding protein,and promotes the translocation of Hu R into the cytoplasm.A high cytoplasmic level of Hu R led to the activation of the Wnt signaling pathway by stabilizing and enhancing low-density lipoprotein receptor-related protein 5/6(Lrp5/6)andβ-catenin expression,thereby stimulating the osteogenic differentiation of BMSCs.Furthermore,overexpression of circ Stag1 in vivo by circ Stag1-loaded adeno-associated virus(circ Stag1-AAV)promoted new bone formation,thereby preventing bone loss in ovariectomized rats.Collectively,we show that circ Stag1 plays a pivotal role in promoting the regeneration of bone tissue via Hu R/Wnt signaling,which may provide new strategies to prevent bone metabolic disorders such as postmenopausal osteoporosis. | Gaoyang Chen Canling Long Shang Wang Zhenmin Wang Xin Chen Wanze Tang Xiaoqin He Zhiteng Bao Baoyu Tan Jin Zhao Yongheng Xie Zhizhong Li Dazhi Yang Guozhi Xiao Songlin Peng | 2022 | Bone Research2022,10,3: | 7 |

| 3 | Replanting of broadleaved trees alters internal nutrient cycles of native and exotic pines in subtropical plantations of China显示文摘Background:The replanting of broadleaved trees in pure coniferous plantations is widely implemented,as mixed plantations are generally more stable and functional.However,the effect of interspecific interactions between broadleaved and coniferous trees on internal nutrient cycles of conifers remains unclear.Methods:We selected pure coniferous plantations of a native(Pinus massoniana)and an exotic(P.elliottii)pine species and their corresponding mixed plantations with broadleaved trees(Schima superba)in subtropical China,and measured the nitrogen(N)and phosphorus(P)contents in the rhizosphere soils,fine roots,twigs,needles and needle litter of pines.We calculated the root capture,needle resorption and translocation of N and P by pines to determine the mobility of nutrients in trees.Results:Although the N and P in the rhizosphere soils increased due to the replanting of broadleaved trees,the N and P contents in the aboveground tissues of the two pine species did not increase in mixed plantations.Mixed planting had a negative effect on the N and P capture of native pine and a positive effect on that of exotic pine.The N and P resorption efficiencies increased in native pine but were unchanged in exotic pine after the replanting of S.superba.Native pine preferentially employed an aboveground nutrient resorption strategy,whereas exotic pine tended to adopt a belowground nutrient capture strategy after replanting.Translocation of N and P in trees was detected,which reflected the trade-offs between root nutrient capture and needle nutrient resorption.Conclusions:The effect of mixed planting varied between the species of native and exotic pines,and the internal nutrient cycles of both pine species might be dominated by interspecific interaction effects on nutrients rather than soil nutrients.Our study highlights the importance of selecting suitable broadleaved species for replanting in coniferous plantations. | Yong Lin Chengkang Xia Gaoyang Wu Fangchao Wang Shengnan Wang Yuanqiu Liu Fusheng Chen | 2022 | Forest Ecosystems2022,9,5: | 2 |

| 16 | Pterostilbene supresses inflammation-induced melanoma metastasis by impeding neutrophil elastase-mediated thrombospondin-1 degradation显示文摘Objective: Chronic inflammation plays a fatal role in tumor metastasis. Pterostilbene(PTE) is a natural dimethylated analogue of resveratrol with anticancer and anti-inflammatory activities. This study aimed to investigate the inhibitory effect of PTE on inflammation-associated metastasis and explore the underlying mechanisms.Methods: Lipopolysaccharide(LPS)-induced lung inflammation and melanoma metastasis models were established in mice. After PTE treatment for four weeks, the organ index, histological changes, proinflammatory cytokines, and the expression and activity of neutrophil elastase(NE), a biomarker of neutrophil influx in the lungs, were analysed. Additionally, direct effects of PTE on NE-induced B16 cell migration were explored in wound healing and Transwell assays, and the expression of thrombospondin-1(TSP-1) and epithelial-mesenchymal transition(EMT) markers were also detected.Results: PTE obviously attenuated the LPS-induced metastasis of circulatory B16 cells to lungs by reducing the number of metastatic nodules on the lung surfaces and the lung weight/body weight ratio. PTE treatment also significantly reduced LPS-activated increase levels of tumor necrosis factor(TNF)-a and interleukin(IL)-6 in the lungs of tumor-bearing mice. In addition, increased expression and enzyme activity of NE and decreased expression of TSP-1 were observed, and these were blocked by PTE. | 17 | Significance of extracellular signal-regulated kinase (ERK) in the development of rectal carcinoma显示文摘Objective: To study the ERK expression and its significance in the development of human rectal carcinoma. Methods: Samples were obtained from 62 patients with rectal carcinoma, including tumor tissue and adjacent normal rectal tissue. Western blot was used to measure the expression of ERK-1 and ERK-2. S-P immunohistochemical method was used to count the microvessel density (MVD). Results: ERK-1 and ERK-2 protein levels were increased in rectal carcinoma tissue compared with those in adjacent normal tissues (t = 2.980 and 2.194, P < 0.01 and 0.05 respectively). The MVD was higher in patients with higher ERK-1 and/or ERK-2 protein levels than that in cases with lower ERK-1 and/or ERK-2 levels.
