Serum osmolality and ions, and gill Na^(+)/K^(+)-ATPase of spottedtail goby Synechogobius ommaturus(R.)in response to acute salinity changes显示文摘This study was carried out to determine the effects of abrupt salinity change on osmoregulatory ability of the spottedtail goby Synechogobius ommaturus.720 juvenile fish(65.3±11.8 g)were transferred to 200 L tanks(with 40 juveniles in each tank),in which salinities were abruptly changed from 10 to 20,30,40,50 and freshwater.Survival rate, serum osmolality,electrolytes(Na^(+),Cl^(-),and K^(+))and gill Na^(+)/K^(+)-ATPase(NKA)activity were assessed successively in 528 h.Results showed serum osmolality, ion concentrations and gill Na^(+)/K^(+)-ATPase activity increased significantly when fish were transferred to salinity 40 and 50 and all fish in these groups died by the end of the experiment.Serum osmolality,Na^(+),Cl^(-)and K^(+)in fish transferred to a salinity of 20,30 and freshwater were not affected and no mortality was detected.Compared with the control group,a significantly decrease of NKA activity happened in the freshwater group, but the activity in 20 and 30 groups was not affected significantly. The results indicated that S.ommaturus could adapt rapidly and maintain homeostasis in a wide range of salinities(from freshwater to salinity 30)and this species may be suitable for aquaculture in estuarine and coastal areas where rapid salinity fluctuations commonly occur. | Chun Shui Yonghai Shi Xueming Hua Zhonghua Zhang Haiming Zhang Genhai Lu Yongde Xie | 2018 | Aquaculture and Fisheries2018,3,2: | 4

Phytoplankton and chlorophyll a relationships with ENSO in Prydz Bay, East Antarctica显示文摘The historical data of phytoplankton and chlorophyll a(Chl a)(1990–2002)obtained during the Chinese National Antarctic Research Expedition(CHINARE)in the Prydz Bay have been integrated. The results showed that the temperature, salinity, nutrients, and oxygen of seawater changed when El Nino/La Nina occurred. The variation of biological communities reflected the response of ecosystem to environmental changes.During El Ni?o period,Chl a concentration and phytoplankton community structure changed significantly, and the relative proportion of diatoms increased while dinoflagellates decreased.During La Ni?a period, the proportion of diatoms decreased, but the golden-brown algae and blue-green algae increased significantly. The variation of phytoplankton population directly affected the biodiversity of the bay, which were also quite sensitive to the marine environment changes.Meanwhile, the satellite remote sensing data of 2002–2011(December–March)have been used to study the temporal connection change of Chl a and phytoplankton in the Prydz Bay.We found that there were significant differences in the monthly variation characteristics of satellite remote sensing Chl a and sea surface temperature(SST),which had some links with sea ice melting and El Ni?o/La Ni?a events. We found that the start time of bloom advanced, lagged or synchronized with the changes of the SST,and we also found the occurrence time of phytoplankton bloom corresponded with the sea ice melting inner bay.To some extent, this study will help us understand the relationships between ENSO events and the phytoplankton bloom in the Southern Ocean. | ZHANG HaiSheng HAN ZhengBing ZHAO Jun YU PeiSong HU ChuanYu SUN WeiPing Yang Dan ZHU GenHai LU Bing Hans-UIrich PETER Walter VETTER | 2014 | Science China Earth Sciences2014,57,12: | 1

Expression of Bmi-1 and EZH2 in tissues adjacent to human epithelial ovarian cancer cells of orthotopic implantation in nude mice显示文摘Objective This study investigated the feasibility of screening residual normal ovarian tissues based on the expression of Bmi-1 and EZH2 in tissues adjacent to orthotopic ovarian carcinomas in nude mice. Methods The human epithelial ovarian cancer cell line OVCAR3 was grown in subcutaneous tissues and the tumor tissues were orthotopically implanted. The expression levels of Bmi-1 and EZH2 were detected by immunohistochemical staining and RT-PCR in cancer tissues, proximal and remote tissues with respect to the cancer tissues, and normal ovarian tissues of nude mice. Results Thirty-five ovarian tissue samples with normal biopsy results were obtained from 40 cases of human epithelial ovarian cancer in the nude mice in which the tumor tissues were orthotopically implanted. Bmi-1 and EZH2 expression levels were lower in proximal paraneoplastic tissue samples than in cancer tissue samples(P < 0.05) and higher than in remote paraneoplastic tissue samples(P < 0.01). No significant difference was found in the expression levels of Bmi-1 and EZH2 using immunohistochemistry among residual normal ovarian tissues obtained from orthotopically implanted models that differed in severity. The expression of Bmi-1 and EZH2 was negative in 20 normal ovarian tissue samples. Conclusion The expression levels of Bmi-1 and EZH2 were reduced with increasing distance from the cancer tissues. Negative expression of these tumor-associated genes can be used as a standard for the screening of normal ovarian tissues adjacent to tumor tissues. Normal ovarian tissues can be obtained from the tissues adjacent to tumors. | Genhai Zhu Lan Hong Shengtan Wang Zhaoxin Yang Chunying Chen Shixuan Wang | 2015 | The Chinese-German Journal of Clinical Oncology2015,14,1: | 0
Absent in melanoma 2 attenuates proliferation and migration and promotes apoptosis of human colorectal cancer cells by activating P38MAPK signaling pathway显示文摘Colorectal cancer(CRC)stands among the top prevalent cancers worldwide and holds a prominent position as a major contributor to cancer-related mortality globally.Absent in melanoma 2(AIM2),a constituent of the interferoninducible hematopoietic interferon-inducible nuclear antigens with 200 amino acid repeats protein family, contributes to both cancer progression and inflammasome activation.Despite this understanding, the precise biological functions and molecular mechanisms governed by AIM2 in CRC remain elusive.Consequently, this study endeavors to assess AIM2’s expression levels, explore its potential antitumor effects, elucidate associated cancer-related processes, and decipher the underlying signaling pathways in CRC.Our findings showed a reduced AIM2 expression in most CRC cell lines.Elevation of AIM2 levels suppressed CRC cell proliferation and migration, altered cell cycle by inhibiting G1/S transition, and induced cell apoptosis.Further research uncovered the participation of P38 mitogen-activated protein kinase(P38MAPK)in AIM2-mediated modulation of CRC cell apoptosis and proliferation.Altogether, our achievements distinctly underscored AIM2’s antitumor role in CRC.AIM2 overexpression inhibited proliferation and migration and induced apoptosis of CRC cells via activating P38MAPK signaling pathway, indicating AIM2 as a prospective and novel therapeutic target for CRC. | ZHI ZHANG XIAOSONG LI YING ZHANG HAO ZHU ZHENGUO QIAO YANG LU XIUWEI MI HUIHUA CAO GENHAI SHEN SONGBING HE | 2024 | Oncology Research2024,32,2: | 0
Comparative Transcriptome Analysis of Seed Germination of a Cotton Variety with High Tolerance to Low Temperature显示文摘Gossypium hirsutum L.is an important cash crop native to the subtropics and is widely cultivated around the world.Low temperature is an important stress that seriously affects seed germination and emergence during planting. In this study, transcriptomic profiles of low-temperature-and normal-temperature-germinated seeds of Xinluzao 25,a variety with low-temperature tolerance and high germination rates, were analyzed and compared. The following results were obtained.(1)A total of 81.06 Gb of clean data were obtained after transcriptome sequencing and assembly, and 76,931 non-redundant Unigene sequences were obtained after data consolidation and concatenation;of these,69,883 Unigene sequences were annotated. In addition,55,463 Unigene transcript sequences(72.2%)were annotated for Gene Ontology(GO)classification, and 26,629 genes were involved in 50 metabolic pathways identified by Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis.(2)Three main pathways related to low-temperature tolerance of seed germination were identified:starch and sucrose metabolism, phenylpropanoid biosynthesis, and cysteine and methionine metabolism.Their main molecular functions involve the regulation of abscisic acid and activities of enzymes such as amylase, peroxidase, and oxidoreductase.During germination at low temperature, more genes were down-regulated than up-regulated genes at the protrusion stage(2 mm),and more genes were up-regulated than down-regulated at the germination stage(30 mm)after protrusion.(3)The enzyme activities at the two stages showed that amylase, peroxidase, catalase, and glutathione reductase had higher activities when the seeds germinated at 15℃.In this study, high expression of amylase, peroxidase, catalase, and glutathione reductase genes may be the main cause of increased tolerance to low temperature. | Genhai Hu Maoni Chao Xiuren Zhou Yuanzhi Fu | 2023 | Phyton-International Journal of Experimental Botany2023,92,9: | 0
