1Clinical observation of salvianolic acid B in treatment of liver fibrosis in chronic hepatitis B显示文摘AIM: To evaluate the clinical efficacy of salvianolic acidB (SA-B) on liver fibrosis in chronic hepatitis B.METHODS: Sixty patients with definite diagnosis of liverfibrosis with hepatitis B were included in the trial.Interferon-γ (IFN-γ) was used as control drug. Thepatients took orally SA-B tablets or received muscularinjection of IFN-γ in the double blind randomized test,The complete course lasted 6 months. The histologicalchanges of liver biopsy specimen before and after thetreatment were the main evidence in evaluation, incombination with the results of contents of serum HA,LN, Ⅳ-C, P-Ⅲ-P, liver ultrasound imaging, andsymptoms and signs.RESULTS: Reverse rate of fibrotic stage was 36.67 % inSA-B group and 30.0 % in IFN-γgroup. Inflammatoryalleviating rate was 40.0 % in SA-B group and 36.67 %in IFN-γ group. The average content of HA and Ⅳ-Cwas significantly lower than that before treatment. Theabnormal rate also decreased remarkably. Overallanalysis of 4 serological fibrotic markers showedsignificant improvement in SA-B group as comparedwith the IFN-γgroup. Score of liver ultrasound imagingwas lower in SA-B group than in IFN-γgroup (HA 36.7 %vs80 %,Ⅳ-C 3.3 % vs23.2 %). Before the treatment,ALT AST activity and total bilirubin content of patientswho had regression of fibrosis after oral administrationof SA-B, were significantly lower than those of patientswho had aggravation of fibrosis after oraladministration of SA-B. IFN-γ showed certain sideeffects (fever and transient decrease of leukocytes,occurrence rates were 50 % and 3.23 %), but SA-Bshowed no side effects.CONCLUSION: SA-B could effectively reverse liverfibrosis in chronic hepatitis B. SA-B was better than IFN-γ in reduction of serum HA content, overall decrease of4 serum fibrotic markers, and decrease of ultrasoundimaging score. Liver fibrosis in chronic hepatitis B withslight liver injury was more suitable to SA-B in anti-fibrotic treatment. 3Effect of multidisciplinary team treatment on outcomes of patients with gastrointestinal malignancy显示文摘AIM: To evaluate the effect of multidisciplinary team (MDT) treatment modality on outcomes of patients with gastrointestinal malignancy in China. METHODS: Data about patients with gastric and colorectal cancer treated in our center during the past 10 years were collected and divided into two parts. Part 1 consisted of the data collected from 516 consecutive complicated cases discussed at MDT meetings in Peking University School of Oncology (PKUSO) from December 2005 to July 2009. Part 2 consisted of the data collected from 263 consecutive cases of resect-able locally advanced rectal cancer from January 2001 to January 2005. These 263 patients were divided into neoadjuvant therapy (NT) group and control group. Patients in NT group received MDT treatment, namely neoadjuvant therapy + surgery + postoperative adjuvant therapy. Patients in control group underwent direct surgery + postoperative adjuvant therapy. The outcomes in two groups were compared. RESULTS: The treatment strategy was altered after discussed at MDT meeting in 76.81% of gastric cancer patients and in 58.33% of colorectal cancer patients before operation. The sphincter-preservation and local control of tumor were better in NT group than in control group. The 5-year overall survival rate was also higher in NT group than in control group (77.23% vs 69.75%, P = 0.049). 6Exome sequencing confirms molecular diagnoses in 38 Chinese families with hereditary spherocytosis显示文摘Hereditary spherocytosis(HS), the most common cause of congenital hemolytic anemia, is caused by deficiency of the erythrocyte membrane proteins. Five causative genes(ANK1, SPTB, SPTA1, SLC4 A1, and EPB42) have been identified. To date,molecular genetic studies have been performed in different populations, including the American, European, Brazilian, Japanese and Korean populations, whereas only a few studies have been described in the Chinese population. Here, by reanalysis of the exome data, we revealed causative mutations and established a definitive diagnosis of HS in all 38 Chinese families. We found 34 novel mutations and four reported mutations in three known HS-causing genes—17 in ANK1, 17 in SPTB and four in SLC4 A1,suggesting that ANK1 and SPTB are the major genes in Chinese patients with HS. All of the ANK1 or SPTB mutations, scattered throughout the entire genes, are non-recurrent; and most of them are null mutations, which might cause HS via a haploinsufficiency mechanism. De novo mutations in ANK1 or SPTB often occur with an unexpected high frequency(87.5% and64.2%, respectively). 7EXPERIMENTAL STUDY OF FLOWFIELD STRUCTURE AROUND AN OGIVE-CYLINDER显示文摘The flowfield structure and their aerodynamic characteristics over an ogive cylinder were studied by means of flow visualization and surface pressure measurement in a water tunnel and a wind tunnel. The existence of multi asymmetric vortices over long slender bodies was experimentally confirmed at large angles of attack and in the subcritical Reynolds number range. The spatial 3 D characteristics of the multi vortices system were analyzed and a physical model was developed. The topological structure of different patterns in cross flow plane was studied and the mechanism governing the formation of asymmetric vortices and multi vortices was discussed from the viewpoint of stability of the topological structure. 8Droplet deposition and efficiency of fungicides sprayed with small UAV against wheat powdery mildew显示文摘Small unmanned aerial vehicle(UAV),a timely and effective pesticide sprayer,can be used to spray pesticide in a high efficiency without harming the crop,and is especially suitable for spraying for small farms and hills in Asian countries.In recent years,it has been chosen as a key and new technique for pesticide application.This paper studied the impact of UAV(UAV N-3)spraying parameters(different working height and different spraying concentrations)on the deposition of droplets on the wheat canopy and the prevention of powdery mildew.Water sensitive paper was used as the sampler to collect the pesticide droplets and image processing software Deposit Scan was used to compute the coverage rate of droplets on the wheat top layer to acquire the proportional distribution of droplets on the wheat lower layer.The experimental results showed the impact of spraying height on the distribution of droplets on the wheat upper layer was quite significant,when the spraying height was 5.0 m and the spraying speed was 4 m/s,the coverage rate of droplets on the wheat lower layer was the largest,as it was 45.6%of that on the upper layer,the droplets distribution was the most uniform,and the coefficient of variation was 33.13%.450 g/hm^(2)(dosage registered)of triadimefon SC(44%)was sprayed by the UAV,the control efficiency reached 55.1%after applying which was better than 20%and 40%of dosage decreased for each hectare,and the applying effect(35.6%)of 20%dosage decreased for each hectare had no significant difference from the applying effect(34.6%)applied by a knapsack-type electric sprayer.At 10 d after applying,the prevention effect realized by UAV was lower than that realized by a knapsack-type electric sprayer,and it may be correlated to the meteorological condition and water amount in the pesticide sprayed.Thus,when UAV spraying was chosen to prevent wheat powdery mildew under a serious disease situation,an auxiliary agent for spraying could be added to prolong the retention of pesticide on the plant surface to extend the pesticide effect.This study can provide a reference for the optimized design,performance upgrade and reasonable application of small UAV sprayers.Weicai Qin Xinyu Xue Shaoming Zhang Wei Gu Baokun Wang 2018International Journal of Agricultural and Biological Engineering2018,11,2:16

11PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. 12Impact of EBV infection and immune function assay for lymphoproliferative disorder in pediatric patients after liver transplantation: A single-center experience显示文摘Background:Post-transplant lymphoproliferative disorder(PTLD)is a lethal complication after pediatric liver transplantation,but information regarding risk factors for the development of PTLD remains unclear.This study was to identify characteristics and risk factors of PTLD.Methods:A total of 705 pediatric patients who underwent liver transplantation between January 2017 and October 2018 were studied.Impact of clinical characteristics and Epstein-Barr virus(EBV)infection on the development of PTLD was evaluated.In addition,ImmuKnow assay was adopted in partial patients to analyze the immune status.Results:Twenty-five(3.5%)patients suffered from PLTD with a median time of 6 months(3–14 months)after transplantation.Extremely high tacrolimus(TAC)level was found in 2 fatal cases at PTLD onset.EBV infection was found in 468(66.4%)patients.A higher peak EBV DNA loads(>9590 copies/mL)within 3 months was a significant indicator for the onset of PTLD.In addition,the ImmuKnow assay demonstrated that overall immune response was significantly lower in patients with EBV infection and PTLD(P<0.0001).The cumulative incidence of PTLD was also higher in patients with lower ATP value(≤187 ng/mL,P<0.05).Conclusions:A careful monitoring of EBV DNA loads and tacrolimus concentration might be supportive in prevention of PTLD in pediatric patients after liver transplantation.In addition,application of the ImmuKnow assay may provide guidance in reducing immunosuppressive agents in treatment of PTLD.Tian Qin Xiang-Qian Gu Seog-Song Jeong Yan-Yan Song Jin-Chuan Liu Jian-Xin Zheng Feng Xue Qiang Xia 2020Hepatobiliary & Pancreatic Diseases International2020,19,1:8

15Cadmium Activates Reactive Oxygen Species-dependent AKT/mT OR and Mitochondrial Apoptotic Pathways in Neuronal Cells显示文摘Objective To examine the role of Cd-induced reactive oxygen species(ROS)generation in the apoptosis of neuronal cells.Methods Neuronal cells(primary rat cerebral cortical neurons and PC12 cells)were incubated with or without Cd post-pretreatment with rapamycin(Rap)or N-acetyl-L-cysteine(NAC).Cell viability was determined by MTT assay,apoptosis was examined using flow cytometry and fluorescence microscopy,and the activation of phosphoinositide 3'-kinase/protein kinase B(Akt)/mammalian target of rapamycin(m TOR)and mitochondrial apoptotic pathways were measured by western blotting or immunofluorescence assays.Results Cd-induced activation of Akt/m TOR signaling,including Akt,m TOR,p70 S6 kinase(p70 S6K),and eukaryotic initiation factor 4E binding protein 1(4E-BP1).Rap,an m TOR inhibitor and NAC,a ROS scavenger,blocked Cd-induced activation of Akt/m TOR signaling and apoptosis of neuronal cells.Furthermore,NAC blocked the decrease of B-cell lymphoma 2/Bcl-2 associated X protein(Bcl-2/Bax)ratio,release of cytochrome c,cleavage of caspase-3 and poly(ADP-ribose)polymerase(PARP),and nuclear translocation of apoptosis-inducing factor(AIF)and endonuclease G(Endo G).Conclusion Cd-induced ROS generation activates Akt/m TOR and mitochondrial pathways,leading to apoptosis of neuronal cells.Our findings suggest that m TOR inhibitors or antioxidants have potential for preventing Cd-induced neurodegenerative diseases.YUAN Yan WANG Yi HU Fei Fei JIANG Chen Yang ZHANG Ya Jing YANG Jin Long ZHAO Shi Wen GU Jian Hong LIU Xue Zhong BIAN Jian Chun LIU Zong Ping 2016Biomedical and Environmental Sciences2016,29,2:8

18Associations of egg consumption with incident cardiovascular disease and all-cause mortality显示文摘Eggs are nutrient-dense while also loaded with abundant cholesterol, thus making the public hesitant about their consumption.We conducted the study to investigate if egg consumption is associated with incident cardiovascular disease(CVD) and all-cause mortality. Using the project of Prediction for Atherosclerotic Cardiovascular Disease Risk in China, we included 102,136 adults free of CVD and assessed their egg consumption with food-frequency questionnaires. CVD endpoints and all-cause mortality were confirmed during follow-ups by interviewing participants or their proxies and checking hospital records/death certificates.The HRs(95% CIs) were calculated using the cohort-stratified Cox regression models. During 777,163 person-years of followup, we identified 4,848 incident CVD and 5,511 deaths. U-shaped associations of egg consumption with incident CVD and allcause mortality were observed. Compared with consumption of 3–<6/week, the multivariable-adjusted HRs(95% CIs) of <1/week and ≥10/week for incident CVD were 1.22(1.11 to 1.35) and 1.39(1.28 to 1.52), respectively. The corresponding HRs(95% CIs) for all-cause mortality were 1.29(1.18 to 1.41) and 1.13(1.04 to 1.24). 19Exosomal piRNA profiling revealed unique circulating piRNA signatures of cholangiocarcinoma and gallbladder carcinoma显示文摘Cholangiocarcinoma(CCA)and gallbladder carcinoma(GBC)are biliary tract cancers with poor five-year survival and high recurrence rates.Both CCA and GBC patients suffer from lack of circulating diagnostic biomarkers at the early stage.Extracellular vesicles,especially exosomes,have been emerged as promising diagnostic sources for cancers due to easy and quick accessibility.Hence,identification of exosomal biomarkers provides a novel strategy for CCA and GBC diagnosis.Here,five CCA patients and four GBC patients were enrolled for exosomal small RNA sequencing.Our data showed that exosomal piwi-interacting RNA(piRNA)populations were altered in the plasma of CCA and GBC patients.In comparison to healthy individuals,694 and 323 piRNAs were upregulated in CCA and GBC,respectively,while 36 and 191 piRNAs were down regulated.Interestingly,sequencing results predicted that piR-2660989,piR-10506469,piR-20548188,piR-10822895,piR-hsa-23209,and piR-18044111 were upregulated in both CCA and GBC plasma.Importantly,we further included blood samples from 50 health individuals,40 CCA patients,and 25 GBC patients and found that piR-10506469 were significantly increased in the exosomes of plasma from both CCA and GBC patients.Moreover,we analyzed the expression levels of differentially expressed exosomal piRNAs in the plasma of CCA and GBC patient before and after surgeries and found that piR-10506469 and piR-20548188 were significantly decreased in patients underwent surgeries.Taken together,our data revealed that exosomal piRNAs those are differentially expressed in CCA and GBC plasma may serve as potential biomarkers for the diagnosis of CCA and GBC.Xinjin Gu Chen Wang Hui Deng Chong Qing Rong Liu Sanhong Liu Xinying Xue 2020Acta Biochimica et Biophysica Sinica2020,52,5:7
20Hybrid CFD/FEM-BEM simulation of cabin aerodynamic noise for vehicles traveling at high speed显示文摘Flow passing a vehicle may lead to the increase of the cabin interior noise level through a variety of mechanisms. These mechanisms include vibrations caused by aerodynamic excitations and reradiation from the glass panels, exterior noise trans-mitted and leaked through door seals including gaps and glass edge, and transmission of airborne noise generated by the interaction of flow with body panels. It is of vital importance to predict both the flow fields and the acoustic sources around the ve-hicle to accurately assess the impact of wind induced noise inside the cabin. In the present study, an unstructured segregated finite volume model was used to calculate the flow fields in which a hexahedron grid is used to simplify the vehicle geometry.A large eddy simulation coupled with a wall function model was applied to predict the exterior transient flow fields. The mean flow quantities were thus calculated along the symmetry plane and the vehicle's side windows. A coupled FEM/BEM method was used to compute the vehicle's interior noise level.
