| 1 | Detection of DNA methylation changes during seed germination in rapeseed (Brassica napus)显示文摘DNA methylation is known to play a crucial role in regulating plant development and or- gan or tissue differentiation. In this study, we as- sessed the extent and pattern of cytosine methylation during rapeseed (Brassica napus L.) seed germina- tion, and compared the methylation level of various tissues in seedling, using the techniques of methyla- tion-sensitive amplified polymorphism (MSAP) and HPLC separation and quantification of nucleosides. In all, 484 bands, each representing a recognition site cleaved by either or both of the isoschizomers, were amplified by 12 pairs of selective primers in DNA ob- tained from dry seeds. A total of 76 sites were found to be differentially digested by the isoschizomers, indicating that approximately 15.7% of 5′-CCGG-3′ sites in the genome were cytosine methylated. Four classes of patterns were observed in a comparative assay of cytosine methylation in the dry and germi- nating seeds; a small number of hypermethylation events occurred at 5′-CCGG-3′ sites in germinating seeds compared with dry seeds, while many more hypomethylation events were detected after seed germination. Differences in DNA methylation level in various tissues were also detected; radicel was less methylated than hypocotyl and cotyledon. These observations were further confirmed by HPLC analy- sis. In addition, sequencing of eleven differentially methylated fragments and the subsequent blast search revealed that cytosine methylated 5′-CCGG- 3′ sequences were equally distributed between cod- ing and non-coding regions. These results clearly demonstrate the power of MSAP technique for large-scale DNA methylation detection in rapeseed genome, and the complexity of DNA methylationchange during seed germination. At present the ultimate bearing ca- pacity of RC structures strengthened with bonded steel plates is calculated mostly using the theory based on the test. Four beams, including one reference beam, two strengthened concrete beams in primary force and secondary force respectively, and one strengthened concrete beam which was not anchored enough, were tested under four-point bending (4PB) in order to get the data of strain of longitudinal bars, bonded bottom steel plate in tension and deflection of beams in the middle span. The experimental program was supported by a three-dimensioned finite analysis using ABAQUS. | 6 | A Nonlinear Observer Approach of SOC Estimation Based on Hysteresis Model for Lithium-ion Battery显示文摘In this paper, a state of charge(SOC) estimation approach for lithium-ion battery based on equivalent circuit model and the input-to-state stability(ISS) theory has been proposed. According to the electrochemical performance of lithiumion battery, the equivalent circuit model with two RC networks is established, which includes hysteresis characteristic in inner electrochemical response process. The nonlinear relation between open circuit voltage(OCV) and SOC is obtained from a rapid test. Exponential fitting method is used to identify the parameters of the model. A novel state observer based on ISS theory is designed for lithium-ion battery SOC estimation. The designed observer is tested on AMESim and Simulink co-simulation. Passive shock wave/boundary layer control(PSBC) for drag reduction was conducted by SC(2)-0714 supercritical wing, with emphases on parameter of porous/slot and bump, such as porous distribution, hole diameter,cavity depth, porous direction and so on. A sequential quadratic programming(SQP) optimization method coupled with adjoint method was adopted to achieve the optimized shape and position of the bumps.Computational fluid dynamics(CFD), force test and oil test with half model all indicate that PSBC with porous, slot and bump generally reduce the drag by weaker lambda shock at supercritical conditions.According to wind tunnel test results for angle of attack of 2? at Mach number M = 0.8, the porous configuration with 6.21% porosity results in a drag reduction of 0.0002 and lift–drag ratio increase of 0.2, the small bump configuration results in a drag reduction of 0.0007 and lift–drag ratio increase of 0.3.Bump normally reduce drag at design point with shock wave position being accurately computed. | 17 | A low-energy ion spectrometer with half-space entrance for three-axis stabilized spacecraft显示文摘A low-energy ion spectrometer(LEIS) for use aboard three-axis stabilized spacecraft has been developed to measure ion energy per charge distribution in three-dimensional space with good energy-, angular-and temporal-resolutions. For the standard top-hat electrostatic analyzer used widely in space plasma detection, three-axis stabilized spacecraft makes it difficult to obtain complete coverage of all possible ion arrival directions. We have designed angular scanning deflectors supplementing to a cylindrically symmetric top-hat electrostatic analyzer to provide a half-space field of view as 360°×90°(–45°–+45°), and fabricated the LEIS flight model for detecting magnetospheric ions in geosynchronous orbit. The performance of this payload has been evaluated in detail by a series of simulation and environmental tests, and the payload has also been calibrated through laboratory experiments using a low-energy ion source.
