1Survival of transplanted neurotrophin-3 expressing human neural stem cells and motor function in a rat model of spinal cord injury显示文摘<正>BACKGROUND:Many methods have been attempted to repair nerves following spinal cord injury, including peripheral nerve transplantation,Schwann cell transplantation,olfactory ensheathing cell transplantation,and embryonic neural tissue transplantation.However,there is a need for improved outcomes. OBJECTIVE:To investigate the repair feasibility for rat spinal cord injury using human neural stem cells(hNSCs) genetically modified by lentivirus to express neurotrophin-3. DESIGN,TIME AND SETTING:In vitro cell biological experiment and in vivo randomized,controlled, genetic engineering experiment were performed at the Third Military Medical University of Chinese PLA and First People's Hospital of Yibin,China from March 2006 to December 2007. MATERIALS:A total of 64 adult,female,Wistar rats were used for the in vivo study.Of them,48 rats were used to establish models of spinal cord hemisection,and were subsequently equally and randomly assigned to model,genetically modified hNSC,and normal hNSC groups.The remaining 16 rats served as normal controls. METHODS:hNSCs were in vitro genetically modified by lentivirus to secrete both green fluorescence protein and neurotrophin-3.Neurotrophin-3 expression was measured by Western blot. Genetically modified hNSC or normal hNSC suspension(5×10~5) was injected into the rat spinal cord following T_(10) spinal cord hemisection.A total of 5μL Dulbecco's-modified Eagle's medium was infused into the rat spinal cord in the model group.Transgene expression and survival of transplanted hNSCs were determined by immunohistochemistry.Motor function was evaluated using the Basso,Beattie,and Bresnahan(BBB) scale. MAIN OUTCOME MEASURES:The following parameters were measured:expression of neurotrophin-3 produced by genetically modified hNSCs,transgene expression and survival of hNSCs in rats,motor function in rats. RESULTS:hNSCs were successfully genetically modified by lentivirus to stably express neurotrophin-3.The transplanted hNSCs primarily gathered at,or around,the injection site two weeks following transplantation,and gradually migrated towards the surrounding tissue. Transplanted hNSCs were observed 7.0-8.0 mm away from the injection site.In addition,hNSCs were observed 10 weeks after transplantation.At week 4,BBB locomotor scores were significantly greater in the genetically modified hNSC and normal hNSC groups,compared with the model group (P<0.05),and scores were significantly greater in the genetically modified hNSC group compared with the normal hNSC group(P<0.05). 2Type I IFN augments IL-27-dependent TRIM25 expression to inhibit HBV replication显示文摘Hepatitis B virus(HBV)can cause chronic hepatitis B,which may lead to cirrhosis and liver cancer.Type I interferon(IFN)is an approved drug for the treatment of chronic hepatitis B.However,the fundamental mechanisms of antiviral action by type I IFN and the downstream signaling pathway are unclear.TRIM25 is an IFN-stimulated gene(ISG)that has an important role in RIG-I ubiquitination and activation.Whether TRIM25 is induced in liver cells by type I IFN to mediate anti-HBV function remains unclear.Here we report that interleukin-27(IL-27)has a critical role in IFN-induced TRIM25 upregulation.TRIM25 induction requires both STAT1 and STAT3.In TRIM25 knockout HepG2 cells,type I IFN production was consistently attenuated and HBV replication was increased,whereas overexpression of TRIM25 in HepG2 cells resulted in elevated IFN production and reduced HBV replication.More interestingly,we found that TRIM25 expression was downregulated in HBV patients and the addition of serum samples from HBV patients could inhibit TRIM25 expression in HepG2 cells,suggesting that HBV might have involved a mechanism to inhibit antiviral ISG expression and induce IFN resistance.Collectively,our results demonstrate that type I IFN-induced TRIM25 is an important factor in inhibiting HBV replication,and the IFN-IL-27-TRIM25 axis may represent a new target for treating HBV infection.Guangyun Tan Qingfei Xiao Hongxiao Song Feng Ma Fengchao Xu Di Peng Na Li Xiaosong Wang Junqi Niu Pujun Gao F Xiao-Feng Qin Genhong Cheng 2018Cellular & Molecular Immunology2018,15,3:13
3Type III interferon-induced CBFβinhibits HBV replication by hijacking HBx显示文摘Hepatitis B virus(HBV)and its associated chronic infection remain serious health threats worldwide.However,there is still no impactful approach for clinical treatment of hepatitis B patients.Therefore,developing a better understanding of the interactions between HBV and its host is particularly important.HBV infection has been reported to induce type-III but not type-I or type-II interferon(IFN).In this study,we identified CBFβ,an HIV enhancer,as an HBV restriction factor that is specifically induced by type-III IFN in the early stages of HBV infection.Type-III IFN-induced IL-10 played an important role in the production of CBFβ.Interestingly,the interaction between CBFβ-and HBV-encoded regulatory protein X(HBx)enhanced the stability of CBFβ,but notably blocked HBx-mediated promotion of HBV replication.CBFβexpression was lower in HBV patients than in healthy persons,and the addition of serum from HBV patients inhibited CBFβexpression in HepG2 cells.On the contrary,HBV via HBsAg inhibited type-III IFN-induced CBFβexpression and decreased the anti-HBV activity of type-III IFN,suggesting that HBV inhibits antiviral interferon-stimulated gene(ISG)expression and induces IFN resistance.Collectively,our results demonstrate that type-III IFN-triggered and IL-10-induced CBFβare crucial factors for inhibiting HBV replication,and the HBx–CBFβ–HBsAg axis reveals a new molecular mechanism of interaction between HBV and its hosts.Fengchao Xu Hongxiao Song Qingfei Xiao Na Li Hong Zhang Genhong Cheng Guangyun Tan 2019Cellular & Molecular Immunology2019,16,4:3
4Improved adaptively robust estimation algorithm for GNSS spoofer considering continuous observation error显示文摘Once the spoofer has controlled the navigation sys-tem of unmanned aerial vehicle(UAV),it is hard to effectively control the error convergence to meet the threshold condition only by adjusting parameters of estimation if estimation of the spoofer on UAV has continuous observation error.Aiming at this problem,the influence of the spoofer’s state estimation error on spoofing effect and error convergence conditions is theoretically analyzed,and an improved adaptively robust estimation algo-rithm suitable for steady-state linear quadratic estimator is pro-posed.It enables the spoofer’s estimator to reliably estimate UAV status in real time,improves the robustness of the estima-tor in responding to observation errors,and accelerates the con-vergence time of error control.Simulation experiments show that the mean value of normalized innovation squared(NIS)is reduced by 88.5%,and the convergence time of NIS value is reduced by 76.3%,the convergence time of true trajectory error of UAV is reduced by 42.3%,the convergence time of estimated trajectory error of UAV is reduced by 67.4%,the convergence time of estimated trajectory error of the spoofer is reduced by 33.7%,and the convergence time of broadcast trajectory error of the spoofer is reduced by 54.8%when the improved algorithm is used.The improved algorithm can make UAV deviate from pre-set trajectory to spoofing trajectory more effectively and more subtly.GAO Yangjun LI Guangyun LYU Zhiwei ZHANG Lundong LI Zhongpan 2022Journal of Systems Engineering and Electronics2022,33,5:2

11Experimental Investigations on NO_(x) Emission and Combustion Dynamics in an Axial Fuel Staging Combustor显示文摘Axial Fuel Staging(AFS)technology is an advanced low-emission combustion method in modem gas turbine,which divides the combustor into two axially arranged combustion zones.For revealing the characteristics of axial staged combustion,an industrial-grade combustor was designed and built.The distribution of temperature and velocity field in the combustor was presented with numerical simulation.And an Atmospheric Combustor Test Rig for axial staged combustion was built.The flow resistance characteristics of the combustor were measured at first.Then the effects of the equivalent ratio and the preheating temperature on the pollutant emission and combustion instability were investigated.The results show that the total pressure recovery coefficient in cold state is always above 98%;starting the secondary combustion at low load can reduce NO emissions by 50%,and can suppress the combustion oscillation amplitude of the combustor.At the design point with φ=0.62 and preheating temperature=400°C,NO emission and CO emission are 15.68 and 4.22 mg/m^(3)(@15%O_(2)).LI Yuze JIA Yuliang JIN Ming ZHU Xutong GE Bing MAO Ronghai REN Lilei CHEN Mingmin JIAO Guangyun 2022Journal of Thermal Science2022,31,1:0
12Large-scale spatial data visualization method based on augmented reality显示文摘Background A task assigned to space exploration satellites involves detecting the physical environment within a certain space.However,space detection data are complex and abstract.These data are not conducive for researchers'visual perceptions of the evolution and interaction of events in the space environment.Methods A time-series dynamic data sampling method for large-scale space was proposed for sample detection data in space and time,and the corresponding relationships between data location features and other attribute features were established.A tone-mapping method based on statistical histogram equalization was proposed and applied to the final attribute feature data.The visualization process is optimized for rendering by merging materials,reducing the number of patches,and performing other operations.Results The results of sampling,feature extraction,and uniform visualization of the detection data of complex types,long duration spans,and uneven spatial distributions were obtained.The real-time visualization of large-scale spatial structures using augmented reality devices,particularly low-performance devices,was also investigated.Conclusions The proposed visualization system can reconstruct the three-dimensional structure of a large-scale space,express the structure and changes in the spatial environment using augmented reality,and assist in intuitively discovering spatial environmental events and evolutionary rules.Xiaoning QIAO Wenming XIE Xiaodong PENG Guangyun LI Dalin LI Yingyi GUO Jingyi REN 2024虚拟现实与智能硬件(中英文)2024,6,2:0

14Prey life stages modulate effects of predation stress on prey lifespan,development,and reproduction in mites显示文摘The non-consumptive effects of predator-induced stress can influence a variety of life-history traits.Many previous studies focused only on short-term effects such as development and reproductive rates.Recent studies have showed that long-term predation stress(given during the whole life of the prey)and short-term predation stress(provided during the immature stage of the prey)could generate completely opposite results:the former could decrease lifespan,whereas the later could increase lifespan.However,it is still unclear whether the advantage is because of the short duration of exposure or the early stage of life during which exposure was exerted.Thus,in this study,the prey(Tyrophagus putrescentiae)was exposed to predation stress from the predator(Neoseiulus cucumeris)during different life stages(larva,protonymph,tritonymph,first 5 d of oviposition,the full lifespan or none of the above).The results showed that the predation stress supplied during larval and protonymphal stage delayed development,reduced fecundity and prolonged lifespan of the prey,while the stress given during tritonymphal stage only reduced lifespan slightly and the stress given during the first 5 d of oviposition did not change lifespan but reduced fecundity.This study indicated that the effects of predation stress are dependent on prey life stage and the predation stress experienced in the early life stages is important to lifespan modulation.Xiaoying Weil Guangyun Li Zhi-Qiang Zhang 2023Insect Science2023,30,3:0
