1SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
2Comparative Analysis of Three Types of Peaches:Identification of the Key Individual Characteristic Flavor Compounds by Integrating Consumers' Acceptability with Flavor Quality显示文摘In order to recognize the key flavor compounds influencing the consumers' choice for honey peach, nectarine, and flat peach and provide important information for orientation breeding, sugars, organic acids, and aroma volatiles in 3 types of peaches were examined by high performance liquid chromatography(HPLC) and gas chromatography–mass spectrometry(GC–MS) and their key individual characteristic flavor compounds were identified by multivariate analysis integrated with consumers' evaluation. No significant differences in sugar and organic acid were observed between 3 types of peaches. In hierarchical clustering analysis(HCA) and principal component analysis(PCA)models, 3 types of peaches could not be differentiated by sugars or organic acids, but could be identified completely by aroma volatiles or by flavor compounds. A partial least squares regression(PLSR) model revealed that the key individual characteristic volatiles in nectarine, honey peach, and flat peach are C9 compounds and terpenic compounds, norisoprenoids and C6 compounds, and benzaldehyde, γ-decalactone, andδ-dodecalactone, respectively. These results suggest that sugars and organic acids form the background flavor of peach fruit and lactones shape the typical peach aroma. 3Upconversion nanoparticles for super-resolution quantification of single small extracellular vesicles显示文摘Although small EVs(sEVs)have been used widely as biomarkers in disease diagnosis,their heterogeneity at single EV level has rarely been revealed.This is because high-resolution characterization of sEV presents a major challenge,as their sizes are below the optical diffraction limit.Here,we report that upconversion nanoparticles(UCNPs)can be used for super-resolution profiling the molecular heterogeneity of sEVs.We show that Er3+-doped UCNPs has better brightness and Tm3+-doped UCNPs resulting in better resolution beyond diffraction limit.Through an orthogonal experimental design,the specific targeting of UCNPs to the tumour epitope on single EV has been cross validated,resulting in the Pearson’s R-value of 0.83 for large EVs and~65%co-localization double-positive spots for sEVs.Furthermore,super-resolution nanoscopy can distinguish adjacent UCNPs on single sEV with a resolution of as high as 41.9 nm.When decreasing the size of UCNPs from 40 to 27 nm and 18 nm,we observed that the maximum UCNPs number on single sEV increased from 3 to 9 and 21,respectively.This work suggests the great potentials of UCNPs approach“digitally”quantify the surface antigens on single EVs,therefore providing a solution to monitor the EV heterogeneity changes along with the tumour progression progress.Guan Huang Yongtao Liu Dejiang Wang Ying Zhu Shihui Wen Juanfang Ruan Dayong Jin 2022eLight2022,2,1:2

9Numerical prediction of thermo-hydrodynamics on the gas-lubricated film in wedge-shaped microchannel显示文摘One wedge-shaped microchannel was established,and the hydrodynamic properties of the wedge-shaped gas film were comprehensively investigated.The Navier-Stokes equations coupled with the full energy equation were adopted to mainly analyze the lubrication hydrodynamics of the gas film,as the horizontal plate was viewed as the adiabatic wall or the horizontal plate temperature was equal to the tilt plate temperature.A higher gas film temperature strengthened the rarefaction effect,and the more rarefied gas weakened the squeeze.Meanwhile,the vertical flow across the gas film could indicate the relation between the velocity vector and the gas film squeeze and expansion.The adiabatic horizontal plate could resist the heat conduction and enhance the rarefaction effect,and thus the direction of motion of the gas molecules was easier to be changed.ZHANG Xueqing ZHANG Yangang CHEN Qinghua LIU Juanfang 2019排灌机械工程学报2019,37,8:0
10The Effect of Send/Receive Dual Channel Parameters on Polarization Parameters Measurement显示文摘The dual-linear polarization radar is what China will popularize gradually following the Doppler radar technology.Following the Doppler radar technology,the dual-linear polarization radar,which has been widely applied to enhancing modem weather radar system,becomes more and more popular in China.Microphysical characteristics of hydrometeors,such as differential reflectivity,specific differential phase,and zero-lag correlation coefficient,are usually provided by the radar.Brought by the angular orientation of the feed,drop canting,and backscatter depolarization,however,inevitable measurement errors exit because of the unwanted horizontal and vertical wave coupling.Since small errors in these sensitive parameters may lead to erroneous judgment,both the factors themselves and their weights of influence should be studied specifically,with the mode of simultaneous transmission and reception of horizontally and vertically polarized waves.The study provides theoretical basis for the design of dual polarization Doppler radar,and will be helpful in the research of cloud,precipitation physics,and weather modification.ZHANG Wenwen YIN Fulian CHAI Juanfang BAI Xuesong 2017Chinese Journal of Electronics2017,26,2:0
11Studies on the Correlation between Germination and cAMP Content of Different Color Chlamydospores in Ustiloginoidea virens显示文摘Ustilaginoidea virens is the causal agent of a serious disease of rice. To reveal the relationship between germination and the 3'-5'-cyclic adenosine monophosphate(c AMP) content from the dormant(black or green-black) and non-dormant(yellow)chlamydospore in Ustiloginoidea virens,this study adopts ultrasonic-bath method and high-performance liquid chromatography(HPLC) method, for extraction c AMP content of the different color chlamydospore. The results demonstrated that, as for the freshly chlamydospores collected from the false smut balls during their germination, c AMP content of yellow chlamydospore appeared a slight growth during 0-12 h, and showed a rapid declining as the germination time extended(12-48 h). Above all, in yellow chlamydospores, the germination rate and the content of c AMP presented a very notably negative correlation(|r|=0.861 9>r0.01=0.834), but the correlation between germination rate and content of c AMP was not obvious in black chlamydospores. 12An Ensemble Kalman Filter Approach Based on Level Set Parameterization for Acoustic Source Identification Using Multiple Frequency Information显示文摘In this paper,a reconstruction problem of the spatial dependent acoustic source from multiple frequency data is discussed.Suppose that the source function is supported on a bounded domain and the piecewise constant intensities of the source are known on the support.We characterize unknown domain by the level set technique.And the level set function can be modeled by a Hamilton-Jacobi system.We use the ensemble Kalman filter approach to analyze the system state.This method can avoid to deal with the nonlinearity directly and reduce the computation complexity.In addition,the algorithm can achieve the stable state quickly with the Hamilton-Jacobi system.From some numerical examples,we show these advantages and verify the feasibility and effectiveness.Xiaomei Yang Zhiliang Deng Juanfang Wang 2020Communications in Mathematical Research2020,36,2:0
