| 3 | Overexpression of microRNA-124 promotes the neuronal differentiation of bone marrow-derived mesenchymal stem cells显示文摘microRNAs(miRNAs) play an important regulatory role in the self-renewal and differentiation of stem cells. In this study, we examined the effects of miRNA-124(miR-124) overexpression in bone marrow-derived mesenchymal stem cells. In particular, we focused on the effect of overexpression on the differentiation of bone marrow-derived mesenchymal stem cells into neurons. First, we used GeneChip technology to analyze the expression of miRNAs in bone marrow-derived mesenchymal stem cells, neural stem cells and neurons. miR-124 expression was substantially reduced in bone marrow-derived mesenchymal stem cells compared with the other cell types. We constructed a lentiviral vector overexpressing miR-124 and transfected it into bone marrow-derived mesenchymal stem cells. Intracellular expression levels of the neuronal early markers β-III tubulin and microtubule-associated protein-2 were significantly increased, and apoptosis induced by oxygen and glucose deprivation was reduced in transfected cells. After miR-124-transfected bone marrow-derived mesenchymal stem cells were transplanted into the injured rat spinal cord, a large number of cells positive for the neuronal marker neurofilament-200 were observed in the transplanted region. The Basso-Beattie-Bresnahan locomotion scores showed that the motor function of the hind limb of rats with spinal cord injury was substantially improved. These results suggest that miR-124 plays an important role in the differentiation of bone marrow-derived mesenchymal stem cells into neurons. | 4 | Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. | 5 | Identification of a marine agarolytic bacterium Agarivorans albus QM38 and cloning and sequencing its beta-agarase genes显示文摘A total of 117 agar-decomposing cultures were isolated from coastal seawater around Qingdao,China.The phenotypic and agarolytic features of an agarolytic isolate,QM38,were investigated.The strain was gram negative,strictly aerobic,curved rod and polar flagellum.On the basis of several phenotypic characters,biochemical and morphological characters and phylogenetic analysis of the gene coding for the 16S rRNA,the strain was identified as Agarivorans albus strain QM38.This strain can liquefy the agar on the solid agar plate.An excellular agarase activity was determined in liquid culture.The enzyme exhibited maximal activity at 40℃,pH 7.6.Its activity was greatly affected by different concentrations of agarose.The highest activity 32 U/ml was achieved in the culture supernatant.The hydrolytic product was analyzed by fluorophore-assisted carbohydrate electrophoresis (FACE).After complete hydrolysis of agarose,a series of agaro-oligosaccharides were produced.The main products of the enzymes were oligosaccharides in the degree of polymerization (DP) of 2,4,6 and 8.Three genes agaD01,agaD02 and agaD03,encoding β-agarases,had been cloned from genomic DNA of Agarivorans albus strain QM38.The open reading frame of agaD01,consisted of 2 988 bp,and shared 95.5%-98.9% identity to the β-agarase genes of some strains of Vibrio and Agarivorans.Gene agaD02 comprised 2 868 bp and encoded a 955-amino-acid protein.It showed 97.4% and 98.7% identity to the β-agarase genes of strain Vibrio sp.PO-303 and strain Vibrio sp.JT0107,respectively.Only partial sequence of agaD03 gene has been cloned.It showed 96.5% identity to β-agarase gene (agaB) of Pseudoalteromonas sp.CY24,and shared 96.8% identity to β-agarase-c gene of Vibrio sp.PO-303. | DU Zongjun WANG Jing YANG Lijun CHEN Guanjun | 2011 | Acta Oceanologica Sinica2011,30,1: | 5 |
| 6 | Isolation and diversity analysis of heterotrophic bacteria associated with sea anemones显示文摘A study was undertaken to investigate the heterotrophic bacterial flora associated with the sea anemones.Samples of the sea anemones Anthopleura midori were collected from the coast of Weihai and bacteria were isolated from these samples.Additionally,high numbers of viable bacteria were obtained from the celom wall and surface of anemone,the community of cultivable bacteria was very diverse.As a result of this isolation,60 strains were obtained,56 of them were selected for identification and characterization by 16S rRNA gene sequence analysis and limited phenotypic testing.Among these isolates,16 strains were phylogenetically related to members of the genus P seudoalteromonas and neighboring taxa.Other isolates included members of the genera Colwellia,Vibrio,Acinetobacter,Pseudomonas,Endozoicomonas,Roseovarius,Paracoccus,Loktanella,Leisingera,Sulfitobacter,Bacillus,Staphylococcus,Plantibacter,Microbacterium,Micrococcus,Joostella,Psychroserpens,Cellulophaga,Krokinobacter,Polaribacter and P sychrobacter.Seven potential novel species were found.Among 60 strains,17 of them can produce proteolytic exoenzyme,20 can produce lipolytic exoenzyme.Strain NQ8 has strong antagonistic effects on some Vibrio strains.This study demonstrates that the culturable fraction of bacteria from the sea anemones Anthopleura midori is diverse and appears to possess much potential as a source for the discovery of novel bioactive materials. | DU Zongjun ZHANG Wanyi XIA Hongjie LU Guoqiang CHEN Guanjun | 2010 | Acta Oceanologica Sinica2010,29,2: | 3 |
| 7 | Review of surface transient charge measurement on solid insulating materials via the Pockels technique显示文摘Surface charges accumulated on solid insulating materials in electrical apparatuses and electronic devices could distort the initial electric field and severely damage the insulation performance.Accurate transient surface charge measurement is critical to determine the mechanisms of charge transport and flashover occurrence,and estimate surface insulation strength under high voltage.The measurement technique based on the Pockels electro-optic effect is considered as the only approach for 2-D dynamic charge distribution behaviour so far,and it has good capability to avoid original electric field distortion caused by conven-tional methods featured by metal probes.This paper reviews recent progress of the non-invasive dynamic surface charge measurement systems on the Pockels technique.We focus especially on typical systems with transmitting and reflecting configurations and the frequently used fast inversion algorithms.It can be inferred that higher spatial-temporal resolution,higher signal-to-noise ratio,and better compatibility for both transparent and opaque materials are the future development trends,which greatly depend on multiple technological breakthroughs at impact-resistant electro-optic crystals,advanced high-speed photography and high-performance inversion algorithms.In addition,the recent studies on transient behaviour of surface discharge,together with the present limitation and future prospects are discussed.It is expected that more ideas will be inspired in transient surface charge measurement by the Pockels technique and other relevant electro-optic methods. | Bo Zhang Jianyi Xue Xingyu Chen Sile Chen Haibao Mu Yang Xu Guanjun Zhang | 2021 | High Voltage2021,6,4: | 2 |
| 8 | Tracking and recognition algorithm for a robot harvesting oscillating apples显示文摘Apple fruits on trees tend to swing because of wind or other natural causes,therefore reducing the accuracy of apple picking by robots.To increase the accuracy and to speed up the apple tracking and identifying process,tracking and recognition method combined with an affine transformation was proposed.The method can be divided into three steps.First,the initial image was segmented by Otsu’s thresholding method based on the two times Red minus Green minus Blue(2R-G-B)color feature;after improving the binary image,the apples were recognized with a local parameter adaptive Hough circle transformation method,thus improving the accuracy of recognition and avoiding the long,time-consuming process and excessive fitted circles in traditional Hough circle transformation.The process and results were verified experimentally.Second,the Shi-Tomasi corners detected and extracted from the first frame image were tracked,and the corners with large positive and negative optical flow errors were removed.The affine transformation matrix between the two frames was calculated based on the Random Sampling Consistency algorithm(RANSAC)to correct the scale of the template image and predict the apple positions.Third,the best positions of the target apples within 1.2 times of the prediction area were searched with a de-mean normalized cross-correlation template matching algorithm.The test results showed that the running time of each frame was 25 ms and 130 ms and the tracking error was more than 8%and 20%in the absence of template correction and apple position prediction,respectively.In comparison,the running time of our algorithm was 25 ms,and the tracking error was less than 4%.Therefore,test results indicate that speed and efficiency can be greatly improved by using our method,and this strategy can also provide a reference for tracking and recognizing other oscillatory fruits. | Qinghua Yang Chen Chen Jiayu Dai Yi Xun Guanjun Bao | 2020 | International Journal of Agricultural and Biological Engineering2020,13,5: | 2 |

| 12 | Superpixel-based segmentation algorithm for mature citrus显示文摘With the decrease of agricultural labors and the increase in production costs,harvesting robots have become a research hotspot in recent years.To guide harvesting robots to pick mature citrus more precisely under variable illumination conditions,an image segmentation algorithm based on superpixel was proposed.Efficient simple linear iterative clustering(SLIC)algorithm which takes similarity of adjacent pixels into account was adopted to segment the images captured under variable illumination conditions into superpixels.The color and texture features of these superpixels were extracted and fused into feature vectors as descriptors to train backpropagation neural networks(BPNN)classifier in the next step.The adjacency information of superpixels was considered by calculating the global-local binary pattern(LBP)in R component images when extracting texture features.To accelerate the classification process,the mean of Cr-Cb image was utilized to find superpixels of interest which were regarded as candidates of citrus superpixels.These candidates were then classified by a pre-trained BPNN model with superpixel-level accuracy of 98.77%and pixel-level accuracy of 94.96%,while the average time to segment one image was 0.4778 s.Therefore,the results indicated that a superpixel-based segmentation algorithm toward citrus images had decent light robustness as well as high accuracy that could guide harvesting robot to pick mature citrus efficiently. | Qinghua Yang Yiqin Chen Yi Xun Guanjun Bao | 2020 | International Journal of Agricultural and Biological Engineering2020,13,4: | 1 |

| 16 | Cytoreductive Surgery and Hyperthermic Intraperitoneal Chemotherapy for Pseudomyxoma Peritonei of Appendiceal Origin-801 Cases from a Single Institution in China显示文摘Aim:As more and more centers has published their treatment results of pseudomyxoma peritonei(PMP)with cytoreductive surgery(CRS)and hyperthermic intraperitoneal chemotherapy(HIPEC),the data from China is missing.Myxoma Department of Aerospace Hospital is the biggest center treating PMP in China.The purpose of this study is to report the early and long-term outcomes for PMP from this single center.Methods:801 appendix-derived PMP out of 1008 consecutive patients treated in Myxoma Department of Aerospace Hospital between 2008 and 2019 were retrospectively analyzed.Results:Complete cytoreductive surgery(CCRS)was achieved in 240(30%)patients with median PCI of 14(1~39),and the rest had maximal tumor debulking(MTD),HIPEC was implemented in 96.3%of CCRS and 78.6%of MTD.The major morbidity(grade III/IV)was 11.4%and the 30-day operative mortality is 0.7%.The 5-and 10-year OS of CCRS was 76.9%and 64.1%,which is significantly higher than MTD(5-,10-year OS as 36.1%,27.1%;p<0.001).On the univariate analysis,all prognostic factors(gender,PSS,interval time,prior chemotherapy,prior HIPEC,Peritoneal Cancer Index(PCI),completeness of cytoreduction(CC),HIPEC,pathology,present of serous ascites)were found to be associated with overall survival except for age.On multivariate analysis,only PCI>20,MTD,high pathologic grade and without HIPEC were independent factors predicting poorer prognosis.Conclusions:CCRS+HIPEC can benefit PMP well with controllable risks.MTD+HIPEC may benefit PMP as well when CCRS cannot be achieved after fully asscessment by an experienced peritoneal maglignacy center,but the surgery should be performed as limited as possible. | Ao Xia Xichao Zhai Lubiao An Bing Wang Guanjun Shi Ying Cai Yiyan Lu Shaojun Pang Feng Chen Hongbin Xu Ruiqing Ma | 2020 | Journal of Oncology Research2020,2,2: | 0 |

| 18 | Thermofluidic investigations of oil natural transformer:Closed-loop modelling and experimental validation显示文摘Excessively high temperatures inside a power transformer can accelerate the ageing rate and even cause unplanned outages.Accurately estimating the hot-spot temperature(HST)of a power transformer is important for regulating the load and ensuring long-term operation.In this study,a closed-loop model of a 110-kV product transformer with an oil natural/air forced(ONAF)cooling mode is built to predict HST,in which the compression ratio of windings,equivalent heat source and radiator geometry are considered.According to the flow field of the winding,the‘dead oil zone’and reverse oil flow are observed in the horizontal oil channels.The accuracy of the proposed model is experimentally validated by temperature-rise tests.The results show that the absolute errors between the measured results and simulation are below 5°C,and the average error is~2°C.Furthermore,the arrangements of windings and radiators are studied to enhance the oil circulation.By adding 5 washers to the high-voltage winding,the average temperature can be decreased by 6°C compared with the case without a washer.Elevating the radiator can also mitigate the characteristic temperatures,and the optimal height difference between the radiator and winding centre is suggested to be 0.66 m. | Yuan Li Yiran Gao Chen Wang Chao Zhu Ying Li Yuankai Wang Guanjun Zhang | 2024 | High Voltage2024,9,1: | 0 |
