2YTH Domain: A Family of N^6-methyladenosine (m^6A) Readers显示文摘Like protein and DNA, different types of RNA molecules undergo various modifications. Accumulating evidence suggests that these RNA modifications serve as sophisticated codes to mediate RNA behaviors and many important biological functions. N^6-methyladenosine(m^6 A)is the most abundant internal RNA modification found in a variety of eukaryotic RNAs, including but not limited to m RNAs, t RNAs, r RNAs, and long non-coding RNAs(lnc RNAs). In mammalian cells, m^6 A can be incorporated by a methyltransferase complex and removed by demethylases, which ensures that the m^6 A modification is reversible and dynamic. Moreover, m^6 A is recognized by the YT521-B homology(YTH) domain-containing proteins, which subsequently direct different complexes to regulate RNA signaling pathways, such as RNA metabolism, RNA splicing, RNA folding, and protein translation. 3Current developments in pharmacological therapeutics for chronic constipation显示文摘Chronic constipation is a common gastrointestinal disease severely affecting the patient's quality of life. The traditional treatment of constipation is the use of laxatives. Recently, several new drugs including lubiprostone, linaclotide and prucalopride have been approved for treatment of chronic constipation. However, a significant unmet medical need still remains, particularly among those patients achieving poor results by current therapies. The 5-HT4 receptor modulators velusetrag and naronapride,the guanylate cyclase C agonist plecanatide and the ileal bile acid transporter inhibitor elobixibat are recognized as the most promising drugs under investigation. 5Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 6Influence of plant architecture on maize physiology and yield in the Heilonggang River valley显示文摘The size and distribution of leaf area determine light interception in a crop canopy and influence overall photosynthesis and yield. Optimized plant architecture renders modern maize hybrids(Zea mays L.) more productive, owing to their tolerance of high plant densities. To determine physiological and yield response to maize plant architecture, a field experiment was conducted in 2010 and 2011. With the modern maize hybrid ZD958, three plant architectures, namely triangle, diamond and original plants, were included at two plant densities, 60,000 and 90,000 plants ha-1. Triangle and diamond plants were derived from the original plant by spraying the chemical regulator Jindele(active ingredients,ethephon, and cycocel) at different vegetative stages. To assess the effects of plant architecture, a light interception model was developed. Plant height, ear height, leaf size,and leaf orientation of the two regulated plant architectures were significantly reduced or altered compared with those of the original plants. On average across both plant densities and years, the original plants showed higher yield than the triangle and diamond plants,probably because of larger leaf area. The two-year mean grain yield of the original and diamond plants were almost the same at 90,000 plants ha-1(8714 vs. 8798 kg ha-1). The yield increase(up to 5%) of the diamonds plant at high plant densities was a result of increased kernel number per ear, which was likely a consequence of improved plant architecture in the top and middle canopy layers. The optimized light distribution within the canopy can delay leaf senescence, especially for triangle plants. The fraction of incident radiation simulated by the interception model successfully reflected plant architecture traits. Integration of canopy openness is expected to increase the simulation accuracy of the present model. 9The effect of low-molecular-weight organic-acids(LMWOAs)on treatment of chromium-contaminated soils by compost-phytoremediation: Kinetics of the chromium release and fractionation显示文摘A soil–plant biological system was developed from chromium(Cr) polluted soil treated by the compost-phytoremediation method. The transformation and migration of the Cr in this system is comprehensively studied in this research. The results illustrated that the co-composting treatment can reduce the Cr availability from 39%(F1 was about 31% of total, F2 was about 8% of total) to less than 2% by stabilizing the Cr. However, herbaceous plants can accumulate the concentrations of Cr from 113.8 to 265.2 mg/kg in the two crops,even though the concentration of soluble Cr in the substrate soil was below 0.1 mg/L.Cr can be assimilated and easily transferred in the tissues of plants because the lowmolecular-weight organic-acids(LMWOAs) derived from the plant root increase the bioavailability of Cr. The amount of extracted Cr dramatically increased when the organic acids were substituted in this order: citric acid > malic acid > tartaric acid > oxalic acid >acetic acid. On average the maximum(147.4 mg/kg) and the minimum(78.75 mg/kg) Cr were extracted by 20 mmol/L citric acid and acetic acid, respectively. The desorption of Cr in different acid solutions can be predicted by the pseudo second-order kinetics. The exchangeable Cr, carbonate-bound Cr, and residual Cr decreased, while Fe–Mn oxide bound Cr and organic bound Cr increased in the soil solid phase. Internal failure is observed as one of the most serious factors, including loss of electrode materials, structure deformation and dendrite growth. It usually incubates from atomic/molecular level and progressively aggravates along with lithiation. Understanding the internal failure is of great importance for developing solutions of failure prevention and advanced anode materials. In this research, different internal failure processes of anode materials for lithium batteries are discussed. 16Non-equilibrium Plasma Assisted Combustion of Low Heating Value Fuels显示文摘This paper describes the effects of non-equilibrium air plasma generated by a dielectric barrier discharge (DBD) on the combustion of low heating value fuels. The experimental results indicate that addition of a very small amount of energy to the air flow in the form of DBD significantly improves the flame stability. Moreover, main combustion characteristics such as flame propagation speed, combustion intensity and lean blow-off limits are also enhanced by the effect of plasma. Some active radicals such as excited O atom and excited N2 molecule are observed by spectrograph in the discharge area. 17Engineering an enthesis-like graft for rotator cuff repair:An approach to fabricate highly biomimetic scaffold capable of zone-specifically releasing stem cell differentiation inducers显示文摘Rotator cuff(RC)attaches to humerus across a triphasic yet continuous tissue zones(bone-fibrocartilage-tendon),termed“enthesis”.Regrettably,rapid and functional enthesis regeneration is challenging after RC tear.The existing grafts bioengineered for RC repair are insufficient,as they were engineered by a scaffold that did not mimic normal enthesis in morphology,composition,and tensile property,meanwhile cannot simultaneously stimulate the formation of bone-fibrocartilage-tendon tissues.Herein,an optimized decellularization approach based on a vacuum aspiration device(VAD)was developed to fabricate a book-shaped decellularized enthesis matrix(O-BDEM).Then,three recombinant growth factors(CBP-GFs)capable of binding collagen were synthesized by fusing a collagen-binding peptide(CBP)into the N-terminal of BMP-2,TGF-β3,or GDF-7,and zone-specifically tethered to the collagen of O-BDEM to fabricate a novel scaffold(CBP-GFs/O-BDEM)satisfying the above-mentioned requirements.After ensuring the low immunogenicity of CBP-GFs/O-BDEM by a novel single-cell mass cytometry in a mouse model,we interleaved urine-derived stem cell-sheets into this CBP-GFs/O-BDEM to bioengineer an enthesis-like graft.Its high-performance on regenerating enthesis was determined in a canine model.These findings indicate this CBP-GFs/O-BDEM may be an excellent scaffold for constructing enthesis-like graft to patch large/massive RC tears,and provide breakthroughs in fabricating graded interfacial tissue.Can Chen Qiang Shi Muzhi Li Yang Chen Tao Zhang Yan Xu Yunjie Liao Shulin Ding Zhanwen Wang Xing Li Chunfeng Zhao Lunquan Sun Jianzhong Hu Hongbin Lu 2022Bioactive Materials2022,7,10:4

19Extended Range(10–30 Days) Heavy Rain Forecasting Study Based on a Nonlinear Cross-Prediction Error Model显示文摘Extended range(10–30 d) heavy rain forecasting is difficult but performs an important function in disaster prevention and mitigation. In this paper,a nonlinear cross prediction error(NCPE) algorithm that combines nonlinear dynamics and statistical methods is proposed. The method is based on phase space reconstruction of chaotic single-variable time series of precipitable water and is tested in 100 global cases of heavy rain. First,nonlinear relative dynamic error for local attractor pairs is calculated at different stages of the heavy rain process,after which the local change characteristics of the attractors are analyzed. Second,the eigen-peak is defined as a prediction indicator based on an error threshold of about 1.5,and is then used to analyze the forecasting validity period. The results reveal that the prediction indicator features regarded as eigenpeaks for heavy rain extreme weather are all reflected consistently,without failure,based on the NCPE model; the prediction validity periods for 1–2 d,3–9 d and 10–30 d are 4,22 and 74 cases,respectively,without false alarm or omission. The NCPE model developed allows accurate forecasting of heavy rain over an extended range of 10–30 d and has the potential to be used to explore the mechanisms involved in the development of heavy rain according to a segmentation scale. 20PhaseⅡstudy of induction chemotherapy followed by concurrent chemoradiotherapy with raltitrexed and cisplatin in locally advanced nasopharyngeal carcinoma显示文摘Objective:For locally advanced nasopharyngeal carcinoma(LA-NPC)patients,high incidences of distant metastases and severe treatment related toxicities are the main obstacles needed to be overcome.Raltitrexed,a specific thymidylate synthase inhibitor with a convenient administration schedule,has an acceptable and manageable toxicity,and possesses radio-sensitizing properties.To investigate the efficacy and safety of raltitrexed and cisplatin induction chemotherapy and concurrent chemoradiotherapy(IC+CCRT)in patients with LA-NPC,a phaseⅡclinical study was conducted.Methods:Sixty eligible patients with LA-NPC were enrolled into this study.A raltitrexed-cisplatin combination was used as part of an IC+CCRT regimen.Raltitrexed-cisplatin IC was given once every 3 weeks(q3 w)for two cycles,followed by raltitrexed-cisplatin based CCRT q3 w for two cycles.Intensity-modulated radiotherapy(IMRT)was given for all enrolled patients.Results:All patients were included in survival analysis according to the intent-to-treat principle.The objective response rate(ORR)3 months after treatment was 98%.The 2-year overall survival(OS)rate was 92%.The median relapse-free survival(RFS)time was 30.5[95%confidence interval(95%CI),28.4-32.3]months.The 2-year RFS rate was 85%.The 2-year local failure-free survival(LFFS)rate was 97%and the 2-year distant metastasis-free survival(DMFS)rate was 88%.Acute toxicities were mostly grade 2 and 3 reactions in bone marrow suppression,gastrointestinal side effect and oropharyngeal mucositis.Only two patients occurred grade 4 acute toxicities,one was bone marrow suppression and the other was dermatitis radiation.Conclusions:The combination of raltitrexed and cisplatin has a comparable efficacy to those in standard firstline therapy.Yuan Wu Xueyan Wei Zilong Yuan Hongbin Xu Yanping Li Ying Li Liu Hu Guang Han Yu Qian Desheng Hu 2020Chinese Journal of Cancer Research2020,32,5:4
