1High-efficiency and multiplex adenine base editing in plants using new TadA variants显示文摘Recently reported adenine base editors(ABEs)exhibit powerful potential for targeted gene correction as well as developing gain-of-function mutants and novel germplasms for both gene function studies and crop breeding.However,editing efficiency varies significantly among different target sites.Here,we investigated the activities of three evolved E.coli adenosine deaminase TadA variants(TadA8e,TadA8.17,and TadA8.20)side-by-side in transgenic rice.We found that TadA8e outperforms TadA8.17 and TadA8.20,and induces efficient A-to-G conversion at all tested sites in the rice genome,including those that were un-editable by ABE7.10 in our previous experiments.Furthermore,V82S/Q154R mutations were incorporated into TadA8e,resulting in a new variant that we named TadA9.Our data show that TadA9 is broadly compatible with CRISPR/SpCas9,CRISPR/SpCas9-NG,and CRISPR/SpRY,as well as CRISPR/ScCas9 nickase systems,achieving comparable or enhanced editing in a larger editing window at diverse PAM sites as compared with TadA8e.Finally,TadA9 was used to simultaneously install novel SNPs in four endogenous herbicide target genes in the commercial rice cultivar Nangeng 46 for potential field application in.weed control.Collectively,we successfully generated a series of novel ABEs that can efficiently edit adenosines in the rice genome.Our findings suggest that TadA9 and TadA8e have great potentials in the development of plant base editors and crop molecular breeding.Daqi Yan Bin Ren Lang Liu Fang Yan Shaofang Li Guirong Wang Wenxian Sun Xueping Zhou Huanbin Zhou 2021Molecular Plant2021,14,5:8
2A nonlinear POD reduced order model for limit cycle oscillation prediction显示文摘As the amplitude of the unsteady flow oscillation is large or large changes occur in the mean background flow such as limit cycle oscillation,the traditional proper orthogonal decomposition reduced order model based on linearized time or frequency domain small disturbance solvers can not capture the main nonlinear features.A new nonlinear reduced order model based on the dynamically nonlinear flow equation was investigated.The nonlinear second order snapshot equation in the time domain for proper orthogonal decomposition basis construction was obtained from the Taylor series expansion of the flow solver.The NLR 7301 airfoil configuration and Goland+ wing/store aeroelastic model were used to validate the capability and efficiency of the new nonlinear reduced order model.The simulation results indicate that the proposed new reduced order model can capture the limit cycle oscillation of aeroelastic system very well,while the traditional proper orthogonal decomposition reduced order model will lose effectiveness.CHEN Gang *,LI YueMing & YAN GuiRong MOE Key Laboratory for Strength & Vibration,College of Aerospace,Xi’an Jiaotong University,Xi’an 710049,China 2010Science China(Physics,Mechanics & Astronomy)2010,53,7:7
3Molecular breeding of water lily: engineering cold stress tolerance into tropical water lily显示文摘Water lilies(order Nymphaeales)are rich in both economic and cultural values.They grow into aquatic herbs,and are divided into two ecological types:tropical and hardy.Although tropical water lilies have more ornamental and medicinal values compared to the hardy water lily,the study and utilization of tropical water lilies in both landscaping and pharmaceutical use is greatly hindered due to their limited planting area.Tropical water lilies cannot survive the winter in areas beyond 24.3°N latitude.Here,the transgenic pipeline through the pollen-tube pathway was generated for water lily for the first time.To improve cold stress tolerance of tropical water lilies,a gene encoding choline oxidase(CodA)driven by a cold stress-inducible promoter was transformed into a tropical water lily through the pollen-tube transformation.Six independent transgenic lines were tested for survival rate during two winter seasons from 2015 to 2017 in Hangzhou(30.3°N latitude).PCR and southern blot detection revealed that the CodA gene had been integrated into the genome.Reverse transcription PCR showed that CodA gene was induced after cold stress treatment,and further quantitative real-time PCR revealed different expressions among six 4 lines and line 3 had the highest expression.Multiple physiological experiments showed that after cold stress treatment,both the conductivity and malondialdehyde(MDA)levels from transgenic plants were significantly lower than those of non-transgenic plants,whereas the content of betaine and the activity of superoxide dismutase,catalase,and peroxidase were higher than those from non-transgenic plants.These results suggest that expression of exogenous CodA gene significantly improved the cold stress tolerance of tropical water lilies through a wide range of physiological alterations.Our results currently expanded a six-latitude cultivating area of the tropical water lilies.These results not only illuminate the bright future for water lily breeding but will also facilitate the functional genomic studies.Cuiwei Yu Guirong Qiao Wenmin Qiu Dongbei Yu Shirong Zhou Yan Shen Guanchun Yu Jing Jiang Xiaojiao Han Mingying Liu Liangsheng Zhang Fei Chen Yuchu Chen Renying Zhuo 2018Horticulture Research2018,5,1:4
4Urban-rural disparities in mild cognitive impairment and its functional subtypes among community- dwelling older residents in central China显示文摘background Substantial variations in the prevalence of mild cognitive impairment(MCI)and its subtypes have been reported,although mostly in geographically defined developed countries and regions.Less is known about MCI and its subtypes in rural areas of less developed central China.Aims The study aimed to compare the prevalence of MCI and its subtypes in residents aged 65 years or older in urban and rural areas of Hubei Province,China.Methods Participants aged 65 years or older were recruited between 2018 and 2019.Inperson structured interviews and clinical and neuropsychological assessments were performed at city health community centres and township hospitals.results Among 2644 participants without dementia,735 had MCI,resulting in a prevalence of 27.8%for total MCI,20.9%for amnestic MCI(aMCI)and 6.9%for non-amnestic MCI(naMCI).The prevalence of MCI in urban and rural areas was 20.2%and 44.1%,respectively.After adjusting for demographic factors,the prevalence of total MCI,aMCI and naMCI differed significantly between rural and urban areas(adjusted odds ratio(OR)2.10,1.44 and 3.76,respectively).Subgroup analysis revealed an association between rural socioeconomic and lifestyle disadvantage and MCI and its subtypes.Conclusions Our findings suggest that the prevalence of MCI among urban residents in central China is consistent with that in other metropolis areas,such as Shanghai,but the prevalence in rural areas is twice that in urban areas.Prospective studies and dementia prevention in China should focus on rural areas.Dan Liu Lin Li Lina An Guirong Cheng Cong Chen Mingjun Zou Bo Zhang Xuguang Gan Lang Xu Yangming Ou Qingming Wu Ru Wang Yan Zeng 2021General Psychiatry2021,34,5:3
5Active Control Law Design for Flutter/LCO Suppression Based on Reduced Order Model Method显示文摘Active stability augmentation system is an attractive and promising technology to suppress flutter and limit cycle oscillation(LCO).In order to design a good active control law,the control plant model with low order and high accuracy must be provided,which is one of the most important key points.The traditional model is based on low fidelity aerodynamics model such as panel method,which is unsuitable for transonic flight regime.The physics-based high fidelity tools,reduced order model(ROM) and CFD/CSD coupled aeroservoelastic solver are used to design the active control law.The Volterra/ROM is applied to constructing the low order state space model for the nonlinear unsteady aerodynamics and static output feedback method is used to active control law design.The detail of the new method is demonstrated by the Goland+ wing/store system.The simulation results show that the effectiveness of the designed active augmentation system,which can suppress the flutter and LCO successfully.Chen Gang Li Yueming Yan Guirong 2010Chinese Journal of Aeronautics2010,23,6:3
6Surface Tension Calculation of Undercooled Alloys显示文摘Based on the Butler equation and extrapolated thermodynamic data of undercooled alloys from those of liquid stable alloys, a method for surface tension calculation of undercooled alloys is proposed. The surface tensions of liquid stable and undercooled Ni-Cu (xNi=0.42) and Ni-Fe (xNi=0.3 and 0.7) alloys are calculated using STCBE (Surface Tension Calculation based on Butler Equation) program. 7Calibration of the solar radio spectrometer显示文摘This paper shows some improvements and new results of calibration of Chinese solar radio spectrometer by analyzing the daily calibration data recorded in the period of 1997-2007. First, the calibration coefficient is fitted for three bands (1.0-2.0 GHz, 2.6-3.8 GHz, 5.2-7.6 GHz) of the spectrometer by using the moving-average method confined by the property of the daily calibration data. By this calibration coefficient, the standard deviation of the calibration result was less than 10 sfu for 95% frequencies of 2.6-3.8 GHz band in 2003. This result is better than that calibrated with the constant coefficient. Second, the calibration coefficient is found in good correlation with local air temperature for most frequencies of 2.6-3.8 GHz band. 8Effects of high modes on the wind-induced response of super high-rise buildings显示文摘For super high-rise buildings,the vibration period of the basic mode is several seconds,and it is very close to the period of the fluctuating wind.The damping of super high-rise buildings is low,so super high-rise buildings are very sensitive to fluctuating wind.The wind load is one of the key loads in the design of super high-rise buildings.It is known that only the basic mode is needed in the wind-response analysis of tall buildings.However,for super high-rise buildings,especially for the acceleration response,because of the frequency amplification of the high modes,the high modes and the mode coupling may need to be considered.Three typical super high-rise projects with the SMPSS in wind tunnel tests and the random vibration theory method were used to analyze the effect of high modes on the wind-induced response.The conclusions can be drawn as follows.First,for the displacement response,the basic mode is dominant,and the high modes can be neglected.Second,for the acceleration response,the high modes and the mode coupling should be considered.Lastly,the strain energy of modes can only give the vibration energy distribution of the high-rise building,and it cannot describe the local wind-induced vibration of high-rise buildings,especially for the top acceleration response.Feng Ruoqiang Guirong Yan Ge Jinming 2012Earthquake Engineering and Engineering Vibration2012,11,3:2
