| 2 | Hepatitis C virus infection of human hepatoma cell line 7721 in vitro显示文摘AIM To establish a cell culture system with long-term replication of hepatitis C virus in vitro.``METHODS Human hepatoma cell line 7721 was tested for its susceptibility to HCV by incubating with a serum from a patient with chronic hepatitis C. Cells and supernatant were harvested at various phases during the culturing periods The presence of HCV RNA, the expression of HCV antigens in cells and/or supernatant were examined by RT-PCR, in situ hybridization and immunohistochemistry respectively.``RESULTS The intracellular HCV RNA was first detected on d 2 after infection and then could be intermittently detected in both cells and supernatant over a period of at least three months. The expression of HCV NS3, CP10antigens could be observed in cells. | 6 | Mature Archean continental crust in the Yangtze craton: Evidence from petrology, geochronology and geochemistry显示文摘Previous studies have shown that the Archean basement is widely distributed throughout the Yangtze craton. To date, however, Archean basement terrains have not been found, except for a few Kongling high-grade metamorphic terrains in the Huangling dome that have been confirmed to be of Archean age. To further understand the basement component and crustal evolution of the Yangtze craton, we carried out a petrological, geochronological and geochemical study of the Jinshan K-feldspar granite emplaced within the Yangpo Group, located in Huji Town, Zhongxiang City, Hubei Province. Results indicate that the zircon SHRIMP U-Pb age of the Jinshan granite is 2655±9 Ma, placing it within the middle Neoarchean. Chemically, this pluton yields abundant silica and alkalis and is depleted in Ca, Mg and Ti. Furthermore, it is enriched in Rb, Th, Ga, Y and Zr, depleted in Sr, Ba, Nb and Ta, and especially lacking in Eu. High ratios of FeO*/MgO (32.0 to 58.7) and 104 ×Ga/Al (3.19 to 3.41) were also found. The pluton exhibits characteristics typical of A-type granites with crustal source magmas. Moreover, the meta-sedimentary rock association of the Yangpo Group, into which the pluton intruded, clearly shows relatively stable depositional environments of a shallow shelf sequence. Therefore, before the middle Neoarchean, the Yangtze craton contained mature continental crust. | 7 | Clinical application of microRNA in gastric cancer in Eastern Asian area显示文摘Recent research has shown that microRNA (miRNA), which is involved in almost every step of gastric carcinogenesis, has broad prospective application in diagnosis and therapy of gastric carcinoma. Eastern Asia (South Korea, Japan and China) has the highest incidence of gastric cancer in the world. There were 988 000 new cases of gastric cancer worldwide and 736 000 deaths in 2008. Approximately 60% of the cases of gastric cancer are found in East Asia (mainly China). We herein provide a brief review of the clinical applications of miRNA, which include the following aspects: (1) miRNA may serve as a potential new generation of tumor markers; (2) a complete miRNA expression profile is highly specific, can reflect the evolutionary lineage and differentiation of tumors, and be used to carry out diversity analysis; (3) detecting specific miRNA expression in peripheral blood will become a new method for diagnosis of gastric cancer; (4) miRNA can predict prognosis of gastric cancer; (5) miRNA has predictive value in determining chemotherapy and radiotherapy resistance; and (6) miRNA could be a type of innovative drug. | 8 | Bone marrow mesenchymal stem cells repair spinal cord ischemia/reperfusion injury by promoting axonal growth and anti-autophagy显示文摘Bone marrow mesenchymal stem cells can differentiate into neurons and astrocytes after transplantation in the spinal cord of rats with ischemia/reperfusion injury. Although bone marrow mesenchymal stem cells are known to protect against spinal cord ischemia/reperfusion injury through anti-apoptotic effects, the precise mechanisms remain unclear. In the present study, bone marrow mesenchymal stem cells were cultured and proliferated, then transplanted into rats with ischemia/reperfusion injury via retro-orbital injection. Immunohistochemistry and immunofluorescence with subsequent quantification revealed that the expression of the axonal regeneration marker, growth associated protein-43, and the neuronal marker, microtubule-associated protein 2, significantly increased in rats with bone marrow mesenchymal stem cell transplantation compared with those in rats with spinal cord ischemia/reperfusion injury. Furthermore, the expression of the autophagy marker, microtubule-associated protein light chain 3B, and Beclin 1, was significantly reduced in rats with the bone marrow mesenchymal stem cell transplantation compared with those in rats with spinal cord ischemia/reperfusion injury. Western blot analysis showed that the expression of growth associated protein-43 and neurofilament-H increased but light chain 3B and Beclin 1 decreased in rats with the bone marrow mesenchymal stem cell transplantation. Our results therefore suggest that bone marrow mesenchymal stem cell transplantation promotes neurite growth and regeneration and prevents autophagy. | 10 | Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold. | 13 | Reproductive traits associated with invasiveness in Conyza sumatrensis显示文摘Conyza sumatrensis (Retz.) E. Walker, a member of Asteraceae, is a highly invasive species. However, its reproduction biology remains poorly known. To understand the role of reproductive traits in successful invasion of the species, we studied several traits of its reproductive system: the miniature capitulum and gynomonoecious sexual system, the biology and phenology of capitula and florets, pollen/ovule ratio, the mating system (self-compatibility), flower visitors, physical traits and dispersal potential of achenes, germination potential of achenes from manually pollinated capitula, and the association of these traits with invasiveness. Our study showed that the reproductive traits of autonomous seed production, versatile mating system of self- and cross-pollination, and generalized pollination system might contribute to the species' successful invasive capability. The invasiveness was further enhanced by the high and rapid production of achenes, as well as the high percentage, rapid germination rate and high dispersal capability of achenes. | 14 | Seroepidemiological Investigation of Lyme Disease and Human Granulocytic Anaplasmosis among People Living in Forest Areas of Eight Provinces in China显示文摘Objective Lyme disease and Human granulocytic anaplasmosis are tick‐borne diseases caused by Borrelia burgdorferi and Anaplasma phagocytophilum respectively. We have investigated infection and co-infection of the two diseases in the population of forest areas of eight provinces in China by measuring seroprevalence of antibodies against B. burgdorferi and A. phagocytophilum. Methods Forest areas in 8 provinces were chosen for investigation using whole sampling and questionnaire survey methods. 3 669 serum samples from people in the forest areas were tested for the presence of antibodies by indirect immunofluorescent assay (IFA). Results Seroprevalence against B. burgdorferi was 3% to 15% and against A. phagocytophilum was 2% to 18% in the study sites in the 8 provinces in China. We also found co-infection of B. burgdorferi and A. phagocytophilum in 7 of the 8 provinces (the exception being the Miyun area in Beijing). The seroprevalence for both B. burgdorferi and A. phagocytophilum was significantly higher among people exposed to ticks than among people who were not exposed to ticks. | 15 | Autophagy inhibition enhances apigenin-induced apoptosis in human breast cancer cells显示文摘Apigenin (4',5,7-trihydroxyflavone) is a member of the flavone subclass of flavonoids present in fruits and vegetables. The involvement of autophagy in the apigenin-induced apoptotic death of human breast cancer cells was investigated. Cell proliferation and viability were assessed by 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) and clonogenic assays. Flow cytometry, fluorescent staining and Western blot analysis were employed to detect apoptosis and autophagy, and the role of autophagy was assessed using autophagy inhibitors. Apigenin dose- and time-dependently repressed the proliferation and clonogenic survival of the human breast cancer T47D and MDA-MB-231 cell lines. The death of T47D and MDA-MB-231 cells was due to apoptosis associated with increased levels of Caspase3, PARP cleavage and Bax/Bcl-2 ratios. The results from flow cytometry and fluorescent staining also verified the occurrence of apoptosis. In addition, the apigenin-treated cells exhibited autophagy, as characterized by the appearance of autophagosomes under fluorescence microscopy and the accumulation of acidic vesicular organelles (AVOs) by flow cytometry. Furthermore, the results of the Western blot analysis revealed that the level of LC3-II, the processed form of LC3-I, was increased. Treatment with the autophagy inhibitor, 3-methyladenine (3-MA), significantly enhanced the apoptosis induced by apigenin, which was accompanied by an increase in the level of PARP cleavage. Similar results were also confirmed by flow cytometry and fluorescence microscopy. These results indicate that apigenin has apoptosis-and autophagy-inducing effects in breast cancer cells. | 16 | GID1 modulates stomatal response and submergence tolerance involving abscisic acid and gibberellic acid signaling in rice显示文摘Plant responses to abiotic stresses are coordinated by arrays of growth and developmental programs.Gibberellic acid(CA) and abscisic acid(ABA) play critical roles in the developmental programs and environmental responses,respectively,through complex signaling and metabolism networks.However,crosstalk between the two phytohormones in stress responses remains largely unknown.In this study,we report that CIBBERELLIN-INSENSITIVE DWARF 1(GID1),a soluble receptor for GA,regulates stomatal development and patterning in rice(Oryza sativa L.).The gid1 mutant showed impaired biosynthesis of endogenous ABA under drought stress conditions,but it exhibited enhanced sensitivity to exogenous ABA.Scanning electron microscope and infrared thermal image analysis indicated an increase in the stomatal conductance in the gid1 mutant under drought conditions.Interestingly,the gid1 mutant had increased levels of chlorophyll and carbohydrates under submergence conditions,and showed enhanced reactive oxygen species(ROS)-scavenging ability and submergence tolerance compared with the wild-type.Further analyses suggested that the function of GID1 in submergence responses is partially dependent on ABA,and GA signaling by GID1 is involved in submergence tolerance by modulating carbohydrate consumption.Taken together,these findings suggest GID1 plays distinct roles in stomatal response and submergence tolerance through both the ABA and GA signaling pathways in rice. | Hao Du Yu Chang Fei Huang Lizhong Xiong | 2015 | Journal of Integrative Plant Biology2015,57,11: | 12 |
| 17 | The calcium-dependent kinase OsCPK24 functions in cold stress responses in rice显示文摘Calcium-dependent protein kinases(CPKs)are serine/threonine protein kinases that function in plant stress responses. Although CPKs are recognized as key messengers in signal transduction, the specific roles of CPKs and the molecular mechanisms underlying their activity remain largely unknown. Here, we characterized the function of Os CPK_(24), a cytosol-localized calciumdependent protein kinase in rice. Os CPK_(24) was universally and highly expressed in rice plants and was induced by cold treatment. Whereas Os CPK_(24) knockdown plants exhibited increased sensitivity to cold compared to wild type(WT), Os CPK_(24)-overexpressing plants exhibited increased cold tolerance. Plants overexpressing Os CPK_(24) exhibited increased accumulation of proline(an osmoprotectant) and glutathione(an antioxidant) and maintained a higher GSH/GSSG(reduced glutathione to oxidized glutathione) ratio during cold stress compared to WT. In addition to these effects in response to cold stress, we observed the kinase activity of Os CPK_(24) varied under different calcium concentrations. Further,Os CPK_(24) phosphorylated Os Grx_(10), a glutathionedependent thioltransferase, at rates modulated by changes in calcium concentration. | 18 | Inulin Can Alleviate Metabolism Disorders in ob/ob Mice by Partially Restoring Leptinrelated Pathways Mediated by Gut Microbiota显示文摘Inulin has been used as a prebiotic to alleviate glucose and lipid metabolism disorders in mice and humans by modulating the gut microbiota. However, the mechanism underlying the alleviation of metabolic disorders by inulin through interactions between the gut microbiota and host cells is unclear. We use ob/ob mice as a model to study the effect of inulin on the cecal microbiota by16 S rRNA gene amplicon sequencing and its interaction with host cells by transcriptomics. The inulin-supplemented diet improved glucose and lipid metabolism disorder parameters in ob/ob mice,alleviating fat accumulation and glucose intolerance. The a diversity of gut microbial community of ob/ob mice was reduced after inulin treatment, while the b diversity tended to return to the level of wild type mice. Interestingly, Prevotellaceae UCG 001(family Prevotellaceae) was obviously enriched after inulin treatment. A comparative analysis of the gene expression profile showed that the cecal transcriptome was changed in leptin gene deficiency mice, whereas the inulin-supplemented diet partially reversed the changes in leptin gene-related signaling pathways, especially AMPK signaling pathway, where the levels of gene expression became comparable to those in wild type mice.Further analysis indicated that Prevotellaceae UCG 001 was positively correlated with the AMPK signaling pathway, which was negatively correlated with markers of glycolipid metabolism disorders. | 20 | Major advances in studies of the physical geography and living environment of China during the past 70 years and future prospects显示文摘The natural environment provides material essentials for human survival and development. The characteristics,processes, regional differentiation and forcing mechanisms of the elements of the natural environment(e.g. geomorphology,climate, hydrology, soil, etc.) are the main objects of research in physical geography. China has a complex natural environment and huge regional differentiation and therefore it provides outstanding reserach opportunities in physical geography. This review summarizes the most important developments and the main contributions of research in the physical geography and human living environment in China during the past 70 years. The major topics addressed are the uplift of the Tibetan Plateau and the evolution of its cryosphere, the development of fluvial systems, the acidification of the vast arid region of the Asian interior, variations in the monsoon and westerly climate systems on multiple timescales, the development of lakes and wetlands, the watershed system model, soil erosion, past human-environment interactions, biogeography, and physical geographic zonality. After briefly introducing international research developments, we review the history of research in physical geography in China, focusing on the major achievements and major academic debates, and finally we summarize the status of current research and the future prospects.
