| 1 | Autophagy inhibition enhances apigenin-induced apoptosis in human breast cancer cells显示文摘Apigenin (4',5,7-trihydroxyflavone) is a member of the flavone subclass of flavonoids present in fruits and vegetables. The involvement of autophagy in the apigenin-induced apoptotic death of human breast cancer cells was investigated. Cell proliferation and viability were assessed by 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) and clonogenic assays. Flow cytometry, fluorescent staining and Western blot analysis were employed to detect apoptosis and autophagy, and the role of autophagy was assessed using autophagy inhibitors. Apigenin dose- and time-dependently repressed the proliferation and clonogenic survival of the human breast cancer T47D and MDA-MB-231 cell lines. The death of T47D and MDA-MB-231 cells was due to apoptosis associated with increased levels of Caspase3, PARP cleavage and Bax/Bcl-2 ratios. The results from flow cytometry and fluorescent staining also verified the occurrence of apoptosis. In addition, the apigenin-treated cells exhibited autophagy, as characterized by the appearance of autophagosomes under fluorescence microscopy and the accumulation of acidic vesicular organelles (AVOs) by flow cytometry. Furthermore, the results of the Western blot analysis revealed that the level of LC3-II, the processed form of LC3-I, was increased. Treatment with the autophagy inhibitor, 3-methyladenine (3-MA), significantly enhanced the apoptosis induced by apigenin, which was accompanied by an increase in the level of PARP cleavage. Similar results were also confirmed by flow cytometry and fluorescence microscopy. These results indicate that apigenin has apoptosis-and autophagy-inducing effects in breast cancer cells. | 5 | The Significance of Minimally Invasive Core Needle Biopsy and Immunohistochemistry Analysis in 235 Cases with Breast Lesions显示文摘OBJECTIVE To evaluate core needle biopsy (CNB) as a mini-mally invasive method to examine breast lesions and discuss theclinical significance of subsequent immunohistochemistry (IHC)analysis.METHODS The clinical data and pathological results of 235 pa-tients with breast lesions, who received CNB before surgery, wereanalyzed and compared. Based on the results of CNB done beforesurgery, 87 out of 204 patients diagnosed as invasive carcinomawere subjected to immunodetection for p53, c-erbB-2, ER and PR.The morphological change of cancer tissues in response to chemo-therapy was also evaluated.RESULTS In total of 235 cases receiving CNB examination, 204were diagnosed as invasive carcinoma, reaching a 100% consistentrate with the surgical diagnosis. Sixty percent of the cases diag-nosed as non-invasive carcinoma by CNB was identified to havethe presence of invading elements in surgical specimens, and simi-larly, 50% of the cases diagnosed as atypical ductal hyperplasia byCNB was confirmed to be carcinoma by the subsequent result ofexcision biopsy. There was no significant difference between theCNB biopsy and regular surgical samples in positive rate of im-munohistochemistry analysis (p53, c-erbB-2, ER and PR; P >0.05).However, there was significant difference in the expression rate ofp53 and c-erbB-2 between the cases with and without morphologi-cal change in response to chemotherapy (P < 0.05). In most caseswith p53 and c-erbB-2 positive, there was no obvious morphologi-cal change after chemotherapy.CONCLUSION CNB is a cost-effective diagnostic method withminimal invasion for breast lesions, although it still has some limi-tations. | 6 | Traumatic Neuromas in Breast Cancer Patients after Mastectomy显示文摘OBJECTIVE Traumatic neuromas are rare benign lesions occurring post-mastectomy,which are usually suspected as tumour recurrences before excision biopsy.We report six cases presenting with palpable nodules post-mastectomy,to emphasize the differential diagnosis of traumatic neuroma from recurrent cancer.METHODS Six cases were reviewed.The age of patients ranged from 33 to 61 years.Nodular masses were found close to the mastectomy scar during follow-up over a period of 2.8 to 8 years.RESULTS In one patient three nodular masses were detected.Five patients had received chemotherapy,three of which had also received ra-diotherapy.Ultrasound examinations showed a well-circumscribed,homoge-neous,hypo-echoic subcutaneous nodular lesion in four cases,and a poorly defined hypo-echoic nodule with good conduction in two cases.No obvious distant metastases had been identified.Each patient underwent surgical excisional biopsy.All of nodules had a diameter less than 1 cm.Histopatho-logical examination showed proliferation of nerve fibre bundles,which were disordered,oriented and well circumscribed in fibro-adipose tissue.CONCLUSION It was concluded that all of the lesions were traumatic neuromas,independent from the initial tumor.Traumatic neuromas occur-ring in mastectomy scars are difficult to distinguish from a tumor recurrence.Although radiological evaluation of the mass with ultrasound is of value,the diagnosis can only be confirmed following a histopathological evaluation. | Xin Wang Xuchen Cao Liansheng Ning | 2007 | Chinese Journal of Clinical Oncology2007,4,3: | 0 |
