| 3 | Genetic diversity of RNA viruses infecting invertebrate pests of rice显示文摘Invertebrate species are a natural reservoir of viral genetic diversity,and invertebrate pests are widely distributed in crop fields.However,information on viruses infecting invertebrate pests of crops is limited.In this report,we describe the deep metatranscriptomic sequencing of 88 invertebrate samples covering all major invertebrate pests in rice fields.We identified 296 new RNA viruses and 13 known RNA viruses.These viruses clustered within 31 families,with many highly divergent viruses constituting potentially new families and genera.Of the identified viruses,13 RNA viruses clustered within the Fiersviridae family of bacteriophages,and 48 RNA viruses clustered within families and genera of mycoviruses.We detected known rice viruses in novel invertebrate hosts at high abundances.Furthermore,some novel RNA viruses have genome structures closely matching to known plant viruses and clustered within genera of several plant virus species.Fortyfive potential insect pathogenic RNA viruses were detected in invertebrate species.Our analysis revealed that host taxonomy plays a major role and geographical location plays an important role in structuring viral diversity.Cross-species transmission of RNA viruses was detected between invertebrate hosts.Newly identified viral genomes showed extensive variation for invertebrate viral families or genera.Together,the large-scale metatranscriptomic analysis greatly expands our understanding of RNA viruses in rice invertebrate species,the results provide valuable information for developing efficient strategies to manage insect pests and virus-mediated crop diseases. | Haoran Wang Shufen Chao Qing Yan Shu Zhang Guoqing Chen Chonghui Mao Yang Hu Fengquan Yu Shuo Wang Liang Lv Baojun Yang Jiachun He Songbai Zhang Liangsheng Zhang Peter Simmonds Guozhong Feng | 2024 | Science China(Life Sciences)2024,67,1: | 0 |
| 4 | Cloning and Functional Analysis of Calcineurin Subunits A and B in Development and Fecundity of Nilaparvata lugens(Stål)显示文摘Serine/threonine phosphatase calcineurin(CN)is a unique but confounding calcium/calmodulin-mediated enzyme,which is composed of a catalytic subunit A(CNA)and a regulatory subunit B(CNB).We cloned six transcripts for CNA named from NlCNA-X1 to NlCNA-X6,one CNB named NlCNB1 and one CNB homologous gene NlCNBH1 from Nilaparvata lugens.All of them are constitutively transcripted in various tissues and developmental stages.The primary structure of the six isoforms showed obvious differences in the length and composition of the amino acid sequence between the two binding domains of Ca^(2+)/calmodulin(CaM)and CNB.Ca^(2+)-binding EF-hand motifs were found in NlCNB1 and NlCNBH1.The specific gene silencing of NlCNA,NlCNB1 and NlCNBH1 respectively by RNAi resulted in drastical reduction in survival rate,female weight,eclosion rate and fecundity of N.lugens.These results showed that NlCNA,NlCNB1 and NlCNBH1 were required for N.lugens growth and reproduction.The negative effects of NlCNB1 silence on nymph mortality(97%),molting malformation(90%)and female sterile(50%)were more serious than those of NlCNA or NlCNBH1.qRT-PCR and enzyme-linked immunosorbent assay(ELISA)analyses indicated that the nymphs with silenced NlCNA,NlCNB1 or NlCNBH1 showed impaired hormone and energy metabolism.In nymphs,the contents of 20-hydroxyecdysone(20E)after NlCNB1 RNAi and phenoloxidase after NlCNA RNAi were particularly decreased.These results suggested that NlCNA is involved in immunity of N.lugens by regulation of phenoloxidase,while NlCNB1 may control the growth and development of N.lugens by 20E signaling pathway in addition to interact with CNA.Injection of 70 ng/μL dsNlCNB1 resulted in 77.0%down regulation of NlCNB1,and the nymph mortality was up to 57.9%at 10 d after injection.Therefore,NlCNB1 could be a potential candidate target used for strategy design in control of N.lugens.Our results revealed the importance of CN in the regulation of the growth and development of N.lugens,which provided a basis for further study of the molecular mechanism of CN. | WANG Weixia ZHU Tingheng WAN Pinjun WEI Qi HE Jiachun LAI Fengxiang FU Qiang | 2022 | Rice science2022,29,2: | 0 |
| 5 | Gelatin-Based Metamaterial Hydrogel Films with High Conformality for Ultra-Soft Tissue Monitoring显示文摘Implantable hydrogel-based bioelectronics(IHB)can precisely monitor human health and diagnose diseases.However,achieving biodegradability,biocompatibility,and high conformality with soft tissues poses significant challenges for IHB.Gelatin is the most suitable candidate for IHB since it is a collagen hydrolysate and a substantial part of the extracellular matrix found naturally in most tissues.This study used 3D printing ultrafine fiber networks with metamaterial design to embed into ultra-low elastic modulus hydrogel to create a novel gelatin-based conductive film(GCF)with mechanical programmability.The regulation of GCF nearly covers soft tissue mechanics,an elastic modulus from 20 to 420 kPa,and a Poisson’s ratio from-0.25 to 0.52.The negative Poisson’s ratio promotes conformality with soft tissues to improve the efficiency of biological interfaces.The GCF can monitor heartbeat signals and respiratory rate by determining cardiac deformation due to its high conformability.Notably,the gelatin characteristics of the biodegradable GCF enable the sensor to monitor and support tissue restoration.The GCF metamaterial design offers a unique idea for bioelectronics to develop implantable sensors that integrate monitoring and tissue repair and a customized method for endowing implanted sensors to be highly conformal with soft tissues. | Yuewei Chen Yanyan Zhou Zihe Hu Weiying Lu Zhuang Li Ning Gao Nian Liu Yuanrong Li Jing He Qing Gao Zhijian Xie Jiachun Li Yong He | 2024 | Nano-Micro Letters2024,16,2: | 0 |
| 6 | Improved rhizoremediation for decabromodiphenyl ether(BDE-209)in E-waste contaminated soils显示文摘An experiment was conducted to improve rhizoremediation for decabromodiphenyl ether(BDE-209)contaminated soil from typical E-waste dismantling areas.Plants of ryegrass(Lolium perenne L.)and rice(Oryza sativa L.)were cultivated in aged-contaminated(initial concentration of 346.3μg BDE-209·kg^(-1))and freshly-spiked(initial concentration of 3127μg BDE-209·kg^(-1))soils,coupling with the agricultural modification strategies of compost addition and/or arbuscular mycorrhizal fungi(AMF)infection,respectively.60 days’growth of ryegrass significantly facilitated the dissipation of BDE-209,with the most effective in its rhizosphere in treatment inoculated with AMF;the BDE-209 dissipation rates achieved 51.9% and 22.8% in rhizosphere,and 43.5% and 19.8% in non-rhizosphere,for aged-contaminated and freshlyspiked soils,respectively.120 days’growth of rice with simultaneous inoculation of AMF and addition of compost was the most effective in facilitating BDE-209 dissipation in agedcontaminated soil,with the removal rates of 53.3% and 48.1% in rhizosphere and nonrhizosphere soils respectively;while for freshly-spiked soils,the most effective removal was achieved by compost addition only,with the BDE-209 dissipation rates of 27.9% and 26.6% in rhizosphere and non-rhizosphere soils,respectively.High throughput sequencing analysis of rhizosphere soil DNA showed that responses in microbial communities and their structure differed with plant species,soil pollution dose,AMF inoculation and/or compost addition.Actinomycetales,Xanthomonadales,Burkholderiales,Sphingomonadales,Clostridiales,Cytophagales,Gemmatimonadales and Saprospirales were the sensitive responders and even possibly potential functional microbial groups during the facilitated removal of BDE-209 in soils.This study illustrates an effective rhizoremediation pattern for removal of BDE-209 in pollution soils,through successive cultivation of rice and followed by ryegrass,with rice growth coupled with AMF inoculation and compost addition,while ryegrass growth coupled with AMF inoculation only. | Jiayin Feng Xinquan Shen Jian Chen Jiachun Shi Jianming Xu Caixian Tang Philip C.Brookes Yan He | 2019 | Soil Ecology Letters2019,1,3: | 0 |
