1Human carboxylesterases:a comprehensive review显示文摘Mammalian carboxylesterases(CEs) are key enzymes from the serine hydrolase superfamily.In the human body, two predominant carboxylesterases(CES1 and CES2) have been identified and extensively studied over the past decade. These two enzymes play crucial roles in the metabolism of a wide variety of endogenous esters, ester-containing drugs and environmental toxicants. The key roles of CES in both human health and xenobiotic metabolism arouse great interest in the discovery of potent CES modulators to regulate endobiotic metabolism or to improve the efficacy of ester drugs. This review covers the structural and catalytic features of CES, tissue distributions, biological functions, genetic polymorphisms, substrate specificities and inhibitor properties of CES1 and CES2, as well as the significance and recent progress on the discovery of CES modulators. The information presented here will help pharmacologists explore the relevance of CES to human diseases or to assign the contribution of certain CES in xenobiotic metabolism. 3Potassium 2-(l-hydroxypentyl)-benzoate attenuates neuroinflammatory responses and upregulates heme oxygenase-1 in systemic lipopolysaccharide-induced inflammation in mice显示文摘A neuroinflammatory response is commonly involved in the progression of many neurodegenerative diseases. Potassium 2-(1-hydroxypentyl)-benzoate(PHPB), a novel neuroprotective compound, has shown promising effects in the treatment of ischemic stroke and Alzheimer's disease(AD). In the present study, the anti-inflammatory effects of PHPB were investigated in the plasma and brain of C57BL/6 mice administered a single intraperitoneal(i.p.) injection of lipopolysaccharide(LPS). Levels of i NOS and the cytokines TNFα, IL-1β and IL-10 were elevated in plasma, cerebral cortex and hippocampus after LPS injection and the number of microglia and astrocytes in cortex and hippocampus were increased. LPS also upregulated the expression of heme oxygenase-1(HO-1) in the cortex and hippocampus. PHPB reduced the levels of i NOS and cytokines in the plasma and brain, decreased the number of microglia and astrocytes and further enhanced the upregulation of HO-1. In addition, PHPB inhibited the LPS-induced phosphorylation of ERK, P38 and JNK. 4CD4^(+)CD25^(+)but not CD4^(+)Foxp3^(+) T cells as a regulatory subset in primary biliary cirrhosis显示文摘Increasing evidence indicates a role for regulatory T cells(Tregs)in the immune response and in autoimmune diseases,but the role of Tregs and cytokines in autoimmune hepatic diseases remains largely unclear and controversial,especially in patients with primary biliary cirrhosis(PBC).This study was undertaken to investigate Tregs and different cytokines in the liver and peripheral blood of PBC patients.We found that these patients demonstrated a reduction of CD4^(+)CD25^(+) T cells but elevated CD4^(+)Foxp3^(+) T cells in peripheral blood mononuclear cells(PBMCs)and CD41 T cells.The percentage of CD4^(+)CD25^(+) T cells in PBMCs was negatively correlated with elevated plasma interferon(IFN)-c levels.A liver-specific analysis showed that the frequency of Foxp31 Tregs,transforming growth factor(TGF)-b1 and IFN-c were increased in PBC patients.Our findings suggest that an imbalance between CD4^(+)CD25^(+) Tregs and cytotoxic cytokines plays a crucial role in the pathogenesis of PBC while the role of Foxp3 needs further investigation.Dandan Wang Huayong Zhang Jun Liang Zhifeng Gu Qiang Zhou Xiangshan Fan Yayi Hou Lingyun Sun 2010Cellular & Molecular Immunology2010,7,6:9
5Facile synthesis of graphene via reduction of graphene oxide by artemisinin in ethanol显示文摘The preparation of reduced graphene oxide(RGO)by chemical reduction of graphene oxide(GO)usually involves highly toxic reducing agents which are harmful to the environment and human health.In this paper,a mediated facile and relative green approach for the preparation of RGO in ethanol using artemisinin as a reducing agent is reported for the first time.The morphology and de-oxidation efficiency of the obtained RGO were characterized by transmission electron microscope(TEM),atomic force microscope(AFM),and X-ray photoelectron spectroscopy(XPS).The results showed that artemisinin can effectively reduce GO into few-layered RGO with a high carbon to oxygen ratio(11.7).The mechanism for elimination of oxygen-containing functional groups decorated on GO nanosheets by artemisinin was proposed.The important features of relatively environmentally friendly and facile operation procedures endow this approach with great promise in the mass production of RGO and various graphene-based materials,especially for biomaterials.Dandan Hou Qinfu Liu Xianshuai Wang Ying Quan Zhichuan Qiao Li Yu Shuli Ding 2018Journal of Materiomics2018,4,3:6

7Identifying changes in the wheat kernel proteome under heat stress using iTRAQ显示文摘Wheat(Triticum aestivum L.) is one of the three major global food crops. Hightemperature stress can affect its yield and quality. Studies of the effect of hightemperature stress on wheat kernel development are important because they can reveal the stability of wheat quality and lead to the genetic improvement of wheat quality traits. In this study, the isobaric tags for relative and absolute quantitation(iTRAQ)method was adopted to analyze changes in the protein expression profile of wheat cultivars under high temperature stress. The protein content of wheat grain increased under heat stress, while the SDS-sedimentation value and starch content decreased.Grain filling was deficient under high temperature stress, which reduced thousandkernel weight but did not affect wheat kernel length. The 207 differentially expressed proteins identified in Gaocheng 8901 under heat stress were associated with energy metabolism, growth and development, and stress response. Gene Ontology enrichment analysis showed that the annotated proteins that were differentially expressed in Gaocheng 8901 under heat stress were involved mainly in stimulus response, abiotic stress response, stress response, and plasma membrane. A set of 78 differentially expressed proteins were assigned to 83 KEGG signaling/metabolic pathways. KEGG pathway enrichment analysis showed that this set of proteins was significantly enriched in members of 51 pathways, and the proteins participated mainly in protein synthesis in the endoplasmic reticulum, starch and sucrose metabolism, and reaction on ribosomes. Five differentially expressed proteins were involved in protein–protein interaction networks that may greatly influence the yield and quality of wheat grain. 8A comparative study of hydrothermal aging effect on cerium and lanthanum doped Cu/SSZ-13 catalysts for NH_(3)-SCR显示文摘Ce-or La-doped Cu/SSZ-13 catalysts were prepared by a hydrothermal method and Cu,Ce or La ions were incorporated through stepwise ion exchange,The catalyst activity was measured for the ammonia selective catalytic reduction reaction.The structure and composition of catalyst were characterized by using X-ray diffraction,scanning electron microscopy,inductively coupled plasma mass spectrometry solid-state NMR,NH_(3)-TPD techniques,and the active components were examined by XPS and XANES.The results indicate that the Ce and La doping can both completely preserve the SCR activity of Cu/SSZ-13 above 300℃,but there is also a decrease of activity below 200℃.On the other hand,Ce doping is beneficial to the formation of framework aluminum,stabilizes molecular sieve framework and Cu active sites of Cu/SSZ-13,thereby improves the catalyst hydrothermal stability.But La doping will decrease the amount of framework aluminum and Cu active sites of Cu/SSZ-13 after hydrothermally aging,even destroy zeolite CHA structure.This is quite harmful to the catalyst hydrothermal stability.Di Deng Shujun Deng Dandan He Zhiheng Wang Zhipeng Chen Yi Ji Guoping Yan Guangjin Hou Licheng Liu Hong He 2021Journal of Rare Earths2021,39,8:5

11Influence of order degree of coaly graphite on its structure change during preparation of graphene oxide显示文摘Coaly graphite is an important natural graphite resource that derived from coal under a natural process that associated with igneous intrusion.Flake graphite is usually used for the chemical synthesis of graphene oxide(GO),the main precursor for preparation of graphene,but few papers pay attention to preparing GO using coaly graphite.In this paper,four kinds of natural coaly graphite with different graphitization degrees were exposed to a modified Hummer’s oxidation method to prepare GO.The flake graphite sample was also used for comparison.The results showed that the structural change process from graphite to GO were significantly affected by the graphitization degree of the original coaly graphite.The relationship between yields of GO and graphitization degrees of the coaly graphite was explored.The mechanism of why the graphite with low graphitization degrees cannot be totally oxidized was proposed.Coaly graphite with a graphitization degree of higher than 80% was easier to be oxidized and yielded the same amount of GO as the flake graphite did,suggesting it is the potential substitute for the flake graphite to produce GO in bulk quantities.Dandan Hou Kuo Li Rujia Ma Qinfu Liu 2020Journal of Materiomics2020,6,3:2
12CoPt-Co hybrid supported on amino modified SiO2 nanospheres as a high performance catalyst for hydrogen generation from ammonia borane显示文摘CoPt-Co hybrids were successfully supported on amino modified SiO2 nanospheres by a chemical reduction method at a temperature of 278 K.The solid carrier i.e.amino modified SiO2,provides numerous anchoring sites for the metal nanoparticles(NPs)to improve the dispersion while reducing the size of metal NPs.The supported NPs displayed a narrow particle size distribution on the SiO2 surface with an average diameter of 12 nm.The XRD results alongside with the binary alloy phase diagram suggest that the resulted NPs are bimetallic,composed of CoPt and amorphous Co.Among the prepared materials,the solid with the specific composition of SiO2@Pt0.1Co0.9was proved to be effective catalyst for ammonia borane(AB)hydrolysis in aqueous solution.The turnover frequency(TOF)value of the supported nanocatalyst was 25.59molH2min^-1·molM^-1,almost twice as that of unsupported Pt0.1Co0.9NPs while the activation energy was 37.05 kJ mol^-1.Furthermore,the SiO2@Pt0.1Co0.9composite manifested high catalytic activity even after five cycles of reuse.Hongming Zhang Dandan Ke Lina Cheng Xuelei Feng Xuewei Hou Jin Wang Yuan Li Shumin Han 2019Progress in Natural Science:Materials International2019,29,1:2

14Prediction of Deformation and Footprint of High-speed Rotating Radial Tires显示文摘In this research,the tire deformations due to rotation are divided into three parts:tread,belt and carcass.Tread deformation is obtained by using a rotating hollow cylinder model,belt expansion is calculated according to the laminated composite shell theory,and a formula of dynamic equilibrium profile is built to investigate carcass deformation.Moreover,a simple method is used to predict the footprint changes caused by tire deformation.Results show that a large deformation is occurring at tire shoulder,a smaller one at tread centre,and the shoulder deformations are mostly caused by carcass.Length of footprint side edges increase with tire rolling speed increasing.The predicted tread,belt,carcass deformations and footprint changes are in good agreement with finite element analysis results.The proposed method and the results will offer a helpful guidance to tire and vehicle engineers.CUI Zhibo CHAI Delong LYU Jiafeng HU Kai ZHANG Chunsheng HOU Dandan WANG Youshan 2020橡胶工业2020,67,5:2
15Sensing carboxylesterase 1 in living systems by a practical and isoformspecific fluorescent probe显示文摘Carboxylesterase 1(CES1), one of the most abundant serine hydrolases in mammals, has drawn much attentions in recent years, owing to this enzyme involves in many physiological processes via hydrolysis of both endogenous esters and xenobiotic esters. Herein, to real-time monitor the activities of CES1 in various biological systems, a practical and iso form-specific fluorescent probe was developed on the basis of the substrate preference of CES1, as well as the structural and optical properties of BODIPY dyes. After screening of a panel of BODIPY ester derivatives, probe 1 displayed the best combination of specificity,sensitivity, enzymatic kinetics and applicability for monitoring CES1 activities in real samples. This probe was successfully used to detect CESl activities in several biological systems including tissue preparations,living cells, tissue slices and zebrafish. Furthermore, the biomedical applications of probe 1 for screening of CES1 inhibitors were also demonstrated using tissue preparations or living cells as enzyme sources. 17Screening and evaluation of antioxidants from lees by micro-injector systems combined with a fluorescent probe,N-borylbenzyloxycarbonyl-3,7-dihydroxyphenoxazine,in living Drosophila显示文摘Over the past few decades, the determination of antioxidant activity by chemical probe has been widely reported, but in vivo evaluation via model organisms of Drosophila melanogaster and rapid discovery system has not been studied adequately. In this study, we determined the antioxidant activity of lees and demonstrated the ability of compounds in lees to scavenge H_2O_2 in vitro and in vivo by different chemical probes. Lees increased the ability of Drosophila against oxidative stress and antioxidant enzyme activity in vivo. Five ingredients of organic acids and flavones in lees extract that rapidly scavenged H_2O_2 were revealed by a post-column-derived HPLC-UV-FLD system based on 4-hydroxyphenylacetic acid(PHPAA)chemiluminescence. Additionally, another fluorescent probe,N-borylbenzyloxycarbonyl-3,7-dihydroxyphenoxazine(NBCD), was selected to evaluate the reactive oxygen species(ROS) scavenging capacity of lees in living Drosophila melanogaster using a microfluidic injection test coupled with microscopic imaging analysis, and similar effects were observed in flies when they were treated with tartaric acid and caffeic acid. 18Clinical study on treatment of chronic viral cholestatic hepatitis with Chishaodanpi decoction显示文摘OBJECTIVE: To observe the therapeutic effect of Chishaodanpi decoction(CSDPD) on chronic viral cholestatic hepatitis.METHODS: A total of 107 subjects with chronic viral cholestatic hepatitis were enrolled in our hospital from March 2007 to November 2012. Patients were randomly divided into treatment(54 cases)and control groups(53 cases). The control group was treated with potassium magnesium aspartate,diammonium glycyrrhizinate, glucurolactone, vitamin C, and lamivudine, once a day. The treatment group was treated with modified CSDPD, 100 m L a time, twice a day, in addition to the treatment given to the control group. The patients in both groups were treated for 8 weeks. The main symptoms and signs were recorded every day throughout the clinical trial. Before and after the trial,changes in liver function including total bilirubin(TBil), direct bilirubin(DBil), total bile acid(TBA),and the activities of alkaline phosphatase(ALP), alanine aminotransferase(ALT), aspartate aminotransferase(AST), and γ-glutamyl transferase(γ-GT),were all detected. Adverse reactions were also recorded.RESULTS: There were no differences in gender, age,disease duration, symptoms, signs, or laboratory findings between the two groups(P>0.05). After an8-week treatment, improvements in jaundice, weakness, poor appetite, abdominal distention, and skin itching were significantly better in the treatment group than in the control group(P<0.05). In the treatment group, 43 patients had a significant response to the treatment, seven patients had a response, and four patients had no response, with 21,12, and 20 patients in the control group, respectively. The total effective rate was 92.6% in the treatment group and 62.3% in the control group, which was a significant difference(P<0.05). The levels of TBil, DBil, TBA, ALP, ALT, AST, and γ-GT in both groups were significantly lower after treatment,and were significantly different between the two groups(P<0.05). A few patients in the treatment group had mild adverse effects such as increased bowel movement frequency and mild stomachache. 19Inhibitory effect of Sumu(Lignum Sappan) plus Fuzi(Radix Aconiti Lateralis Praeparata) on a lung carcinoma model显示文摘OBJECTIVE: To investigate the inhibitory effect of Sumu(Lignum Sappan) plus Fuzi(Radix Aconiti Lateralis Praeparata)(SF) on the growth and metastasis of Lewis lung carcinoma.METHODS: A lung carcinoma model was established by subcutaneously inoculating Lewis lung carcinoma cells into C57 BL/6 mice. The mice were randomly divided into four groups of 13 mice(control, low-dose, moderate-dose, and high-dose), and gavaged once-daily for 21 consecutive days. The rates of tumor inhibition, metastasis, and metastasis inhibition were observed. The differential expressions of s P-selectin and vascular endothelial growth factor C(VEGFC) were compared between the treatment groups and the control group.RESULTS: The tumor weights differed significantly between the treatment groups and the control group(P < 0.05).
