2A Study on Anti-oxidative Activity of Soybean Peptideswith Linoleic Acid Peroxidation Systems显示文摘Soybean bioactive peptides(SBPs) were prepared from the isolated soybean protein by proteolysis with an alkaline protease, alcalase, at 50 ℃ and pH=8.0. The dependence of hydrolysis time on hydrolysis degree and molecular weight distribution were examined. The hydrolysate was fractionated on a Sephadex G-25 column and the anti-oxidative activities of the fractions were detected by the method of pyrogallol auto-oxidation. The average chain length of soybean peptides that have anti-oxidative activity was estimated to be about 7. The anti-oxidative properties of the soybean peptide were also studied by using linoleic acid peroxidation systems. The optimal condition of the peroxidation system was set up, Vc/Cu 2+ as the inducer at pH=7.4 and 25 ℃. 5An Improved Barcoded Oligonucleotide Primers-based Next-generation Sequencing Approach for Direct Identification of Viral Pathogens in Clinical Specimens显示文摘Objective To provide a feasible and cost-effective next-generation sequencing(NGS) method for accurate identification of viral pathogens in clinical specimens, because enormous limitations impede the clinical use of common NGS, such as high cost, complicated procedures, tremendous data analysis, and high background noise in clinical samples. Methods Viruses from cell culture materials or clinical specimens were identified following an improved NGS procedure: reduction of background noise by sample preprocessing, viral enrichment by barcoded oligonucleotide(random hexamer or non-ribosomal hexanucleotide) primer-based amplification, fragmentation-free library construction and sequencing of one-tube mixtures, as well as rapid data analysis using an in-house pipeline. Results NGS data demonstrated that both barcoded primer sets were useful to simultaneously capture multiple viral pathogens in cell culture materials or clinical specimens and verified that hexanucleotide primers captured as many viral sequences as hexamers did. Moreover, direct testing of clinical specimens using this improved hexanucleotide primer-based NGS approach provided further detailed genotypes of enteroviruses causing hand, foot, and mouth disease(HFMD) and identified other potential viruses or differentiated misdiagnosis events. 6The Effects of Different Sex-Linked Dwarf Variations on Chinese Native Chickens显示文摘Variants in chicken growth hormone receptor (GHR) gene lead to sex-linked dwarf (SLD) chickens, but effects of different variants are distinct. In this study, 11 SLD chicken breeds or strains including 3 Chinese native breeds and 8 breeding strains were studied in order to investigate the effects of different sex-linked dwarf variations on growth performance. The results showed that there were three reasons which could lead to dwarfism in the 11 breeds or strains. Firstly, an about 1.7 kb deletion of growth hormone receptor (GHR) gene leads to dwarfism in Jiangxi dwarf chicken, strains GF24, GF26, N308, N309, and N310. Secondly, a T354C mutation in exon 5 of the GHR gene leads to dwarfism in strains N301 and N305. Thirdly, an unknown variant leads to dwarfism in Guizhou Yellow Dwarf chicken and Yixing Bantam chicken. In addition, all individuals of N303 had the 1.7 kb deletion of the GHR gene, and additionally, some of them also carried the T354C mutation. As far as the performance of individuals were compared among T354C homozygote, deletion homozygote, and heterozygote carrying both T354C and deletion, it was found that the T354C's impacts on body weight of Chinese chickens were maximum, the body weight of chickens with homozygote T354C was 92.12% of those with heterozygote, and the difference of the body weight between deletion homozygote and heterozygote was not significant. 7Nusap1 is essential for neural crest cell migration in zebrafish显示文摘Microtubules play important roles in mitotic spindle assembly and chromosome segregation to maintain normal cell cycle progression.A number of microtubule-associated proteins have been identified in epithelial and neural cell cultures;however,their physiological significance is not well characterized due to the lack of appropriate in vivo animal models.Nucleolar spindleassociated protein(NuSAP)is a microtubule-binding protein and is reported to be involved in mitosis by cell culture studies.In this report,we identified the zebrafish homologue of human NuSAP and investigated its expression profile and functions.Using in situ hybridization,we demonstrated that transcripts of zebrafish nusap1 are specifically expressed in the retina,forebrain,hindbrain and neural crest.When the in vivo expression of nusap1 was knocked down through antisense oligonucleotide morpholino technology,the morphants of nusap1 showed impaired morphogenesis in the trunk and yolk extension,implying the involvement of Nusap1 in cell migration.Mechanistic studies revealed that nusap1 morphants have an altered expression pattern of neural crest markers crestin and sox9b,but normal expression of blood vessel and notochord markers gata1 and shh.In addition,nusap1 mRNA injection caused serious apoptosis in retina and hindbrain tissue,and these phenotypes can be rescued by co-injection of morpholino against nusap1.These observations not only suggest a role for Nusap1 in connecting apoptosis with cell migration,but also provide strong evidences that Nusap1 is potentially involved in morphogenesis in vertebrates.Jing Nie Hua Wang Fuchu He Huizhe Huang 2010Protein & Cell2010,1,3:7
8Risk Factors for Strangulated Ovarian Hernia in Female Infants:the Role of Ovarian Volume显示文摘The risk factors associated with strangulated ovarian hernia (SOH)in female patients (<1 year old)were identified.A retrospective analysis was conducted regarding the data from 2006 to 2017.The patients were divided into 2groups:SOH group (n=9)and non-SOH group (n=23). Patient demographics,clinical signs,preoperative examinations and intraoperative findings were compared between the two groups,and risk factors for SOH were tested using a binary logistic regression model.To explore whether greater ovary was more likely to be twisted,leading to SOH, all the patients were divided into ovary volume <5cm^3 and ≥5cm^3 groups and the association between ovarian volume and ovary torsion was assessed.Among a total of 32 female patients (<1year old)with incarcerated ovarian herniation,9 patients developed SOH.The single variate analysis revealed that times of manual reduction,ovarian volume,ovary with or without multiple cysts,ovary torsion or not and angle of ovary torsion were found to be significant factors associated with SOH.The multivariate analysis showed ovarian volume was evidenced as an independent risk factor for SOH.Furthermore,the incidence of ovary torsion was significantly higher in ovarian volume ≥5cm^3 group than in ovarian volume <5cm^3 group,indicating that larger ovary was more likely to result in ovary torsion,leading to SOH.Our study demonstrated that the odds of SOH increased with increasing ovarian volume in female patients (<1 year old)because the relatively greater ovary at this age was more likely to be incarcerated and twisted,leading to SOH.Yu CHEN Xiang-zhi PENG Wei LU Kai ZHENG Jian GUO Hua NIE Xiao-jie SONG Yan ZHANG Jing YANG 2018Current Medical Science2018,38,6:6
9Chromatin accessibility analysis reveals that TFAP2A promotes angiogenesis in acquired resistance to anlotinib in lung cancer cells显示文摘Anlotinib,a multitarget tyrosine kinase inhibitor,is effective as a third-line treatment against non-small cell lung cancer(NSCLC).However,acquired resistance occurs during its administration.To understand the molecular mechanisms of anlotinib resistance,we characterized chromatin accessibility in both the parental and anlotinib-resistant lung cancer cell line NCI-H1975 through ATAC-seq.Compared with the parental cells,we identified 2666 genomic regions with greater accessibility in anlotinib-resistant cells,in which angiogenesis-related processes and the motifs of 21 transcription factors were enriched.Among these transcription factors,TFAP2A was upregulated.TFAP2A knockdown robustly diminished tumor-induced angiogenesis and partially rescued the anti-angiogenic activity of anlotinib.Furthermore,transcriptome analysis indicated that 2280 genes were downregulated in anlotinib-resistant cells with TFAP2A knocked down,among which the PDGFR,TGF-β,and VEGFR signaling pathways were enriched.Meanwhile,we demonstrated that TFAP2A binds to accessible sites within BMP4 and HSPG2.Collectively,this study suggests that TFAP2A accelerates anlotinib resistance by promoting tumor-induced angiogenesis.Le-le Zhang Jun Lu Rui-qi Liu Min-juan Hu Yi-ming Zhao Sheng Tan Shu-yuan Wang Bo Zhang Wei Nie Yu Dong Hua Zhong Wei Zhang Xiao-dong Zhao Bao-hui Han 2020Acta Pharmacologica Sinica2020,41,10:6
10Research Data Management Implementation at Peking University Library:Foster and Promote Open Science and Open Data显示文摘Research Data Management(RDM)has become increasingly important for more and more academic institutions.Using the Peking University Open Research Data Repository(PKU-ORDR)project as an example,this paper will review a library-based university-wide open research data repository project and related RDM services implementation process including project kickoff,needs assessment,partnerships establishment,software investigation and selection,software customization,as well as data curation services and training.Through the review,some issues revealed during the stages of the implementation process are also discussed and addressed in the paper such as awareness of research data,demands from data providers and users,data policies and requirements from home institution,requirements from funding agencies and publishers,the collaboration between administrative units and libraries,and concerns from data providers and users.The significance of the study is that the paper shows an example of creating an Open Data repository and RDM services for other Chinese academic libraries planning to implement their RDM services for their home institutions.The authors of the paper have also observed since the PKU-ORDR and RDM services implemented in 2015,the Peking University Library(PKUL)has helped numerous researchers to support the entire research life cycle and enhanced Open Science(OS)practices on campus,as well as impacted the national OS movement in China through various national events and activities hosted by the PKUL.Hua Nie Pengcheng Luo Ping Fu 2021Data Intelligence2021,3,1:5
11Abnormal Metabolic Connectivity in Rats at the Acute Stage of Ischemic Stroke显示文摘Stroke at the acute stage is a major cause of disability in adults, and is associated with dysfunction of brain networks. However, the mechanisms underlying changes in brain connectivity in stroke are far from fully elucidated. In the present study, we investigated brain metabolism and metabolic connectivity in a rat ischemic stroke model of middle cerebral artery occlusion(MCAO)at the acute stage using18 F-fluorodeoxyglucose positron emission tomography. Voxel-wise analysis showed decreased metabolism mainly in the ipsilesional hemisphere, and increased metabolism mainly in the contralesional cerebellum. We used further metabolic connectivity analysis to explore the brain metabolic network in MCAO.Compared to sham controls, rats with MCAO showed most significantly reduced nodal and local efficiency in the ipsilesional striatum. In addition, the MCAO group showed decreased metabolic central connection of the ipsilesional striatum with the ipsilesional cerebellum, ipsilesional hippocampus, and bilateral hypothalamus. 12Protective effects of medical ozone combined with traditional Chinese medicine against chemically-induced hepatic injury in dogs显示文摘AIM: To investigate the protective effect of medical ozone (O3) combined with Traditional Chinese Medicine (TCM) Yigan Fuzheng Paidu Capsules (YC) against carbon tetrachloride (CCl4)-induced hepatic injury in dogs. METHODS: Thirty healthy dogs were divided randomly into fi ve groups (n = 6 in each group), namely control, oleanolic acid tablet (OAT), O3, YC and O3 + YC, given either no particular pre-treatment, oral OAT, medical ozone rectal insulfflation every other day, oral YC, or oral YC plus medical ozone rectal insulfflation every other day, respectively, for 30 consecutive days. After pre-treatment, acute hepatic injury was induced in all dogs with a single-dose intraperitoneal injection of CCl4. General condition and survival time were recorded. The biochemical and hematological indexes of alanine aminotransferase (ALT), aspartate aminotransferase/ alanine aminotransferase (AST/ALT), serum total bilirubin (TBIL), prothrombin time (PT), blood ammonia (AMMO), and blood urea nitrogen (BUN) were measured after CCl4 injection. Hepatic pathological changes were also observed.RESULTS: Compared to the other four groups, the changes of group O3 + YC dogs’ general conditions (motoricity, mental state, eating, urination and defecation) could be better controlled. In group O3 + YC the survival rates were higher (P < 0.05 vs group control). AST/ALT values were kept within a normal level in group O3 + YC. Hepatic histopathology showed that hepatic injury in group O3 + YC was less serious than those in the other four groups. 14Matrix Effects and Behaviors of Elements in Oxalic Acid Medium by Inductively Coupled Plasma Mass Spectrometry显示文摘The influence of different concentration of oxalic acid matrix on elemental inductively coupled plasma mass spectrometry (ICP-MS) has been investigated. It has been proved that the sensitivity of analytes can be significantly enhanced by adding small amounts of oxalic acid medium with adjusted nebulizer flow-rate gas,especially for the elements with ionization potential between 9 and 11 eV. 15断肢再植术后感染患者甲襞微循环及血流指标的变化观察显示文摘目的分析断肢再植术后感染患者甲襞微循环及血流变指标的变化,为断肢再植术后感染患者的诊治及预防控制提供依据。方法选取2012年1月-2015年9月收治的56例断肢再植术后感染患者为观察组,同时间段56例断肢再植术后未发生感染患者为对照组,将两组患者的甲襞微循环及血流变指标进行检测与比较,并比较观察组中浅层感染及深部感染者的检测结果。结果观察组甲襞微循环加权积分中的襻周积分、流态积分、管襻积分及总积分分别为(1.64±0.21)、(2.31±0.27)、(1.25±0.14)及(3.32±0.32),其均高于对照组(0.88±0.12)、(1.18±0.15)、(0.64±0.06)及(2.30±0.24),其他甲襞微循环指标及血流变指标也均高于对照组,且观察组中深部感染患者的检测结果高于浅层感染的患者,各个比较指标间差异均有统计学意义(P<0.05)。结论断肢再植术后感染患者甲襞微循环及血流变指标均较高,且浅部感染与深部感染患者的检测结果也存在明显差异,因此应重视对此类患者进行微循环状态的改善。张全英 朱玉花 李琼 李中锋 王丽娜 NIE Xing-guo QIN Hua 2016中华医院感染学杂志2016,26,13:4
16Quantification of Trace Amounts of Impurities in High Purity Cobalt by High Resolution Inductively Coupled Plasma Mass Spectrometry显示文摘An analytical method using high resolution inductively coupled plasma mass spectrometry (HR-ICP-MS) for rapid simultaneous determination of Be, Mg, Al, Ti, V, Cr, Mn, Fe, Ni, Cu, Zn, Ga, Ge, As, Se, Mo, Ag, Cd, Sn, Sb, Ba, Pt, Au and Pb in high purity cobalt was described. Sample digestions were performed in closed microwave vessels using HNO3 and HCl. The matrix effects due to the presence of excess HCl and Co were evaluated. The usefulness of high mass resolution for overcoming some spectral interference was demonstrated. The optimum conditions for the determination was tested and discussed. Correction for matrix effects, Sc, Rh and Bi were used as internal standards. The detection limits is 0.003-0.57 μg/g, the recovery ratio is 92.2%-111.2%, and the RSD is less than 3.6%. The method is accurate, quick and convenient. 17Effect of calcium citrate on bone integration in a rabbit femur defect model显示文摘Objective:To explore effect of calcium citrate on bone integration in a rabbit femur defect model, and to compare the bone formation with different sizes by radiological and histological study. Methods:Twenty-four male Japanese white rabbits were randomly divided into three groups (Group A.B.C)in this study.Under anesthesia,defects of four sizes(1.2,1.5,2.0 and 2.5 mm) were created in each of the rabbits.Commercially pure calcium citrate powder was placed inside the medullary compartment of the femur(Experimental),while in the contralateral femur(Control) nothing was implanted.The defects were analyzed using radiography and histological analysis by using Imagepro-Plus 6.0 software after animal was sacrificed at 4th(Group A),6th(Group B) and 8th(Group C) weeks postoperatively.Four samples were analyzed for each size of defect and each healing period.Results:The histological and the radiologic evaluation were performed after sacrification of all rabbits on postoperative 4th and 6th weeks,It showed significant difference between the experimental group and the control group when these defects were less than or equal to 2.0 mm.No statistical difference was observed when these defects were larger than 2.0 mm at all healing periods except at the 4th week.Conclusions:Calcium citrate affects the early periods of hone defects healing mechanism in Japanese white rabbits positively,especially when the defect is not too large,We suggest further studies on calcium citrate to determine the effects of various dosages,administration ways and the experimental time on the bone defects.Wei Zhang Wei Wang Qing-Yu Chen Zhong-Qin Lin Shao-Wen Cheng Dong-Quan Kou Xiao-Zhou Ying Yue Shen Xiao-Jie Cheng Peng-Fei Nie Xiu-Cui Li Ferdinand An Rompis Hang Huang Hua Zhang Zhong-Lin Mu Lei Peng 2012Asian Pacific Journal of Tropical Medicine2012,5,4:3

19Primary human hepatocyte transplantation in the therapy of hepatic failure:2 cases report显示文摘Liver failure is the end stage of hepatopathy with unfavorable prognosis.In two patients with liver failure,viable primary human hepatocytes,obtained from resected liver tissue of patients with hepatolithiasis.were transplanted into the spleen by interventional therapy through femoral arterial cannula.After transplantation,the patients’ clinical symptoms and liver function were significantly improved.However,their bilirubin increased within six days following transplantation.One suffered from hepatic coma and give up treatment and the other patient died forteen days after transplantation.It is technically safe to treat liver failure by intrasplenic transplantation of adult hepatocytes and the clinical efficacy has been confirmed.How to make transplanted hepatic cells proliferate and functionally survive is the key point to maintain continuous improvement of the recipient’s hepatic function.Zhuo-Ri Li Xian-Hai Mao Xiao-Xuan Hu Sheng-Dan Nie Yong-Zhong Shi Hua Xiang Jian-Hui Yang Ou-De Cao 2012Asian Pacific Journal of Tropical Medicine2012,5,2:3
