2Hepatitis B virus X protein modulates the apoptosis of hepatoma cell line induced by TRAIL显示文摘The purpose of this study is to observe the effects of HBx on the apoptosis of hepatoma cells induced by TNF-related apoptosis-inducing ligand (TRAIL) and to study prelimi- nary molecular mechanisms for its effects. In order to set up a model in vitro, BEL7402-HBx cell line, stably expressing HBx mRNA, was established by stable transfection of pcDNA-HBx, which contains HBx gene, into hepatoma cell line BEL7402. Control cell line BEL7402-cDNA3, stably transfected with pcDNA3, was set up simultaneously as a control. Trypan blue exclusion test, caspase 3 activity detection and TUNEL assay were performed to detect the apoptosis of BEL7402, BEL7402-cDNA3, BEL7402-HBx induced by TRAIL. The expression of TRAIL recep- tors in three groups was analyzed by Flow cytometry. In addition, phosphorothioated antisense oligonucleotide against the translation initial region of HBx gene (PS-asODNs/HBx) was used to block the expression of HBx in HepG2.2.15 cells and to further confirm the effects of HBx on TRAIL-induced apoptosis. Trypan blue exclusion test indicated that TRAIL had a dose-dependent cytotoxicity on BEL7402, BEL7402-cDNA3 and BEL7402-HBx cells. Under treatment of the same concentration of TRAIL, BEL7402-HBx had a higher apoptosis rate and a higher level of Caspase 3 activation than BEL7402 and BEL7402-cDNA3. TUENL assay showed that the apoptosis rate of BEL7402-HBx induced by 10 μg/L TRAIL was 41.4%±7.2%, signifi- cantly higher than that of BEL7402 and BEL7402-cDNA3 cells. Blockade of HBx expression in Hep G2.2.15 cells partly inhibited the apoptosis induced by TRAIL. The introduction or blockade of HBx did not change the expression pattern of TRAIL receptors. The present study firstly con- firms the effects of HBx on TRAIL- induced apoptosis from two different points and it is not re- lated with the expression level of TRAIL receptors. 3Dark solitons in WS_2 erbium-doped fiber lasers显示文摘Tungsten disulfide(WS_2) is a type of anisotropic-layered compound and has broadband saturable absorption features as saturable absorbers(SAs). With WS_2-based SAs, dark solitons in erbium-doped fiber(EDF) lasers are first obtained. For the generated dark solitons, the center wavelength is measured to be 1530 nm, and the repetition rate is about 116.5 MHz. A series of optical spectra is exhibited. 4Using an Improved Clustering Method to Detect Anomaly Activities显示文摘In this paper, an improved k-means based clustering method (IKCM) is proposed. By refining the initial cluster centers and adjusting the number of clusters by splitting and merging procedures, it can avoid the algorithm resulting in the situation of locally optimal solution and reduce the number of clusters dependency. The IKCM has been implemented and tested. We perform experiments on KDD-99 data set. The comparison experiments with H-means+also have been conducted. 7Study on rolling process optimization of high carbon steel wire显示文摘The existing problems in the manufacture of SWRH82B high carbon steel wire were discussed by sampling and testing the microstructure and properties of the steel from the workshop. To solve the problems, the experimental parameters for thermal simula- tion were optimized, and the thermal simulating experiments were carried out on a Gleeble1500 thermal simulator. The process pa- rameters for the manufacture were optimized after analysis of the data, and the productive experiments were performed after the wa- ter box in front of the no-twist blocks was reconstructed, to control the temperature of the loop layer. The results from the productive experiments showed that the cooling rate of 10-15°C/s was reasonable before phase transformation, about 5°C/s during phase trans- formation, and 600-620°C was the suitable starting temperature for phase transformation. 8Temporal-spatial distributions of road silt loadings and fugitive road dust emissions in Beijing from 2019 to 2020显示文摘Road silt loading(sL)is an important parameter in the fugitive road dust(FRD)emissions.In this study,the improved Testing Re-entrained Aerosol Kinetic Emissions from Roads(TRAKER)combined with the AP-42 method was firstly developed to quickly measure and estimate the sLs of paved roads in Beijing,China.The annual average sLs in Beijing was 0.59±0.31 g/m^(2)in 2020,and decreased by 22.4%compared with that in 2019.The seasonal variations of sLs followed the order of spring>winter>summer>autumn in the two years.The seasonal mean road sLs on the same type road in the four seasons presented a decline trend from^(2)019 to 2020,especially on the Expressway,decreasing 47.4%-72.7%.The road sLs on the different type roads in the same season followed the order of Major arterial∼Minor arterial∼Branch road>Express road,and Township road∼Country highway>Provincial highway∼National highway.The emission intensities of PM10 and PM_(2.5)from FRD in Beijing in 2020 were lower than those in 2019.The PM10 and PM_(2.5)emission intensities at the four planning areas in the two years all presented the order of the capital functional core area>the urban functional expansion area>the urban development new area>the ecological conservation and development area.The annual emissions of PM10 and PM_(2.5)from FRD in Beijing in 2020 were 74,886 ton and 18,118 ton,respectively,decreasing by∼33.3%compared with those in 2019.Haibin Wang Lihui Han Tingting Li Song Qu Yuncheng Zhao Shoubin Fan Tong Chen Haoran Cui Junfang Liu 2023Journal of Environmental Sciences2023,,10:1

13Development of P genome-specific SNPs and their application in tracing Agropyron cristatum introgressions in common wheat显示文摘As an important wild relative of wheat, Agropyron cristatum has been successfully used for wheat improvement. Currently, a few useful agronomic traits of A. cristatum, such as high grain number per spike and resistance to diseases, have been transferred into common wheat.However, the effective detection of small A. cristatum segmental introgressions in common wheat is still difficult. The objective of this study was to identify A. cristatum-specific single nucleotide polymorphisms(SNPs) for the detection of small alien segments in wheat. The transcriptome sequences of A. cristatum were aligned against wheat coding DNA sequences(CDS) for SNP calling. As a result, we discovered a total of 167,613 putative SNPs specific to the P genome of A. cristatum compared with the common wheat genomes. Among 230 selected SNPs with functional annotations related to inflorescence development and stress resistance,68 were validated as P genome-specific SNPs in multiple wheat backgrounds using Kompetitive Allele Specific PCR(KASP) assays. Among them, 55 SNPs were assigned to six homoeologous groups of the P genome using wheat-A. cristatum addition lines, and 6 P-specific SNP markers were further physically mapped on different segments of chromosome 6 P in 6 P translocation lines. The P genome-specific SNPs were also validated by Sanger sequencing and used to detect the P chromatin in wheat-A. cristatum cryptic introgression lines. Two SNP markers(Unigene20217-182 and Unigene20307-1420) were detected in two wheat-A. cristatum introgression lines that showed enhanced grain number per spike and high resistance to powdery mildew. 18EUV-dependence of Venusian dayside ionopause altitude:VEX and PVO observations显示文摘The Venusian dayside ionosphere, similar to other planetary ionospheres, is produced primarily by ionization of its neutral upper atmosphere due to solar extreme ultraviolet (EUV) radiation. It has become clear that the expansion of the ionosphere may be strongly controlled by the EUV level, as exhibited in data collected by the Pioneer Venus Orbiter (PVO) during one solar cycle (1978 1992). However, the EUV-dependence of the Venusian dayside ionopause altitude, which defines the outer boundary of the ionosphere, remains obscure because the PVO crossed the dayside ionopause only during the solar maximum;its periapsis lifted too high during the solar minimum. Recently, during the period 2006 2014, which included the longest and quietest solar minimum of the past several decades, Venus Express (VEX) provided measurements of the photoelectron boundary (PEB) over the northern high-latitude region. Since the photoelectron boundary is closely related to the ionopause, we have an opportunity to analyze the EUV effect on the dayside ionopause by combining PVO and VEX observations. We have evaluated and then reduced the orbit bias effect in data from both PVO and VEX, and then used the results to derive a relationship between solar EUV level and the dayside ionopause altitude. 20Introgression of chromosome 6PL terminal segment from Agropyron cristatum to increase both grain number and grain weight in wheat显示文摘Agropyron cristatum(2n=4x=28,PPPP),which harbours many high-yield and disease-resistance genes,is a promising donor for wheat improvement.Narrow genetic diversity and the trade-off between grain weight and grain number have become bottlenecks for increasing grain yield in wheat.In this study,a novel translocation line,WAT650l,was derived from the chromosome 6P addition line 4844–12,which can simultaneously increase both grain number per spike(GNS)and thousand-grain weight(TGW).Cytological analysis and molecular marker analysis revealed that WAT650l was a 5BL.5BS-6PL(bin 12–17)translocation line.Assessment of agronomic traits and analysis of the BC4F2 and BC5F2 populations suggested that the 6PL terminal chromosome segment in WAT650l resulted in increased grain number per spike(average increased by 14.07 grains),thousand-grain weight(average increased by 4.31 g),flag leaf length,plant height,spikelet number per spike and kernel number per spikelet during the two growing seasons of 2020–2021 and 2021–2022.Additionally,the increased GNS locus and high-TGW locus of WAT650l were mapped to the bins 16–17 and 12–13,respectively,on chromosome 6PL by genetic population analysis of three translocation lines.In summary,we provide a valuable germplasm resource for broadening the genetic base of wheat and overcoming the negative relationship between GNS and TGW in wheat breeding.Yida Lin Shenghui Zhou Xuezhong Liang Bing Han Junli Yang Baojin Guo Jinpeng Zhang Haiming Han Weihua Liu Xinming Yang Xiuquan Li Lihui Li 2023The Crop Journal2023,11,3:1
