| 3 | Functional assembly of root-associated microbial consortia improves nutrient efficiency and yield in soybean显示文摘Root-associated microbes are critical for plant growth and nutrient acquisition. However, scant information exists on optimizing communities of beneficial root-associated microbes or the mechanisms underlying their interactions with host plants. In this report, we demonstrate that rootassociated microbes are critical influencers of host plant growth and nutrient acquisition. Three synthetic communities(SynComs) were constructed based on functional screening of 1,893 microbial strains isolated from root-associated compartments of soybean plants. Functional assemblage of SynComs promoted significant plant growth and nutrient acquisition under both N/P nutrient deficiency and sufficiency conditions.Field trials further revealed that application of SynComs stably and significantly promoted plant growth, facilitated N and P acquisition, and subsequently increased soybean yield. Among the tested communities, SynCom1 exhibited the greatest promotion effect, with yield increases of up to 36.1% observed in two field sites. Further RNA-seq implied that SynCom application systemically regulates N and P signaling networks at the transcriptional level, which leads to increased representation of important growth pathways, especially those related to auxin responses. | 5 | NO66 overexpression rescues ethanol-induced cell apoptosis in human AC16 cardiomyocytes by suppressing PTEN and activating the PI3K/Akt signaling显示文摘Previously,Nucleolar protein 66(NO66)was reported to be closely associated with alcohol exposure-induced injury.However,the role of NO66 in alcohol-induced cytotoxicity remains unclear.In this study,we explored the potential effect and mechanism of NO66 on ethanol-induced apoptosis in human AC16 cardiomyocytes.The AC16 cell lines with NO66 and phosphatase and tensin homolog(PTEN)overexpression were constructed.Cell counting kit-8(CCK-8),lactate dehydrogenase(LDH)assay,Annexin V-FITC/PI staining,and flow cytometry were used to evaluate the cell viability,membrane damage,and apoptosis,respectively.Quantitative real-time PCR(qRT-PCR)and western blot analysis were applied to measure mRNA and protein expression.The results showed that acute ethanol exposure markedly augmented cytotoxicity and reduced NO66 level in AC16 cardiomyocytes.Overexpression of NO66 partially reversed ethanol-induced apoptosis.NO66 upregulation reversed the decrease in phosphorylation of protein kinase B(Akt)and B-cell lymphoma-2/Bcl-2-associated x(Bcl-2/Bax)ratio and the increase in PTEN,p53,and caspase-3 activity induced by ethanol treatment.Meanwhile,the application of PI3K inhibitor(LY294002)and PTEN overexpression attenuated the inhibition efficiency of NO66 on cell apoptosis.In addition,PTEN overexpression weakened the effect of NO66 on PI3K/Akt activation,without affecting the level of NO66.Our data suggested that NO66 overexpression might play an anti-apoptotic role in ethanol-induced cell injury via reducing PTEN and upregulating the PI3K/Akt pathway. | Hanyu Deng Bo Yu Yang Yu Ge Tian Liu Yang | 2020 | Acta Biochimica et Biophysica Sinica2020,52,10: | 3 |
| 6 | Transferring quantum entangled states between multiple single-photon-state qubits and coherent-state qubits in circuit QED显示文摘We present a way to transfer maximally-or partially-entangled states of n single-photon-state(SPS)qubits onto ncoherent-state(CS)qubits,by employing 2nmicrowave cavities coupled to a superconducting flux qutrit.The two logic states of a SPS qubit here are represented by the vacuum state and the single-photon state of a cavity,while the two logic states of a CS qubit are encoded with two coherent states of a cavity.Because of using only one superconducting qutrit as the coupler,the circuit architecture is significantly simplified.The operation time for the state transfer does not increase with the increasing of the number of qubits.When the dissipation of the system is negligible,the quantum state can be transferred in a deterministic way since no measurement is required.Furthermore,the higher-energy intermediate level of the coupler qutrit is not excited during the entire operation and thus decoherence from the qutrit is greatly suppressed.As a specific example,we numerically demonstrate that the high-fidelity transfer of a Bell state of two SPS qubits onto two CS qubits is achievable within the present-day circuit QED technology.Finally,it is worthy to note that when the dissipation is negligible,entangled states of n CS qubits can be transferred back onto n SPS qubits by performing reverse operations.This proposal is quite general and can be extended to accomplish the same task,by employing a natural or artificial atom to couple 2nmicrowave or optical cavities. | Qi-Ping Su Hanyu Zhang Chui-Ping Yang | 2021 | Frontiers of physics2021,16,6: | 2 |
| 7 | Comparative analysis of membranous and other nephropathy subtypes and establishment of a diagnostic model显示文摘This study aimed to compare clinical features between membranous nephropathy (MN) and nonmembranous nephropathy (non-MN), to explore the clinically difTerential diagnosis of these two types, and to establish a diagnostic model of MN. After renal biopsy was obtained, 798 patients were divided into two groups based on their examination results: primary MN group (n = 248) and non-MN group (n = 550). Their data were statistically analyzed. Logistic regression analysis indicated that anti-PLA2R antibodies, IgG, and Cr were independently correlated with MN, and these three parameters were then used to establish the MN diagnostic model. A receiver operating characteristic (ROC) curve confirmed that our diagnostic model could distinguish between patients with and without MN, and their corresponding sensitivity, specificity, and AUC were 79.9%, 89.4%, and 0.917, respectively. The cutoff value for this combination in MN diagnosis was 0.34. Hereinto, Streptococcus mutans(S. mutans)is the most important cariogenic bacteria. In this work, MIC behavior of a new 304-Cu SS in presence of S. mutans was studied by the observations using scanning electron microscopy(SEM) and confocal laser scanning microscopy(CLSM) including live/dead staining, extracellular polymeric substance(EPS)staining and pitting corrosion, electrochemical test, and X-ray photoelectron spectroscopy(XPS). Above results showed that 304-Cu SS possessed excellent biofilm inhibition ability and presented lower corrosion current density(icorr), larger polarization resistance(Rp) and charge transfer resistance(Rct) in the presence of S. mutans, indicating that 304-Cu SS had a better MIC resistance against S. mutans. | 10 | Pre-Drilling Prediction Techniques on the High-Temperature High-Pressure Hydrocarbon Reservoirs Offshore Hainan Island,China显示文摘Decreasing the risks and geohazards associated with drilling engineering in high-temperature high-pressure(HTHP) geologic settings begins with the implementation of pre-drilling prediction techniques(PPTs). To improve the accuracy of geopressure prediction in HTHP hydrocarbon reservoirs offshore Hainan Island, we made a comprehensive summary of current PPTs to identify existing problems and challenges by analyzing the global distribution of HTHP hydrocarbon reservoirs, the research status of PPTs, and the geologic setting and its HTHP formation mechanism. Our research results indicate that the HTHP formation mechanism in the study area is caused by multiple factors, including rapid loading, diapir intrusions, hydrocarbon generation, and the thermal expansion of pore fluids. Due to this multi-factor interaction, a cloud of HTHP hydrocarbon reservoirs has developed in the Ying-Qiong Basin, but only traditional PPTs have been implemented, based on the assumption of conditions that do not conform to the actual geologic environment, e.g., Bellotti's law and Eaton's law. In this paper, we focus on these issues, identify some challenges and solutions, and call for further PPT research to address the drawbacks of previous works and meet the challenges associated with the deepwater technology gap. | 11 | Quantification of traffic-induced compaction based on soil and agricultural implement parameters显示文摘Vehicle-induced soil compaction occurs when agricultural machinery is working in the fields.The accumulated soil compaction could destroy soil structure and inhibit crop growth.The low degree of visualization of soil compaction has always been an important reason for restricting the development of compaction alleviation technology.Therefore,the main objective of this study was to predict soil compaction based on soil and agricultural implement parameters.The component of soil compaction prediction includes traffic-induced stress transmission evaluation and the quantitative relationship between soil stress and bulk density.The modified FRIDA model was used to elucidate the soil stress propagation,which has been validated by previous studies.The Bailey formula was used to establish the intrinsic relationship between soil stress and bulk density.The soil uniaxial compression test was applied to obtain the parameters of the Bailey formula,and soil samples were prepared with three different levels of water content.After fitting with the Bailey formula,under the condition that the soil moisture contents were 16%,20%,and 24%,the fitting coefficients of soil bulk density were respectively 0.980,0.959,and 0.975,which were close to 1.The results indicated that the Bailey formula could be used to calculate soil bulk density based on the stress conditions of the soil.To verify the practicality of the soil compaction prediction model,a field experiment was carried out in Zhuozhou City,Hebei Province,China.The treatment was set for 1,3,5,7,and 9 times compaction with two different loads of compaction equipment.The results showed that the fit coefficient between the predicted and measured values of soil bulk density was greater than 0.641.The slope of the equation was greater than 0.782,proving that the soil bulk density prediction model based on agricultural implements and soil parameters has a good predictive effect on soil bulk density.The soil compaction evaluation model can provide a theoretical basis to further understand the soil compaction mechanism,allowing rational measures of soil compaction alleviation to be made. | Xianliang Wang Xiangcai Zhang Xiaona Lin Luming Sha Hanyu Yang Zhaoyang Guo Zeyu Chen Rongyan Sun | 2020 | International Journal of Agricultural and Biological Engineering2020,13,5: | 1 |
| 12 | PXR activation impairs hepatic glucose metabolism partly via inhibiting the HNF4α-GLUT2 pathway显示文摘Drug-induced hyperglycemia/diabetes is a global issue. Some drugs induce hyperglycemia by activating the pregnane X receptor(PXR), but the mechanism is unclear. Here, we report that PXR activation induces hyperglycemia by impairing hepatic glucose metabolism due to inhibition of the hepatocyte nuclear factor 4-alpha(HNF4 a)-glucose transporter 2(GLUT2) pathway. The PXR agonists atorvastatin and rifampicin significantly downregulated GLUT2 and HNF4 a expression, and impaired glucose uptake and utilization in HepG2 cells. Overexpression of PXR downregulated GLUT2 and HNF4 a expression, while silencing PXR upregulated HNF4 a and GLUT2 expression.Silencing HNF4 a decreased GLUT2 expression, while overexpressing HNF4 a increased GLUT2 expression and glucose uptake. Silencing PXR or overexpressing HNF4 a reversed the atorvastatininduced decrease in GLUT2 expression and glucose uptake. In human primary hepatocytes, atorvastatin downregulated GLUT2 and HNF4 a mRNA expression, which could be attenuated by silencing PXR. Silencing HNF4 a downregulated GLUT2 mRNA expression. These findings were reproduced with mouse primary hepatocytes. Hnf4 a plasmid increased Slc2 a2 promoter activity. Hnf4 a silencing or pregnenolone-16 a-carbonitrile(PCN) suppressed the Slc2 a2 promoter activity by decreasing HNF4 a recruitment to the Slc2 a2 promoter. Liver-specific Hnf4 a deletion and PCN impaired glucose tolerance and hepatic glucose uptake, and decreased the expression of hepatic HNF4 a and GLUT2. In conclusion, PXR activation impaired hepatic glucose metabolism partly by inhibiting the HNF4 aGLUT2 pathway. | 19 | Predicting the structure and stability of titanium oxide electrides显示文摘The search for new inorganic electrides has attracted significant attention due to their potential applications in transparent conductors,battery electrodes,electron emitters,as well as catalysts for chemical synthesis.However,only a few inorganic electrides have been successfully synthesized thus far,limiting the variety of electride examples.Here,we show the stabilization of inorganic electrides in the Ti-rich Ti–O system through first-principles calculations in conjunction with swarm-intelligence-based CALYPSO method for structure prediction.Besides the known Ti-rich stoichiometries of Ti_(2)O,Ti_(3)O,and Ti_(6)O,two hitherto unknown Ti_(4)O and Ti_(5)O stoichiometries are predicted to be thermodynamically stable at certain pressure conditions.We found that these Tirich Ti–O compounds are primarily zero-dimensional electrides with excess electrons confined in the atom-sized lattice voids or between the cationic layers playing the role as anions.The underlying mechanism behind the stabilization of electrides has been rationalized in terms of the excess electrons provided by Ti atoms and their accommodation of excess electrons by multiple cavities and layered atomic packings.The present results provide a viable direction for searching for practical electrides in the technically important Ti–O system. | Xin Zhong Meiling Xu Lili Yang Xin Qu Lihua Yang Miao Zhang Hanyu Liu Yanming Ma | 2018 | npj Computational Materials2018,,1: | 0 |
