| 1 | SlMYB75,an MYB-type transcription factor, promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality. However, the use of a single gene to impact several metabolic pathways is difficult. Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits. In these engineered fruits, the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage, and these SlMYB75-OE tomatoes displayed a series of physiological changes, including delayed ripening and increased ethylene production. In addition to anthocyanin, the total contents of phenolics, flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times, respectively, compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles, such as aldehyde,phenylpropanoid-derived and terpene volatiles, were significantly increased in SlMYB75-OE fruits, with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment, transcriptomic profiling indicated that the genes involved in the ethylene signaling, phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes. The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality. | Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li | 2019 | Horticulture Research2019,6,1: | 39 |
| 2 | rMVP: A Memory-efficient, Visualization-enhanced, and Parallel-accelerated Tool for Genome-wide Association Study显示文摘Along with the develoipment of high-throughput sequencing technologies, both sample size and SNP number are increasing rapidly in genome-wide association studies(GWAS),and the associated computation is more challenging than ever. Here,we present a memory-efficient,visualization-enhanced,and parallel-accelerated R package called“r MVP”to address the need for improved GWAS computation.r MVP can 1)effectively process large GWAS data,2)rapidly evaluate population structure,3)efficiently estimate variance components by Efficient Mixed-Model Association e Xpedited(EMMAX),Factored Spectrally Transformed Linear Mixed Models(Fa ST-LMM),and Haseman-Elston(HE)regression algorithms,4)implement parallel-accelerated association tests of markers using general linear model(GLM),mixed linear model(MLM),and fixed and random model circulating probability unification(Farm CPU)methods,5)compute fast with a globally efficient design in the GWAS processes, and 6)generate various visualizations of GWASrelated information.Accelerated by block matrix multiplication strategy and multiple threads, the association test methods embedded in r MVP are significantly faster than PLINK,GEMMA,and Farm CPU_pkg.r MVP is freely available at http://gffzz188fe103f8f1460aspcbffc56k5566kpw.ffgz.tsg.suse.edu.cn/xiaolei-lab/r MVP. | Lilin Yin Haohao Zhang Zhenshuang Tang Jingya Xu Dong Yin Zhiwu Zhang Xiaohui Yuan Mengjin Zhu Shuhong Zhao Xinyun Li Xiaolei Liu | 2021 | Genomics, Proteomics & Bioinformatics2021,19,4: | 12 |
| 3 | Natural flavonoid luteolin promotes the differentiation of porcine myoblasts through activation of PI3K/Akt/mTOR signaling显示文摘A key step in the manufacturing of cultured meat is to produce myotubes by induced myogenic differentiation. The development of effective,low-cost,and food-safe components that promote the in vitro differentiation of myoblasts is essential for the industrialization of cultured meat.Flavonoids are a class of plant secondary metabolites with various biological activities, but their effects on the regulation of myoblast behaviors are a lack of study. In this study,we selected four representative flavonoids including luteolin, chrysin, apigenin, and genistein, and investigated their effects on porcine myoblasts in the aspects of proliferation, migration, and differentiation. The results showed that four flavonoids all had relatively low cell toxicity but weak ability to promote the proliferation of porcine myoblasts.A positive effect of luteolin was observed in the migration and differentiation of porcine myoblasts,as indicated by improved migration rate and fusion index,as well as upregulated expression of Myogenin and MyHC.Pharmacological inhibition of PI3K activity attenuated the efficacy of luteolin on porcine myoblasts, and further analysis showed that luteolin increased the phosphorylation of PI3K,Akt,and mTOR,indicating the activation of the PI3K/Akt/mTOR signaling pathway. In conclusion, these findings showed that luteolin promoted the migration and differentiation of porcine myoblast via the activation of PI3K/Akt/mTOR signaling, providing biological evidence for its application in cultured meat production. | Xin Guan Zihe Pan Zhuoya Xu Sidun Zhang Haohao Tang Guocheng Du Jingwen Zhou | 2022 | Food Bioscience2022,47,3: | 1 |
| 4 | Large-area periodic lead halide perovskite nanostructures for lenticular printing laser displays显示文摘Lenticular printing technique provides a promising way to realize stereoscopic displays, especially, when microscopic optical structures are integrated into light-emitting materials/devices.Here,we fabricated large-area periodic structures with a spatial resolution at a wavelength scale from hybrid perovskite materials via a space-confined solution growth method. It takes advantages of both high refractive index contrast and high luminescence brightness, which allows the optical modulation on not only the reflection of illumination, but also the light emission from hybrid perovskites. The distributed feedback within these periodic structures significantly improves the degree of polarization and directionality of laser actions while their threshold is also reduced. These findings enable us to present a prototype of lenticular printing laser displays that vary emission colors at different view angles, which may find applications in creating high-resolution and high-contrast holographical images. | Min Wang Haohao Li Chenghu Dai Ji Tang Baipeng Yin Hong Wang Jingwen Li Yuchen Wu Chuang Zhang Yong Sheng Zhao | 2021 | Science China Chemistry2021,64,4: | 0 |
