1Characterization of the receptor-binding domain(RBD)of 2019 novel coronavirus:implication for development of RBD protein as a viral attachment inhibitor and vaccine显示文摘The outbreak of Coronavirus Disease 2019(COVID-19)has posed a serious threat to global public health,calling for the development of safe and effective prophylactics and therapeutics against infection of its causative agent,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),also known as 2019 novel coronavirus(2019-nCoV).The CoV spike(S)protein plays the most important roles in viral attachment,fusion and entry,and serves as a target for development of antibodies,entry inhibitors and vaccines.Here,we identified the receptor-binding domain(RBD)in SARS-CoV-2 S protein and found that the RBD protein bound strongly to human and bat angiotensin-converting enzyme 2(ACE2)receptors.SARS-CoV-2 RBD exhibited significantly higher binding affinity to ACE2 receptor than SARS-CoV RBD and could block the binding and,hence,attachment of SARS-CoV-2 RBD and SARS-CoV RBD to ACE2-expressing cells,thus inhibiting their infection to host cells.SARS-CoV RBD-specific antibodies could crossreact with SARS-CoV-2 RBD protein,and SARS-CoV RBD-induced antisera could cross-neutralize SARS-CoV-2,suggesting the potential to develop SARS-CoV RBD-based vaccines for prevention of SARS-CoV-2 and SARS-CoV infection.Wanbo Tai Lei He Xiujuan Zhang Jing Pu Denis Voronin Shibo Jiang Yusen Zhou Lanying Du 2020Cellular & Molecular Immunology2020,17,6:35

4Powder injection molding of pure titanium显示文摘An improved wax-based binder was developed for powder injection molding of pure titanium. A critical powder loading of 69 vol.% and a pseudo-plastic flow behavior were obtained by the feedstock based on the binder. The injection molding, debinding, and sintering process were studied. An ideal control of carbon and oxygen contents was achieved by thermal debinding in vacuum atmosphere (10-3 Pa). The mechanical properties of as-sintered specimens were less than those of titanium made by the conventional press-sintering process. 5Abnormal expression of bHLH3 disrupts a flavonoid homeostasis network,causing differences in pigment composition among mulberry fruits显示文摘Mulberry fruits with high concentrations of anthocyanins are favored by consumers because of their good taste,bright color,and high nutritional value.However,neither the regulatory mechanism controlling flavonoid biosynthesis in mulberry nor the molecular basis of different mulberry fruit colors is fully understood.Here,we report that a flavonoid homeostasis network comprising activation and feedback regulation mechanisms determines mulberry fruit color.In vitro and in vivo assays showed that MYBA-bHLH3-TTG1 regulates the biosynthesis of anthocyanins,while TT2L1 and TT2L2 work with bHLH3 or GL3 and form a MYB-bHLH-WD40(MBW)complex with TTG1 to regulate proanthocyanidin(PA)synthesis.Functional and expression analyses showed that bHLH3 is a key regulator of the regulatory network controlling mulberry fruit coloration and that MYB4 is activated by MBW complexes and participates in negative feedback control of the regulatory network to balance the accumulation of anthocyanins and proanthocyanidins.Our research demonstrates that the interaction between bHLH3 and MYB4 in the homeostasis regulatory network ensures that the fruits accumulate desirable flavonoids and that this network is stable in pigmentrich mulberry fruits.However,the abnormal expression of bHLH3 disrupts the balance of the network and redirects flavonoid metabolic flux in pale-colored fruits,resulting in differences in the levels and proportions of anthocyanins,flavones,and flavonols among differently colored mulberry fruits(red,yellow,and white).The results of our study reveal the molecular basis of the diversity of mulberry fruit colors.Han Li Zhen Yang Qiwei Zeng Shibo Wang Yiwei Luo Yan Huang Youchao Xin Ningjia He 2020Horticulture Research2020,7,1:8

7A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice,rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines.Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge,even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases.Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu 2022Cell Research2022,32,3:5

12Study on Intervention of Blood Sugar and Blood Lipid in Diabetic Mice by Abrus Cantoniensis and Armeniaca mume Sieb and Their Compatibility显示文摘Objective:The study aims to explore the interventional effect of Abrus cantonensis and Armeniaca mume Sieb and their compatibility on blood glucose and blood lipid in diabetic mice and to explore the optimal ratio of efficacy of their compatibility.Methods:SPF Kunming male mice were fed with high sugar and high fat diet for 6 weeks and then intraperitoneally injected with STZ solution to establish a diabetes model.The successfully modeled mice were randomly divided into 6 groups according to their body weight:model group,Abrus Cantoniensis group,Armeniaca mume Sieb group,as well as Abrus Cantoniensis and Armeniaca mume Sieb group divided into 1:2,1:1 and 2:1 groups according to volume ratio with 10 mice in each group.Another 10 healthy mice were taken as blank group and intraperitoneally injected with sodium citrate buffer.The corresponding solution was given intragastric administration by body mass once a day for 20 days.After the last administration,fasting was performed for 12 hours.Serum was collected by the method of eyeball blood collection to determine the contents of fasting blood glucose,total cholesterol Triacylglycerol,triglyceride,low density lipoprotein and high-density lipoprotein.Results:Compared with Abrus Cantoniensis and Armeniaca mume Sieb groups,Glu,TG,TC and LDL-C of the three compatibility groups significantly decreased,with the most significant decrease in the compatibility group 1:2,while there was no significant difference in HDL-C among all groups.Conclusion:Abrus Cantoniensis and Armeniaca mume Sieb and their combination have synergistic effects on the improvement of blood glucose and lipid levels in mice with high glucose and high fat combined with STZ-induced diabetes mellitus model.Among all the experimental groups,the compatibility 1:2 group had the best effect.Yixin Zhang Shibo Zhao Sitong Lu Danni Lu Yedong Shen Changhuan He Zhonggang Wu Suoyi Huang 2021TMR Pharmacology Research2021,1,4:1

14Tunable light emission from carbon dots by controlling surface defects显示文摘Carbon dots(CDs)are metal-free fluorescent materials that can be used in optical and electronic devices,but few studies have focused on one-step synthesis routes for CDs with tunable color and high photoluminescence quantum yield(PLQY).Herein,CDs with tunable light emission were synthesized using a novel amide-assisted solvothermal approach.The as-prepared CDs were well dispersed and homogeneous,with average diameters of approximately 2.0–4.0 nm,depending on the dopants.Owing to the surface states with different ratios of nitrogen-and oxygen-related species,different CDs can exhibit blue,green,red,or white emission with relatively high PLQYs of 61.6%,41.3%,29.1%and 19.7%,respectively.XPS measurements,in conjunction with DFT calculations,indicate that nitrogen substitution(pyridinic/pyrrolic nitrogen)dominates the blue emission,while introducing oxygen functional groups lowered the LUMO energy level,which resulted in redder emission.In addition,the CDs are demonstrated as a bioimaging probe in both in vitro and in vivo assays,and the white light CDs have been demonstrated to be potential fluorescent materials for white-light-emitting diode(WLED).Huijun Li Sancan Han Bowen Lyu Ting Hong Shibo Zhi Ling Xu Fengfeng Xue Liman Sai Junhe Yang Xianying Wang Bin He 2021Chinese Chemical Letters2021,32,9:1

20“Three medicines and three formulas”in COVID-19:from bench to bedside显示文摘Chinese guideline has proven effective in the fight against coronavirus disease 2019(COVID-19)during the global spread of the epidemic.Traditional Chinese medicine(TCM)is widely recognized for its effectiveness in alleviating symptoms,inhibiting disease progress,reducing mortality,and improving the cure rate of patients with COVID-19.During the pandemic,“three medicines and three formulas”(TMTFs)stood out from hundreds of others in registered clinical studies,which became highly recommended as TCM for COVID-19 treatment.The TMTFs not only effectively relieve the clinical symptoms of fever,cough,fatigue,and phlegm,but also substantially shorten the time of nucleic acid negative conversion,improve lung computed tomography imaging features and inflammation,ameliorate clinical biochemical indicators,and reduce sequelae.Their potential pharmacological mechanisms are mainly related to the crosstalk between viral toxicity,endothelial damage,cytokine storm,immune response,and microthrombus.In brief,the clinical effects and the potential mechanisms of TMTFs on COVID-19 were systematically analyzed and summarized covering the entirety of disease development,including virus invasion and replication,immune response and cytokine storm,and acute respiratory distress syndrome and multiple organ dysfunction syndrome.This review provides a theoretical basis and reference for the in-depth understanding of the positive role of TMTFs in COVID-19 treatment.Guangxu Xiao Simeng Wang Ruijiao Wang Li Tan Shibo Ma Shuang He Guanwei Fan Yan Zhu Ming Lyu 2023Acupuncture and Herbal Medicine2023,3,4:1
