| 1 | Construction, expression and characterization of the engineered antibody against tumor surface antigen, p185^(c-erbB-2)显示文摘The c-erbB-2 proto-oncogene encodes a 185kDa protein p185, which belongs to epidermal growth factorreceptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis forcertain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibitsproliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. Inorder to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed thesingle-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichiacoli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cellimmunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain(ECD) of p185. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion moleculewith a homodimer form and the recombinant product was expressed in mammalian cells. In a series ofsubsequent analysis this fusion protein showed identical antigen binding site and activity with the parentantibody. | 2 | Syntheses of amphiphilic biodegradable copolymers of poly(ethyl ethylene phosphate) and poly(3-hydroxybutyrate) for drug delivery显示文摘Biodegradable and amphiphilic triblock copolymers poly(ethyl ethylene phosphate)-poly(3-hydroxybutyrate)-poly(ethyl ethylene phosphate) (PEEP-b-PHB-b-PEEP) have been successfully synthesized through ring-opening polymerization. The structures are confirmed by gel permeation chromatography and NMR analyses. Crystallization investigated by X-ray diffraction reveals that the block copolymer with higher content of poly(ethyl ethylene phosphate) (PEEP) is more amorphous, showing decreased crystallizability. The obtained copolymers self-assemble into biodegradable nanoparticles with a coreshell micellar structure in aqueous solution, verified by the probe-based fluorescence measurements and transmission electronic microscopy (TEM) observation. The hydrophobic poly(3-hydroxybutyrate) (PHB) block serves as the core of the micelles and the micelles are stabilized by the hydrophilic PEEP block. The size and size distribution are related to the compositions of the copolymers. Paclitaxel (PTX) has been encapsulated into the micelles as a model drug and a sustained drug release from the micelles is observed. | 3 | Multi-isotopic system geochronology of low temperature eclogite from Huangzhen,Southern Dabie Terrain显示文摘Zircon U-Pb thermal ionization mass-spectrometer (TIMS) and secondary ion mass-spectrometer (SIMS) dating, mica and amphibole 40Ar-39Ar dating and mineral Sm-Nd isotopic compositions of Huangzhen low temperature eclogite and country granitic gneiss are carried out. The zircon U-Pb weighted average SIMS age is (231.6±9.7) Ma for one eclogite. The mica Ar-39Ar isochron age is (232.6±2.1) Ma and the lowest plateau age is (221.7±2.4) 40 Ma from same sample. U-Pb TIMS concordant ages from other eclogite zircons are from (221.3 ±1.4) Ma to (222.5±2.3) Ma. U-Pb SIMS low intercept age from country granitic gneiss is (221 ±35) Ma. The retrograde amphibole Ar-39Ar isochron age is (205.9±1.0) Ma. Except for mica, 40 which may contain excess Ar, all the ages represent peak and retrograde metamorphism of low 40 temperature eclogites. It is indicated that the Huangzhen low temperature eclogites differ from Xiongdian low temperature eclogites of north of the Northern Dabie Terrain in metamorphic ages. Huangzhen low temperature eclogites share one coherent HP-UHP terrain with high temperature eclogites from Southern Dabie Terrain and they may have differences in subduction depth and cooling rates during exhumation. | CHEN Daogong1, E. | 4 | A high time resolution multi-gap resistive plate chamber显示文摘Research on a single cell Multi-gap Resistive Plate Chamber (MRPC) was carried out for the Time Of Flight detector (TOF) R and D of the STAR experi- ment. The property of the single chamber was tested with the test beam facility T10 at CERN in Geneva. The MRPC was scanned with the test beam to get the efficiency and time resolution at different position of the active area. The HV plateau was also measured. The time resolution is 65 ps which satisfies the requirement of STAR. | 7 | Encounter probability analysis of typhoon and plum rain in the Taihu Lake Basin显示文摘It is of great significance to study the encounter possibility of typhoon and plum rain in the Taihu Lake Basin in order to adjust the schemes of design storm and utilize flood resource and make the decision of real-time scheduling for flood control in the Basin. Based upon the existing research findings of description of encounter phenomena of typhoon and plum rain, the initial time of typhoon affecting the Taihu Lake Basin, the start time of plum rain season and the end time of plum rain season were selected as the characteristic indices of typhoon and plum rain, respectively. According to their synchronous data from 1954 to 2009, the distribution functions of the above-mentioned three random variables were identified and a formula expressing the encounter possibility of typhoon and plum rain was derived. In view of the complexity and solution of this formula, joint distribution functions of typhoon and plum rain were constructed by the use of Gumbel Copula function on the basis of the adaptability comparison among three functions (Gumbel Copula, Clayton Copula and Frank Copula) in Archimedean functions. A new and simple ignition criterion, heat release rate model and mass loss rate model are proposed based on experiments. Combined with these models, the zone-model of fire can be used to predict fire development in a single compartment according to the species of combustibles, their quantities and distribution. The system consists of five essential portions: knowledge-base , inference engine, database, knowledge-acquisition module and explanatory interface. The system is implemented in Turbo PROLOG artificial intelligence language. Compared with other spectral interpretation systems this system is of the following advantages, i. e. the system has a friendly user interface, two kinds of methods for managing toowledge-base, several useful explanatory facilities such as why and how should explanation be made, etc. ; in addition, it can be run on the IBM PC/XT and its compatible microcomputers. | 13 | Metal-organic framework MIL-53(Al): synthesis, catalytic performance for the Friedel-Crafts acylation, and reaction mechanism显示文摘Metal-organic framework MIL-53(Al) was synthesized by a solvothermal method using aluminum nitrate as the aluminium source and 1,4-benzenedicarboxylic acid(H2BDC) as the organic ligand. The structure of samples was characterized by X-ray diffraction(XRD) and Fourier transform infrared spectroscopy(FT-IR). The catalytic activity and recyclability of MIL-53(Al) catalyst for the Friedel-Crafts acylation reaction of indole with benzoyl chloride were evaluated. The reaction conditions were optimized and a reaction mechanism was suggested. The results showed that the MIL-53(Al) catalyst exhibited good catalytic activity and recyclability for the Friedel-Crafts acylation reaction. When the molar ratio of indole and MIL-53(Al) catalyst was 1:0.06(n1:ncatalyst), the molar ratio of indole and benzoyl chloride was 1:3, and the solvent was dichloromethane, the conversion of indole could reach 97.1% and the selectivity of 3-acylindole could reach 81.1% at 25 °C after 8 h. The catalyst can be reused without significant degradation in catalytic activity. Their one-and two-photon absorption (TPA) and excited fluorescence properties have been experimentally investigated. The two-photon absorption cross-sections of two compounds were estimated by two-photon excited fluorescence technique using 200 fs, 76 MHz, Ti:sapphire laser, which are 22 and 154 GM for BCSPC and BCPBC, respectively. The optimal excitation wavelengths are 780 nm for both BCSPC and BCPBC.
