1Response of soil heat-water processes to vegetation cover on the typical permafrost and seasonally frozen soil in the headwaters of the Yangtze and Yellow Rivers显示文摘The response of soil temperature and moisture to vegetative cover in the active layer of permafrost and seasonally frozen soil were assessed and compared. Soil temperature and moisture, under a range of vegetation covers (92%, 65% and 30%) in the permafrost and vegetation covers (95%, 70%―80%, 40%―50% and 10%) in the seasonally frozen soil, were measured on a daily basis. A decline in vege-tation cover led to a decrease in the integral of freezing depth of active permafrost layer, but an increase in seasonally frozen soil. The maximum invasion depth and duration of the negative isotherm during the frozen period and of the positive isotherm during the non-frozen period clearly increased when vegetation cover declined. With a reduction of vegetation cover, the soil moisture in the active layer of the permafrost decreased for depths of 0.20―0.60 m, but increased for depths of 0.60―0.80 m, while for seasonally frozen soil, soil moisture of the entire profile (0.10―1.20 m) increased. 2Neddylation of PTEN regulates its nuclear import and promotes tumor development显示文摘PTEN tumor suppressor opposes the PI3K/Akt signaling pathway in the cytoplasm and maintains chromosomal integrity in the nucleus.Nucleus–cytoplasm shuttling of PTEN is regulated by ubiquitylation,SUMOylation and phosphorylation,and nuclear PTEN has been proposed to exhibit tumor-suppressive functions.Here we show that PTEN is conjugated by Nedd8 under high glucose conditions,which induces PTEN nuclear import without effects on PTEN stability.PTEN neddylation is promoted by the XIAP ligase and removed by the NEDP1 deneddylase.We identify Lys197 and Lys402 as major neddylation sites on PTEN.Neddylated PTEN accumulates predominantly in the nucleus and promotes rather than suppresses cell proliferation and metabolism.The nuclear neddylated PTEN dephosphorylates the fatty acid synthase(FASN)protein,inhibits the TRIM21-mediated ubiquitylation and degradation of FASN,and then promotes de novo fatty acid synthesis.In human breast cancer tissues,neddylated PTEN correlates with tumor progression and poor prognosis.Therefore,we demonstrate a previously unidentified pool of nuclear PTEN in the Nedd8-conjugated form and an unexpected tumor-promoting role of neddylated PTEN.Ping Xie Zhiqiang Peng Yujiao Chen Hongchang Li Mengge Du Yawen Tan Xin Zhang Zhe Lu Chun-Ping Cui Cui Hua Liu Fuchu He Lingqiang Zhang 2021Cell Research2021,31,3:11
3Gα_s Relays Sphingosine-1-Phosphate Receptor 1 Signaling to Stabilize Vascular Endothelial-Cadherin at Endothelial Junctions to Control Mouse Embryonic Vascular Integrity显示文摘Sphingosine-1-phosphate receptor 1(S1PR1),a G protein-coupled receptor(GPCR),controls vascular stability by stabilizing vascular endothelial(VE)-cadherin junctional localization and inhibiting vascular endothelial growth factor receptor 2(VEGFR2) signaling.However,the molecular mechanisms that link S 1PR1 signaling to intracellular effectors remain unknown,In this study,we demonstrate that the heterotrimeric G protein subfamily member Gα_s,encoded by GNAS,acts as a relay mediator of S1PR1 signaling to control vascular integrity by stabilizing VE-cadherin at endothelial junctions.The endothelial cell-specific deletion of Gαs in mice causes early embryonic lethality with massive hemorrhage and a disorganized vasculature.The immunostalning results revealed that Gα_s deletion remarkably reduces the junctional localization of VE-cadherin,whereas the mural cell coverage of the vessels is not impaired.In addition,we found that Gαs depletion blocks the S1PRl-activation induced VE-cadherin stabilization at junctions,supporting that Gα_s acts downstream of S1PR1 signaling,Thus,our results demonstrate that Gα_s is an essential mediator to relay S1PR1 signaling and maintain vascular integrity.Ximing Shao Ke Liu Yi Fan Zhihao Ding Min Chen Minyan Zhu Lee S.Weinstein Hongchang Li Huashun Li 2015Journal of Genetics and Genomics2015,42,11:8

5Ultrasound-guided percutaneous microwave ablationfor small liver cancers adjacent to large vessels:long-term outcomes and strategies显示文摘Objective The aim of the study was to evaluate the long-term efficacy and safety of percutaneous microwave ablation(MWA) for small hepatic cancers adjacent to large vessels and to investigate the treatment strategies.Methods From March 2009 to July 2015,a total of 86 patients with 94 tumors underwent ultrasound(US)-guided percutaneous MWA,with pathologically proven or clinically diagnosed liver cancers measuring ≤ 3 cm in diameter and located ≤ 10 mm from a major vessel(n = 94).Regular follow-up after MWA was performed to assess treatment efficacy and perioperative complications.Results The complete ablation rate at 1 month after MWA was 93.3%(84/90).The 6-,9-,12-,24-,36-,48-,60-,72-,and 84-month local recurrence rates were 2.4%,2.4%,3.7%,6.6%,8.4%,8.4%,8.4%,8.4%,and 8.4%,respectively.There were no major complications.The perioperative special complication rate was 5.32%(5/94),including 3 cases of moderate liver function damage and 2 cases of limited sub-capsular hematoma.Conclusion Percutaneous MWA for small hepatic cancers adjacent to large vessels is feasible,effective,and safe with an acceptable rate of complications.The key point is to strictly follow operative indications and adopt proper treatment strategies.Ruobing Liu Kaiyan Li Hongchang Luo Wei Zhang Tingting Zhang Meng Gao Wenhui Zha Xinwu Cui Youbin Deng 2017Oncology and Translational Medicine2017,3,2:3

7The deubiquitinase OTUD1 inhibits colonic inflammation by suppressing RIPK1-mediated NF-κB signaling显示文摘The E3 ubiquitin ligase(E3)-mediated ubiquitination and deubiquitinase(DUB)-mediated deubiquitination processes are closely associated with the occurrence and development of colonic inflammation.Ovarian tumor deubiquitinase 1(OTUD1)is involved in immunoregulatory functions linked to infectious diseases.However,the effect of OTUD1 on intestinal immune responses during colonic inflammatory disorders such as inflammatory bowel disease(IBD)remains unclear.Here,we show that loss of OTUD1 in mice contributes to the pathogenesis of dextran sulfate sodium(DSS)-induced colitis via excessive release of proinflammatory cytokines.In addition,bone marrow transplantation experiments revealed that OTUD1 in hematopoietic cells plays a dominant role in protection against colitis.Mechanistically,OTUD1 physically interacts with receptor-interacting serine/threonine-protein kinase 1(RIPK1)and selectively cleaves K63-linked polyubiquitin chains from RIPK1 to inhibit the recruitment of NF-κB essential modulator(NEMO).Moreover,the expression of OTUD1 in mucosa samples from ulcerative colitis(UC)patients was lower than that in mucosa samples from healthy controls.Furthermore,we demonstrate that the UC-associated OTUD1 G430V mutation abolishes the ability of OTUD1 to inhibit RIPK1-mediated NF-κB activation and intestinal inflammation.Taken together,our study unveils a previously unexplored role of OTUD1 in moderating intestinal inflammation by inhibiting RIPK1-mediated NF-κB activation,suggesting that the OTUD1-RIPK1 axis could be a potential target for the treatment of IBD.Bo Wu Lihua Qiang Yong Zhang Yesheng Fu Mengyuan Zhao Zehui Lei Zhe Lu Yan-Ge Wei Hongmiao Dai Yingwei Ge Mingqiu Liu Xuemei Zhou Zhiqiang Peng Hongchang Li Chun-Ping Cui Jing Wang Hui Zheng Cui Hua Liu Lingqiang Zhang 2022Cellular & Molecular Immunology2022,19,2:3

9Tuning the nucleation and decomposition of Li2O_(2) by fluorine-doped carbon vesicles towards high performance Li-O_(2) batteries显示文摘Li-O_(2) batteries provide an attractive and potential strategy for energy conversion and storage with high specific energy densities.However,large over-potential in oxygen evolution reactions (OER) caused by the decomposition obstacles of Li_(2)O_(2) seriously impedes its electrochemical performances.Herein,a novel N,O,S and F co-doping vesicular carbon was prepared by self-template pyrolysis method and used in LiO_(2) battery to tune the nucleation and decomposition of Li_(2)O_(2).The introduction of F in the carbon matrix with suitable content can regulate the adsorption of intermediates,through which the morphology of Li_(2)O_(2) can be controlled to film,favorable to its decomposition in charge process.The cathode based on the optimized F doped carbon vesicle exhibits improved electrochemical performances including a low over-potential,large capacity and a long-term stability.Density functional theory (DFT) results show that F and C in C–F bond hasve a strong interaction to Li and O in Li_(2)O_(2),respectively,which can enhance the transfer of electrons from Li_(2)O_(2) to the carbon matrix to generate hole polaron and thus accelerate the delithiation and decomposition of Li_(2)O_(2).This work provides a new sight into understanding the mechanism of nucleation and decomposition of Li_(2)O_(2) for the development of high-performance Li-O_(2) batteries.Shiyu Ma Hongchang Yao Zhongjun Li Qingchao Liu 2022Journal of Energy Chemistry2022,31,7:2

14Microbiologically Influenced Corrosion of Q235 Carbon Steel by Aerobic Thermoacidophilic Archaeon Metallosphaera cuprina显示文摘The effect of thermoacidophilic archaeon Metallosphaera cuprina(M.cuprina)on the corrosion of Q235 carbon steel in its culture medium was investigated in this work.In the sterile culture medium,the carbon steels showed uniform corrosion morphologies and almost no corrosion products covered the sample surface during 14 days of immersion test.In the presence of M.cuprina,some corrosion pits appeared on the surface of carbon steels in the immersion test,exhibiting typical localized corrosion morphologies.Moreover,the sample surfaces were covered by a large number of insoluble precipitates during the immersion.After 14 days,the thickness of precipitates reached approximately 50μm.The results of weight loss test and electrochemical test demonstrated that the carbon steels in the M.cuprina-inoculated culture medium had higher corrosion rate than that in the sterile culture medium.The oxygen concentration cell caused by M.cuprina biofilms resulted in localized corrosion behavior,and the ferrous oxidation ability of M.cuprina accelerated the anodic dissolution of carbon steels,thus promoting the corrosion process of carbon steels.Hongchang Qian Shangyu Liu Wenlong Liu Pengfei Ju Dawei Zhang 2022Acta Metallurgica Sinica(English Letters)2022,35,2:1

16TGA factors promote plant root growth by modulating redox homeostasis or response显示文摘To identify novel regulators of stem cell renewal,we mined an existing but little explored cell typespecific transcriptome dataset for the Arabidopsis root.A member of the TGA family of transcription factors,TGA8,was found to be specifically expressed in the quiescent center(QC).Mutation in TGA8 caused a subtle root growth phenotype,suggesting functional redundancy with other TGA members.Using a promoter::HGFP transgenic approach,we showed that all TGA factors were expressed in the root,albeit at different levels and with distinct spatial patterns.Mutant analyses revealed that all TGA factors examined contribute to root growth by promoting stem cell renewal,meristem activity,and cell elongation.Combining transcriptome analyses,histochemical assays,and physiological tests,we demonstrated that functional redundancy exists among members of cladesⅡandⅤor those in cladesⅠandⅢ.These two groups of TGA factors act differently,however,as their mutants responded to oxidative stress differently and quantitative reverse transcription polymerase chain reaction assays showed they regulate different sets of genes that are involved in redox homeostasis.Our study has thus uncovered a previously unrecognized broad role and a mechanistic explanation for TGA factors in root growth and development.Xiaochen Hu Liyun Yang Mengfei Ren Lin Liu Jing Fu Hongchang Cui 2022Journal of Integrative Plant Biology2022,64,8:1

18Basic fibroblast growth factor improves learning and memory functions in chronic stress mice显示文摘Four weeks of uncertain stress was used to establish an animal model of chronic stress. Basic fibroblast growth factor was injected daily for 15 days following stress induction. Cell morphology in the hippocampal CA3 region of chronic stress mice revealed cell damage. Nitric oxide content and calcium concentration were significantly increased in the hippocampus, and learning and memory functions were significantly decreased. After basic fibroblast growth factor intervention, Ca2+ overload was decreased and neuronal damage was relieved in hippocampal neurons, which improved learning and memory functions in chronic stress mice. 20Integration of scRNA and bulk RNA-sequence to construct the 5-gene molecular prognostic model based on the heterogeneity of thyroid carcinoma endothelial cell显示文摘Thyroid cancer(TC)is a kind of cancer with high heterogeneity,which leads to significant difference in prognosis.The prognostic molecular processes are not well understood.Cancer cells and tumor microenvironment(TME)cells jointly determine the heterogeneity.However,quite a little attention was paid to cells in the TME in the past years.In this study,we not only reveal that endothelial cells(ECs)are strongly associated with the progress of papillary thyroid cancer(PTC)using single-cell RNA-seq(scRNA-seq)data downloaded from Gene Expression Omnibus(GEO)and WGCNA,but also screen 5 crucial genes of ECs:CLDN5,ABCG2,NOTCH4,PLAT,and TMEM47.Fur-thermore,the 5-gene molecular prognostic model is constructed,which can predict how well a patient will do on PD-L1 blockade immunotherapy for TC and evaluate prognosis.Quantitative real-time polymerase chain reaction(qRT-PCR)analysis demonstrates that PLAT is decreased in TC and the increase of PLAT can restrain the migratory capacity of TC cells.Meanwhile,in TC cells,PLAT suppresses VEGFa/VEGFR2-mediated human umbilical vascular endothelial cell(HUVEC)proliferation and tube formation.Totally,we construct the 5-gene molecular prognostic model from the perspective of EC and provide a new idea for immunotherapy of TC.Zhaoxian Ni Shan Cong Hongchang Li Jiazhe Liu Qing Zhang Chuanchao Wei Gaofeng Pan Hui He Weiyan Liu Anwei Mao 2024Acta Biochimica et Biophysica Sinica2024,56,2:0
