A human circulating immune cell landscape in aging and COVID-19显示文摘Age-associated changes in immune cells have been linked to an increased risk for infection. However,a global and detailed characterization of the changes that human circulating immune cells undergo with age is lacking. Here,we combined scRNA-seq,mass cytometry and sCATAC-seq to compare immune cell types in peripheral blood collected from young and old subjects and patients with COVID-19.We found that the immune cell landscape was reprogrammed with age and was characterized by T cell polarization from naive and memory cells to effector, cytotoxic, exhausted and reg-ulatory cells, along with increased late natural killer cells,age-associated B cells, inflammatory monocytes and age-associated dendritic cells. In addition, the expression of genes, which were implicated in coron-avirus susceptibility, was upregulated in a cell subtype-specific manner with age.Notably,COVID-19 promoted age-induced immune cell polarization and gene expression related to inflammation and cellular senes-cence.Therefore,these findings suggest that a dysreg-ulated immune system and increased gene expression associated with SARS-CoV-2 susceptibility may at least partially account for COVID-19 vulnerability in the elderly. | Yingfeng Zheng Xiuxing Liu Wenqing Le Lihui Xie He Li Wen Wen Si Wang Shuai Ma Zhaohao Huang Jinguo Ye Wen Shi Yanxia Ye Zunpeng Liu Moshi Song Weiqi Zhang Jing-Dong J.Han Juan Carlos lzpisua Belmonte Chuanle Xiao Jing Qu Hongyang Wang Guang-Hui Liu Wenru Su | 2020 | Protein & Cell2020,11,10: | 10

Experimental study on enhancement of the metastatic potential of portal vein tumor thrombus-originated hepatocellular carcinoma cells using portal vein serum显示文摘Objective: Portal vein metastasis of hepatocellular carcinoma(HCC) results in a poor prognosis and seriously affects the survival rate of patients. The mechanism underlying the formation of portal vein tumor thrombus(PVTT) is complex and is not yet fully understood. This study was conducted to investigate the impact of portal vein blood on the proliferation, metastasis, invasion and apoptosis of PVTT cells and to explore its possible mechanisms, which was expected to lay a foundation for ascertaining the mechanism underlying the portal vein metastasis of HCC.Methods: Peripheral blood and portal vein blood were collected from patients with HCC, and the sera from these two sources were used to culture the PVTT-originated HCC cell line CSQT-2. The cells were collected after 24 h, and flow cytometry was performed to detect cell proliferation, cell cycle stages and apoptosis. Transwell migration and invasion assays were applied to detect the metastasis and invasion of the cells in each group. The changes in the expression of MMP-2 and MMP-9 in cells were detected via Western blotting. The contents of IL-12, IFN-γ, IL-1β, IL-2 and TNF-α in the two groups of sera were quantified using corresponding kits. Results: Compared with the group of cells cultured with peripheral serum, the cells cultured with portal vein serum showed significantly lower apoptosis(P<0.01), significantly enhanced cell metastasis and invasion(P<0.01), whereas cell proliferation and the stages of the cell cycle did not differ significantly(P>0.05). A significantly increased expression level of MMP-2 has been observed in tumor cells treated portal vein serum. In addition, compared with peripheral serum, the content of IL-12 was significantly decreased in portal vein serum(P<0.05), while the contents of IFN-γ, IL-1β, IL-2, and TNF-α did not differ significantly(P>0.05). Conclusions: Portal vein serum from HCC patients could inhibit the apoptosis of PVTT-originated HCC cells and promote cell metastasis and invasion. This effect may be related to the lower level of IL-12 in portal vein serum. | Yufu Tang Hongming Yu Long Zhang Kang Wang Weixing Guo Jie Shi Shupeng Liu Mengchao Wu Hongyang Wang Shuqun Cheng | 2014 | Chinese Journal of Cancer Research2014,26,5: | 2

Phosphate modified carbon nanotubes for oxidative dehydrogenation of n-butane显示文摘Catalytic performance of phosphate-modified carbon nanotube(PoCNT) catalysts for oxidative dehydrogenation(ODH) of n-butane has been systematically investigated. The Po CNT catalysts are characterized by SEM, TEM, XPS and TG techniques. We set the products selectivity as a function of butane conversion over various phosphate loading, and it is found that the PoCNT catalyst with the 0.8% phosphate weight loading(0.8PoCNT) exhibits the best catalytic performance. When the phosphate loading is higher than 0.8 wt%, the difference of catalytic activity among the PoCNT catalysts is neglectable. Consequently, the ODH of n-butane over the 0.8PoCNT catalyst is particularly discussed via changing the reaction conditions including reaction temperatures, residence time and n-butane/O_2 ratios. The interacting mechanism of phosphate with the oxygen functional groups on the CNT surface is also proposed. | Yajie Zhang Rui Huang Zhenbao Feng Hongyang Liu Chunfeng Shi Junfeng Rong Baoning Zong Dangsheng Su | 2016 | Journal of Energy Chemistry2016,25,3: | 1
Downregulation of metabotropic glutamate receptors mGluR5 and glutamate transporter EAAC1 in the myenteric plexus of the diabetic rat ileum显示文摘Objective: To study the morphologic abnormalities of the myenteric plexus in diabetic rats and to explore the mechanism of their effect on gastrointestinal motility. Methods: Forty rats were randomly divided into a diabetic group and a control group, Gastric emptying and small intestine transit rates were measured and histologic and molecular changes in glutamatergic nerves in the ileal myenteric plexus were observed, mGluR5 receptor and EAAC1 transporter changes in the diabetic rats were studied using fluorescence immunohistochemistry and RT-PCR. Results:Eighteen weeks after the establishment of the diabetic rats model, gastric emptying and small intestine transit rates were found to be significantly delayed in the diabetic group when compared with the control group. The density of glutamatergic ganglia and neurons in the ileal myenterie plexus were significantly decreased in the diabetic group when compared with control group(P < 0.05) and the mGluR5 receptors and EAAC1 transporters were downregulated in the diabetic rats(P < 0.05). Conclusion: Decreased glutamatergic enteric ganglia and neurons and decreased mGluR5 receptors and EAAC1 transporters in the intestinal myenteric plexus is one of the mechanisms of diabetic gastroenteropathy in rats. | Yanhua Bai Jun Zhang Hongyang Shi Fei Dai | 2008 | Journal of Nanjing Medical University2008,22,6: | 0
Nanogenerator-based bidirectional pressure sensor array and its demonstration in underwater invasive species detection显示文摘Lakes areas, which cause catastrophic damages in both commercial fishery and ecological systems. However, current assessment strategies may pose challenges for lake-wide abundance estimation and non-target anadromous species preservation. Here,we demonstrate an efficacious species-specific non-destructive sensing system based on porous ferroelectret nanogenerator for in-situ monitoring of lamprey spawning migration using their unique suction behavior.Simulations show that the porous structure enables a redistribution of surface charges under bidirectional deformations, which allows the detection of both positive and negative pressures. The quasi-piezoelectric effect is further validated by quantitative analysis in a wide pressure range of−50 to 60 kPa,providing detailed insights into transduction working principles. For reliable lamprey detection,a 4×4-pixel sensor array is developed and integrated with a complementary metal-oxide-semiconductor(CMOS)based signal processing array thus constituting a sensing panel capable of recording oral suction patterns in an underwater environment. | Yunqi Cao Hongyang Shi Xiaobo Tan Nelson Sepúlveda | 2023 | Nano Research2023,16,9: | 0
ZmMS39 encodes a callose synthase essential for male fertility in maize(Zea mays L.)显示文摘Callose contributes to many biological processes of higher plants including pollen development, cell plate and vascular tissue formation,as well as regulating the transport function of plasmodesmata. The functions of callose synthase genes in maize have been little studied. We describe a maize male-sterile mutant 39(ms39)characterized by reduced plant height. In this study,we confirmed using CRISPR/Cas9 technology that a mutation in Zm00001d043909(ZmCals12),encoding a callose synthase,is responsible for the male sterility of the ms39 mutant.Compared with male-fertile plants, callose deposition around the dyads and tetrads in ms39 anthers was significantly reduced.Increased cell autophagy observed in ms39 anthers may have been due to the premature programmed cell death of tapetal cells, leading to collapse of the anther wall structure.Disordered glucose metabolism in ms39 may have intensified autophagy in anthers.Evaluation of the ms39 gene on maize heterosis by paired-crossed experiment with 11 maize inbred lines indicated that ms39 can be used for maize hybrid seed production. | Qunkai Niu Ziwen Shi Peng Zhang Shuai Su Bin Jiang Xiaowei Liu Zhuofan Zhao Suzhi Zhang Qiang Huang Chuan Li Tao Yu Hongyang Yi Tingzhao Rong Moju Cao | 2023 | The Crop Journal2023,11,2: | 0
