1A human circulating immune cell landscape in aging and COVID-19显示文摘Age-associated changes in immune cells have been linked to an increased risk for infection.However,a global and detailed characterization of the changes that human circulating immune cells undergo with age is lacking.Here,we combined scRNA-seq,mass cytometry and sCATAC-seq to compare immune cell types in peripheral blood collected from young and old subjects and patients with COVID-19.We found that the immune cell landscape was reprogrammed with age and was characterized by T cell polarization from naive and memory cells to effector,cytotoxic,exhausted and reg-ulatory cells,along with increased late natural killer cells,age-associated B cells,inflammatory monocytes and age-associated dendritic cells.In addition,the expression of genes,which were implicated in coron-avirus susceptibility,was upregulated in a cell subtype-specific manner with age.Notably,COVID-19 promoted age-induced immune cell polarization and gene expression related to inflammation and cellular senes-cence.Therefore,these findings suggest that a dysreg-ulated immune system and increased gene expression associated with SARS-CoV-2 susceptibility may at least partially account for COVID-19 vulnerability in the elderly.Yingfeng Zheng Xiuxing Liu Wenqing Le Lihui Xie He Li Wen Wen Si Wang Shuai Ma Zhaohao Huang Jinguo Ye Wen Shi Yanxia Ye Zunpeng Liu Moshi Song Weiqi Zhang Jing-Dong J.Han Juan Carlos lzpisua Belmonte Chuanle Xiao Jing Qu Hongyang Wang Guang-Hui Liu Wenru Su 2020Protein & Cell2020,11,10:10

3High electrocaloric effect in barium titanate-sodium niobate ceramics with core-shell grain assembly显示文摘Electrocaloric effect(ECE)is promising in realizing solid-state cooling as an alternative to the conventional refrigeration with environmentally harmful coolant and low efficiency.High ECE in lead-free ferroelectric ceramics is highly desirable for the EC cooling.In this work,different from the researches that tune the ECE by conventional compositional design or external stress engineering,we fabricated the(1-x)BaTiO_(3)-xNaNbO_(3)(BTO-xNN)lead-free ceramics with a core-shell grain structure arising from the inhomogeneous stoichiometry of element distribution,leading to the internal compressing stress in the grains.It is interesting that the phase transition behavior,including the phase transition temperature and the diffusion property,is regulated by the core-shell grain structure induced internal stress,which can be capitalized on for the favorable ECE.Cooperated with 0.02 NN,a high ECE,e.g.adiabatic temperature change(ΔT)of 3.6 K and isothermal entropy change(ΔS)of 4.5 J kg^(-1) K^(-1),is attained in the BTO ceramic.As the internal stress further increases with more NN,the BTO-0.06NN exhibits an extremely stable ECE with a variety rate below ±4% in a wide temperature range from 300 K to 360 K.This work provides a novel approach to explore pronounced ECE in lead-free ferroelectrics for eco-friendly refrigeration.Chao Zhang Quanpei Du Wenru Li Dong Su Meng Shen Xiaoshi Qian Bing Li Haibo Zhang Shenglin Jiang Guangzu Zhang 2020Journal of Materiomics2020,6,3:4

5Aging weakens Th17 cell pathogenicity and ameliorates experimental autoimmune uveitis in mice显示文摘Aging-induced changes in the immune system are associated with a higher incidence of infection and vaccination failure.Lymph nodes,which filter the lymph to identify and fight infections,play a central role in this process.However,careful characterization of the impact of aging on lymph nodes and associated autoimmune diseases is lacking.We combined single-cell RNA sequencing(scRNA-seq)with flow cytometry to delineate the immune cell atlas of cervical draining lymph nodes(CDLNs)of both young and old mice with or without experimental autoimmune uveitis(EAU).We found extensive and complicated changes in the cellular constituents of CDLNs during aging.When confronted with autoimmune challenges,old mice developed milder EAU compared to young mice.Within this EAU process,we highlighted that the pathogenicity of T helper 17 cells(Th17)was dampened,as shown by reduced GM-CSF secretion in old mice.The mitigated secretion of GMCSF contributed to alleviation of IL-23 secretion by antigen-presenting cells(APCs)and may,in turn,weaken APCs’effects on facilitating the pathogenicity of Th17 cells.Meanwhile,our study further unveiled that aging downregulated GM-CSF secretion through reducing both the transcript and protein levels of IL-23R in Th17 cells from CDLNs.Overall,aging altered immune cell responses,especially through toning down Th17 cells,counteracting EAU challenge in old mice.He Li Lei Zhu Rong Wang Lihui Xie Jie Ren Shuai Ma Weiqi Zhang Xiuxing Liu Zhaohao Huang Binyao Chen Zhaohuai Li Huyi Feng Guang-Hui Liu Si Wang Jing Qu Wenru Su 2022Protein & Cell2022,13,6:1

9Severe Global Environmental Issues Caused by Canada’s Record-Breaking Wildfires in 2023显示文摘Due to the record-breaking wildfires that occurred in Canada in 2023,unprecedented quantities of air pollutants and greenhouse gases were released into the atmosphere.The wildfires had emitted more than 1.3 Pg CO_(2)and 0.14 Pg CO_(2)equivalent of other greenhouse gases(GHG)including CH4 and N_(2)O as of 31 August.The wildfire-related GHG emissions constituted more than doubled Canada’s planned cumulative anthropogenic emissions reductions in 10 years,which represents a significant challenge to climate mitigation efforts.The model simulations showed that the Canadian wildfires impacted not only the local air quality but also that of most areas in the northern hemisphere due to long-range transport,causing severe PM_(2.5)pollution in the northeastern United States and increasing daily mean PM_(2.5)concentration in northwestern China by up to 2μg m-3.The observed maximum daily mean PM_(2.5)concentration in New York City reached 148.3μg m-3,which was their worst air quality in more than 50 years,nearly 10 times that of the air quality guideline(i.e.,15μg m-3)issued by the World Health Organization(WHO).Aside from the direct emissions from forest fires,the peat fires beneath the surface might smolder for several months or even longer and release substantial amounts of CO_(2).The substantial amounts of greenhouse gases from forest and peat fires might contribute to the positive feedback to the climate,potentially accelerating global warming.To better understand the comprehensive environmental effects of wildfires and their interactions with the climate system,more detailed research based on advanced observations and Earth System Models is essential.Zhe WANG Zifa WANG Zhiyin ZOU Xueshun CHEN Huangjian WU Wending WANG Hang SU Fang LI Wenru XU Zhihua LIU Jiaojun ZHU 2024Advances in Atmospheric Sciences2024,41,4:0
