| 2 | Targeting EZH1/2 induces cell cycle arrest and inhibits cell proliferation through reactivation of p57CDKN1C and TP53INP1 in mantle cell lymphoma显示文摘Objective: To explore the effect of dysregulation of epigenetic regulator EZH1 and EZH2 on the proliferation in MCL and the underlying mechanisms.Methods: In this study, we elucidated the role of EZH1 and EZH2 overexpression by immunohistochemistry and correlated them to clinical outcome in 41 MCL patients.Quantitative real-time PCR and Western blot were applied to confirm the level of EZH1 and EZH2 in well-characterized MCL cell lines which were compared to those of na?ve B cells.Then we manipulated the expression of EZH1 and EZH2 in MCL cells using CRISPR/Cas9 system to directly investigate their functional roles in MCL.We also evaluated the effect of two small molecule selective inhibitors, EPZ005687 and UNC1999, on MCL cell proliferation, cell cycle distribution and apoptosis in vitro.Finally, we performed RNA-sequencing(RNA-Seq) and Chromatin immunoprecipitation(ChIP) assay to further gain insight into the underlying molecular mechanisms.Results: We found that EZH2 protein is overexpressed in approximately half of this cohort of MCL cases.More importantly, the overexpression of EZH2 is associated with poor OS in the patients.Nevertheless, simple EZH2 depletion in vitro has little impact on the viability of MCL cells, predominantly because of the consequent up-regulation of EZH1.Consistently, UNC1999, a dual EZH1/2 inhibitor, unlike the EZH2 selective inhibitor EPZ005687, exerts a potent inhibitory effect on MCL cells.Furthermore, we discover CDKN1C and TP53 INP1 as the two important cell cycle regulators, the expression of which are repressed by EZH1/2 mediated epigenetic regulation and are restored by EZH1/2 dual inhibition.Conclusions: Our study suggests that EZH2 participates in the pathogenesis of MCL which may serve as a potential biomarker for prognosis prediction.The dual inhibition of EZH1/2 is a promising therapeutic strategy for MCL. | Wei Li Chengfeng Bi Yating Han Tian Tian Xianhuo Wang Huijing Bao Xiaoying Xu Xuhan Zhang Lu Liu Weiwei Zhang Hai Gao Huaqing Wang Huilai Zhang Bin Meng Xi Wang Kai Fu | 2019 | Cancer Biology & Medicine2019,16,3: | 2 |

| 4 | Brain-derived neurotrophic factor and Bcl-2 expression in rat brain areas following chronic morphine treatment显示文摘The ventral tegmental area and the locus coeruleus are associated with psychological and physical dependence of opioid addiction. To date, very little is known about brain-derived neurotrophic factor (BDNF) and Bcl-2 gene and protein changes following morphine addiction. The present study utilized immunohistochemistry and in situ hybridization techniques, which revealed that there were increased BDNF levels, but decreased Bcl-2 levels in the prefrontal cortex, locus coeruleus, hippocampus, and the ventral tegmental area during morphine-dependence formation and abstinence. However, the levels of BDNF remained unchanged, and Bcl-2 expression was increased in the nucleus accumbens. | 5 | Bidirectional high sidelobe suppression silicon optical phased array显示文摘An optical phased array(OPA),the most promising non-mechanical beam steering technique,has great potential for solid-state light detection and ranging systems,holographic imaging,and free-space optical communications.A high quality beam with low sidelobes is crucial for long-distance free-space transmission and detection.However,most previously reported OPAs suffer from high sidelobe levels,and few efforts are devoted to reducing sidelobe levels in both azimuthal(φ)and polar(θ)directions.To solve this issue,we propose a Y-splitter-assisted cascaded coupling scheme to realize Gaussian power distribution in the azimuthal direction,which overcomes the bottleneck in the conventional cascaded coupling scheme and significantly increases the sidelobe suppression ratio(SLSR)in theφdirection from 20 to 66 dB in theory for a 120-channel OPA.Moreover,we designed an apodized grating emitter to realize Gaussian power distribution in the polar direction to increase the SLSR.Based on both designs,we experimentally demonstrated a 120-channel OPA with dual-Gaussian power distribution in bothφandθdirections.The SLSRs inφandθdirections are measured to be 15.1 d B and 25 dB,respectively.Furthermore,we steer the beam to the maximum field of view of 25°×13.2°with a periodic 2λpitch(3.1μm).The maximum total power consumption is only 0.332 W with a thermo-optic efficiency of 2.7 m W∕π. | HUAQING QIU YONG LIU XIANSONG MENG XIAOWEI GUAN YUNHONG DING HAO HU | 2023 | Photonics Research2023,11,4: | 0 |
| 6 | Energy‑efcient integrated silicon optical phased array显示文摘An optical phased array(OPA)is a promising non-mechanical technique for beam steering in solid-state light detection and ranging systems.The performance of the OPA largely depends on the phase shifter,which afects power consumption,insertion loss,modulation speed,and footprint.However,for a thermo-optic phase shifter,achieving good performance in all aspects is challenging due to trade-ofs among these aspects.In this work,we propose and demonstrate two types of energy-efcient optical phase shifters that overcome these trade-ofs and achieve a well-balanced performance in all aspects.Additionally,the proposed round-spiral phase shifter is robust in fabrication and fully compatible with deep ultraviolet(DUV)processes,making it an ideal building block for large-scale photonic integrated circuits(PICs).Using the high-performance phase shifter,we propose a periodic OPA with low power consumption,whose maximum electric power consumption within the feld of view is only 0.33 W.Moreover,we designed Gaussian power distribution in both the azimuthal(ψ)and polar(θ)directions and experimentally achieved a large sidelobe suppression ratio of 15.1 and 25 dB,respectively. | Huaqing Qiu Yong Liu Xiansong Meng Xiaowei Guan Yunhong Ding Hao Hu | 2023 | Frontiers of Optoelectronics2023,16,3: | 0 |
| 7 | Effects of asthma on stability of Wisconsin card sorting test measures A twin study显示文摘BACKGROUND: The four measures used to assess the stability of the Wisconsin card sorting test (WCST), that is, the number of preservative errors, percentage of preservative errors, number of categories completed, and the number of trials to complete the first category, reflect the function of the frontal lobe. OBJECTIVE: This study was designed to investigate the effects of asthma on the stability of WCST measurements through the use of a twin study, and to analyze whether egg-type difference exists. DESIGN: A cohort study. SETTING: Mental Health Center, the First Affiliated Hospital of Chongqing Medical University. PARTICIPANTS: Fifty-nine pairs of twins, aged 6–16 years, were primarily selected from schools between August 2005 and February 2007 and the WCST and Zygosity identification test was applied. Twins with achromatopsia, severe upper limb diseases, somatic diseases, or mental disorders were excluded. According to disease history of asthmatic attack, children were assigned into asthma (n = 16) and non-asthma (n = 43) groups. METHODS: Four WCST measurements were determined in the 59 pairs of twins, and egg-type differences were identified in conjunction. RESULTS: All 59 pairs of twins were included in the final analysis. Among the pairs of twins, 28 (48.5 %) were monozygotic twins, and 31 (52%) were dizygotic twins. Among the monozygotic and dizygotic twins, the number of preservative errors and percentage of preservative errors were significantly higher in the asthma group than in the non-asthma group (P < 0.05). However, there was no significant difference of each corresponding measure between the monozygotic and dizygotic twins (P > 0.05).
