1Cd uptake in rice cultivars treated with organic acids and EDTA显示文摘A pot experiment was conducted to examine the activity of antioxidant enzymes, the content of malondialdehyde (MDA), proline and protein, and Cd uptake in different rice cultivars exposed to Cd (0, 1 and 5 mg/kg) in the presence of organic acids and ethylenediamine tetraacetic acid (EDTA). The results showed the increase in activity of dismutase (SOD), contents of proline and protein but a decline in activities of peroxidase (POD) and catalase (CAT), and MDA content for cultivars Xiushui63 and IIyou527. The resistance to Cd was higher in Xiushui63 than that in IIyou527 under the same Cd treatment. Cadmium contents in grain, straw and roots of both cultivars were markedly reduced in the presence of organic acids and EDTA. 2Influence of leptin on luteinizing hormone and follicle stimulating hormone secreted from cultured rat anterior pituitary cells显示文摘BACKGROUND: Leptin may regulate reproductive function via release of hypothalamic neuropeptide Y. However, it is unknown whether this regulatory effect is limited to the hypothalamus. OBJECTIVE: To detect the effect of different dosages of leptin on luteinizing hormone (LH) and follicle stimulating hormone (FSH) secretion from in vitro cultured rat anterior pituitary cells. DESIGN: Contrast study based on cells. SETTING: This study was performed in the Basic Institute of Chengde Medical College, Chengde City, Hebei Province, China from March to June 2007. MATERIALS: Eighteen female Wistar rats of three months of age, weighing 200-220 g, and of clean grade were used. Leptin was provided by Peprotech Company, DMEM culture medium by Invitrogen Company, and the radioimmunological kit by Beijing Zhongshan Jinqiao Biotechnology Co., Ltd. METHODS: Three glandular organs were regarded as one group for culture of anterior pituitary cells. In the control group, saline was added to the culture medium instead of leptin. In the leptin group, leptin was prepared into different concentrations of 1×10-12, 1×10-11, 1×10-9, 1×10-7, and 1×10-6 mol/L for stimulation of cultured cells. The culture supernatant was obtained at three hours after additional of saline/leptin. MAIN OUTCOME MEASURES: Contents of LH and FSH were detected by radioimmunology. RESULTS: Following leptin stimulation, LH release increased with increasing concentrations of leptin up to 1×10-9 mol/L, where LH release peaked. LH release then progressively decreased with increasing leptin concentrations (P < 0.01). LH release in the leptin (1×10-12, 1×10-11, 1×10-7, and 1×10-6 mol/L) groups was significantly higher than that in the control group (P < 0.01). FSH content in the leptin (1×10-11, 1×10-9, and 1×10-7 mol/L) groups was significantly higher than that in the control group (P < 0.01). 6Redox-dependent regulation of end-binding protein 1 activity by glutathionylation显示文摘Cytoskeletal proteins are susceptible to glutathionylation under oxidizing conditions,and oxidative damage has been implicated in several neurodegenerative diseases.End-binding protein 1(EB1)is a master regulator of microtubule plus-end tracking proteins(+TIPs)and is critically involved in the control of microtubule dynamics and cellular processes.However,the impact of glutathionylation on EB1 functions remains unknown.Here we reveal that glutathionylation is important for controlling EB1 activity and protecting EB1 from irreversible oxidation.In vitro biochemical and cellular assays reveal that EB1 is glutathionylated.Diamide,a mild oxidizing reagent,reduces EB1 comet number and length in cells,indicating the impairment of microtubule dynamics.Three cysteine residues of EB1 are glutathionylated,with mutations of these three cysteines to serines attenuating microtubule dynamics but buffering diamide-induced decrease in microtubule dynamics.In addition,glutaredoxin 1(Grx1)deglutathionylates EB1,and Grx1 depletion suppresses microtubule dynamics and leads to defects in cell division orientation and cell migration,suggesting a critical role of Grx1-mediated deglutathionylation in maintaining EB1 activity.Collectively,these data reveal that EB1 glutathionylation is an important protective mechanism for the regulation of microtubule dynamics and microtubule-based cellular activities.Miao Chen Jian Wang Yang Yang Tao Zhong Peng Zhou Huixian Ma Jingrui Li Dengwen Li Jun Zhou Songbo Xie Min Liu 2021Science China(Life Sciences)2021,64,4:0

13Boosting electrocatalytic selectivity in carbon dioxide reduction:The fundamental role of dispersing gold nanoparticles on silicon nanowires显示文摘Carbon dioxide electrochemical reduction(CO_(2)RR)has been recognized as an efficient way to mitigate CO_(2)emissions and alleviate the pressure on global warming and associated environmental consequences.Gold(Au)is reported as stable and active electrocatalysts to convert CO_(2)to CO at low overpotential due to its moderate adsorption strength of^(*)COOH and^(*)CO.The request for improved catalytic performance,however,is motivated by current unsatisfied catalytic selectivity because of the side hydrogen evolution reaction.In this context,the design of Au based binary catalysts that can boost CO selectivity is of great interest.In the present work,we report that Au nanoparticles can be feasibly dispersed and anchored on silicon nanowires to form Au-Si binary nanomaterials.The Au-Si may stably drive CO_(2)RR with a CO Faraday efficiency of 95.6%at−0.6 V vs.RHE in 0.5 mol/L KHCO_(3)solution.Such selectivity outperforms Au particles by up to 61%.Controlled experiments illustrate that such catalytic enhancement can chiefly be ascribed to electronic effects of binary catalysts.Theoretical calculations reveal that spontaneously produced silicon oxide may not only inhibit hydrogen evolution reaction,but also stabilize the key intermediate^(*)COOH in CO formation.Fan Liao Xing Fan Huixian Shi Qing Li Mengjie Ma Wenxiang Zhu Haiping Lin Youyong Li Mingwang Shao 2022Chinese Chemical Letters2022,33,9:0
14Exploration of Hanshi Zufei prescription for treatment of COVID-19 based on network pharmacology显示文摘Objective:Network pharmacology combines drug and disease targets with biological information networks based on the integrity and systematicness of the interactions between drugs and disease targets.This study aims to explore the molecular basis of Hanshi Zufei formula for treatment of COVID-19 based on network pharmacology and molecular docking techniques.Methods:Using TCMSP,the chemical constituents and molecular targets of Atractylodis Rhizoma,Citri Reticulatae Pericarpium,Magnoliae Officinalis Cortex,Pogostemonis Herba,Tsaoko Fructus,Ephedrae Herba,Notopterygii Rhizoma et Radix,Zingiberis Rhizoma Recens,and Arecae Semen were investigated.The predicted targets of novel coronavirus were screened using the NCBI and Gene Cards databases.To further screen the drug-disease core targets network,the corresponding target proteins were queried using multiple databases(Biogrid,DIP,and HPRD),a protein interaction network graph was constructed,and the network topology was analyzed.The molecular docking studies were also performed between the network’s top 15 compounds and the coronavirus(SARS-Co V-2)3 CL hydrolytic enzyme and angiotensin conversion enzymeⅡ(ACE2).Results:The herb-active ingredient-target network contained nine drugs,86 compounds,and 49 drugdisease targets.Gene ontology(GO)enrichment analysis resulted in 1566 GO items(P<0.05),among which 1438 were biological process items,35 were cell composition items,and 93 were molecular function items.Fourteen signal pathways were obtained by enrichment screening of the KEGG pathway database(P<0.05).The molecular docking results showed that the affinity of the core active compounds with the SARS-CoV-23 CL hydrolase was better than for the other compounds.Conclusion:Several core compounds can regulate multiple signaling pathways by binding with 3 CL hydrolase and ACE2,which might contribute to the treatment of COVID-19.Xinrui Li Zishuai Wen Mingdong Si Yuxin Jia Huixian Liu Yuguang Zheng Donglai Ma 2022Chinese Herbal Medicines2022,14,2:0
