1Long non-coding RNA LINC-PINT attenuates paclitaxel resistance in triple-negative breast cancer cells via targeting the RNA-binding protein NONO显示文摘The treatment of triple-negative breast cancer(TNBC)relies largely on chemotherapies.However,it is frequent that TNBC patients develop resistance to the chemotherapy drugs.Generation of drug-resistant cell lines facilitates the identification of drug resistance.Here,we established two paclitaxel(PTX)-resistant TNBC cancer cell lines using an intermittent and stepwise method and found that long non-coding RNA long intergenic non-protein-coding RNA p53-induced transcript(LINC-PINT)was significantly decreased in PTX-resistant cancer cells.Ectopic expression of LINC-PINT sensitized both PTX-resistant TNBC and wild-type TNBC to PTX.Moreover,RNA immunoprecipitation showed that LINC-PINT bound to RNA-binding protein NONO.Overexpression of LINC-PINT resulted in the degradation of NONO in a proteasome-dependent manner and vice versa.Knockdown of NONO with siRNA sensitized TNBC to PTX.We further analyzed the expression level of LINC-PINT and NONO in patient samples via online database and found that LINC-PINT and NONO may function antagonistically in all types of breast cancers.Taken together,our data illustrated a tumor suppressor role of LINC-PINT in sensitizing TNBC to chemotherapies via destabilizing NONO.Jinghua Chen Meiqin Zhu Liqiu Zou Junxian Xia Jiacheng Huang Quantong Deng Ruilian Xu 2020Acta Biochimica et Biophysica Sinica2020,52,8:4

3Strain heterogeneity,cooccurrence network,taxonomic composition and functional profile of the healthy ocular surface microbiome显示文摘Background:There is growing evidence indicating that the microbial communities that dwell on the human ocular surface are crucially important for ocular surface health and disease.Little is known about interspecies interactions,functional profiles,and strain heterogeneity across individuals in healthy ocular surface microbiomes.Methods:To comprehensively characterize the strain heterogeneity,cooccurrence network,taxonomic composition and functional profile of the healthy ocular surface microbiome,we performed shotgun metagenomics sequencing on ocular surface mucosal membrane swabs of 17 healthy volunteers.Results:The healthy ocular surface microbiome was classified into 12 phyla,70 genera,and 140 species.The number of species in each healthy ocular surface microbiome ranged from 6 to 47,indicating differences in microbial diversity among individuals.The species with high relative abundances and high positivity rates were Streptococcus pyogenes,Staphylococcus epidermidis,Propionibacterium acnes,Corynebacterium accolens,and Enhydrobacter aerosaccus.A correlation network analysis revealed a competitive interaction of Staphylococcus epidermidis with Streptococcus pyogenes in ocular surface microbial ecosystems.Staphylococcus epidermidis and Streptococcus pyogenes revealed phylogenetic diversity among different individuals.At the functional level,the pathways related to transcription were the most abundant.We also found that there were abundant lipid and amino acid metabolism pathways in the healthy ocular surface microbiome.Conclusion:This study explored the strain heterogeneity,cooccurrence network,taxonomic composition,and functional profile of the healthy ocular surface microbiome.These findings have important significance for the future development of probiotic-based eye therapeutic drugs.Yutong Kang Shudan Lin Xueli Ma Yanlin Che Yiju Chen Tian Wan Die Zhang Jiao Shao Jie Xu Yi Xu Yongliang Lou Meiqin Zheng 2021Eye and Vision2021,8,1:3
4Clinical Analysis of 240 Patients with HLA-B27 Associated Acute Anterior Uveitis显示文摘Purpose: To analyze the prevalence of HLA-B27 associated acute anterior uveitis and to identify its clinical features. Methods: A total of 240 patients with HLA-B27 associated acute anterior uveitis,who were admitted to Zhejiang Ophthalmologic Hospital between December 2006 and October 2012, were retrospectively analyzed.The age of onset,sex,affected eyes,HLA-B27 antigen detection,recurrence,joint involvement, and surgical complications were investigated. Results:The average age of onset was 37.0 ±12.0 years and the ratio of male to female patients was 2.4:1. Most cases had alternate unilateral or bilateral involvement. Among all participants, 234 cases (97.5%) were HLA-B27 positive, and 124 cases (51.7%) had spondyloarthropathies (SpA), dominated by 108 cases with ankylosing spondylitis. (AS,45.0%),and mostly seen in male subjects(P<0.05).Six patients were HLA-B27 negative(2.5%)and on statistical significance was noted between male and female patients (P >0.05).A total of 193 cases (80.4%) presented with complications, mainly fibrinous exudation, posterior synechia, and vitreous opacity. Conclusion: HLA-B27 that is associated acute anterior uveitis with a relatively high incidence and recurrence presents with more severe clinical features than does idiopathic acute anterior uveitis,and it often accompanies systemic arthritic diseases. 5Automatic search method for multiple differentials and its application on MANTIS显示文摘Multiple differential cryptanalysis is one of the extensions of classic differential cryptanalysis. In this paper, we present a generic automatic search method for clustering multiple differentials on a target block cipher. Our search method has two steps. Firstly, the sets of input and output differences will be determined.With these sets, we get different multiple differentials. Then for each one of these multiple differentials, we enumerate and record all satisfied differential trails, which leads to a more accurate evaluation of the multiple differentials distinguisher. Among these different multiple differentials distinguishers, we can choose the best one for key recovery attack. We demonstrate our search method by applying it on the part of differentials of the block cipher MANTIS. As a result, we find a new 10-round multiple differentials distinguisher with probability 2^(-55.98) and an 11-round multiple differentials distinguisher with probability 2^(-63.71), which is the longest distinguisher for MANTIS so far as we know. This new 10-round distinguisher can lead to a better signal-to-noise ratio, so we derive an improved key recovery attack on MANTIS-6 with the complexity of about 2^(51.79) chosen-plaintext queries, 2^(51.91) encryptions and data-time product 2^(103.70), which is better than the previous best one with data-time product 2^(110.61). Aiming at exploring the gap between the performance of multiple differential attack and the security margin on MANTIS, we also use the 11-round distinguisher to derive a key recovery attack on MANTIS-7 with the complexity of about 2^(61.86) chosen-plaintext queries,2^(102.92) encryptions and data-time product 2^(164.78). 8Cooperative neural-adaptive fault-tolerant output regulation for heterogeneous nonlinear uncertain multiagent systems with disturbance显示文摘This paper investigates the cooperative output regulation problem for heterogeneous nonlinear uncertain multiagent networked systems subject to actuator failure,bounded matched or mismatched disturbances or disturbances produced by a given linear exosystem.Accurate information about nonlinearity,actuator failure and disturbance may be completely unknown.First,a distributed finite-time observer is designed to estimate the dynamics of the exosystem on a finite-time interval over a communication digraph.Then,a neural-adaptive control protocol is proposed.It is shown that(i)closed-loop multiagent systems are asymptotically stable,with output synchronization errors that tend to zero in the absence of mismatched disturbance,and(ii)the states of the closed-loop multiagent systems and the output synchronization errors are bounded in the presence of mismatched disturbance.Finally,a simulation example is given to demonstrate the effectiveness of the proposed control strategy.Shanling DONG Guanrong CHEN Meiqin LIU Zheng-Guang WU 2021Science China(Information Sciences)2021,64,7:1

18Systematic study of the transfer of amino acids across the water/1,2-dichloroethane interface facilitated by dibenzo-18-crown-6显示文摘Facilitated ion transfer reactions of 20 amino acids with dibenzo-18-crown-6 (DB18C6) at the water/1,2-dichloroethane (W/DCE) interfaces supported at the tips of micro- and nano-pipets were investigated systematically using cyclic voltammetry. It was found that there were only 10 amino acids, that is, Leu, Val, Ile, Phe, Trp, Met, Ala, Gly, Cys, Gln (in brief), whose protonated forms as cations can give well-defined facilitated ion transfer voltammograms within the potential window, and the reaction pathway was proven to be consistent with the transfer by interfacial complexation/dissociation (TIC/TID) mechanisms. The association constants of DB 18C6 with different amino acids in the DCE (β0), and the kinetic parameters of reaction were evaluated based on the steady-state voltammetry of micro- or nano-pipets, respectively. The experimental results demonstrated that the selectivity of complexation of protonated amino acid by DB18C6 compared with that of alkali metal cations was low, which can be attributed to the vicinal effect arising from steric hindrance introduced by their side group and the steric bulk effect by lipophilic stabilization.
