| 1 | Mesenchymal stem cell therapy in retinal and optic nerve diseases: An update of clinical trials显示文摘Retinal and optic nerve diseases are degenerative ocular pathologies which lead to irreversible visual loss. Since the advanced therapies availability, cell-based therapies offer a new all-encompassing approach. Advances in the knowledge of neuroprotection, immunomodulation and regenerative properties of mesenchymal stem cells(MSCs) have been obtained by several preclinical studies of various neurodegenerative diseases. It has provided the opportunity to perform the translation of this knowledge to prospective treatment approaches for clinical practice. Since 2008, several first steps projecting new treatment approaches, have been taken regarding the use of cell therapy in patients with neurodegenerative pathologies of optic nerve and retina. Most of the clinical trials using MSCs are in Ⅰ/Ⅱ phase, recruiting patients or ongoing, and they have as main objective the safety assessment of MSCs using various routes of administration. However, it is important to recognize that, there is still a long way to go to reach clinical trials phase Ⅲ-Ⅳ. Hence, it is necessary to continue preclinical and clinical studies to improve this new therapeutic tool. | 2 | Preclinical evaluation of azathioprine plus buthionine sulfoximine in the treatment of human hepatocarcinoma and colon carcinoma显示文摘AIM: To evaluate the efficacy and the safety of azathioprine (AZA) and buthionine sulfoximine (BSO) bylocalized application into HepG2 tumor in vivo.METHODS: Different hepatoma and colon carcinoma cell lines (HepG2, HuH7, Chang liver, LoVo, RKO, SW-48, SW-480) were grown in minimal essencial medium supplemented with 10% fetal bovine serum and 1% antibiotic/antimycotic solution and maintained in a humidified 37 ℃ incubator with 5% CO2. These cells were pretreated with BSO for 24 h and then with AZA for different times. We examined the effects of this combination on some proteins and on cellular death. We also studied the eff icacy and the safety of AZA (6 mg/kg per day) and BSO (90 mg/kg per day) in HepG2 tumor growth in vivo using athymic mice. We measured safety by serological markers such as aminotransferases and creatine kinase.RESULTS: The in vitro studies revealed a new mechanism of action for the AZA plus BSO combination in the cancer cells compared with other thiopurines (6-mercaptopurine, 6-methylmercaptopurine, 6-thioguanine and 6-methylthioguanine) in combination with BSO. The cytotoxic effect of AZA plus BSO in HepG2 cells resulted from necroptosis induction in a mitochondrial-dependent manner. From kinetic studies we suggest that glutathione (GSH) depletion stimulates c-Jun amino-terminal kinase and Bax translocation in HepG2 cells with subsequent deregulation of mitochondria (cytochrome c release, loss of membrane potential), and proteolysis activation leading to loss of membrane integrity, release of lactate dehydrogenase and DNA degradation. Some of this biochemical and cellular changes could be reversed by N-acetylcysteine (a GSH replenisher). | 5 | Toric人工晶状体在合并高度角膜散光白内障中的应用(英文)显示文摘目的:评估Bi-Flex toric人工晶状体在合并散光的白内障中的应用。方法:回顾性研究包括16例22眼合并2.50D以上散光的白内障患者。测量了术前与术后的裸眼视力、最佳矫正视力、客观及主观验光、角膜散光仪和角膜地形图测量的散光值。术后人工晶状体屈光轴位置使用向量进行分析。结果:术后,主观验光散光值从4.05D±1.53D显著降低至1.35D±0.86D,(P<0.05)。屈光轴位置指标J_0,从术前的-0.81±2.02D降至-0.12±0.62D(P<0.05)。裸眼视力和最佳矫正视力术后1mo时均显著改善(P<0.05),分别为0.24±0.19和0.06±0.08。人工晶状体屈光轴旋转程度平均为2.95±5.25度,86.36%的患眼旋转程度小于10度。术前、术后角膜散光计测量值未见显著差异(J_0和J_(45)均有P>0.05)。结论:对于合并大于2.50D散光的白内障患者,Bi-Flex toric人工晶状体植入是安全有效的治疗方法。 | Irene Simo Rafael Freiria Laura Remón | 2018 | 国际眼科杂志2018,18,2: | 2 |

| 16 | Home range variability,spatial aggregation,and excursions of Akodon azarae and Oligoryzomys flavescens in Pampean agroecosystems显示文摘Rodents are reservoirs of various types of hantavirus,some of which are agents of hantavirus pulmonary syndrome in humans.Each hantavirus is associated with a single rodent host species but successive spill-over events may eventually lead to host-switching and new species’becoming host of a given pathogen.This study aims to gain an understanding of the spatial ecology of two hantavirus-host species,Akodon azarae,and Oligoryzomys flavescens,by identifying factors modulating their home range sizes and stability,and by evaluating intra-and interspecific spatial aggregation for these species and a third one—Oxymycterus rufus—living in sympatry.For this,eleven capture-mark-recapture surveys were carried out,spanning 22 months.We found that A.azarae males have larger and more mobile home ranges than females,independently of the season.Consequently,males could likely have a more relevant role in the transmission of hantavirus because of their greater exposure both to a higher number of contacts between individuals and viral contamination of the environment.Contrasting,O.flavescens individuals showed negligible displacements of their home range through time,which could limit the range of hantavirus spread in host populations.Since O.flavescens is host to Lechiguanas hantavirus(pathogenic to humans)this result encompasses epidemiological relevance,for it may imply the existence of local foci of infection.Additionally,individuals of both species performed excursions outside their home ranges.These events could enable hantavirus spread over distances beyond the normal range of movements and lead to new hantavirus outbreaks in formerly non-infected rodent populations,favoring the persistence of the virus in nature. | Irene GOROSITO Ailén BENÍTEZ Maria BUSCH | 2020 | Integrative Zoology2020,15,5: | 1 |
