| 1 | Domain structure and defects of highly ordered Bi_4Si_3O_(12) micro-crystals显示文摘Highly ordered Bi4Si3O12 micro-crystals were prepared at normal atmosphere. Phase identification of the prepared crystals was accomplished by X-ray diffractometer (XRD). Domain structure and defects were characterized by environmental scanning electron microscopy (ESEM). XRD shows that the obtained micro-crystals are of eulytite structure with chemical formulation of Bi4Si3O12. A highly ordered growth pattern is confirmed due to the faster growth of the {124} faces than that of the {204} faces by ESEM. The growing process of the domain structure is of pollen parent and filial generation pattern. The filial generations of Bi4Si3O12 crystals are generated from the pollen parent. Cracks generate from the defect areas and propagate along the {124} faces due to their lower binding energy under a proper temperature gradient, contributing to the total transcrystalline fracture. It is confirmed that the generation and development of the voids in the crystal grains can be developed when unmatched dimensions of the two opposite faces are formed. | 2 | Five-year follow-up of 23 asymmetrical Parkinson's disease patients treated with unilateral subthalamic nucleus stimulation显示文摘In this study, 23 asymmetrical Parkinson's disease patients were treated with unilateral deep brain stimulation of the subthalamic nucleus and followed up for 5 years. At 5 years after stimulation treatment, Unified Parkinson's Disease Rating Scale II, III and axial symptom scores in the off-drug condition were significantly increased compared those at baseline. However, total Unified Parkinson's Disease Rating Scale II, III and axial symptom scores were significantly lower with stimulation-on compared with the synchronous stimulation-off state in off-drug condition, and the motor symptoms of contralateral side limbs were effectively controlled. Only low Hoehn-Yahr stage was correlated with good long-term postoperative improvement in motor symptoms. The mean levodopa-equivalent daily dose after stimulation treatment was significantly lower than that before treatment, but dyskinesias became worse. | 3 | Escape from abluminal LRP1-mediated clearance for boosted nanoparticle brain delivery and brain metastasis treatment显示文摘Breast cancer brain metastases(BCBMs)are one of the most difficult malignancies to treat due to the intracranial location and multifocal growth.Chemotherapy and molecular targeted therapy are extremely ineffective for BCBMs due to the inept brain accumulation because of the formidable bloodbrain barrier(BBB).Accumulation studies prove that low density lipoprotein receptor-related protein 1(LRP1)is promising target for BBB transcytosis.However,as the primary clearance receptor for amyloid beta and tissue plasminogen activator,LRP1 at abluminal side of BBB can clear LRP1-targeting therapeutics.Matrix metalloproteinase-1(MMP1)is highly enriched in metastatic niche to promote growth of BCBMs.Herein,it is reported that nanoparticles(NPs-K-s-A)tethered with MMP1-sensitive fusion peptide containing HER2-targeting K and LRP1-targeting angiopep-2(A),can surmount the BBB and escape LRP1-mediated clearance in metastatic niche.NPs-K-s-A revealed infinitely superior brain accumulation to angiopep-2-decorated NPs-A in BCBMs bearing mice,while comparable brain accumulation in normal mice.The delivered doxorubicin and lapatinib synergistically inhibit BCBMs growth and prolongs survival of mice bearing BCBMs.Due to the efficient BBB penetration,special and remarkable clearance escape,and facilitated therapeutic outcome,the fusion peptide-based drug delivery strategy may serve as a potential approach for clinical management of BCBMs. | Naveed Ullah Khan Jiang Ni Xiufeng Ju Tongtong Miao Haiyan Chen Liang Han | 2021 | Acta Pharmaceutica Sinica B2021,11,5: | 3 |

| 5 | Mechanical properties and influence mechanism of confined concrete arches in high-stress tunnels显示文摘Deep underground projects(e.g., coal mines), are often faced with complex conditions such as high stress and extremely soft rock. The strength and rigidity of the traditional support system are often insufficient,which makes it difficult to meet the requirements of ground control under complex conditions. As a new support form with high strength and rigidity, the confined concrete arch plays an important role in controlling the rock deformation under complex conditions. The section shape of the tunnel has an important impact on the mechanical properties and design of the support system. However, studies on the mechanical properties and influence mechanism of the new confined concrete arch are rarely reported. To this end, the mechanical properties of traditional U-shaped steel and new confined concrete arches are compared and comparative tests on arches of circular and straight-leg semicircular shapes in deep tunnels are conducted. A large mechanical testing system for underground engineering support structure is developed. The mechanical properties and influence mechanism of confined concrete arches with different section shapes under different loading modes and cross-section parameters are systematically studied. Test results show that the bearing capacity of the confined concrete arch is 2.10 times that of the U-shaped steel arch, and the bearing capacity of the circular confined concrete arch is 2.27 times that of the straight-leg semicircular arch. Among the various influencing factors and their engineering parameters,the lateral stress coefficient has the greatest impact on the bearing capacity of the confined concrete arch,followed by the steel pipe wall thickness, steel strength, and core concrete strength. Subsequently, the economic index of bearing capacity and cost is established, and the optimization design method for the confined concrete arch is proposed. | 9 | Identification of serum metabolites enhancing inflammatory responses in COVID-19显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is characterized by a strong production of inflammatory cytokines such as TNF and IL-6,which underlie the severity of the disease.However,the molecular mechanisms responsible for such a strong immune response remains unclear.Here,utilizing targeted tandem mass spectrometry to analyze serum metabolome and lipidome in COVID-19 patients at different temporal stages,we identified that 611 metabolites(of 1,039)were significantly altered in COVID-19 patients.Among them,two metabolites,agmatine and putrescine,were prominently elevated in the serum of patients;and 2-quinolinecarboxylate was changed in a biphasic manner,elevated during early COVID-19 infection but levelled off.When tested in mouse embryonic fibroblasts(MEFs)and macrophages,these 3 metabolites were found to activate the NF-κB pathway that plays a pivotal role in governing cytokine production.Importantly,these metabolites were each able to cause strong increase of TNF and IL-6 levels when administered to wildtype mice,but not in the mice lacking NF-κB.Intriguingly,these metabolites have little effects on the activation of interferon regulatory factors(IRFs)for the production of type I interferons(IFNs)for antiviral defenses.These data suggest that circulating metabolites resulting from COVID-19 infection may act as effectors to elicit the peculiar systemic inflammatory responses,exhibiting severely strong proinflammatory cytokine production with limited induction of the interferons.Our study may provide a rationale for development of drugs to alleviate inflammation in COVID-19 patients. | Chen-Song Zhang Bingchang Zhang Mengqi Li Xiaoyan Wei Kai Gong Zhiyong Li Xiangyang Yao Jianfeng Wu Cixiong Zhang Mingxia Zhu Lei Zhang Xiufeng Sun Yi-Hong Zhan Zhengye Jiang Wenpeng Zhao Wei Zhong Xinguo Zhuang Dawang Zhou Hai-Long Piao Sheng-Cai Lin Zhanxiang Wang | 2022 | Science China(Life Sciences)2022,65,10: | 1 |

| 16 | Electro-elastic features of YBa3(PO_(4))3 and YbBa3(PO_(4))3 crystals with pure face-shear mode for acoustic wave sensor applications显示文摘The fundamental shear horizontal(SH0)waves have been proved to be a very useful method for nondestructive testing due to their non-dispersive characteristics.Currently,although several methods have been proposed to generate SH0 waves,it is still a challenge to obtain pure SH0 wave mode efficiently.Herein,in this study,non-centrosymmetric YBa3(PO_(4))3(YBP)and YbBa3(PO_(4))3(YbBP)crystals with pure face-shear mode were grown via the traditional Czochralski pulling method at temperatures above 1800℃.High resolution X-ray diffraction was carried out to evaluate the crystalline quality.Results showed that the YBP and YbBP crystals were of good quality,with the full width at half-maxima being on the order of 42.500 and 52.300,respectively.Moreover,the full set of electro-elastic constants for the YBP and YbBP crystals were determined and collated using a combination of impedance and pulse-echo methods.The piezoelectric coefficient(d14)of the YBP and YbBP crystals were determined to be 10.6 pC/N and 11.4 pC/N,respectively,corresponding to the electromechanical coupling coefficient(k14)equal to 15.4%and 15.9%,exhibiting large and pure face-shear vibration mode.Collectively,these results indicate that the YBP and YbBP crystals are promising for acoustic wave sensing application. | Guangda Wu Lingkai Kong Mengdi Fan Fapeng Yu Chao Jiang Xiufeng Cheng Xian Zhao | 2021 | Journal of Materiomics2021,7,4: | 0 |
| 17 | Deep brain stimulation of the subthalamic nucleus treats Parkinson's disease through enhancing metabolic activity of the corpus striatum Verification by single photon emission computed tomography and positron emission tomography显示文摘BACKGROUND:Subthalamic nucleus deep brain stimulation (STN DBS) for Parkinson's disease (PD) has achieved good effects,but to date the mechanism of STN DBS remains poorly understood. STN DBS may increase dopamine levels or metabolic activity of the corpus striatum. OBJECTIVE:To validate the effects of STN DBS on dopamine metabolism and glucose metabolism in the corpus striatum of hemiparkinsonian monkeys using single photon emission computed tomography (SPECT) and position emission tomography (PET). DESIGN,TIME AND SETTING:A controlled animal study was performed at the Neurosurgery Laboratory,Changhai Hospital of the Second Military Medical University of Chinese PLA between January 2004 and December 2007. METHODS:Hemiparkinsonism was induced in adult Rhesus Macaque monkeys,which exhibit similar characteristics of PD in humans,through unilateral internal carotid artery infusion of 1-methy-4-phenyl-1,2,3,6-tetrahydropyrindine. Following model establishment,stimulation electrodes were implanted in the right STN,and chronic high-frequency stimulation (60 μs pulse width,130 Hz frequency,and 1.5-2.0 V pressure) was performed. MAIN OUTCOME MEASURES:The changes in dopamine transporter (DAT),D2 receptor (D2R),and glucose metabolism in the corpus striatum following STN DBS were observed using SPECT and PET. RESULTS:SPECT examination showed that DAT specific binding in the right corpus striatum was increased at 3 months after DBS compared with prior to stimulation,and D2R specific binding in the right corpus striatum gradually decreased near levels on the left (non-electrode-implanted) side within 3 months after DBS. PET examination showed that the glucose metabolism in the right corpus striatum was markedly increased at 3 months after effective DBS. Hemiparkinsonism monkeys showed improved left limb rigidity,increased activities,and stable gait under chronic high-frequency stimulation.
