1Salivary protease spectrum biomarkers of oral cancer显示文摘Proteases are important molecules that are involved in many physiological and pathological processes of the human body,such as growth,apoptosis and metastasis cancer cells.They are potential targets in cancer diagnosis and biotherapy.In this study,we analyzed the salivary protease spectrum of patients with oral squamous cell carcinoma (OSCC),oral benign masses and chronic periodontitis,as well as that of health,using human protease array kits,enzyme-linked immunosorbent assay,western blot and immunofluorescence.The salivary protease spectrum was found to be associated with oral diseases.For example,the saliva of patients with OSCC contained increased numbers of proteases than those of other oral diseases and health.The levels of matrix metalloproteinase (MMP)-1,MMP-2,MMP-10,MMP-12,A disintegrin and metalloprotease (ADAM)9,A disintegrin and metalloprotease with thrombospondin type 13 motifs (ADAMST13),cathepsin V and kallikrein 5 in the saliva of patients with OSCC were significantly increased compared with those of other groups.Taking MMP-1,cathepsin V,kallikrein 5 and ADAM9 as biomarkers of OSCC,cutoff values were199,11.34,9.29 and 202.55 pg·mL?1,respectively.From the area under the curve,sensitivity and specificity,the combination of cathepsin V/kallikrein5/ADAM9 was an optimal biomarker for diagnosing OSCC.Thus,analysis of the salivary protease spectrum may be an innovative and cost-efficient approach to evaluating the health status of the oral cavity.Specifically,increases in cathepsin V,kallikrein 5 and ADAM9 may be useful biomarkers in the screening and diagnosis of OSCC.Yun Feng Qian Li Jiao Chen Ping Yi Xin Xu Yaping Fan Bomiao Cui Yu Yu Xiaoying Li Yue Du Qianming Chen Lingling Zhang Jingjing Jiang Xuedong Zhou Ping Zhang 2019International Journal of Oral Science2019,11,2:2

3Antibody-engineered red blood cell interface for high-performance capture and release of circulating tumor cells显示文摘Circulating tumor cells(CTCs),as important liquid biopsy target,can provide valuable information for cancer progress monitoring and individualized treatment.However,current isolation platforms incapable of balancing capture efficiency,specificity,cell viability,and gentle release have restricted the clinical applications of CTCs.Herein,inspired by the structure and functional merits of natural membrane interfaces,we established an antibody-engineered red blood cell(RBC-Ab)affinity interface on microfluidic chip for high-performance isolation and release of CTCs.The lateral fluidity,pliability,and anti-adhesion property of the RBC microfluidic interface enabled efficient CTCs capture(96.5%),high CTCs viability(96.1%),and high CTCs purity(average 4.2-log depletion of leukocytes).More importantly,selective lysis of RBCs by simply changing the salt concentration was utilized to destroy the affinity interface for efficient and gentle release of CTCs without nucleic acid contamination.Using this chip,CTCs were successfully detected in colon cancer samples with 90%sensitivity and 100%specificity(20 patients and 10 healthy individuals).After the release process,KRAS gene mutations of CTCs were identified from all the 5 cancer samples,which was consistent with the results of tissue biopsy.We expect this RBC interface strategy will inspire further biomimetic interface construction for rare cell analysis.Haicong Shen Rui Su Jiao Peng Lin Zhu Kunyue Deng Qi Niu Yanling Song Liu Yang Lingling Wu Zhi Zhu Chaoyong Yang 2022Bioactive Materials2022,7,5:1

10An antibody cocktail with broadened mutational resistance and effective protection against SARS-CoV-2显示文摘Neutralizing antibodies have been proven to be highly effective in treating mild and moderate COVID-19 patients,but continuous emergence of SARS-CoV-2 variants poses significant challenges.Antibody cocktail treatments reduce the risk of escape mutants and resistance.In this study,a new cocktail composed of two highly potent neutralizing antibodies(HB27 and H89Y)was developed,whose binding epitope is different from those cocktails that received emergency use authorization.This cocktail showed more potent and balanced neutralizing activities(IC_(50)0.9–11.3 ng mL^(-1))against a broad spectrum of SARS-CoV-2 variants over individual HB27 or H89Y antibodies.Furthermore,the cocktail conferred more effective protection against the SARS-CoV-2 Beta variant in an aged murine model than monotherapy.It was shown to prevent SARS-CoV-2 mutational escape in vitro and effectively neutralize 61 types of pseudoviruses harbouring single amino acid mutation originated from variants and escape strains of Bamlanivimab,Casirivimab and Imdevimab with IC_(50)of 0.6–65 ng mL^(-1).Despite its breadth of variant neutralization,the HB27+H89Y combo and EUA cocktails lost their potencies against Omicron variant.Our results provide important insights that new antibody cocktails covering different epitopes are valuable tools to counter virus mutation and escape,highlighting the need to search for more conserved epitopes to combat Omicron.Chunyun Sun Hang Chi Fei Yuan Jing Li Ji Yang Aihua Zheng Fei Wang Lingling Sun Yanjing Zhang Ping Hu Lihua Jiao Yongqiang Deng Liangzhi Xie 2023Science China(Life Sciences)2023,66,1:0
11Promoting oral mucosal wound healing using a DCS-RuB_(2)A_(2) hydrogel based on a photoreactive antibacterial and sustained release of BMSCs显示文摘The high occurrence rate and difficulties in symptom control are listed as the major problems of oral mucosal disease by medical professionals.Following the development of oral mucosal lesions,the oral microenvironment changes,immunity declines,and continuous bacterial stimulation causes wound infection.Traditional antibacterial drugs are ineffective for oral mucosal lesions.To overcome this problem,a light-responsive antibacterial hydrogel containing sustained-release BMSCs was inspired by the trauma environment in the oral cavity,which is different from that on the body surface since it mostly remains under dark conditions.In the absence of light,the hydrogel seals the wound to form a barrier,exerts a natural bacteriostatic effect,and prevents invasion by foreign bacteria.Simultaneously,mesenchymal stem cells are presented,and the released growth factors and other substances have excellent anti-inflammatory and angiogenic effects,which result in rapid repair of the damaged site.Under light conditions,after photo-induced shedding of the hydrogel,RuB_(2)A exerts an antibacterial effect accompanied by degradation of the hydrogel.Results in a rat oral mucosal repair model demonstrate that DCS-RuB_(2)A_(2)-BMSCs could rapidly repair the oral mucosa within 4 days.Sequencing data provide ideas for further analysis of the intrinsic molecular mechanisms and signaling pathways.Taken together,our results suggest that this light-responsive antibacterial hydrogel loaded with BMSCs can be used for rapid wound repair and may advance the development of therapeutic strategies for the treatment of clinical oral mucosal defects.Wenxin Qi Naijun Dong Lingling Wu Xueqi Zhang He Li Hao Wu Natalie Ward Jian Yu He Liu Jiao Wang Xiaoyong Deng Robert Chunhua Zhao 2023Bioactive Materials2023,,5:0
12Structure and electrochemical performance of hydrogen storage alloy La_(0.7)Mg_(0.3)Ni_(2.875)Co_(0.525)Mn_(0.1)-boron composite显示文摘The structure and electrochemical characteristics of La0.7Mg0.3Ni2.875Co0.525Mn0.1-boron composite was studied systematically. The AB3 type hydrogen storage alloys La0.7Mg0.3Ni2.875Co0.525Mn0.1 were successfully synthesized by means of inter-media alloy La2Mg17. The alloys were composited with boron at different weight rate. From the XRD analyses, each alloy of this series is mainly composed of (La,Mg)Ni3 phase and the LaNi5 phase, and the phase abundance of each phase varies with the boron weight rate, moreover, after composition, the c and cell volumes of (La,Mg)Ni3 phase increase, and the LaNi5 phase keep the same, which indicate that the boron may enter (La,Mg)Ni3 phase. The electrochemical studies show that the maximum discharge capacity of the composites decreases, but the cycling life improved. 13WheatCENet:A Database for Comparative Co-expression Network Analysis of Allohexaploid Wheat and Its Progenitors显示文摘Genetic and epigenetic changes after polyploidization events could result in variable gene expression and modified regulatory networks.Here,using large-scale transcriptome data,we constructed co-expression networks for diploid,tetraploid,and hexaploid wheat species,and built a platform for comparing co-expression networks of allohexaploid wheat and its progenitors,named WheatCENet.WheatCENet is a platform for searching and comparing specific functional coexpression networks,as well as identifying the related functions of the genes clustered therein.Functional annotations like pathways,gene families,protein-protein interactions,microRNAs(miRNAs),and several lines of epigenome data are integrated into this platform,and Gene Ontology(GO)annotation,gene set enrichment analysis(GSEA),motif identification,and other useful tools are also included.Using WheatCENet,we found that the network of WHEAT ABERRANT PANICLE ORGANIZATION I(WAPOI)has more co-expressed genes related to spike development in hexaploid wheat than its progenitors.We also found a novel motif of CCWWWWWWGG(CArG)specifically in the promoter region of WAPO-Al,suggesting that neofunctionalization of the WAPO-AI gene affects spikelet development in hexaploid wheat.WheatCENet is useful for investigating co-expression networks and conducting other analyses,and thus facilitates comparative and functional genomic studies in wheat.Zhongqiu Li Yiheng Hu Xuelian Ma Lingling Da Jiajie She Yue Liu Xin Yi Yaxin Cao Wenying Xu Yuannian Jiao Zhen Su 2023Genomics, Proteomics & Bioinformatics2023,21,2:0
