1Reconstruction of lateral root formation through single-cell RNA sequencing reveals order of tissue in itiation显示文摘Postembryonic organogenesis is critical for plant development.Underground,lateral roots(LRs)form the bulk of mature root systems,yet the ontogeny of the LR primordium(LRP)is not clear.In this study,we performed the single-cell RNA sequencing through the first four stages of LR formation in Arabidopsis.Our analysis led to a model in which a single group of precursor cells,with a cell identity different from their pericycle origins,rapidly reprograms and splits into a mixed ground tissue/stem cell niche fate and a vascular precursor fate.The ground tissue and stem cell niche fates soon separate and a subset of more specialized vascular cells form sucrose transporting phloem cells that appear to connect to the primary root.We did not detect cells resembling epidermis or root cap,suggesting that outer tissues may form later,preceding LR emergence.At this stage,some remaining initial precursor cells form the primordium flanks,while the rest create a reservoir of pluripotent cells that are able to replace the LR if damaged.Laser ablation of the central and lateral LRP regions showed that remaining cells restart the sequence of tissue initiation to form a LR.Collectively,our study reveals an ontological hierarchy for LR formation with an early and sequential split of main root tissues and stem cells.Laura Serrano-Ron Pablo Perez-Garcia Alvaro Sanchez-Corrionero Inmaculada Gude Javier Cabrera Pui-Leng Ip Kenneth D.Birnbaum Miguel A.Moreno-Risueno 2021Molecular Plant2021,14,8:2

7Analyzing the genomic and transcriptomic architecture of milk traits in Murciano-Granadina goats显示文摘Background:In this study,we aimed to investigate the molecular basis of lactation as well as to identify the genetic factors that influence milk yield and composition in goats.To achieve these two goals,we have analyzed how the mRNA profile of the mammary gland changes in seven Murciano-Granadina goats at each of three different time points,i.e.78 d(T1,early lactation),216 d(T2,late lactation)and 285 d(T3,dry period)after parturition.Moreover,we have performed a genome-wide association study(GWAS)for seven dairy traits recorded in the 1st lactation of 822 Murciano-Granadina goats.Results:The expression profiles of the mammary gland in the early(T1)and late(T2)lactation were quite similar(42 differentially expressed genes),while strong transcriptomic differences(more than one thousand differentially expressed genes)were observed between the lactating(T1/T2)and non-lactating(T3)mammary glands.A large number of differentially expressed genes were involved in pathways related with the biosynthesis of amino acids,cholesterol,triglycerides and steroids as well as with glycerophospholipid metabolism,adipocytokine signaling,lipid binding,regulation of ion transmembrane transport,calcium ion binding,metalloendopeptidase activity and complement and coagulation cascades.With regard to the second goal of the study,the performance of the GWAS allowed us to detect 24 quantitative trait loci(QTLs),including three genome-wide significant associations:QTL1(chromosome 2,130.72-131.01 Mb)for lactose percentage,QTL6(chromosome 6,78.90-93.48 Mb)for protein percentage and QTL17(chromosome 17,11.20 Mb)for both protein and dry matter percentages.Interestingly,QTL6 shows positional coincidence with the casein genes,which encode 80%of milk proteins.Conclusions:The abrogation of lactation involves dramatic changes in the expression of genes participating in a broad array of physiological processes such as protein,lipid and carbohydrate metabolism,calcium homeostasis,cell death and tissue remodeling,as well as immunity.We also conclude that genetic variation at the casein genes has a major impact on the milk protein content of Murciano-Granadina goats.Dailu Guan Vincenzo Landi María Gracia Luigi-Sierra Juan Vicente Delgado Xavier Such Anna Castelló Betlem Cabrera Emilio Mármol-Sánchez Javier Fernández-Alvarez JoséLuis Ruiz de la Torre Casañas Amparo Martínez Jordi Jordana Marcel Amills 2020Journal of Animal Science and Biotechnology2020,11,3:0

9The cold-induced factor CBF3 mediates root stem cell activity,regeneration,and developmental responses to cold显示文摘Plant growth and development involve the specification and regeneration of stem cell niches(SCNs).Although plants are exposed to disparate environmental conditions,how environmental cues affect developmental programs and stem cells is not well understood.Root stem cells are accommodated in meristems in SCNs around the quiescent center(QC),which maintains their activity.Using a combination of genetics and confocal microscopy to trace morphological defects and correlate them with changes in gene expression and protein levels,we show that the cold-induced transcription factor(TF)C-REPEAT BINDING FACTOR 3(CBF3),which has previously been associated with cold acclimation,regulates root development,stem cell activity,and regeneration.CBF3 is integrated into the SHORT-ROOT(SHR)regulatory network,forming a feedback loop that maintains SHR expression.CBF3 is primarily expressed in the root endodermis,whereas the CBF3 protein is localized to other meristematic tissues,including root SCNs.Complementation of cbf3-1 using a wild-type CBF3 gene and a CBF3 fusion with reduced mobility show that CBF3 movement capacity is required for SCN patterning and regulates root growth.Notably,cold induces CBF3,affecting QC activity.Furthermore,exposure to moderate cold around 10℃–12℃promotes root regeneration and QC respecification in a CBF3-dependent manner during the recuperation period.By contrast,CBF3 does not appear to regulate stem cell survival,which has been associated with recuperation from more acute cold(-4℃).We propose a role for CBF3 in mediating the molecular interrelationships among the cold response,stem cell activity,and development.Pablo Perez-Garcia Ornella Pucciariello Alvaro Sanchez-Corrionero Javier Cabrera Cristina del Barrio Juan Carlos del Pozo Mariano Perales Krzysztof Wabnik Miguel A.Moreno-Risueno 2023Plant Communications2023,4,6:0
