| 1 | Method for prevention and control of spontaneous combustion of coal seam and its application in mining field显示文摘Spontaneous combustion of coal seam has been and continues to be a big problem in coal mines. It could pose great threat to the safety of the whole mine and all miners, especially when it occurs in or nearby coal mines. Besides, environment of area surrounded mines during combustion can be threatened where large amount of toxic gases including CO_2, CO, SO_2 and H_2S can be leased by fire in mine. Hence, it is important and significant for scholars to study the controlling and preventing of the coal seam fire. In this paper, the complicated reasons for the occurrence and development of spontaneous combustion in coal seam are analysed and different models under various air leakage situations are built as well. Based on the model and approximately calculation, the difficulty of fire extinguishment in coal seam is pointed out as the difficulty and poor effect to remove the large amount of heat released. Detailed measurements about backfilling and case analyses are also provided on the basis of the recent ten years' practice of controlling spontaneous combustion in coal seams in China. A technical fire prevention and control method has been concluded as five steps including detection, prevention, sealing, injection and pressure adjustment. However, various backfill materials require different application and environmental factors, so in this paper, analyses and discussion about the effect and engineering application of prevention of spontaneous combustion are provided according to different backfilling technologies and methods. | 2 | Femtosecond-laser sharp shaping of millimeter-scale geometries with vertical sidewalls显示文摘As femtosecond(fs)laser machining advances from micro/nanoscale to macroscale,approaches capable of machining macroscale geometries that sustain micro/nanoscale precisions are in great demand.In this research,an fs laser sharp shaping approach was developed to address two key challenges in macroscale machining(i.e.defects on edges and tapered sidewalls).The evolution of edge sharpness(edge transition width)and sidewall tapers were systematically investigated through which the dilemma of simultaneously achieving sharp edges and vertical sidewalls were addressed.Through decreasing the angle of incidence(AOI)from 0◦to−5◦,the edge transition width could be reduced to below 10µm but at the cost of increased sidewall tapers.Furthermore,by analyzing lateral and vertical ablation behaviors,a parameter-compensation strategy was developed by gradually decreasing the scanning diameters along depth and using optimal laser powers to produce non-tapered sidewalls.The fs laser ablation behaviors were precisely controlled and coordinated to optimize the parameter compensations in general manufacturing applications.The AOI control together with the parameter compensation provides a versatile solution to simultaneously achieve vertical sidewalls as well as sharp edges of entrances and exits for geometries of different shapes and dimensions.Both mm-scale diameters and depths were realized with dimensional precisions below 10µm and surface roughness below 1µm.This research establishes a novel strategy to finely control the fs laser machining process,enabling the fs laser applications in macroscale machining with micro/nanoscale precisions. | Qiuchi Zhu Peixun Fan Nan Li Timothy Carlson Bai Cui Jean-François Silvain Jerry L Hudgins Yong Feng Lu | 2021 | International Journal of Extreme Manufacturing2021,3,4: | 6 |
| 3 | Regulation of hepatic micro RNA expression by hepatocyte nuclear factor 4 alpha显示文摘AIM To uncover the role of hepatocyte nuclear factor 4 alpha(HNF4α) in regulating hepatic expression of micro RNAs.METHODS Microarray and real-time PCR were used to determine hepatic expression of micro RNAs in young-adult mice lacking Hnf4α expression in liver(Hnf4α-Liv KO). Integrative genomics viewer software was used to analyze the public chromatin immunoprecipitation-sequencing datasets for DNA-binding of HNF4α, RNA polymerase-Ⅱ, and histone modifications to loci of micro RNAs in mouse liver and human hepatoma cells. Dual-luciferase reporter assay was conducted to determine effects of HNF4α on the promoters of mouse and human micro RNAs as well as effects of micro RNAs on the untranslated regions(3'UTR) of two genes in human hepatoma cells. RESULTS Microarray data indicated that most micro RNAs remained unaltered by Hnf4α deficiency in Hnf4α-Liv KO mice. However, certain liver-predominant micro RNAs were down-regulated similarly in young-adult male and female Hnf4α-Liv KO mice. The down-regulation of mi R-101, mi R-192, mi R-193 a, mi R-194, mi R-215, mi R-802, and mi R-122 as well as induction of mi R-34 and mi R-29 in male Hnf4α-Liv KO mice were confirmed by real-timePCR. Analysis of public chromatin immunoprecipitationsequencing data indicates that HNF4α directly binds to the promoters of mi R-101, mi R-122, mi R-194-2/mi R-192 and mi R-193, which is associated with histone marks of active transcription. Luciferase reporter assay showed that HNF4α markedly activated the promoters of mouse and human mi R-101b/mi R-101-2 and the mi R-194/mi R-192 cluster. Additionally, mi R-192 and mi R-194 significantly decreased activities of luciferase reporters for the 3'UTR of histone H3F3 and chromodomain helicase DNA binding protein 1(CHD1), respectively, suggesting that mi R-192 and mi R-194 might be important in chromosome remodeling through directly targeting H3F3 and CHD1.CONCLUSION HNF4α is essential for hepatic basal expression of a group of liver-enriched micro RNAs, including mi R-101, mi R-192, mi R-193 a, mi R-194 and mi R-802, through which HNF4α may play a major role in the post-transcriptional regulation of gene expression and maintenance of the epigenome in liver. | Hong Lu Xiaohong Lei Jerry Liu Curtis Klaassen | 2017 | World Journal of Hepatology2017,9,4: | 3 |
