1Genome wide abnormal DNA methylome of human blastocyst in assisted reproductive technology显示文摘Proper reprogramming of parental DNA methylomes is essential for mammalian embryonic development.However,it is unknown whether abnormal methylome reprogramming occurs and is associated with the failure of embryonic development.Here we analyzed the DNA methylomes of 57 blastocysts and 29 trophectoderm samples with different morphological grades during assisted reproductive technology(ART) practices.Our data reveal that the global methylation levels of high-quality blastocysts are similar(0.30 ± 0.02,mean ± SD).while the methylation levels of low-quality blastocysts are divergent and away from those of high-quality blastocysts.The proportion of blastocysts with a methylation level falling within the range of 0.30 ± 0.02 in different grades correlates with the live birth rate for that grade.Moreover,abnormal methylated regions are associated with the failure of embryonic development.Furthermore,we can use the methylation data of cells biopsied from trophectoderm to predict the blastocyst methylation level as well as to detect the aneuploidy of the blastocysts.Our data indicate that global abnormal methylome reprogramming often occurs in human embryos,and suggest that DNA methylome is a potential biomarker in blastocyst selection in ART.Guoqiang Li Yang Yu Yong Fan Congru Li Xiaocui Xu Jialei Duan Rong Li Xiangjin Kang Xin Ma Xuepeng Chen Yuwen Ke Jie Yan Ying Lian Ping Liu Yue Zhao Hongcui Zhao Yaoyong Chen Xiaofang Sun Jianqiao Liu Jie Qiao Jiang Liu 2017Journal of Genetics and Genomics2017,44,10:5

4A sustained release of BMP2 in urine-derived stemcells enhances the osteogenic differentiation and the potential of bone regeneration显示文摘Cell-based tissue engineering is one of the optimistic approaches to replace current treatments for bone defects.Urine-derived stem cells(USCs)are obtained non-invasively and become one of the promising seed cells for bone regeneration.An injectable BMP2-releasing chitosan microspheres/type I collagen hydrogel(BMP2-CSM/Col I hydrogel)was fabricated.USCs proliferated in a time-dependent fashion,spread with good extension and interconnected with each other in different hydrogels both for 2D and 3D models.BMP2 was released in a sustained mode for more than 28 days.Sustained-released BMP2 increased the ALP activities and mineral depositions of USCs in 2D culture,and enhanced the expression of osteogenic genes and proteins in 3D culture.In vivo,the mixture of USCs and BMP2-CSM/Col I hydrogels effectively enhanced bone regeneration,and the ratio of new bone volume to total bone volume was 38%after 8weeks of implantation.Our results suggested that BMP2-CSM/Col I hydrogels promoted osteogenic differentiation of USCs in 2D and 3D culture in vitro and USCs provided a promising cell source for bone tissue engineering in vivo.As such,USCs-seeded hydrogel scaffolds are regarded as an alternative approach in the repair of bone defects.Shuang Wu Zhao Chen Xi Yu Xin Duan Jialei Chen Guoming Liu Min Gong Fei Xing Jiachen Sun Shishu Huang Zhou Xiang 2022Regenerative Biomaterials2022,9,1:0

6Effect of Sr^(2+)on 3D gel-printed Sr_(3-x)Mg_(x)(PO_(4))_(2)composite scaffolds for bone tissue engineering显示文摘Porous magnesium strontium phosphate(Sr_(3-x)Mg_(x)(PO_(4))_(2))(x=2,2.5,3)composite scaffolds were successfully prepared by three dimension gel-printing(3DGP)method in this study.The results show that Sr_(0.5)Mg_(2.5)(PO_(4))_(2)scaffolds had good compressive strength,and Sr_(1.0)Mg_(2.0)(PO_(4))_(2)scaffolds had good degradation rate in vitro.The weight loss rate of Sr_(1.0)Mg_(2.0)(PO_(4))_(2)scaffolds soaked in simulated body fluid(SBF)or 6 weeks was 6.96%,and pH value varied between 7.50 and 8.61,which was within the acceptable range of human body.Preliminary biological experiment shows that MC3T3-E1 cells had good adhesion and proliferation on the surface of Sr_(3-x)Mg_(x)(PO_(4))_(2)scaffolds.Compared with pure Mg3(PO_(4))_(2)scaffolds,strontium doped scaffolds had excellent comprehensive properties,which explain that Sr_(3-x)Mg_(x)(PO_(4))_(2)composite scaffolds can be used for bone tissue engineering.Hongyuan Liu Jialei Wu Siqi Wang Jing Duan Huiping Shao 2023International Journal of Minerals,Metallurgy and Materials2023,30,11:0
