| 3 | Identification of a Stable Quantitative Trait Locus for Percentage Grains with White Chalkiness in Rice (Oryza sativa)显示文摘High chalkiness is a major problem in many rice-producing areas of the world, especially in hybrid rice (Oryza sativa L.) in China. We previously showed a major quantitative trait locus for the percentage of grains with white chalkiness (QTLqPGWC-8) in the interval G1149-R727 on chromosome 8 using a chromosome segment substitution line (CSSL). Here, we selected the line-CSSL50 harboring the QTLqPGWC-8 allele from the CSSLs derived from a cross between Asominori (as a recurrent parent) and IR24 (as a donor parent), which had higher percentage chalkiness, markedly different from that of Asominori. There were also significant differences in starch granules, appearance of amylose content (AAC) and milling qualities between Asominori and CSSL50, but not in grain size or thousand grain weight (TGW). The BC4F2 and BC4F3 populations from a cross between CSSL50 and Asominori were used for fine mapping of qPGWC-8. We narrowed down the location of this QTL to a 142 kb region between Indel markers 8G-7 and 8G-9. QTLqPGWC-8 accounted for 50.9% of the difference in PGWC between the parents. | 4 | Simultaneous heterotrophic nitrification and aerobic denitrification at high initial phenol concentration by isolated bacterium Diaphorobacter sp. PD-7显示文摘A strain capable of phenol degradation, heterotrophic nitrification and aerobic denitrification was isolated from activated sludge of coking-plant wastewater ponds under aerobic condition. Based on its morphology, physiology,biochemical analysis and phylogenetic characteristics, the isolate was identified as Diaphorobacter sp. PD-7.Biodegradation tests of phenol showed that the maximum phenol degradation occurred at the late phase of exponential growth stages, with 1400 mg·L-1phenol completely degraded within 85 h. Diaphorobacter sp. PD-7accumulated a vast quantity of phenol hydroxylase in this physiological phase, ensuring that the cells quickly utilize phenol as a sole carbon and energy source. The kinetic behavior of Diaphorobacter sp. PD-7 in batch cultures was investigated over a wide range of initial phenol concentrations(0–1400 mg·L-1) by using the Haldane model,which adequately describes the dynamic behavior of phenol biodegradation by strain Diaphorobacter sp. PD-7. At initial phenol concentration of 1400 mg·L-1, batch experiments(0.25 L flask) of nitrogen removal under aerobic condition gave almost entirely removal of 120.69 mg·L-1ammonium nitrogen within 75 h, while nitrate nitrogen removal reached 91% within 65 h. | 8 | Diagenesis and porosity evolution of sandstone reservoirs in the East II part of Sulige gas field, Ordos Basin显示文摘It is becoming an important controlling factor of gas exploration and exploitation in the east part of Sulige gas field in the Ordos Basin where the reservoir of main gas formations is tight sandstones. Employing experimental methods of slice identification, casting slice, scan electron microscope, and X-ray diffractions, we studied the characteristics of petrology and diagenesis on reservoirs in Shan1 section of Shanxi formation and He8 section of Shihezi formation of the Permian system in the East II part of Sulige gas field. The results include: (1) the main sandstones in these areas are dominated by lithic sandstone and lithic silicarenite with low grade of maturity; (2) the diagenesis of sandstone in these areas mainly include compaction, cementation, corrosion and alteration. | 10 | Depositional and Diagenetic Controls on Sandstone Reservoirs with Low Porosity and Low Permeability in the Eastern Sulige Gas Field, China显示文摘In order to determine the genesis and the factors that control the low-porosity and lowpermeability sandstone reservoirs in the eastern Sulige Gas Field in the Ordos Basin, systematic studies on the sedimentary facies and diagenesis were conducted by means of analysis of cores, thin sections, fluid inclusions, X-ray diffraction, cathode luminescence and scanning electron microscope. It was found that the sand bodies of the major gas reservoirs in the Shan1 section(P1S1) and the He8 section(P2H8) were formed during the Permian as sedimentary facies such as braided-channel bars, braided-river channels and point bars of a meandering river. Four types of diagenetic facies developed subsequently: in order from the best to the poorest properties these are type A(weak compaction, early calcite cement-chlorite film facies), type B(moderate compaction, quartz overgrowth-feldspar corrosion-kaolinite filling facies), type C(strong compaction, late calcite cement-quartz corrosion facies) and type D(matrix filling and strong compaction facies). This diagenesis is undoubtedly the main reason for the poor reservoir properties of sandstone reservoirs, but the sedimentary facies are the underlying factors that greatly affect the diagenesis and thus the reservoir performance. Favorable diagenetic facies developed mainly in relatively small lithofacies such as braided-river channels, channel bars and point bars. The vertical distribution of the physical properties and the diagenetic facies of the reservoirs are related to the stratigraphic succession. Most of the sandstones between mudstones and thin beds of sandstone are unfavorable diagenetic facies. Analyses indicate that siliceous cementation can hardly be stopped by hydrocarbon filling. Authigenic chlorite could hardly protect the primary porosity. It not only occupies pore space, but also blocks pathways through sandstone reservoirs, so that it has significant influence on the permeability. | 14 | Neocryptotanshinone inhibits lipopolysaccharide-induced inflammation in RAW264.7 macrophages by suppression of NF-κB and iNOS signaling pathways显示文摘Neocryptotanshinone(NCTS) is a natural product isolated from traditional Chinese herb Salvia miltiorrhiza Bunge. In this study, we investigated its anti-inflammatory effects in lipopolysaccharide(LPS)-stimulated mouse macrophage(RAW264.7) cells. MTT results showed that NCTS partly reversed LPS-induced cytotoxicity. Real-time PCR results showed that NCTS suppressed LPS-induced mRNA expression of inflammatory cytokines, including tumor necrosis factor α(TNFα), interleukin-6(IL-6) and interleukin-1β(IL-1β). Moreover, NCTS could decrease LPS-induced nitric oxide(NO)production. Western blotting results showed that NCTS could down-regulate LPS-induced expression of inducible nitric oxide synthase(iNOS), p-IκBα, p-IKKβ and p-NF-κB p65 without affecting cyclooxygenase-2(COX-2). | 17 | GW5-Like, a homolog of GW5, negatively regulates grain width, weight and salt resistance in rice显示文摘Grain size is an important determinant of yield potential in crops. We previously demonstrated that natural mutations in the regulatory sequences of qSW5/GW5 confer grain width diversity in rice. However, the biological function of a GW5 homolog, named GW5-Like(GW5 L), remains unknown. In this study, we report on GW5 L knockout mutants in Kitaake, a japonica cultivar(cv.)considered to have a weak gw5 variant allele that confers shorter and wider grains. GW5 L is evenly expressed in various tissues, and its protein product is localized to the plasma membrane. Biochemical assays verified that GW5 L functions in a similar fashion to GW5. It positively regulates brassinosteroid(BR) signaling through repression of the phosphorylation activity of GSK2. Genetic data show that GW5 L overexpression in either Kitaake or a GW5 knockout line, Kasaorf3(indica cv. Kasalath background), causes more slender, longer grains relative to the wild-type. We also show that GW5 L could confer salt stress resistance through an association with calmodulin protein OsCa M1-1. | 18 | NUMERICAL ANALYSIS OF THE MECHANICS OF SOIL REINFORCEMENT显示文摘Roots affect the soil stress and strain and contribute to soil reinforcement. An analysis of stress and strain in the root-soil composite is presented which combines roots, soils, and an interface element by using the Duncan-Chang E- model and a nonlinear-interface-element model. In this research, a finite element numerical analysis method was applied to simulate the stress field resulting in a composite of Pinus tabulaeformis roots and soils. Results show that roots can transmit the stress from surface soils to deep soils, and can reduce soil stress within the surface soil layer; results also show that the effect of soil reinforcement by roots is limited to surface soils. | 20 | Identification and fine mapping of two blast resistance genes in rice cultivar 93-11显示文摘Rice blast, caused by Magnaporthe oryzae, is a major disease of rice almost worldwide. The Chinese indica cultivar 93-11 is resistant to numerous isolates of the blast fungus in China, and can be used as broad-spectrum resistance resource, particularly in japonica rice breeding programs. In this study, we identified and mapped two blast resistance genes, Pi60(t) and Pi61(t), in cv. 93-11 using F2 and F3 populations derived from a cross between the susceptible cv. Lijiangxintuanheigu(LTH) and resistant cv. 93-11 and inoculated with M. oryzae isolates from different geographic origins. Pi60(t) was delimited to a 274 kb region on the short arm of chromosome 11, flanked by InDel markers K1-4 and E12 and cosegregated with InDel markers B1 and Y10. Pi61(t) was mapped to a 200 kb region on the short arm(near the centromere) of chromosome 12, flanked by InDel markers M2 and S29 and cosegregating with InDel marker M9. In the 274 kb region of Pi60(t), 93-11 contains six NBS-LRR genes including the two Pia/ PiCO39 alleles(BGIOSGA034263 and BGIOSGA035032) which are quite close to the two Pia/ PiCO39 alleles(SasRGA4 and SasRGA5) in Sasanishiki and CO39, with only nine amino acids differing in the protein sequences of BGIOSGA035032 and SasRGA5. In the 200 kb region of Pi61(t), 93-11 contains four NBS-LRR genes, all of which show high identities in protein sequence with their corresponding NBS-LRR alleles in susceptible cv. Nipponbare. Comparison of the response spectra and physical positions between the target genes and other R genes in the same chromosome regions indicated that Pi60(t) could be Pia/PiCO39 or its allele, whereas Pi61(t) appears to be different from Pita, Pita-2, Pi19(t), Pi39(t) and Pi42(t) in the same R gene cluster.
