| 2 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 14 | Relationship between Geological Structures and Hydrocarbon Enrichment of Different Depressions in the Bohai Bay Basin显示文摘The Bohai Bay Basin contains many depressions with varying degrees of hydrocarbon enrichment associated with the geological structures of different depressions. This study discussed the relationship between the geological structures and hydrocarbon enrichment of the depressions in the Bohai Bay Basin. Based on the Paleogene strata distribution and the length to width ratio of different depressions, their geological structures are divided into three types in plan-view: open(length/width < 2), narrow(length/width > 4) and transitional types(length/width 2–4). In cross section, the geological structures can be divided into dustpan I, dustpan II and double-faulted types. Based on tectonic evolution and sedimentary characteristics, the depressions are classified into early-formed, inherited and late-formed categories. Generally, narrow depressions are mainly located in the northeast and southwest of the Bohai Bay Basin, while open depressions are dominantly distributed in the central area of the basin; late-formed depressions are mainly around the Bohai sea area, and early-formed depressions are mostly located in the periphery of the basin. Geological structures of the depressions control the formation of the source, reservoir and cap rocks as well as hydrocarbon accumulation setting, and further influence the pay zones and oil-bearing sequence. In detail, dustpan II and doublefaulted depressions mainly have A-type sags, which often possess better hydrocarbon generation conditions than dustpan I ones; hydrocarbons in open dustpan II depressions tend to accumulate in the central uplift areas or buried hill, while those in narrow dustpan I depressions always accumulate in gentle slope belts. The oil-bearing sequence for different evolutional depressions corresponds well with the sedimentary strata of the main development stages of depressions. In early-formed depressions, hydrocarbons are mainly enriched in deeply buried reservoirs, while in late-formed depressions hydrocarbons are abundant in the relatively shallow traps. | 15 | High expression levels of IKK alpha and IKK beta are necessary for the malignant properties of liver cancer显示文摘IKK-NF-kappa B signaling is regarded as an important factor in hepatocarcinogenesis and a potential target for liver cancer therapy.Therefore,in this study,we analyzed the expression of mRNAs encoding components and targets of NF-kappa B signaling including IKK alpha,IKK beta,RANK,RANKL,OPG,CyclinD3,mammary serine protease inhibitor(Maspin),CyclinD1,c-FLIP,Bcl-x1,Stat3,Cip1 and Cip2 by real-time PCR in 40 patients with liver cancer.After statistical analysis,7 indices including IKK alpha,IKK beta,RANK,Maspin,c-FLIP,Cip2 and cyclinD1 were found to show significant differences between tumor tissue and its corresponding adjacent tissue.When IKK alpha and IKK beta were downregulated in the hepatocellular carcinoma(HCC) cell lines of MHCC-97L and MHCC-97H in vitro,the numbers of BrdU positive cells were decreased in both IKK alpha and IKK beta knockdown cells.Levels of apoptosis were also investigated in IKK alpha and IKK beta knockdown cells.The growth of HCC was inhibited in the subcutaneous implantation model,and lung metastatogenesis was also significantly inhibited in the kidney capsule transplantation model.Downregulation of IKK alpha and IKK beta in HCC cultured in vitro revealed that increased Maspin,OPG and RANKL expression was associated with metastasis of HCC.These findings were associated with downregulation of Bcl-XL and c-FLIP,which may be the reason for increased apoptosis.The therapeutic effect of IKK alpha and IKK beta downregulation depends on extent of NF-kappa B inhibition and the malignant nature of the HCC.We anticipate that IKK-targeted gene therapy can be used in the treatment of HCC,a cancer that is notoriously resistant to radiation and chemotherapy. | Jiang,RQ Xia,YX Li,J Deng,L Zhao,L Shi,J Wang,XH Sun,BC Nanjing Med Univ,Liver Transplantat Ctr,Affiliated Hosp 1,Nanjing,Jiangsu Prov,Peoples R China Nanjing Med Univ,Ctr Canc,Nanjing,Jiangsu Prov,Peoples R China | 2010 | 南京医科大学学报(自然科学版)2010,30,5: | 13 |

| 17 | Effect of high-molecular-weight glutenin subunit deletion on soft wheat quality properties and sugar-snap cookie quality estimated through near-isogenic lines显示文摘High-molecular-weight glutenin subunits(HMW-GSs) play a critical role in determining the viscoelastic properties of wheat dough. The HMW-GSs are encoded by Glu-A1, Glu-B1, and Glu-D1 loci on the long arms of chromosomes 1A, 1B, and 1D, respectively. In the present study, four near-isogenic lines with different HMW-GS deletions and compositions at the Glu-A1 and Glu-D1 loci in Yangmai 18 background were used for quality analysis. Deletion in Glu-D1 showed much weaker gluten quality and dough strength than null Glu-A1 genotype and wild genotype(WT), based on the measurements of sodium dodecyl sulfate(SDS)-sedimentation, lactic acid solvent retention capacity(SRC), gluten index, development time, stability time, and alveograph P and L values. The deletion of Glu-D1 did not significantly affect grain hardness, grain protein content, water SRC, sodium carbonate SRC, and sucrose SRC. Double null genotype in Glu-A1 and Glu-D1 and single null genotype in Glu-D1 showed significantly higher cookie diameter, crispness, and lower cookie height compared with single null genotype in Glu-A1 and WT. | 20 | U-Pb zircon geochronology, geochemistry and kinetics of the Huaniushan A-type granite in Northwest China显示文摘The Huaniushan granite is located at the Beishan orogenic belt, northwestern China. At the contact zone between the granite and marble, a hydrothermal Pb-Zn and skarn Au deposit is formed. LA-ICP-MS zircon U-Pb dating yielded a weighted mean 206Pb/238U age of 229.5±2.6 Ma (MSDW=0.93) for the Huaniushan granite, imply-ing its Late Triassic intrusion. Geochemistry analyses show that the Huaniushan granite is enriched in Si, K, Na, and REE, and depleted in Mg and Ca, with contents of SiO2 (70.8% to 74.4%), Na2O+K2O (8.8% to 10.2%), CaO (0.93% to 1.44%), and MgO (0.14% to 0.48%). REE is characterized by obvious negative Eu anomaly. Rb, Th, U, K, Pb, Nb, Zr and Hf elements are rich in the granite while Ba, Sr, P, Ti and Eu are deplete. The granite has a high (Zr+Nb+Ce+Y) abundance and 104 Ga/Al ratios. Petrology, major and trace elements data all indicate that the Hua-niushan granite is A-type granite which intruded in a post-collisional extensional tectonic setting. The magma was dominantly sourced from partial melting of crustal intermediate-felsic igneous rocks.
