1Intestinal lysozyme liberates Nod1 ligands from microbes to direct insulin trafficking in pancreatic beta cells显示文摘Long-range communication between intestinal symbiotic bacteria and extra-intestinal organs can occur through circulating bacterial signal molecules, through neural circuits, or through cytokines or hormones from host cells. Here we report that Nod1 ligands derived from intestinal bacteria act as signal molecules and directly modulate insulin trafficking in pancreatic beta cells. The cytosolic peptidoglycan receptor Nod1 and its downstream adapter Rip2 are required for insulin trafficking in beta cells in a cell-autonomous manner. Mechanistically, upon recognizing cognate ligands, Nod1 and Rip2 localize to insulin vesicles, recruiting Rab1a to direct insulin trafficking through the cytoplasm. Importantly, intestinal lysozyme liberates Nod1 ligands into the circulation, thus enabling long-range communication between intestinal microbes and islets. The intestine-islet crosstalk bridged by Nod1 ligands modulates host glucose tolerance. 2Detection of 2-thiaadamantanes in the oil from Well TZ-83 in Tarim Basin and its geological implication显示文摘Thermochemical sulfate reduction(TSR)causes increasing of sulfur content in the oils,reduction of oil quality,and produces a toxic and corrosive H2S which may take great adverse effect on health and equipment.Although there are several ways to identify presence of TSR in gas field,yet in the case of oil with low H2S content,it is still very difficult to identify TSR.2-thiaadamantanes is one of the most effective indicators of TSR.This paper for the first time discovers 2-thiaadamantanes in the oil from Well TZ-83 in Tarim Basin.A silver nitrate silica gel chromatography was used to enrich the oil from low 2-thiaadamantanes content to the level that GC/MS and GC/MS/MS can detect.2-thiaadamantanes were assigned by comparison of mass spectra with published data,and the fragmental ions in mass spectra were confirmed by MRM mode of GC/MS/MS.H2S gas produced in Tazhong area is thought to be gen-erated from TSR since 2-thiaadamantanes were present in the oil.JIANG NaiHuang ZHU GuangYou ZHANG ShuiChang WANG ZhengJun 2008Chinese Science Bulletin2008,53,3:9

11A Case Report:Severe Bullous Skin Reaction Induced by Anti-PD-1 Antibody(Toripalimab)Therapy in a Patient with Thymic Carcinoma显示文摘The development of immune checkpoint inhibitors,such as those targeting programmed cell death protein 1(PD-1),represents a major breakthrough in cancer therapy.Although immune checkpoint blockade therapy has a favorable risk/benefit ratio,it causes significant immune-related adverse events(irAEs),such as cutaneous reactions,in particular,severe bullous skin reactions and toxic epidermal necrolysis.Here,we report a case of a 51-year-old woman with malignant thymoma who developed a severe bullous skin reaction(characterized by a systemic rash,bullae,epidermal desquamation,and Stevens-Johnson syndrome)as a result of treatment with the PD-1 inhibitor toripalimab.The patient was treated with high doses of glucocorticoid,intravenous immunoglobulin,and intensive care,and eventually recovered from the severe irAEs.The intravenous injection of anti-PD-1 antibodies induces cutaneous reactions,which are associated with higher mortality rates.High doses of glucocorticoid combined with intravenous immunoglobulin are effective in alleviating such irAEs.Thus,improving the level of care and preventing skin infections can effectively reduce the risk of death.Chong TENG Xiaowei SONG Chengjuan FAN Yashuang ZHAO Lina DU Siqi MAN Yuanyuan HU Zhengjun JIANG Tao XIN 2022Clinical Cancer Bulletin2022,1,3:0

13Cancer cells corrupt normal epithelial cells through miR-let-7c-rich small extracellular vesicle-mediated downregulation of p53/PTEN显示文摘Tumor volume increases continuously in the advanced stage, and aside from the self-renewal of tumor cells, whether the oncogenic transformation of surrounding normal cells is involved in this process is currently unclear. Here, we show that oral squamous cell carcinoma(OSCC)-derived small extracellular vesicles(s EVs) promote the proliferation, migration, invasion, and epithelial-mesenchymal transition(EMT) of normal epithelial cells but delay their apoptosis. In addition, nuclear-cytoplasmic invaginations and multiple nucleoli are observed in s EV-treated normal cells, both of which are typical characteristics of premalignant lesions of OSCC. Mechanistically, mi Rlet-7c in OSCC-derived s EVs is transferred to normal epithelial cells, leading to the transcriptional inhibition of p53 and inactivation of the p53/PTEN pathway. In summary, we demonstrate that OSCC-derived s EVs promote the precancerous transformation of normal epithelial cells, in which the mi R-let-7c/p53/PTEN pathway plays an important role.
