1Maize Genic Male-Sterility Genes and Their Applications in Hybrid Breeding: Progress and Perspectives显示文摘As one of the most important crops, maize not only has been a source of the food, feed, and industrial feedstock for biofuel and bioproducts, but also became a model plant system for addressing fundamental questions in genetics. Male sterility is a very useful trait for hybrid vigor utilization and hybrid seed production. The identification and characterization of genic male-sterility (GMS) genes in maize and other plants have deepened our understanding of the molecular mechanisms controlling anther and pollen development, and enabled the development and efficient use of many biotechnology-based male-sterility (BMS) systems for crop hybrid breeding. In this review, we summarize main advances on the identification and characterization of GMS genes in maize, and con struct a putative regulatory network controlling maize anther and pollen development by comparative genomic analysis of GMS genes in maize, Arabidopsis, and rice. Furthermore, we discuss and appraise the features of more than a dozen BMS systems for propagating male-sterile lines and producing hybrid seeds in maize and other plants. Finally, we provide our perspectives on the studies of GMS genes and the development of novel BMS systems in maize and other plants. 2Epidemiological and risk analysis of the H7N9 subtype influenza outbreak in China at its early stage显示文摘Dozens of human cases infected with H7N9 subtype avian influenza virus (AIV) have been confirmed in China since March, 2013. Distribution data of sexes, ages, professions and regions of the cases were analyzed in this report. The results showed that the elderly cases, especially the male elderly, were significantly more than expected, which is different from human cases of H5N1 avian influenza and human cases of the pandemic H1N1 influenza. The outbreak was rated as a Grade Ⅲ (severe) outbreak, and it would evolve into a Grade IV (very severe) outbreak soon, using a method reported previously. The H7N9 AIV will probably circulate in humans, birds and pigs for years. Moreover, with the driving force of natural selection, the virus will probably evolve into highly pathogenic AIV in birds, and into a deadly pandemic influenza virus in humans. Therefore, the H7N9 outbreak has been assumed severe, and it is likely to become very or extremely severe in the future, highlighting the emergent need of forceful scientific measures to eliminate any infected animal flocks. 3ZmMs30 Encoding a Novel GDSL Lipase Is Essential for Male Fertility and Valuable for Hybrid Breeding in Maize显示文摘Genic male sterility (GMS) is very useful for hybrid vigor utilization and hybrid seed production. Although a large number of GMS genes have been identified in plants, little is known about the roles of GDSL lipase members in anther and pollen development. Here, we report a maize GMS gene, ZmMs30, which encodes a novel type of GDSL lipase with diverged catalytic residues. Enzyme kinetics and activity assays show that ZmMs30 has lipase activity and prefers to substrates with a short carbon chain. ZmMs30 is specifically expressed in maize anthers during stages 7-9. Loss of ZmMs30 function resulted in defective anther cuticle, irregular foot layer of pollen exine, and complete male sterility. Cytological and lipidomics analyses demonstrate that ZmMs30 is crucial for the aliphatic metabolic pathway required for pollen exine formation and anther cuticle development. Furthermore, we found that male sterility caused by loss of ZmMs30 function was stable in various inbred lines with different genetic background, and that it didn't show any negative effect on maize heterosis and production, suggesting that ZmMs30 is valuable for crossbreeding and hybrid seed production. We then developed a new multi-control sterility system using ZmMs30 and its mutant line, and demonstrated it is feasible for generating desirable GMS lines and valu. able for hybrid maize seed production. 4SIS accuracy and service performance of the BDS-3 basic system显示文摘On December 27,2018,the basic system of the third-generation BeiDou navigation satellite system(BDS-3)completed the deployment of its constellation of 18 MEO networking satellites as well as the construction of the operation control system(OCS)and began to provide basic navigation services to users worldwide.Compared with BDS-2,BDS-3 aims to offer users better navigation signals and higher precision with a series of new technologies.For example,the spaceborne atomic clock of BDS-3 is upgraded for higher performance,the Ka-band inter-satellite link is adopted for inter-satellite ranging and communication,and new B1C and B2a signals are broadcast in addition to B1I and B3I signals(compatible with BDS-2).In addition,a 9-parameter model based on a spherical harmonic function is employed for ionospheric delay corrections.Using the observation data from 18 satellites of the basic system,this paper conducts a comprehensive evaluation of the pseudorange measurement characteristics,signal-in-space(SIS)accuracy of navigation messages and global service capability of BDS-3.The results indicate that the pseudorange measurement multipath effect and observation noise of BDS-3 satellites are better than those of BDS-2;additionally,with the support of inter-satellite links,the user range error(URE)of the BDS-3 satellite broadcast ephemeris is better than 10 cm,the precision of the broadcast clock parameter is better than 1.5 ns,and the SIS accuracy is better than 0.6 m overall.Different from the traditional Klobuchar model,the BeiDou global broadcast ionospheric delay correction model(BDGIM)can provide ionospheric delay corrections better than 70%for worldwide single-frequency users.The service capability evaluation of the basic system consists mainly of the accuracy improvement of the B1I and B3I signals according to BDS-2 as well as the global positioning accuracy of the new signals.These results prove that the BDS-3 basic system has achieved the design goal;that is,both the horizontal and the vertical global positioning accuracies are better than 10 m(95%).In the future,6 MEO satellites as well as 3 GEO satellites and 3 IGSO satellites for regional enhancement purposes will be deployed for full operation;consequently,BDS-3 will definitely provide a higher SIS accuracy and better service capability.JinPing Chen XiaoGong Hu ChengPan Tang ShanShi Zhou YuFei Yang JunYang Pan Hui Ren YueXin Ma QiuNing Tian Bin Wu Yang Yu 2020Science China(Physics,Mechanics & Astronomy)2020,63,6:14
5Efficient Synthesis of Graphene Nanoribbons Sonochemically Cut from Graphene Sheets显示文摘We report a facile approach to synthesize narrow and long graphene nanoribbons (GNRs) by sonochemically cutting chemically derived graphene sheets (GSs). The yield of GNRs can reach ~5 wt% of the starting GSs. The resulting GNRs are several micrometers in length, with ~75% being single-layer, and ~40% being narrower than 20 nm in width. A chemical tailoring mechanism involving oxygen-unzipping of GSs under sonochemical conditions is proposed on the basis of experimental observations and previously reported theoretical calculations;it is suggested that the formation and distribution of line faults on graphite oxide and GSs play crucial roles in the formation of GNRs. 6Normal Structure and Function of Endothecium Chloroplasts Maintained by ZmMs33-Mediated Lipid Biosynthesis in Tapetal Cells Are Critical for Anther Development in Maize显示文摘Genic male sterility(GMS)is critical for heterosis utilization and hybrid seed production.Although GMS mutants and genes have been studied extensively in plants,it has remained unclear whether chloroplast-associated photosynthetic and metabolic activities are involved in the regulation of anther development.In this study,we characterized the function of ZmMs33/ZmGPAT6,which encodes a member of the glycerol-3-phosphate acyltransferase(GPAT)family that catalyzes the first step of the glycerolipid synthetic pathway.We found that normal structure and function of endothecium(En)chloroplasts maintained by ZmMs33-mediated lipid biosynthesis in tapetal cells are crucial for maize anther development.ZmMs33 is expressed mainly in the tapetum at early anther developmental stages and critical for cell proliferation and expansion at late stages.Chloroplasts in En cells of wild-type anthers function as starch storage sites before stage 10 but as photosynthetic factories since stage 10 to enable starch metabolism and carbohydrate supply.Loss of ZmMs33 function inhibits the biosynthesis of glycolipids and phospholipids,which are major components of En chloroplast membranes,and disrupts the development and function of En chloroplasts,resulting in the formation of abnormal En chloroplasts containing numerous starch granules.Further analyses reveal that starch synthesis during the day and starch degradation at night are greatly suppressed in the mutant anthers,leading to carbon starvation and low energy status,as evidenced by low trehalose-6-phosphate content and a reduced ATP/AMP ratio.The energy sensor and inducer of autophagy,SnRK1,was activated to induce early and excessive autophagy,premature PCD,and metabolic reprogramming in tapetal cells,finally arresting the elongation and development of mutant anthers.Taken together,our results not only show that ZmMs33 is required for normal structure and function of En chloroplasts but also reveal that starch metabolism and photosynthetic activities of En chloroplasts at different developmental stages are essential for normal anther development.These findings provide novel insights for understanding how lipid biosynthesis in the tapetum,the structure and function of En chloroplasts,and energy and substance metabolism are coordinated to maintain maize anther development.Taotao Zhu Ziwen Li Xueli An Yan Long Xiaofeng Xue Ke Xie Biao Ma Danfeng Zhang Yijian Guan Canfang Niu Zhenying Dong Quancan Hou Lina Zhao Suowei Wu Jinping Li Weiwei Jin Xiangyuan Wan 2020Molecular Plant2020,13,11:9
7Estimation of lunar titanium content: Based on absorption features of Chang’E-1 interference imaging spectrometer (ⅡM)显示文摘Two linear regression models based on absorption features extracted from CE-1 IIM image data are presented to discuss the relationship between absorption features and titanium content. We computed five absorption parameters (Full Wave at Half Maximum (FWHM), absorption position, absorption area, absorption depth and absorption asymmetry) of the spectra collected at Apollo 17 landing sites to build two regression models, one with FWHM and the other without FWHM due to the low relation coefficient between FWHM and Ti content. Finally Ti content measured from Apollo 17 samples and Apollo 16 samples was used to test the accuracy. The results show that the predicted values of the model with FWHM have many singular values and the result of model without FWHM is more stable. 8Experimental Study on the Characteristics of CO_2 Hydrate Formation in Porous Media below Freezing Point显示文摘Porous medium has an obvious effect on the formation of carbon dioxide hydrate. In order to study the characteristics of CO2 hydrate formation in porous medium below the freezing point, the experiment of CO2 hydrate formation was conducted in a high-pressure 1.8-L cell in the presence of porous media with a particle size of 380 μm, 500 μm and 700 μm, respectively. The test results showed that the porous medium had an important influence on the process of CO2 hydrate formation below the freezing point. Compared with porous media with a particle size of 500 μm and 700 μm, respectively, the average hydrate formation rate and gas storage capacity of carbon dioxide hydrate in the porous medium with a particle size of 380 μm attained 0.016 14 mol/h and 65.094 L/L, respectively. 9Design and fabrication of superconducting HEB mixer显示文摘This paper describes the design and fabrication of superconducting hot electron bolometer (HEB) mixer based on ultra-thin superconducting NbN films. The high quality films were epitaxially grown on high resistance Si substrates. The device was fabricated by magnetron sputtering, electron beam lithography (EBL), reactive ion etching (RIE), lithography, and so on. The device's resistance-tempera-ture (R-T) curves and current-voltage (I-V) curves were studied. The results of THz response of the device are presented. Y-factor technique was used to measure the device's noise temperature. 10A facile and general approach for the synthesis of fluorescent silica nanoparticles doped with inert dyes显示文摘A general and facile approach was developed for the synthesis of almost monodisperse fluorescent silica nanoparticles (NPs) doped with inert dyes, which are organic fluorophores that are strongly fluorescent but are hydrophobic or lack a covalent binding group. The prepared NPs were mesoporous and the dye molecules were encapsulated in the pores via hydrophobic interaction with the CTAB template. 12Maternal supply of cysteamine alleviates oxidative stress and enhances angiogenesis in porcine placenta显示文摘Background:Oxidative stress in placenta is associated with the occurrence of adverse pregnancy outcomes in sow,but there are few satisfactory treatment strategies for these conditions.This study investigated the potential of cysteamine(CS)as an antioxidant protectant for regulating the reproductive performance,redox status,and placental angiogenesis of sows.Methods:The placental oxidative stress status and vascular density of piglets with different birth weights:<1.0 kg(low birth weight,LBW)and 1.4–1.6 kg(normal birth weight,NBW)were evaluated,followed by allotting 84 sows to four treatments(n=21)and feeding them with a basal diet supplemented with 0,100,300,or 500 mg/kg of CS from d 85 of gestation to d 21 of lactation,respectively.Placenta,serum,and colostrum samples of sows or piglets were collected,and the characteristics of sows and piglets were recorded.Furthermore,the in vivo results were validated using porcine vascular endothelial cells(PVECs).Results:Compared with the NBW placentae,the LBW placentae showed increased oxidative damage and were vulnerable to angiogenesis impairment.Particularly,H2O2-induced oxidative stress prompted intracellular reactive oxygen species generation and inhibited the tube formation and migration of PVECs as well as the expression of vascular endothelial growth factor-A(VEGF-A)in vitro.However,dietary CS supplementation can alleviate oxidative stress and improve the reproductive performance of sows.Specifically,compared with the control group,dietary 100 mg/kg CS could(1)decrease the stillbirth and invalid rates,and increase both the piglet birth weight in the low yield sows and the placental efficiency;(2)increase glutathione and reduce malondialdehyde in both the serum and the colostrum of sows;(3)increase the levels of total antioxidant capacity and glutathione in LBW placentae;(4)increase the vascular density,the mRNA level of VEGF-A,and the immune-staining intensity of platelet endothelial cell adhesion molecule-1 in the LBW placentae.Furthermore,the in vitro experiment indicated that CS pre-treatment could significantly reverse the NADPH oxidase 2-ROS-mediated inactivation of signal transducer and activator of transcription-3(Stat3)signaling pathway induced by H2O2 inhibition of the proliferation,tube formation,and migration of PVECs.Meanwhile,inhibition of Stat3 significantly decreased the cell viability,tube formation and the VEGF-A protein level in CS pretreated with H_(2)O_(2)-cultured PVECs.Conclusions:The results indicated that oxidative stress and impaired angiogenesis might contribute to the occurrence of LBW piglets during pregnancy,but CS supplementation at 100 mg/kg during late gestation and lactation of sows could alleviate oxidative stress and enhance angiogenesis in placenta,thereby increasing birth weight in low yield sows and reducing stillbirth rate.The in vitro data showed that the underlying mechanism for the positive effects of CS might be related to the activation of Stat3 in PVECs.Shuangbo Huang Zifang Wu Zihao Huang Xiangyu Hao Longmiao Zhang Chengjun Hu Jianfu Wei Jinping Deng Chengquan Tan 2022Journal of Animal Science and Biotechnology2022,13,1:4
13An in situ ATR-FTIR study of mixed collectors BHA/DDA adsorption in ilmenite-titanaugite flotation system显示文摘This paper researched the enhanced flotation separation performance of ilmenite and titanaugite using the mixed collector benzhydroxamic acid/dodecylamine(BHA/DDA).The interface assembly mechanism was mainly investigated through in situ attenuated total reflectance Fourier transform infrared(ATRFTIR)spectroscopy combined with the two-dimensional correlation spectroscopy(2D-COS)and X-ray photoelectron spectroscopy(XPS).It has been found that BHA/DDA mixed collectors successfully separate ilmenite from titanaugite at a molar ratio of 8:1.Zeta potential experiments suggested that,in the presence of mixed collector system,the BHA-DDA complex adsorbed on the ilmenite surface via the chemically adsorbed BHA and the electrostatically adsorbed DDA,however,the complex adsorbed on the surface of titanaugite unstably.According to in situ ATR-FTIR combined with 2D-COS and XPS results,the interface assembly mechanism of BHA/DDA is summarized as:the function group of BHA molecules first binds to the metal sites on minerals to form bidentate ligand,then DDA co-adsorbed with BHA on the surface of minerals through hydrogen bonding.DDA may change the adsorption modes of some BHA on the ilmenite surface from four-membered ring to five-membered ring,while the modes on the titanaugite surface is true opposite.Finally,recommended adsorption configurations of the BHA/DDA complex on the two mineral surfaces are proposed.Liping Luo Houqin Wu Longhua Xu Jinping Meng Jiahui Lu Huan Zhou Xiaomei Huo Lingyun Huang 2021International Journal of Mining Science and Technology2021,31,4:4
14Insights into the enhanced structure stability and electrochemical performance of Ti^(4+)/F^(-) co-doped P2-Na_(0.67)Ni_(0.33)Mn_(0.67)O_(2) cathodes for sodium ion batteries at high voltage显示文摘P2-Na_(0.67)N_(i0.33)Mn_(0.67)O_(2)is considered as a promising cathode material for sodium-ion battery (SIBs)because of its high capacity and discharge potential.However,its practical use is limited by Na^(+)/vacancy ordering and P2-O2 phase transition.Herein,a Ti^(4+)/F^(-) co-doping strategy is developed to address these issues.The optimal P2-Na_(0.67)Ni_(0.33)Mn_(0.37)Ti_(0.3)O_(1.9)F_(0.1) exhibits much enhanced sodium storage performance in the high voltage range of 2.0–4.4 V,including a cycling stability of 77.2%over 300cycles at a rate of 2 C and a high-rate capability of 87.7 m Ah g^(-1) at 6 C.Moreover,the P2-Na_(0.67)Ni_(0.33)Mn_(0.37)Ti_(0.3)O_(1.9)F_(0.1) delivers reversible capacities of 82.7 and 128.1 m Ah g^(-1) at-10 and 50℃ at a rate of 2 C,respectively.The capacity retentions over 200 cycles at-10℃ is 94.2%,implying more opportunity for practical application.In-situ X-ray diffraction analysis reveals that both P2-O2 phase transitions and Na^(+)/vacancy ordering is suppressed by Ti^(4+)/F^(-) co-doping,which resulting in fast Na^(+) diffusion and stable phase structure.The hard carbon//P2-Na_(0.67)Ni_(0.33)Mn_(0.37)Ti_(0.3)O_(1.9)F_(0.1) full cell exhibits a high energy density of 310.2 Wh kg^(-1) and remarkable cyclability with 82.1%retention after 300 cycles at 1 C in the voltage range of 1.5–4.2 V.These results demonstrate that the co-doping Ti^(4+)/F^(-) is a promising strategy to improve the electrochemical properties of P2-Na_(0.67)Ni_(0.33)Mn_(0.67)O_(2),providing a facile tactic to develop high performance cathode materials for SIBs.Pengfei Zhou Jing Zhang Zhennan Che Zuhao Quan Ju Duan Xiaozhong Wu Junying Weng Jinping Zhao Jin Zhou 2022Journal of Energy Chemistry2022,31,4:3
15Evaluation of genetic diversity and relationships within and between two breeds of duck based on microsatellite markers显示文摘The genetic diversity of two natural populations(M,N) of Beijing duck(Anas platyrhynchos) and 11 artificially selected lines of Beijing duck(A,B,E-L,O) from China Gold Star Duck Production Ltd.,along with two Cherry Valley duck lines(C and D) from the British Cherry Valley Livestock Division,was evaluated using 18 microsatellite markers covering 16 linkage groups.A phylogenetic tree of the 15 populations of duck,formed of four main branches,was constructed from Nei's D_A genetic distance.The mean genetic differentiation index(F_(ST)) in all loci,Nei's standard genetic distance(Ds),and the genetic distance D_A between the Beijing duck and the Cherry Valley duck were 0.075,0.143 and 0.142,respectively.These results demonstrated a high degree of genetic similarity between the two breeds and supported the hypothesis that the Cherry Valley duck was derived from the Beijing duck.The F_(ST) matrix of seven clusters of Beijing duck suggested that the efficiency of selection was not significant to some extent and should be supplemented by marker -assisted selection.Fei Wu Yinghua Huang Ying Ma Shengqiang Hu Jinping Hao Ning Li 2009Progress in Natural Science:Materials International2009,19,11:3
16Biological porous carbon encapsulated polyethylene glycol-based phase change composites for integrated electromagnetic interference shielding and thermal management capabilities显示文摘The development of functional composites with excellent thermal management capabilities and electro-magnetic interference(EMI)shielding has become extremely urgent for keeping up with the continuous improvement of the operating speed and efficiency for electronic equipment.In this study,the biolog-ical wood-derived porous carbon(WPC)was determined as the supporting material to encapsulating polyethylene glycol(PEG),and a series of WPC/PEG/Fe_(3)O_(4) phase change composites(PCCs)with excel-lent shape stability,EMI shielding and thermal management capabilities were prepared via a simple vac-uum impregnation method.The Fe_(3)O_(4) magnetic particles modified PCCs have greatly improved the EMI shielding effectiveness(SE).The EMI SE of WP-4(7.5 wt.% Fe_(3)O_(4) in PEG)can be up to 55.08 dB between 8.2−12.4 GHz,however,the WP-0 without Fe_(3)O_(4) addition is only 40.08 dB.Meanwhile,the absorption ratio of electromagnetic waves(EMW)has also increased from 75.02%(WP-0)to 85.56%(WP-4),which effectively prevents secondary pollution.In addition,after wrapping a thin layer of polydimethylsiloxane resin(PDMS),the obtained WP-4 can maintain a high heat storage capacity(109.52 J/g)and good wa-ter stability.In short,the prepared WPC/PEG/Fe_(3)O_(4) PCCs have great potential application value in the thermal management and electromagnetic shielding requirements for electronic devices.Shuang Liu Mengjie Sheng Hao Wu Xuetao Shi Xiang Lu Jinping Qu 2022Journal of Materials Science & Technology2022,,18:3
17Rhodojaponin Ⅵ indirectly targets Cav2.2 channels via N-ethylmaleimide-sensitive fusion protein to alleviate neuropathic pain显示文摘Neuropathic pain is a chronic disease that severely afflicts the life and emotional status of patients,but currently available treatments are often ineffective.Novel therapeutic targets for the alleviation of neuropathic pain are urgently needed.Rhodojaponin Ⅵ,a grayanotoxin from Rhododendron molle,showed remarkable antinociceptive efficacy in models of neuropathic pain,but its biotargets and mechanisms are unknown.Given the reversible action of rhodojaponin Ⅵ and the narrow range over which its structure can be modified,we perforwmed thermal proteome profiling of the rat dorsal root ganglion to determine the protein target of rhodojaponin Ⅵ.N-Ethylmaleimide-sensitive fusion(NSF) was confirmed as the key target of rhodojaponin Ⅵ through biological and biophysical experiments.Functional validation showed for the first time that NSF facilitated trafficking of the Cav2.2 channel to induce an increase in Ca2+current intensity,whereas rhodojaponin Ⅵ reversed the effects of NSF.In conclusion,rhodojaponin Ⅵ represents a unique class of analgesic natural products targeting Cav2.2 channels via NSF.Keliang Chen Tao Wang Yong Li Jun Wu Cheng-Xiao Zhao Sheng Liu Fengrun Sun Yehong Fang Jiahuan Hu Jinping Hu Chong-Jing Zhang Haibo Yu Chao Ma Shi-Shan Yu 2023Acta Pharmaceutica Sinica B2023,13,3:2
18Unraveling specific role of carbon matrix over Pd/quasi-Ce-MOF facilitating toluene enhanced degradation显示文摘Metal organic frameworks(MOFs)derivatives represented by quasi-MOFs have excellent physical and chemical properties and can be applied for the catalytic combustion of volatile organic compounds(VOCs).In this work,Pd/quasi-Ce-BTC synthesized by simple one-step Npyrolysis was applied to the oxidation of toluene,showing excellent toluene catalytic activity(T_(90)=175℃,30000 mL/(g·h)).Microscopic analyses indicate the formation and interaction of a carbon matrix composite quasi-MOF structure interface.The results show that the amorphous carbon matrix formed during the partial pyrolysis of Ce-BTC significantly improves the adsorption and activation capacity of toluene in the reaction,and constructs a reductive system to maintain high concentrations of Ce^(3+)and Pd^(0),which can facilitate the activation and utilization of oxygen in reaction.Quasi in-situ XPS proves that carbon matrix is indirectly involved in the activation and storage of oxygen,and Pd^(0)is the crucial active site for the activation of oxygen.Stability and water resistance tests display good stability of Pd/quasi-Ce-BTC.This work provides a potential method for designing quasi-MOF catalysts towards VOCs effective abatement.Chunjing Su Zhi Li Mengqi Mao Wenhua Ye Jinping Zhong Quanming Ren Hairong Cheng Haomin Huang Mingli Fu Junliang Wu Yun Hu Daiqi Ye Haihong Xu 2022Journal of Rare Earths2022,40,11:2
19Prediction of VIGS efficiency by the Sfold program and its reliability analysis in Gossypium hirsutum显示文摘Genetic transformation in some plant species,including cotton(Gossypium hirsutum), is hampered by laborious and time-consuming processes and often unachievable. Virus-induced gene silencing(VIGS) by double-stranded RNAs can serve as a reverse-genetics tool to determine gene function. However, knockdown levels vary greatly when using a tobacco rattle virus-based vector that carries different c DNA fragments of a gene. How to choose the optional target fragment for high interference efficiency is very challenging. Addressing this challenge requires increasing the efficacy of small interference RNA(si RNA) in target fragment. Here, we describe a method to assess VIGS efficiency by comparing the following parameters of si RNA in target sequence: the disruption energy of the target(DGdisruption), the differential stability of si RNA duplex ends(DSSE), and the internal stability at positions 9–14 of the si RNA antisense strand(AIS), which are calculated by Sfold program(http://gffzz01a2382fbe924301h9w5660knc0k56pu6.ffgz.tsg.suse.edu.cn). We find that the si RNAs with low DGdisruption, high DSSE and high AIS have high activity and easily result in high VIGS efficiency by experimentally testing the actual knockdown levels of the four target genes, Gh PDS,Gh CLA1, Gh AOS1, and Gh CXE1 via choosing different target sequences for each gene.
