1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2Addition of hydrogen peroxide for the simultaneous control of bromate and odor during advanced drinking water treatment using ozone显示文摘Complete removal of the characteristic septic/swampy odor from Huangpu River source water could only be achieved under an ozone dose as high as 4.0 mg/L in an ozone-biological activated carbon(O3-BAC) process, which would lead to the production of high concentrations of carcinogenic bromate due to the high bromide content. This study investigated the possibility of simultaneous control of bromate and the septic/swampy odor by adding H2O2 prior to the O3-BAC process for the treatment of Huangpu River water. H2O2 addition could reduce the bromate concentration effectively at an H2O2/O3(g/g) ratio of 0.5 or higher. At the same time, the septic/swampy odor removal was enhanced by the addition of H2O2, although optimization of the H2O2/O3 ratio was required for each ozone dose. At an ozone dose of 2.0 mg/L, the odor was removed completely at an H2O2/O3 ratio of 0.5. 5Shaking table experimental study of recycled concrete frame-shear wall structures显示文摘In this study, four 1/5 scaled shaking table tests were conducted to investigate the seismic performance of recycled concrete frame-shear wall structures with different recycled aggregates replacement rates and concealed bracing detail. The four tested structures included one normal concrete model, one recycled coarse aggregate concrete model, and two recycled coarse and fi ne aggregate concrete models with or without concealed bracings inside the shear walls. The dynamic characteristics, dynamic response and failure mode of each model were compared and analyzed. Finite element models were also developed and nonlinear time-history response analysis was conducted. The test and analysis results show that the seismic performance of the recycled coarse aggregate concrete frame-shear wall structure is slightly worse than the normal concrete structure. The seismic resistance capacity of the recycled concrete frame-shear wall structure can be greatly improved by setting up concealed bracings inside the walls. 7Quantitative method to determine the regional drinking water odorant regulation goals based on odor sensitivity distribution:Illustrated using 2-MIB显示文摘Taste and odor(T/O) in drinking water often cause consumer complaints and are thus regulated in many countries. However, people in different regions may exhibit different sensitivities toward T/O. This study proposed a method to determine the regional drinking water odorant regulation goals(ORGs) based on the odor sensitivity distribution of the local population. The distribution of odor sensitivity to 2-methylisoborneol(2-MIB) by the local population in Beijing, China was revealed by using a normal distribution function/model to describe the odor complaint response to a 2-MIB episode in 2005, and a 2-MIB concentration of12.9 ng/L and FPA(flavor profile analysis) intensity of 2.5 was found to be the critical point to cause odor complaints. Thus the Beijing ORG for 2-MIB was determined to be 12.9 ng/L. Based on the assumption that the local FPA panel can represent the local population in terms of sensitivity to odor, and that the critical FPA intensity causing odor complaints was 2.5, this study tried to determine the ORGs for seven other cities of China by performing FPA tests using an FPA panel from the corresponding city. ORG values between 12.9 and 31.6 ng/L were determined, showing that a unified ORG may not be suitable for drinking water odor regulations. 8Safety and effectiveness of Traditional Chinese Medicinal herbs for diabetic foot:a systematic review and Meta-analysis显示文摘OBJECTIVE: To evaluate the safety and effectiveness of traditional Chinese medicinal herbs(TCMHs) as an adjunctive treatment for diabetic foot(DF).METHODS: The sources used were Pub Med(1966 to August 2015), the Cochrane Library(1988 to August 2015), the Excerpta Medica Database(1974 to August 2015), Wiley(1807 to August 2015), Ovid(1988 to August 2015), Clinical Trials.gov(1993 to August 2015), the Cochrane Central Register of Controlled Trials(1966 to August 2015), China Science and Technology Journal Database(1994 to August2015), Chi CTR(2007 to August 2015), Sino Med(1978 to August 2015), the China National Knowledge Infrastructure(1984 to August 2015), Wanfang Data Knowledge Service Platform(1998 to August 2015), and the Traditional Chinese Medical Literature Analysis and Retrieval System(TCMLARS)(1984 to August 2015). Studies were identified and selected, and the data were extracted by two independent reviewers. The Cochrane Risk of Bias tool was used to assess the quality of studies. Revman 5.2 software was used for data synthesis and analysis.RESULTS: Sixteen studies were included based on the selection criteria. Of these, seven studies had low bias risk and one had high bias risk. In the overall analysis, TCMHs resulted in a significantly higher total effective rate(OR 5.08; 95% CI 3.50 to7.36; P < 0.000 01), cure rate(OR 2.12; 95% CI1.63 to 2.77; P < 0.000 01), and shorter time to ulcer healing(SMD-0.64; 95% CI-0.89 to-0.40;P < 0.000 01) when compared with non-TCMHs treated DF. The analysis also revealed that significantly fewer amputations occurred in TCMHs patients(OR 0.36; 95% CI 0.20 to 0.65; P = 0.0007).Sensitivity analysis indicated that the findings of the Meta-analysis were robust to study quality,and the funnel plot of the Egger test showed no publication bias.CONCLUSION: TCMHs intervention appears to be more effective for DF, with a similar safety profile,when compared with non-TCMHs treatments, although this result requires further verification with more well-designed studies.Chen Shuo Ma Jianwei Xu Limei Niu Tianhui Dong Jing Liu Wenjun Han Qi 2017Journal of Traditional Chinese Medicine2017,37,6:5
9Impact of 5-Fu/oxaliplatin on mouse dendritic cells and synergetic effect with a colon cancer vaccine显示文摘Objective: The aim of the present study was to investigate the effects of 5-fluorouracil(5-Fu) and oxaliplatin on the function and activation pathways of mouse dendritic cells(DCs), and to clarify whether 5-Fu/oxaliplatin combined with the CD1d-MC38/α-galactosylceramide(α-GC) tumor vaccine exhibits synergistic effects on the treatment of colon cancer in mice.Methods: The combination of the Toll like receptor(TLR) ligands and/or 5-Fu/oxaliplatin was added into myeloid-derived DCs in vitro culture. DC phenotypic changes were detected by flow cytometry, and the secretion of DC cytokines was detected by cytometric bead array(CBA). A MC38 mouse colon cancer model was constructed and the DCs were isolated from the spleen, tumor tissue and lymph nodes following intraperitoneal injection of 5-Fu/oxaliplatin. The cell phenotypes were detected by flow cytometry. The tumor infiltrating leukocytes,splenocytes and lymph node cells were co-cultured with the dead MC38 tumor cells, and the secretion levels of interferon-γ(IFN-γ) were detected. 5-Fu/oxaliplatin combined with our previously developed CD1d-MC38/α-GC tumor vaccine was used to inhibit the growth of MC38 colon cancer in mice, and the tumor growth rate and survival time were recorded.Results: 5-Fu/oxaliplatin exerted no significant effect on the expression of the stimulating phenotypes of DCs in vitro, while it could reduce the expression of programmed death ligand 1/2(PD-L1/L2) and promote interleukin-12(IL-12) secretion by DCs. Furthermore 5-Fu/oxaliplatin was beneficial to the differentiation of T-helper 1(Th1) cells. 5-Fu/oxaliplatin further enhanced the stimulating phenotypic expression of DCs in tumor bearing mice, decreased PD-L1/L2 expression, and specifically activated the lymphocytes. 10Syndecan-2, negatively regulated by miR-20b-5p, contributes to 5-fluorouracil resistance of colorectal cancer cells via the JNK/ERK signaling pathway显示文摘5-Fluorouracil (5-FU) resistance has been long considered as an obstacle to the efficacy of chemotherapy in colorectal cancer (CRC). In this study, we demonstrated the role of miR-20b-5p-regulated syndecan-2 (SDC2) in 5-FU resistance of CRC cells. 5-FU-resistant SW480 CRC cells were established by treatment of SW480 cells with stepwise increase of 5-FU concentration. The results showed that SDC2 was expressed significantly higher in SW480/5-FU cells than in SW480/WT cells as revealed by quantitative real-time polymerase chain reaction and western blot analysis. MTT assay and BrdU assay showed that SDC2 overexpression led to increased cell survival rate, while SDC2 knockdown reversed the drug resistance of SW480/5-FU cells. Wound healing and transwell invasion assays revealed that knockdown of SDC2 inhibited the migratory and invasive ability of SW480/5-FU cells. Moreover, animal experiments indicated that si-SDC2 plays a suppressive role in tumor growth in vivo. We also confirmed that miR-20b-5p interacted with SDC2, which reversed the effect of SDC2 in SW480/5-FU cells via the c-Jun N-terminal kinase (JNK)/extracellular regulated protein kinases (ERK) signaling pathway. 11SARS-CoV-2 nsp12 attenuates type Ⅰ interferon production by inhibiting IRF3 nuclear translocation显示文摘SARS-CoV-2 is the pathogenic agent of COVID-19,which has evolved into a global pandemic.Compared with some other respiratory RNA viruses,SARS-CoV-2 is a poor inducer of type Ⅰ interferon(IFN).Here,we report that SARS-CoV-2 nsp12,the viral RNA-dependent RNA polymerase(RdRp),suppresses host antiviral responses.SARS-CoV-2 nsp12 attenuated Sendai virus(SeV)-or poly(I:C)-induced IFN-β promoter activation in a dose-dependent manner.It also inhibited IFN promoter activation triggered by RIG-I,MDA5,MAVS,and IRF3 overexpression.Nsp12 did not impair IRF3 phosphorylation but suppressed the nuclear translocation of IRF3.Mutational analyses suggested that this suppression was not dependent on the polymerase activity of nsp12.Given these findings,our study reveals that SARS-CoV-2 RdRp can antagonize host antiviral innate immunity and thus provides insights into viral pathogenesis.Wenjing Wang Zhuo Zhou Xia Xiao Zhongqin Tian Xiaojing Dong Conghui Wang Li Li Lili Ren Xiaobo Lei Zichun Xiang Jianwei Wang 2021Cellular & Molecular Immunology2021,18,4:4
12Research on seismic performance of shear walls with concrete filled steel tube columns and concealed steel trusses显示文摘In order to further improve the seismic performance of RC shear walls, a new composite shear wall with concrete filled steel tube (CFT) columns and concealed steel trusses is proposed. This new shear wall is a double composite shear wall; the first composite being the use of three different force systems, CFT, steel truss and shear wall, and the second the use of two different materials, steel and concrete. Three 1/5 scaled experimental specimens: a traditional RC shear wall, a shear wall with CFT columns, and a shear wall with CFT columns and concealed steel trusses, were tested under cyclic loading and the seismic performance indices of the shear walls were comparatively analyzed. Based on the data from these experiments, a thorough elastic-plastic finite element analysis and parametric analysis of the new shear walls were carried out using ABAQUS software. The finite element results of deformation, stress distribution, and the evolution of cracks in each phase were compared with the experimental results and showed good agreement. A mechanical model was also established for calculating the load-carrying capacity of the new composite shear walls. 13Origin and Source of Deep Natural Gas in Nanpu sag, Bohai Bay Basin, China显示文摘Natural gas exploration in Nanpu sag, Bohai Bay Basin, has achieved breakthroughs in recent years, and a number of natural gas and condensate wells with high yield have been found in several structures in the beach area. Daily gas production of single wells is up to 170,000 m3, and high-yield wells are mainly distributed in?the Nanpu No. 1 structural belt.?Studies have shown that these natural gases are mainly hydrocarbon gases, with methane content about 80% to 90% and ethane 6%-9%, so they are mainly wet gas; and non-hydrocarbons are at a low level.?Carbon isotopes of methane range from -42‰ to -36‰, and ethane from -28‰ to -26‰. Calculated maturity based on the relationship between δ13C and Ro of natural gas, the gases are equivalent to those generated from organic matter when Ro is 1.0%-1.7% (mainly 1.25%-1.32%). The natural gas is oil-type gas generated from the source rocks at mature to high mature stage, associated with condensate, so carbon isotopes of the gases are heavier. Natural gas in the Nanpu No.1 structural belt is mainly associated gas with condensate. The analysis of the origin and source of natural gas and condensate, combined with the monomer hydrocarbon carbon isotopes and biomarker, indicated that the main source rocks in the Nanpu No.1 structural belt were Es3 (the lower member of the Shahejie Formation), followed by Es1 (the upper member of the Shahejie Formation).?The high-mature hydrocarbons from source rocks in the deep sag mainly migrated through deep inherited faults into shallow traps and accumulated to form oil and gas pools. 14The genome of the recretohalophyte Limonium bicolor provides insights into salt gland development and salinity adaptation during terrestrial evolution显示文摘Halophytes have evolved specialized strategies to cope with high salinity.The extreme halophyte sea lavender(Limonium bicolor)lacks trichomes but possesses salt glands on its epidermis that can excrete harmful ions,such as sodium,to avoid salt damage.Here,we report a high-quality,2.92-Gb,chromosome-scale L.bicolor genome assembly based on a combination of Illumina short reads,single-molecule,real-time long reads,chromosome conformation capture(Hi-C)data,and Bionano genome maps,greatly enriching the genomic information on recretohalophytes with multicellular salt glands.Although the L.bicolor genome contains genes that show similarity to trichome fate genes from Arabidopsis thaliana,it lacks homologs of the decision fate genes GLABRA3,ENHANCER OF GLABRA3,GLABRA2,TRANSPARENT TESTA GLABRA2,and SIAMESE,providing a molecular explanation for the absence of trichomes in this species.We identified key genes(LbHLH and LbTTG1)controlling salt gland development among classical trichome homologous genes and confirmed their roles by showing that their mutations markedly disrupted salt gland initiation,salt secretion,and salt tolerance,thus offering genetic support for the long-standing hypothesis that salt glands and trichomes may share a common origin.In addition,a whole-genome duplication event occurred in the L.bicolor genome after its divergence from Tartary buckwheat and may have contributed to its adaptation to high salinity.The L.bicolor genome resource and genetic evidence reported in this study provide profound insights into plant salt tolerance mechanisms that may facilitate the engineering of salt-tolerant crops.Fang Yuan Xi Wang Boqing Zhao Xiaojing Xu Miao Shi Bingying Leng Xinxiu Dong Chaoxia Lu Zhongtao Feng Jianrong Guo Guoliang Han Haikuan Zhang Jianwei Huang Min Chen Bao-Shan Wang 2022Molecular Plant2022,15,6:3
15Low soil temperature reducing the yield of drip irrigated rice in arid area by influencing anther development and pollination显示文摘Drip irrigation can produce high rice yields with significant water savings;therefore,it is widely used in arid area water-scarce northern China.However,high-frequency irrigation of drip irrigation with low temperature well water leads to low root zone temperature and significantly reduce the rice yield compared to normal temperature water irrigated rice,for example,reservoir water.The main purpose of this paper is to investigate the effects of low soil temperature on the yield reduction of drip irrigated rice in the spike differentiation stage.The experiment set the soil temperatures at 18℃,24℃and 30℃under two irrigation methods(flood and drip irrigation),respectively.The results showed that,at the 30℃soil temperature,drip irrigation increased total root length by 53%but reduced root water conductivity by 9%compared with flood irrigation.Drip irrigation also increased leaf abscisic acid and proline concentrations by 13%and 5%,respectively.These results indicated that drip irrigated rice was under mild water stress.In the 18℃soil temperature,drip irrigation reduced hydraulic conductivity by 58%,leaf water potential by 40%and leaf net photosynthesis by 25%compared with flood irrigation.The starch concentration in male gametes was also 30%less in the drip irrigation treatment than in the flood irrigation treatment at soil temperature 18℃.Therefore,the main reason for the yield reduction of drip irrigated rice was that the low temperature aggravates the physiological drought of rice and leads to the decrease of starch content in male gametes and low pollination fertilization rate.Low temperature aggravates physiological water deficit in drip irrigated rice and leads to lower starch content in male gametes and low pollination fertilization rate,which is the main reason for the reduced yield of drip irrigated rice.Overall,the results indicated that the low soil temperatures aggravated the water stress that rice was under in the drip irrigated environment,causing declines both in the starch content of male gametes and in pollination rate.Low temperature will ultimately affect the rice yield under drip irrigation.ZHANG Jun DONG Peng ZHANG Haoyu MENG Chaoran ZHANG Xinjiang HOU Jianwei WEI Changzhou 2019Journal of Arid Land2019,11,3:3
16Oil shale resources in China and their utilization显示文摘The unconventional oil and gas resources presented in oil shales have meant these potential sources of hydrocarbons,which has become a research focus. China contains abundant oil shale resources,ranking fourth in the world,with ca. 7 254. 48 × 10~8 t within 24 provinces,including 48 basins and 81 oil shale deposits. About 48% of the total oil shale resources are concentrated in the eastern resource region,with a further 22% in the central resource region. 65% of the total quantity of oil shale resources is present at depths of 0-500 m,with 17% of the total resources being defined as high-quality oil shales yielding more than 10% oil by weight.Chinese oil shale resources are generally hosted by Mesozoic sediments that account for 78% of the total resources. In terms of the geographical distribution of these resources,some 45% are located in plain regions,and different oil shale basins have various characteristics. The oil shale resources in China represent a highly prospective future source of hydrocarbons. 17Trace Amounts of Triple-Functional Additives Enable Reversible Aqueous Zinc-Ion Batteries from a Comprehensive Perspective显示文摘Although their cost-effectiveness and intrinsic safety,aqueous zinc-ion batteries suffer from notorious side reactions including hydrogen evolution reaction,Zn corrosion and passivation,and Zn dendrite formation on the anode.Despite numerous strategies to alleviate these side reactions have been demonstrated,they can only provide limited performance improvement from a single aspect.Herein,a triple-functional additive with trace amounts,ammonium hydroxide,was demonstrated to comprehensively protect zinc anodes.The results show that the shift of electrolyte pH from 4.1 to 5.2 lowers the HER potential and encourages the in situ formation of a uniform ZHS-based solid electrolyte interphase on Zn anodes.Moreover,cationic NH^(4+)can preferentially adsorb on the Zn anode surface to shield the“tip effect”and homogenize the electric field.Benefitting from this comprehensive protection,dendrite-free Zn deposition and highly reversible Zn plating/stripping behaviors were realized.Besides,improved electrochemical performances can also be achieved in Zn//MnO_(2)full cells by taking the advantages of this triple-functional additive.This work provides a new strategy for stabilizing Zn anodes from a comprehensive perspective.Ruwei Chen Wei Zhang Quanbo Huang Chaohong Guan Wei Zong Yuhang Dai Zijuan Du Zhenyu Zhang Jianwei Li Fei Guo Xuan Gao Haobo Dong Jiexin Zhu Xiaohui Wang Guanjie He 2023Nano-Micro Letters2023,15,6:2

20Analysis and seismic tests of composite shear walls with CFST columns and steel plate deep beams显示文摘A composite shear wall concept based on concrete filled steel tube(CFST)columns and steel plate(SP)deep beams is proposed and examined in this study.The new wall is composed of three different energy dissipation elements:CFST columns;SP deep beams;and reinforced concrete(RC)strips.The RC strips are intended to allow the core structural elements-the CFST columns and SP deep beams-to work as a single structure to consume energy.Six specimens of different configurations were tested under cyclic loading.The resulting data are analyzed herein.In addition,numerical simulations of the stress and damage processes for each specimen were carried out,and simulations were completed for a range of location and span-height ratio variations for the SP beams.The simulations show good agreement with the test results.The core structure exhibits a ductile yielding mechanism characteristic of strong column-weak beam structures,hysteretic curves are plump and the composite shear wall exhibits several seismic defense lines.The deformation of the shear wall specimens with encased CFST column and SP deep beam design appears to be closer to that of entire shear walls.Establishing optimal design parameters for the configuration of SP deep beams is pivotal to the best seismic behavior of the wall.The new composite shear wall is therefore suitable for use in the seismic design of building structures.Dong Hongying Cao Wanlin Wu Haipeng Zhang Jianwei Xu Fangfang 2013Earthquake Engineering and Engineering Vibration2013,12,4:1
