1Molecular Characterization of a Leaf Senescence-Related Transcription Factor BrWRKY75 of Chinese Flowering Cabbage显示文摘WRKY is a plant-specific transcription factor(TF) involved in the regulation of many biological processes; however, its role in leaf senescence of leafy vegetables remains unknown. In the present work, a WRKY TF, termed Br WRKY75 was isolated from Chinese flowering cabbage [Brassica rapa L. ssp. chinensis(L.) Mokino var. utilis Tsen et Lee]. Analysis of deduced amino acid sequence and the phylogenetic tree showed that Br WRKY75 has high homology with WRKY75 from Brassica oleracea and Arabidopsis thaliana, and belongs to the II c sub-group. Sub-cellular localization and transcriptional activity analysis revealed that Br WRKY75 is a nuclear protein with transcriptional repression activity, and was up-regulated during leaf senescence. Electrophoretic mobility shift assay confirmed that Br WRKY75 directly bound to the W-box(TTGAC) cis-element. 2Morphological Characteristics of Sand Waves in the Middle Taiwan Shoal Based on Multi-beam Data Analysis显示文摘A DTM map of the study area in the Taiwan Shoal was drawn based on precise and highdensity data acquired in a field survey by a multi-beam sounding system(R2Sonic2024). We identified sand waves in the study area at water depths of 13.89–49.12 m; the main sand waves had heights of 5–25 m, lengths of 0.1–2.0 km, and crest lines 0.1–6.5 km long. The spatial distribution of the sand waves on the seabed is dense in the north and sparse in the south and the directions range between 50°–80° and 90°–135°. Between the main sand waves, secondary sand waves develop with heights of 0.1–5.0 m and lengths of 10–100 m, which are difficult to detect by satellite remote sensing. By comparing the evolution structures of the secondary and main sand waves, we identified three evolution modes of the secondary sand waves: parallel, oblique, and divergent modes according to the relative crest directions. Suitable water depth, reciprocating current speeds between 40 and 100 cm/s, and abundant sediment supply create favorable conditions for the formation of linear sand waves. 3Role of Co Content on Densification and Microstructure of WC–Co Cemented Carbides Prepared by Selective Laser Melting显示文摘WC–Co cemented carbides, well-known as the conventional tooling materials, have not been successfully produced by one step additive manufacturing processes such as selective laser melting(SLM) yet. The microstructure evolution as well as WC grain growth behavior has rarely been investigated in detail during SLM process. In this study, the WC–Co cemented carbides with different Co contents(12–32 wt%) were prepared by optimized SLM processes for comparative investigation of densification behavior, microstructure characterization and mechanical property. The increase in Co content in feedstock carbide granules can improve the densification behavior during SLM process. The SLM processed WC-12 Co shows larger average WC grain size and higher percentage of coarser WC grains as compared with both WC-20 Co and WC-32 Co. The microstructure characterization, combined with finite element simulation, shows the WC grain growth mechanisms include agglomeration and dissolution-deposition of WC during SLM process and agglomeration of WC is an important mechanism especially for WC–Co cemented carbides with Co content as low as 12 wt%. 4Internal structures and high-velocity frictional properties of Longmenshan fault zone at Shenxigou activated during the 2008 Wenchuan earthquake显示文摘This paper reports internal structures of a wide fault zone at Shenxigou,Dujiangyan,Sichuan province,China,and high-velocity frictional properties of the fault gouge collected near the coseismic slip zone during the 2008 Wenchuan earthquake.Vertical offset and horizontal displacement at the trench site were 2.8 m(NW side up)and 4.8 m(right-lateral),respectively.The fault zone formed in Triassic sandstone,siltstone,and shale about 500 m away from the Yingxiu-Beichuan fault,a major fault in the Longmenshan fault system.A trench survey across the coseismic fault,and observations of outcrops and drill cores down to a depth of 57 m revealed that the fault zone consists of fault gouge and fault breccia of about0.5 and 250–300 m in widths,respectively,and that the fault strikes N62°E and dips 68° to NW.Quaternary conglomerates were recovered beneath the fault in the drilling,so that the fault moved at least 55 m along the coseismic slip zone,experiencing about 18 events of similar sizes.The fault core is composed of grayish gouge(GG) and blackish gouge(BG) with very complex slip-zone structures.BG contains low-crystalline graphite of about 30 %.High-velocity friction experiments were conducted at normal stresses of 0.6–2.1 MPa and slip rates of 0.1–2.1 m/s.Both GG and BG exhibit dramatic slip weakening at constant high slip rates that can be described as an exponential decay from peak friction coefficient lpto steadystate friction coefficient lssover a slip-weakening distance Dc.Deformation of GG and BG is characterized by overlapped slip-zone structures and development of sharp slickenside surfaces,respectively.Comparison of our data with those reported for other outcrops indicates that the high-velocity frictional properties of the Longmenshan fault zones are quite uniform and the high-velocity weakening must have promoted dynamic rupture propagation during the Wenchuan earthquake.Yu Wang Shengli Ma Toshihiko Shimamoto Lu Yao Jianye Chen Xiaosong Yang Honglin He Jiaxiang Dang Linfeng Hou Tetsuhiro Togo 2014Earthquake Science2014,27,5:3
5A Banana PHD-Type Transcription Factor MaPHD1 Represses a Cell Wall-Degradation Gene MaXTH6 during Fruit Ripening显示文摘Plant homeobox domain(PHD)-type transcription factors(TFs) are involved in a variety of biological processes. However, its involvement in commercially important fruit ripening process remains largely unclear. In the present work, the characterization of a PHD-type TF termed MaPHD1 from banana fruit is reported. Multiple alignments of the deduced amino acid sequence revealed that Ma PHD1 showed a high homology with Arabidopsis thaliana Alfin1-like proteins belonging to plant-specific sub-family of PHD finger proteins. MaPHD1 was found localized in the nucleus and exhibited trans-repression ability. It was down-regulated by ethylene and ripening. Electrophoretic Mobility Shift Assay(EMSA) and transient expression analysis demonstrated that Ma PHD1 directly bound to the G-rich motifs in the promoter of Ma XTH6, which is associated with cell wall degradation, and subsequently repressed its expression. These findings suggest that MaPHD1 may be negatively associated with banana fruit ripening, at least in part, by the direct suppression of Ma XTH6. 7Methionine oxidation and reduction of the ethylene signaling component MaEIL9 are involved in banana fruit ripening显示文摘The ethylene insensitive 3/ethylene insensitive3-like(EIN3/EIL)plays an indispensable role in fruit ripening.However,the regulatory mechanism that links post-translational modification of EIN3/EIL to fruit ripening is largely unknown.Here,we studied the expression of 13 MaE IL genes during banana fruit ripening,among which MaE IL9 displayed higher enhancement particularly in the ripening stage.Consistent with its transcript pattern,abundance of MaE IL9 protein gradually increased during the ripening process,with maximal enhancement in the ripening.DNA affinity purification(DAP)-seq analysis revealed that MaE IL9 directly targets a subset of genes related to fruit ripening,such as the starch hydrolytic genes MaA MY3D and MaB AM1.Stably overexpressing MaE IL9 in tomato fruit hastened fruit ripening,whereas transiently silencing this gene in banana fruit retarded the ripening process,supporting a positive role of MaEIL9 in fruit ripening.Moreover,oxidation of methionines(Met-129,Met-130,and Met-282)in MaEIL9 resulted in the loss of its DNA-binding capacity and transcriptional activation activity.Importantly,we identified MaEIL9 as a potential substrate protein of methionine sulfoxide reductase A MaMsrA4,and oxidation of Met-129,Met-130,and Met-282in MaEIL9 could be restored by MaMsrA4.Collectively,our findings reveal a novel regulatory network controlling banana fruit ripening,which involves MaMsrA4-mediated redox regulation of the ethylene signaling component MaEIL9.Lisha Zhu Lin Chen Chaojie Wu Wei Shan Danling Cai Zengxiang Lin Wei Wei Jianye Chen Wangjin Lu Jianfei Kuang 2023Journal of Integrative Plant Biology2023,65,1:1
8Tumor-repopulating cell-derived microparticles elicit cascade amplification of chemotherapy-induced antitumor immunity to boost anti-PD-1 therapy显示文摘Immune checkpoint blockade(ICB)therapy,particularly antibodies targeting the programmed death receptor 1(PD-1)and its ligand(PD-L1),has revolutionized cancer treatment.However,its efficacy as a standalone therapy remains limited.Although ICB therapy in combination with chemotherapy shows promising therapeutic responses,the challenge lies in amplifying chemotherapy-induced antitumor immunity effectively.This relies on efficient drug delivery to tumor cells and robust antigen presentation by dendritic cells(DCs).Here,we developed tumor-repopulating cell(TRC)-derived microparticles with exceptional tumor targeting to deliver doxorubicin(DOX@3D-MPs)for improve anti-PD-1 therapy.DOX@3D-MPs effectively elicit immunogenic tumor cell death to release sufficient tumor antigens.Heat shock protein 70(HSP70)overexpressed in DOX@3D-MPs contributes to capturing tumor antigens,promoting their phagocytosis by DCs,and facilitating DCs maturation,leading to the activation of CD8+T cells.DOX@3D-MPs significantly enhance the curative response of anti-PD-1 treatment in large subcutaneous H22 hepatoma,orthotopic 4T1 breast tumor and Panc02 pancreatic tumor models.These results demonstrate that DOX@3D-MPs hold promise as agents to improve the response rate to ICB therapy and generate long-lasting immune memory to prevent tumor relapse.Nana Bie Tuying Yong Zhaohan Wei Qingle Liang Xiaoqiong Zhang Shiyu Li Xin Li Jianye Li Lu Gan Xiangliang Yang 2023Signal Transduction and Targeted Therapy2023,8,11:1

15Mitogen-activated protein kinase 14-mediated phosphorylation of MaMYB4 negatively regulates banana fruit ripening显示文摘Mitogen-activated protein kinase(MAPK/MPK)cascades play crucial parts in plant growth,development processes,immune ability,and stress responses;however,the regulatory mechanism by which MAPK affects fruit ripening remains largely unexplored.Here,we reported that MaMPK14 cooperated with MaMYB4 to mediate postharvest banana fruit ripening.Transient overexpression of individual MaMPK14 and MaMYB4 in banana fruit delayed fruit ripening,confirming the negative roles in the ripening.The ripening negative regulator MaMYB4 could repress the transcription of genes associated with ethylene biosynthesis and fruit softening,such as MaACS1,MaXTH5,MaPG3,and MaEXPA15.Furthermore,MaMPK14 phosphorylated MaMYB4 at Ser160 via a direct interaction.Mutation at Ser160 of MaMYB4 reduced its interaction with MaMPK14 but did not affect its subcellular localization.Importantly,phosphorylation of MaMYB4 by MaMPK14 enhanced the MaMYB4-mediated transcriptional inhibition,binding strength,protein stability,and the repression of fruit ripening.Taken together,our results delineated the regulation pathway of MAPK module during banana fruit ripening,which involved the phosphorylation modification of MaMYB4 mediated by MaMPK14.Yingying Yang Chaojie Wu Wei Shan Wei Wei Yating Zhao Jianfei Kuang Jianye Chen Yueming Jiang Wangjin Lu 2023Horticulture Research2023,10,1:0
16Carbon Doping Triggered Efficient Electrochemical Hydrogen Evolution of Cross-Linked Porous Ru-MoO_(2) Via Solid-Phase Reaction Strategy显示文摘The defect-free structure of Mo-based materials is a“double-edged sword”,which endows the material with excellent stability,but limits its chemical versatility and application in electrochemical hydrogen evolution reaction(HER).Carbon doping engineering is an attractive strategy to effectively improve the performance of Mo-based catalyst and maintain their stability.Herein,we report a cross-linked porous carbon-doped MoO_(2)(C–MoO_(2))-based catalyst Ru/C–MoO_(2) for electrochemical HER,which is prepared by the convenient redox solid-phase reaction(SPR)of porous RuO_(2)/Mo_(2)C composite precursor.Theoretical studies reveal that due to the presence of carbon atoms,the electronic structure of C–MoO_(2) has been properly adjusted,and the loaded small Ru nanoparticles provide a fast water dissociation rate and moderate H adsorption strength.In electrochemical studies under a pH-universal environment,Ru/C–MoO_(2) electrocatalyst exhibits a low overpotential at a current density of 10 mA cm^(-2) and has a low Tafel slope.Meanwhile,Ru/C-MoO_(2) has excellent stability for more than 100 h at an initial current density of 100 mA cm^(-2).Jialin Cai Jianye Yang Xin Xie Jie Ding Leyan Liu Wanyu Tian Yushan Liu Zhiyong Tang Baozhong Liu Siyu Lu 2023Energy & Environmental Materials2023,6,1:0
